Description

Antioxidant and antiaging composition containing flos Pyraricae extract, tea polyphenols and peptides, and its preparation method
Technical Field
The invention belongs to the technical field of cosmetics, and particularly relates to an antioxidant and anti-aging composition containing a common malpighia herb extract, tea polyphenol and peptides and a preparation method thereof.
Background
The skin undergoes changes of deterioration, aging and also a great change in its function. The factors of skin aging are divided into endogenous aging and exogenous aging, wherein the endogenous aging refers to a natural and inevitable aging process of the skin along with the aging process of a human body; exogenous aging mainly refers to photoaging, and the photochemical aging process of skin caused by ultraviolet rays (including environmental pollution and aging factors).
Endogenous aging causes skin aging in the same manner as other organs, and skin is affected by gravity, facial expression, hormonal changes and genetic factors, so that the skin is slowly and irreversibly degenerated, and endogenous aging mainly causes fine wrinkles and thinning of the epidermis.
Extrinsic aging is the combined result of all extrinsic factors that can lead to skin aging, such as lifestyle patterns, smoking, uv irradiation, and environmental pollution, which accumulate over time and is characterized by deep wrinkles, skin laxity, and pigmentation.
Photoaging is one of the major factors causing extrinsic skin aging, and it is currently believed that ultraviolet radiation is the major cause of photoaging. Endogenous aging skin changes are observed in the skin at non-light exposed sites, whereas photoaging-induced skin changes are significantly more pronounced at light exposed sites, such as the hands, face and neck, than in the aging manifestations caused by endogenous factors. There is a long-term accumulation of sunlight in daily life, which is unavoidable, and once photoaging begins, collagen fibers decrease, and the skin becomes flaccid and wrinkles appear. The dermal layer of the skin is a connective tissue containing a large amount of extracellular matrix (ECM) proteins mainly composed of collagen and elastin, and other components of the ECM, in addition to these fibrous proteins, are Proteoglycan (PG) and glycosaminoglycans (GAGs), which are amorphous, surrounding and embedding the fiber and cell matrix in the dermis. Collagen type I (COL 1) in dermal collagen fibers accounts for about 90% of collagen fibers, and gradually decreases as skin aging progresses due to low synthesis and high degradation, so that collagen fibers are broken, disordered, and agglomerated in photoaged skin. Studies have shown that photoaging causes a decrease in collagen stability by altering proline and strong proline residues in the dermis, and the density and morphology of collagen and elastin in the skin are markers for determining whether skin aging occurs.
In photoaging studies, it has been shown that ultraviolet radiation induces the expression of Matrix Metalloproteinases (MMPs) in human skin. MMPs function to degrade extracellular matrix proteins such as collagen, elastin, proteoglycans, and the like. MMP is a zinc-containing endopeptidase with a broad range of specific substrates that mediate the degradation of different components in the ECM. In the skin, keratinocytes of the epidermis and fibroblasts of the dermis secrete mainly MMP1 (collagenase 1), MMP1 degrades type I and type III collagen fibers into specific protein fragments at a single site in the triple helix, and other MMPs (e.g., gelatinases) hydrolyze these collagen fragments further, eventually leading to damage to the collagen-rich dermis.
With the age, the metabolism function of the skin is reduced, the function of an in-vivo antioxidant system is declined, the activity of antioxidant enzyme is continuously reduced, free radicals are excessively accumulated, clearance disorder occurs, and the accumulation of in-vivo oxidative irreversible damage is initiated; in addition, long-term over-irradiation by ultraviolet rays can also cause problems of oxidative stress, inflammatory reaction, telomere damage, collagen synthesis reduction and the like, leading to a series of aging damage and accelerating skin aging. The aging skin is mainly characterized by: loose skin, wrinkles, pigmentation, etc.
At present, most of anti-aging cosmetics on the market start from improving skin collagen, however, the collagen cosmetics are prepared by coating single collagen on the surface of skin, and the single anti-aging way is often poor in effect, so that the anti-aging effect is far from the expectation.

The purpose of the invention can be realized by the following technical scheme:
an antioxidant and anti-aging composition containing a western pear flower extract, tea polyphenol and peptides comprises the following raw materials in percentage by mass:
american pear flower extract solution: 1-20%, tea polyphenol: 1-5%, palmitoyl tripeptide-5 solution: 1-10%, arginine/lysine polypeptide solution: 1-10%, acetyl hexapeptide-8 solution: 1-10%, carnosine: 0.01-0.5%, oligopeptide-3 solution: 0.1-5%, dipeptide diaminobutyrylbenzyl amide diacetate solution 0.1-5%, phenoxyethanol solution: 0.5-5%, butanediol: 1-30%, citric acid: 0.01-0.5%, xanthan gum: 0.01-0.5% and the balance of ionized water.
The American pear flower extract solution comprises 69.4% of water, 20% of butanediol, 10% of American pear flower extract and 0.6% of phenoxyethanol; further, the mass percent of the solution of the American pear flower extract is 1-10%.
The extract of Actinidia chinensis planch has antioxidant effect, and can scavenge hydroxyl free radical and inhibit lipid peroxidation. Pear flower is rich in nutrients such as vitamins, amino acids, minerals, etc., and its extract also contains a large amount of polyphenols such as arbutin, chlorogenic acid, etc., and flavonoids beneficial to skin. Researchers detected arbutin, glycosides, hydroquinone, flavonoids and the like in pear flowers of avocados. In another study, the investigators tested polyphenols in the pear flower extract and identified a phenolic 4-O-Z-coumaroyl rosin that was not reported in previous related studies. Researchers have shown that phenolics in pear flowers play a non-negligible role in the antioxidant capacity of pear flower extracts. According to the experimental results, the hydroquinone content in various polyphenol substances is the highest, and the antioxidant activity is the strongest. Another study indicated that the arbutin content in pear flower was relatively low, but the chlorogenic acid content was highest, compared to the buds, leaf buds, and young fruits.
Chlorogenic acid is an effective phenolic antioxidant with antioxidant capacity stronger than that of caffeic acid, p-hydroxybenzoic acid, ferulic acid, syringic acid, butylated hydroxyanisole and tocopherol. Chlorogenic acid has antioxidant effect because it contains a certain amount of R-OH groups, and can form hydrogen radicals with antioxidant effect to eliminate the activity of hydroxyl radicals and superoxide anions, etc., thereby protecting tissue from damage due to oxidation.
Arbutin is one of the important components in pear flower extracts known at present. The whitening effect and safety of use of arbutin on skin have been extensively studied and confirmed. Arbutin can effectively inhibit the generation of melanin of skin cells, and has certain antioxidant capacity. In addition, arbutin can be hydrolyzed under the influence of skin environment and flora to generate hydroquinone with good oxidation resistance, so that the oxidation resistance effect of arbutin is further improved. Arbutin and hydroquinone detected in the pear flower extract can contribute to the pear flower extract to have good whitening and oxidation resistance.
The extraction method of the American pear flower extract comprises the following operations:
chopping dry American pear flowers, selecting ethanol with the mass percent of 70% as an extraction solvent, soaking the American pear flowers in the extraction solvent for 0.5h, then carrying out reflux extraction, wherein the reflux extraction temperature is 70-80 ℃, the extraction frequency is 2 times, the extraction time is 2 hours each time, and the material-liquid ratio of the first reflux extraction is 1:15, the material-liquid ratio of the second reflux extraction is 1:10; filtering the extracts, and mixing the filtrates; and concentrating the filtrate under reduced pressure, and vacuum drying the concentrated solution to obtain the American pear flower extract.
Tea polyphenol is a natural antioxidant with strong oxidation resistance, has strong electron supply capacity, can be combined with free radicals in skin to be converted into inert substances, and stops chain reaction of the free radicals. Has strong scavenging function to free radicals.
The arginine/lysine polypeptide solution comprises 0.0024% of arginine/lysine polypeptide and 99.9976% of deionized water.
Arginine/lysine polypeptide: the venom is from marine mollusk conus, and has strong biological activity. Such neurotoxin peptides are collectively referred to as conopeptides. Conopeptides derived from conus have the advantages of multiple types, small molecules, strong effect, wide target spot, strong selectivity and the like, and can specifically block a voltage-gated sodium ion channel, so that the internal flow of sodium ions is blocked, and the action potential of muscles cannot be formed, thereby effectively preventing and reducing wrinkles. The arginine/lysine polypeptide synthesized by artificial scientific sequencing through a biological simulation synthesis technology can effectively relax muscles under the condition of not numbing muscle nerve conduction, thereby achieving the effect of removing wrinkles.

Palmitoyl tripeptide-5 can promote synthesis of collagen (type I, type II and type IV) and glucosamine in human fibroblasts, strengthen connective tissues, enhance skin compactness, promote cell activity, supplement collagen in skin, and improve skin elasticity.
The acetyl hexapeptide-8 solution comprises 0.1% of acetyl hexapeptide, 96.87% of deionized water, 3% of 1, 2-hexanediol and 0.03% of ethyl hexyl glycerol.
The acetyl hexapeptide-8 can block the transportation and release of acetylcholine of nerve cell bodies, weaken nerve transmission and further reduce muscle contraction to achieve the aim of reducing wrinkles.
The dipeptide diaminobutyrylbenzylamide diacetate solution comprises dipeptide diaminobutyrylbenzylamide diacetate 0.1%, deionized water 96.87%, 1, 2-hexanediol 3% and ethylhexylglycerin 0.03%.
