Abstract

The present invention relates to active peptides having anti-relaxant, firming effects and cosmetics comprising the same. Specifically, the invention discloses SEQ ID NO 1 and SEQ ID NO:2, alone or in combination, for anti-relaxing, tightening activity and a cosmetic product comprising said peptides, wherein said product comprises in weight%: 1 and/or SEQ ID NO:2, 0.0005-0.001 of peptide, 100.5-2.5 of coenzyme Q, 2.0-4.0 of rosemary extract, 2.0-4.0 of 1, 3-propylene glycol, 1.5-3.5 of rice bran oil, 2.0-4.0 of soybean lecithin, 5.0-15.0 of resveratrol, 1.0-2.0 of ferulic acid, 1.0-2.0 of polyglutamic acid, 0.5-1.5 of dendrobium officinale extract, 0.5-1.5 of hyaluronic acid, 0.4-0.8 of carbomer and the balance of water. The cosmetic has the effects of resisting relaxation, tightening skin, resisting skin aging and the like.

Technical Field
The present invention relates to non-natural peptides with anti-relaxing, tightening effects and their use in cosmetic and personal care compositions.
Background
Skin aging is a phenomenon of skin aging caused by natural factors or non-natural factors. After birth, the skin tissue of a person is developed increasingly, the function of the person is active gradually, and the person starts to degenerate when the person reaches a certain age, and the degeneration is gradually performed unconsciously. The growth period of skin tissues generally ends at about 25 years old, which is called as 'bending angle of skin' by some people, since then, the growth and the aging are carried out simultaneously, the elastic fibers of the skin gradually become thick, and the aging of the skin is slowly and obviously in the early aging period of 40-50 years, although the aging degree can be different from person to person.
Skin aging is the result of the combined action of the intrinsic physiological mechanism of human aging and the damage caused by external wind, sunshine, environmental pollution and the like. Reasonable nutrition can partially inhibit physiological aging, but is difficult to protect the skin from the external environment. Chinese invention patent CN105407866 discloses the use of a combination comprising collagen, preferably marine collagen or a hydrolysed form thereof, and vitamin B3, said use having at least one skin benefit selected from the group consisting of lightening wrinkles, promoting skin plumping or elastic condition of the skin, improving skin firmness and smoothness, improving skin hydration and improving skin radiance. The invention also relates to a combination of active agents comprising at least collagen or a hydrolysed form thereof, vitamin B3 and a tremella extract and to a related cosmetic method.
Chinese patent CN104903346 discloses a synthetic peptide, which has 20-39 amino acid residues and has the function of preventing and/or alleviating skin aging.
Chinese invention patent CN104394876 discloses a composition for preventing skin aging, which comprises tea water derived from fresh tea leaves inactivated by enzyme.
Chinese patent No. 103957998 discloses a skin anti-aging composition, in particular a skin anti-aging composition comprising myricetin or a glycoside thereof and at least one LXR α agonist skin aging refers to the appearance or manifestation of any one or more of wrinkles or sagging, poor skin barrier such as dryness, scalp itching or uneven skin tone such as age spots.

With the improvement of the living standard, people, especially women, pay more and more attention to the external appearance of the people, so that the beauty cosmetics capable of resisting relaxation and tightening the skin are expected by people at present.
Disclosure of Invention
The present inventors have conducted diligent studies and screened peptide libraries in order to develop novel peptides having anti-relaxation, tightening effects. Finally, the inventors have found novel peptides having excellent anti-relaxation, tightening efficacy, ultimately achieving the objects of the present invention.
It is an object of the present invention to provide peptides having anti-relaxing, tightening effects.
It is another object of the present invention to provide a composition for anti-sagging, anti-firming.
It is yet another object of the present invention to provide a method for resisting sagging and tightening.
To achieve the object of the present invention, in a first aspect of the present invention, there is provided a peptide selected from the group consisting of:
(i) a peptide comprising or consisting of amino acid residues as follows:
Ala-Ala-Leu-Met-Asn-Cys-Cys-Glu-His-Ile-Met-Ala (AALMNCCEHIMA) (SEQ ID NO: 1); or
(ii) A peptide comprising or consisting of amino acid residues as follows:
Cys-Met-Ser-Tyr-Thr-Thr-Met-Ile-Asn-Arg-Arg-Thr(CMSYTTMINRRT)(SEQ IDNO:2)。

According to a third aspect of the invention, there is provided the use of a peptide of SEQ ID NO. 1 of the invention for anti-relaxation, anti-compaction.
According to a preferred aspect of the present invention, there is provided SEQ ID NO 1 and SEQ ID NO:2 for use in anti-relaxation and anti-firming.
The present invention also provides a cosmetic for anti-sagging, firming, comprising:
(a) individual peptides or combinations of peptides of the invention;
(b) an antioxidant;
(C) a humectant;
(d) a vegetable oil;
(e) lecithin;
(f) a preservative;
(g) essential oil; and
(h) and (5) purifying the water.
These and other aspects of the present invention will become apparent to those skilled in the art upon reading the following detailed description of the invention, including the exemplary embodiments and examples.
Two peptides are claimed in the present invention, namely peptide (i) having SEQ ID NO:1 or peptide (ii) having SEQ ID NO: 2. When these peptides are used in anti-relaxing, tightening compositions, the inhibition provides anti-relaxing, tightening benefits.
In addition, by experimentation, it was not only demonstrated that each of these two peptides reduces sebum secretion, but also that they interact synergistically when used together.
As used herein, all terms are intended to have their ordinary meaning in the art unless otherwise specifically defined. The term "amino acid" is intended to include naturally occurring amino acids as well as non-naturally occurring amino acids, and broadly includes all small molecules having one carboxyl group and at least one primary or secondary amine group capable of forming a peptide bond. The term "peptide" is intended to include all molecules having at least one peptide bond, and thus includes dipeptides, tripeptides, oligopeptides and polypeptides having up to about 20 amino acid residues. The peptides of the invention are provided in a cosmetically acceptable carrier. The carrier may be hydrophobic or hydrophilic. Suitable hydrophobic carriers include, for example, waxy nonionic materials commonly used in cosmetics, such as esters and ethers of fatty alcohols and fatty acids, with carbon chain lengths of C4 to C22, preferably C8 to C18, most preferably C12 to C18.
Examples of fatty hydrophobic carriers include isopropyl myristate, isopropyl palmitate, octyl palmitate, isopropyl lanolate, acetylated lanolin alcohol, benzoate esters of C12-C15 alcohols, cetyl octanoate, cetyl palmitate, myristyl myristate, myristyl lactate, cetyl acetate, propylene glycol dicaprylate/decanoate, decyl oleate, acetylated lanolin, heptanoic stearate, diisostearate malate, octyl hydroxystearate, isopropyl isostearate, and the like.
Suitable hydrophilic carrier solutions may be, for example, glycols and alkoxylated glycols commonly used in cosmetics, including ethylene glycol, diethylene glycol, triethylene glycol, propylene glycol, dipropylene glycol, and the like.
The anti-laxative, firming composition may be formulated as a nutritional liquid, serum, cosmetic, cream, lotion, serum, spray, stick, and other forms known to those skilled in the art. Creams and lotions are currently the preferred product forms.
In a cosmetically acceptable carrier, the concentration of the peptide may range from 1. mu.M to 200. mu.M, preferably from 10ppb to 150. mu.M, more preferably from 20. mu.M to 100. mu.M, most preferably from 36. mu.M to 50. mu.M.
The anti-sagging, tightening composition typically comprises the above-described carrier solution at a level of from about 0.01% to about 90% by weight, preferably from about 0.1% to about 50% by weight, more preferably from about 0.1% to about 20% by weight, and most preferably from about 1% to about 10% by weight.
Examples of the antioxidant include butylhydroxyanisole, dibutylhydroxytoluene, sodium hydrogen sulfite, ascorbic acid and salts thereof, glutathione peroxidase, glutathione-S-transferase, catalase, superoxide dismutase, thioredoxin, taurine, thiotaurine, hypotaurine, vitamin E, and the like. Among them, ascorbic acid, glutathione, vitamin E and taurine are preferable. These antioxidants may be used in combination of 1 or more than 2, and preferably are a combination of ascorbic acid and taurine. When the antioxidant is added to the composition for external application to the skin (for example, a cosmetic) of the present invention, the addition ratio thereof is not particularly limited, but is usually 0.001 to 6% by weight, preferably 0.01 to 3% by weight, and more preferably 0.2 to 2% by weight, based on the total amount of the composition for external application to the skin.
Examples of the humectant include amino acids such as alanine, serine, leucine, isoleucine, threonine, glycine, proline, hydroxyproline, aspartic acid, arginine, and theanine, and derivatives thereof; polyhydric alcohols such as ethylene glycol, 1, 3-propanediol, 1, 3-butanediol, diethylene glycol, triethylene glycol, tetraethylene glycol, dipropylene glycol, di-1, 3-propanediol, and polyethylene glycol; glycol ethers such as ethylene glycol monomethyl ether, ethylene glycol monoethyl ether, ethylene glycol monopropyl ether, diethylene glycol monomethyl ether, diethylene glycol monoethyl ether, diethylene glycol monopropyl ether, diethylene glycol monobutyl ether, propylene glycol monoethyl ether, propylene glycol monopropyl ether, dipropylene glycol monoethyl ether, and dipropylene glycol monopropyl ether; sugar alcohols such as mannitol, sorbitol, erythritol, xylitol, and trehalose; phospholipids such as lecithin and hydrogenated lecithin; mucopolysaccharides such as sodium hyaluronate, sodium acetylhyaluronate, heparin analogues, sodium chondroitin sulfate, etc.; high molecular compounds such as collagen, elastin, keratin, chitin, chitosan, gelatin, polyglutamic acid, etc.; sodium lactate, sodium pyrrolidone carboxylate, etc.; chamomile extract, aloe vera extract, witch hazel extract, rosemary extract, thyme extract, tea extract, perilla extract, and other plant extracts. The vegetable oil is selected from rose hip oil, sweet almond oil, rose oil, jojoba oil, wheat germ oil, carrot oil, avocado oil, castor oil, avocado oil, coconut oil, palm oil, cocoa butter, palm kernel oil, soybean oil, rice bran oil, aloe oil, veronica officinalis, chenopodium album butter, shea butter, peanut oil, corn oil, macadamia nut oil, Chinese chestnut oil, seabuckthorn oil, linseed oil, passion flower oil, calendula, mango kernel oil and rose hip oil, and is preferably one or a combination of two or more of sweet almond oil, jojoba oil and aloe oil. Preferably, the vegetable oil in the composition of the invention is jojoba oil. When the vegetable oil is added to the composition for external application to the skin (e.g., cosmetic) of the present invention, the addition ratio thereof is not particularly limited, but is usually 0.001 to 10% by weight, preferably 0.5 to 8% by weight, and more preferably 1.0 to 8.0% by weight, based on the total amount of the composition for external application to the skin (e.g., cosmetic).
As preservative, it is selected from benzyl alcohol, phenethyl alcohol, phenoxyethanol and lauroyl arginine ethyl ester, preferably lauroyl arginine ethyl ester. When a preservative is added to the composition for external application to the skin (e.g., cosmetic) of the present invention, the addition ratio is not particularly limited, but is usually 0.001 to 5% by weight, preferably 0.1 to 5% by weight, and more preferably 0.2 to 3% by weight, based on the total amount of the composition for external application to the skin (e.g., cosmetic).
As the lecithin, it is preferably soybean lecithin. When lecithin is added to the composition for external application to skin (e.g., a cosmetic) of the present invention, the addition ratio is not particularly limited, but is usually 0.01 to 5% by weight, preferably 0.1 to 5% by weight, and more preferably 0.2 to 2% by weight, based on the total amount of the composition for external application to skin (e.g., a cosmetic).
Preferably, the cosmetic comprises 2.0-4.0 percent of ascorbic acid, 0.5-2.5 percent of taurine, 0.5-2.5 percent of jojoba oil, 2.0-4.0 percent of rosemary extract, 2.0-4.0 percent of 1, 3-propylene glycol, 1.5-3.5 percent of rice bran oil, 2.0-4.0 percent of soybean lecithin, 5.0-12.0 percent of resveratrol, 0.4-0.8 percent of carbomer and 2.0-4.0 percent of glucan in percentage by weight.
More preferably, the cosmetic of the invention comprises 0.0005 to 0.001% by weight of the amino acid sequence of SEQ ID NO:1 peptide and/or seq id NO:2 peptides. Also preferably, the cosmetic of the present invention further comprises a suitable amount of ethyl lauroyl arginate and the balance of water.
Further preferably, the cosmetic of the present invention comprises, in wt%:
SEQ ID NO:1 peptide 0.001, coenzyme Q100.5-2.5, rosemary extract 2.0-4.0, 1, 3-propylene glycol 2.0-4.0, rice bran oil 1.5-3.5, soybean lecithin 2.0-4.0, resveratrol 5.0-15.0, ferulic acid 1.0-2.0, polyglutamic acid 1.0-2.0, dendrobium officinale extract 0.5-1.5, hyaluronic acid 0.5-1.5 and carbomer 0.4-0.8, and the balance of water.
Still more preferably, the cosmetic of the present invention comprises, in% by weight:
SEQ ID NO:2, 0.001 of peptide, 100.5-2.5 of coenzyme Q, 2.0-4.0 of rosemary extract, 2.0-4.0 of 1, 3-propylene glycol, 1.5-3.5 of rice bran oil, 2.0-4.0 of soybean lecithin, 5.0-15.0 of resveratrol, 1.0-2.0 of ferulic acid, 1.0-2.0 of polyglutamic acid, 0.5-1.5 of dendrobium officinale extract, 0.5-1.5 of hyaluronic acid, 0.4-0.8 of carbomer and the balance of water.
Still further preferably, the cosmetic of the present invention comprises, in wt%:
SEQ ID NO:1, peptide 0.0005 of SEQ ID NO:2, 0.0005 of peptide, 100.5-2.5 of coenzyme Q, 2.0-4.0 of rosemary extract, 2.0-4.0 of 1, 3-propylene glycol, 1.5-3.5 of rice bran oil, 2.0-4.0 of soybean lecithin, 5.0-15.0 of resveratrol, 1.0-2.0 of ferulic acid, 1.0-2.0 of polyglutamic acid, 0.5-1.5 of dendrobium officinale extract, 0.5-1.5 of hyaluronic acid, 0.4-0.8 of carbomer and the balance of water.
As used herein, "comprising" is synonymous with "including," "containing," or "characterized by," and is inclusive or open-ended and does not exclude additional unrecited elements or method steps. The term "comprising" in any of the expressions herein, particularly in describing the method, use or product of the invention, is to be understood as including those products, methods and uses which consist essentially of and consist of the recited components or elements or steps. The invention illustratively described herein suitably may be practiced in the absence of any element or elements, limitation or limitations which is not specifically disclosed herein.
The instructions contained in the anti-sagging, tightening product of the present invention relating to the anti-sagging, tightening product may contain the following: effects and efficacies (e.g. anti-sagging, tightening), methods of application (e.g. even application to the skin, e.g. face and neck), and possible side effects, etc.

Examples
The following examples are intended to illustrate the invention in a non-limiting manner.
Example 1: effect on type I collagen Synthesis
Human skin fibroblasts were plated on 48-well plates at 1 × 105cell/ml, cultured for one day so that cell fusion reaches about 60%, and cultured on FBS-free medium for another day. Preparing DMEM medium without FBS, and dividing into A, B, C, D IVGroup A, group A with deionized water, group B with the peptide of SEQ ID NO:1 so that the concentration was 10ppm, group C with the peptide of SEQ ID NO:2 so that the concentration was 10ppm, and group D with the peptides of SEQ ID NO:1 and SEQ ID NO:2, respectively, so that the concentrations were 5ppm, respectively. Collecting cultured human skin fibroblasts, washing with phosphate buffer solution, transferring into above four groups of culture medium, and culturing in CO2The culture was carried out in an incubator for 48 hours. To evaluate the expression of Procollagen, the culture broth obtained by the above culture method was quantitatively analyzed using a Type I Procollagen C-peptide Kit (Procollagen Type I C-peptide (PIP) EIA Kit, Cat #. MK101, Takara, Japan). In a coated 96-well plate provided in the kit, an anti-collagen antibody as a primary antibody was treated with the supernatant at 37 ℃ for 90 minutes, then washed 3 times with TPBS (0.1% Tween 20in PBS), and after an anti-mouse IgG (mouse IgG) as a secondary antibody was reacted for 90 minutes, washed 3 times with TPBS. After an alkaline phosphatase substrate solution (1mg/mL p-nitrophenyl phosphate-diethanolamine buffer) was reacted at room temperature for 30 minutes, absorbance at a wavelength of 405nm was measured with a microplate reader.

According to the data, compared with a blank control, the two peptides can obviously increase the synthesis of the type I collagen, and the two peptides obtain an effect which is obviously better than that of a single peptide under the condition that the total protein concentration is the same, so that the two peptides have a synergistic effect on promoting the synthesis of the type I collagen.

Skin tightening efficacy test:
volunteers 30 (age 30 to 50 years)
Grouping: randomly dividing 30 persons into 3 groups of 10 persons, each of which was given two test articles, one of which was a control and the other was the firming cream prepared in examples 2 to 4, respectively;
the application method comprises cleaning face in the morning and evening every day, applying appropriate amount of firming cream on the left face, and applying control containing no active ingredient on the right face.
Cycle of 28 days
The measuring instrument comprises a German CK MPA580 elasticity tester and a VISIA skin analysis tester of CANFIELD
Volunteers recorded each person's experimental data using MPA580 elasticity tester, VISIA skin analysis tester on day 28. The experimental data are as follows in table 2:

As can be seen from the table, the firming creams of the examples 2, 3 and 4 have obvious firming effect, and the firming effect of the peptide disclosed by the invention is proved to have certain practical effect on skin firming. However, after four weeks of tightening cream using example 4, the improvement in skin elasticity and wrinkle depth was superior to that of examples 2 and 3, showing a significant synergistic tightening effect.
The present invention has been illustrated by the description of the preferred embodiments above, but it is to be understood that variations and modifications can be made on these embodiments by the skilled person without departing from the spirit or scope of the present invention as expressed in the following claims.

Claims (9)

1. A peptide, referred to as "peptide.04", consisting of SEQ ID NO: 1.
2. A combination of peptides comprising SEQ ID NO:1 and SEQ ID NO: 2.
3. A composition for anti-relaxation, tightening comprising the peptide of claim 1, or a combination comprising the peptide of claim 2.
4. Use of the peptide of claim 1 for the preparation of a composition for anti-relaxation, tightening.
5. Use according to claim 4, wherein the composition is in the form of a cosmetic product.
6. A cosmetic product having anti-sagging, tightening efficacy, comprising: SEQ ID NO:1 and SEQ ID NO:2, coenzyme Q10, rosemary extract, 1, 3-propanediol, rice bran oil, soy lecithin, resveratrol, ferulic acid, polyglutamic acid, dendrobium officinale extract, hyaluronic acid and carbomer.
7. The cosmetic according to claim 6, comprising in weight%:
SEQ ID NO:1 peptide 0.001, coenzyme Q100.5-2.5, rosemary extract 2.0-4.0, 1, 3-propylene glycol 2.0-4.0, rice bran oil 1.5-3.5, soybean lecithin 2.0-4.0, resveratrol 5.0-15.0, ferulic acid 1.0-2.0, polyglutamic acid 1.0-2.0, dendrobium officinale extract 0.5-1.5, hyaluronic acid 0.5-1.5 and carbomer 0.4-0.8, and the balance of water.
8. The cosmetic according to claim 6, comprising in weight%:
SEQ ID NO:2, 0.001 of peptide, 100.5-2.5 of coenzyme Q, 2.0-4.0 of rosemary extract, 2.0-4.0 of 1, 3-propylene glycol, 1.5-3.5 of rice bran oil, 2.0-4.0 of soybean lecithin, 5.0-15.0 of resveratrol, 1.0-2.0 of ferulic acid, 1.0-2.0 of polyglutamic acid, 0.5-1.5 of dendrobium officinale extract, 0.5-1.5 of hyaluronic acid, 0.4-0.8 of carbomer and the balance of water.
9. The cosmetic according to claim 6, comprising in weight%:
