Experimental example 2 pharmacodynamic experiment of the foot bath medicine for promoting animal wound healing
2.1 design of wound models
The mice were ICR mice, purchased from Dundao technologies, inc., and all were male, with a weight specification of 25-30g, and the weight was approximately 30-35g when tested after 5 days of free feeding.
After a mouse is anesthetized by using 10% chloral hydrate according to the dose of 0.003mL/g, 2cm x 4cm body hair on the back is removed, the body hair is cleaned by normal saline, the body hair is sterilized by using 75% alcohol, and then a round whole layer of skin with the diameter of 1cm is cut at the position 2cm beside the two sides of the spinal column on the back of the mouse and is deep to the subcutaneous part (skin to muscular layer) without damaging fat and fascia on the spinal column or abdominal muscles, and the left side and the right side of the spinal column of the mouse are respectively cut. The postoperative wound was sterilized with 75% alcohol and covered with sterile gauze.
2.2 animal grouping and wound treatment
Mice were randomly divided into 6 groups of 6 mice after molding.
In experimental groups 1-4, the left wounds of four groups of mice were dropped with 0.2ml of a solvent (0.01M PH7.2 Tris hydrochloride buffer) as a control, the right wounds were given with the solutions of the Chinese medicinal composition prepared in examples 1-4, respectively, after a few minutes, the dropped liquid in the wounds were absorbed, dried, packed with sterile gauze, placed in a clean cage, raised in cages, given clean drinking water, and taken freely, and changed every day.
In the control group 5, 0.2ml of a solvent (0.01M PH7.2 tris (hydroxymethyl) aminomethane hydrochloride buffer solution) was dropped into the left wound and the right wound of the mouse, and after a few minutes, the liquid dropped into the wound was absorbed and dried, the wound was wrapped with a sterile gauze, and the wound was placed in a clean cage and raised in cages, and clean drinking water was given to the cage for free feeding and changing the dressing every day.
In the control group 6, 0.2ml of solvent (0.01M PH7.2 tris (hydroxymethyl) aminomethane hydrochloride buffer) was dropped into the left side wound of each mouse as a control, and 0.2ml of a commercially available Chinese medicinal composition was dropped into the right side wound, and a Mayinglong hemorrhoid ointment aqueous solution was selected, wrapped with sterile gauze after the liquid dropped into the wound was absorbed and dried for several minutes, and then placed in a clean cage for cage rearing, and given clean drinking water, and the mice were freely eaten and changed each day.
(1) The healing time of the left and right wounds of the experimental groups 1-4 and the left and right wounds of the control group 5 and 6 were recorded, respectively, and the average value of each group was recorded and calculated, and the results are shown in table 2.
(2) Wound healing rates were recorded for each group of mice respectively: the healing conditions of the wounds on the left side and the right side of the mouse are contrastively observed after 1,3,5,7, 10 and 15 days after the drug administration, the area of the wound surface is measured, and the healing rate is calculated. The healing rate = { (original wound area-present wound area) ÷ original wound area } × 100%. The average of each group was calculated and the results are shown in table 3.
2.3 test results and analysis
Table 2 mean wound healing time in groups of wounded mice
TABLE 3 average wound healing rates at different times for each group of wounded mice
Referring to table 2, it can be seen that in the mice of the control group 5, the left side wound and the right side wound are not applied with the traditional Chinese medicine composition of the present invention, and the healing time is 14 days to 15 days. The experimental groups 1 to 4 have an obvious effect of promoting the wound healing of the mouse after applying the traditional Chinese medicine composition prepared in the embodiments 1 to 5 of the present invention to the mouse, and specifically, the left side wound of the mouse of the experimental groups 1 to 4 is not applied with the traditional Chinese medicine composition of the present invention, the healing time is 13 days to 15 days, and the right side wound applied with the traditional Chinese medicine composition of the present invention is 9 days to 11 days, so that the application of the traditional Chinese medicine composition of the present invention can promote the wound healing, increase the wound healing speed, and effectively shorten the healing time of the wound. The traditional Chinese medicine composition has obvious effects of relieving pain, diminishing inflammation and promoting wound healing.
