2.1 design of wound models
The mice were ICR mice, purchased from Dundao technologies, inc., and all were male, with a weight specification of 25-30g, and the weight was approximately 30-35g when tested after 5 days of free feeding.
After a mouse is anesthetized by using 10% chloral hydrate according to the dose of 0.003mL/g, 2cm x 4cm body hair on the back is removed, the body hair is cleaned by normal saline, the body hair is sterilized by using 75% alcohol, and then a round whole layer of skin with the diameter of 1cm is cut at the position 2cm beside the two sides of the spinal column on the back of the mouse and is deep to the subcutaneous part (skin to muscular layer) without damaging fat and fascia on the spinal column or abdominal muscles, and the left side and the right side of the spinal column of the mouse are respectively cut. The postoperative wound was sterilized with 75% alcohol and covered with sterile gauze.
2.2 animal grouping and wound treatment
Mice were randomly divided into 6 groups of 6 mice after molding.
In experimental groups 1-4, the left wounds of four groups of mice were dropped with 0.2ml of a solvent (0.01M PH7.2 Tris hydrochloride buffer) as a control, the right wounds were given with the solutions of the Chinese medicinal composition prepared in examples 1-4, respectively, after a few minutes, the dropped liquid in the wounds were absorbed, dried, packed with sterile gauze, placed in a clean cage, raised in cages, given clean drinking water, and taken freely, and changed every day.
In the control group 5, 0.2ml of a solvent (0.01M PH7.2 tris (hydroxymethyl) aminomethane hydrochloride buffer solution) was dropped into the left wound and the right wound of the mouse, and after a few minutes, the liquid dropped into the wound was absorbed and dried, the wound was wrapped with a sterile gauze, and the wound was placed in a clean cage and raised in cages, and clean drinking water was given to the cage for free feeding and changing the dressing every day.
In the control group 6, 0.2ml of solvent (0.01M PH7.2 tris (hydroxymethyl) aminomethane hydrochloride buffer) was dropped into the left side wound of each mouse as a control, and 0.2ml of a commercially available Chinese medicinal composition was dropped into the right side wound, and a Mayinglong hemorrhoid ointment aqueous solution was selected, wrapped with sterile gauze after the liquid dropped into the wound was absorbed and dried for several minutes, and then placed in a clean cage for cage rearing, and given clean drinking water, and the mice were freely eaten and changed each day.
(1) The healing time of the left and right wounds of the experimental groups 1-4 and the left and right wounds of the control group 5 and 6 were recorded, respectively, and the average value of each group was recorded and calculated, and the results are shown in table 2.
(2) Wound healing rates were recorded for each group of mice respectively: the healing conditions of the wounds on the left side and the right side of the mouse are contrastively observed after 1,3,5,7, 10 and 15 days after the drug administration, the area of the wound surface is measured, and the healing rate is calculated. The healing rate = { (original wound area-present wound area) ÷ original wound area } × 100%. The average of each group was calculated and the results are shown in table 3.
2.3 test results and analysis
Table 2 mean wound healing time in groups of wounded mice
