Test example 2 pharmacodynamic experiments of the foot bath drug on promoting wound healing in animals
2.1 design of wound model
The mice were ICR mice, purchased from Chengdu laboratory animal technologies Inc., were male, had body weights of 25-30g, and were tested after 5 days of free feeding, with body weights ranging from about 30-35 g.
After the mice were anesthetized with 10% chloral hydrate at a dose of 0.003mL/g, the body hair of 2cm x 4cm on the back was removed, the body was cleaned with physiological saline, and then sterilized with 75% alcohol, and then round full-thickness skin with a diameter of 1cm was excised at the side of the back spine of the mice at a position of 2cm, and the depth was reached to the subcutaneous (skin to muscular layer) without damaging fat and fascia on the side of the spine or on the abdominal muscle, and one wound was left and right on each side of the spinal column of the mice. The wound after operation is sterilized with 75% alcohol and covered with sterile gauze.
2.2 grouping of animals and wound treatment
Mice were randomly divided into 6 groups of 6 mice each after molding.
The left wounds of the mice in the experimental groups 1-4 and the four groups are respectively dripped with 0.2ml of solvent (0.01M PH7.2 tris hydrochloride buffer solution) as a control, the right wounds are respectively given with the traditional Chinese medicine composition solution prepared in the examples 1-4, after a few minutes, the liquid dripped into the wounds is absorbed, dried, wrapped by sterile gauze, placed in a clean cage for cage feeding, and fed with clean drinking water, taken freely, and changed for each day.
The left wound and the right wound of the mice in the control group 5 are dripped with 0.2ml of solvent (0.01M PH7.2 tris hydrochloride buffer solution), after a few minutes, the dripped liquid in the wounds is absorbed, dried and wrapped by sterile gauze, and the mice are placed in a clean cage for cage feeding, drinking water and free feeding, and changing medicine every day.
The left side wounds of the mice in the control group 6 are respectively dripped with 0.2ml of solvent (0.01M PH7.2 tris hydrochloride buffer solution) as a control, the right side wounds are respectively dripped with 0.2ml of a commercially available traditional Chinese medicine composition, an aqueous solution of the Makino hemorrhoids ointment is selected, after a few minutes, liquid dripped into the wounds is absorbed, dried and wrapped by sterile gauze, and the mixture is placed in a clean cage for cage feeding, and is free to take food for daily dressing.
(1) The healing time of the left and right wounds of experimental groups 1 to 4 and the healing time of the left and right wounds of control group 5 and control group 6 were recorded, and the average value of each group was recorded and calculated, and the results are shown in table 2.
(2) Wound healing rates of mice in each group were recorded separately: and (3) comparing and observing the wound healing conditions of the left side and the right side of the mice after 1,3,5,7, 10 and 15 days after the administration, measuring the area of the wound surface, and calculating the healing rate. Healing rate = { (primary wound area-current wound area)/(primary wound area } ×100%). The average value of each group was calculated and the results are shown in Table 3.
2.3 test results and analysis
TABLE 2 average wound healing time for groups of wounded mice
TABLE 3 average wound healing Rate of various groups of wounded mice at different times
Referring to Table 2, it can be seen that the mice of the control group 5 had neither left wound nor right wound applied the Chinese medicinal composition of the present invention, and the healing time was 14 to 15 days. The traditional Chinese medicine composition prepared in the embodiment 1-5 of the invention is applied to mice in the experimental group 1-4, so that the traditional Chinese medicine composition has obvious effect on promoting wound healing of the mice, specifically, the traditional Chinese medicine composition of the invention is not applied to left wounds of the mice in the experimental group 1-4, the healing time is 13-15 days, and the healing time of right wounds to which the traditional Chinese medicine composition of the invention is applied is 9-11 days, so that the traditional Chinese medicine composition can promote wound healing, increase wound repair speed and effectively shorten the healing time of wound surfaces. The traditional Chinese medicine composition has obvious effects of easing pain, diminishing inflammation and promoting wound healing.
The commercial Chinese medicinal composition of the control group 6 has a healing time of 12 days, which is shorter than that of the control group 5, but is inferior to the experimental groups 1-4 related to the invention, so that the Chinese medicinal composition related to the invention has better effect.
Referring to Table 4, it can be seen that the mice of test group 1-4 had a significantly slower healing rate of the left wound than the right wound, for example, on day 7, the mice of test group 3 had a healing rate of 36.8% of the left wound, and the right wound had reached 71.8%, and it was seen that the right wound to which the Chinese medicinal composition of the present invention was applied had a faster healing rate.
In contrast, when the experimental group is compared with the control group, for example, the data of the control group 4 and the control group 5 show that the healing rate of the left wound and the right wound of the mice in the control group is only 67.1% at 15 days, the healing rate of the left wound of the mice in the experimental group 4 is 65.7% at 15 days, and the healing rate of the right wound of the mice in the experimental group reaches 99.6% which is higher than that of the mice in the control group 5.
Comparing the data of the control group 6 with the experimental group 3 and the control group 5, the traditional Chinese medicine composition of the control group 6 has a certain effect of promoting wound healing, for example, the wound healing rate of the right side of the control group 6 reaches 84.9% on the 15 th day, which is higher than the wound healing rate of the control group 5 by 67.1%; at this time, the healing rate of the right side of the experimental group 3 reaches 99.6%, which is higher than that of the control group 6, and therefore, the traditional Chinese medicine composition has better effect.
In summary, the traditional Chinese medicine composition for relieving pain and bleeding takes pure traditional Chinese medicines as raw materials, the raw materials are synergistically enhanced, and cortex phellodendri, herba violae, platycodon grandiflorum, cassia twig and raw liquorice added in the raw materials contain rich amino acids and active enzymes, so that the immunity can be improved, and the wound healing capacity of a patient is improved; the berberine contained in the phellodendron can effectively obtain antifungal, antiviral and antibacterial effects, so that the traditional Chinese medicine composition has good antibacterial and anti-inflammatory effects, and the added alum and phellodendron cooperate to enhance the hemostatic effect of the composition; further, the platycodin rich in platycodon grandiflorum is matched with berberine, so that the antibacterial effect of the berberine is enhanced; the ligusticum chuanxiong hort contains alkaloid ligustrazine, and glycyrrhizin in raw liquorice and cassia twig aldehyde in cassia twig are matched, so that the antibacterial, antiviral and analgesic effects of the cassia twig are enhanced. The traditional Chinese medicine composition performs antibiosis and antiphlogosis through multiple dimensions, and various components are cooperated to form an effective component for antibiosis, antiphlogosis and analgesia, so that pain is effectively improved, analgesia and antiphlogosis are promoted to heal wounds. Solves the problems of poor product effect and high cost in the traditional Chinese medicine field for relieving pain after anorectal operation.
