Test example III, skin repair Effect test
1. Experimental sample
The anti-acne cream containing the antibacterial peptide composition prepared in examples 4 to 6 and comparative examples 1 to 6 was tested for skin moisture, elasticity, antioxidation and whitening effects.
2. Experimental method
2.1 skin moisture content (MMV) test
45 volunteers are selected and randomly divided into 9 groups, 5 persons in each group mark the inner sides of the left arm and the right arm of the volunteers with test areas of 3cm multiplied by 3cm, and the same arm can mark a plurality of areas at intervals of 1cm. Test samples were randomly distributed on the left and right arms. Test area and pair using capacitive skin assayAnd (5) measuring the illumination area. The blank value of each test area was measured first and then at a rate of 2.0mg/cm 2 The test product was uniformly coated in the test area using latex fingerstall. After 2h of application, the skin moisture content of the test and control areas was measured and the average value was recorded. The MMV value calculation method is as follows:
skin moisture content increase rate (%) = (post-application moisture measurement-pre-application moisture measurement)/pre-application moisture measurement x 100%
2.2 skin elasticity test
Selecting 100 volunteers, randomly dividing into 10 groups of 10 persons, wherein 1 group is blank group, using water as test sample, sequentially and circularly marking left and right cheeks of a subject, randomly distributing test and control substances on the left and right cheeks, smearing corresponding samples on a test area and a control area for 2 times in the morning and evening every day, and respectively smearing 2.0mg/cm every time 2 After 4 weeks of continuous use, the measured parts were measured using a skin elasticity tester Revisclometer RV600 and the average value was recorded.
The skin elasticity value is expressed by the ratio of the rebound quantity of the skin without negative pressure to the maximum stretching quantity with negative pressure, and the closer the ratio is to 1, the better the skin elasticity is. The test adopts the elasticity parameter R value as an evaluation basis, is used for evaluating the change and influence of a sample on skin elasticity, and calculates the elasticity change rate by the following method:
elastic change rate = R value after sample application/R value before sample application
2.3 antioxidant test
The free activity of sample clearance was determined using the 1,1-diphenyl-2-picrydrazyl (DPPH) method. The samples of examples 4 to 6 and comparative examples 1 to 6 were prepared as solutions having a concentration of 1mg/ml using purified water as a diluent, 1.0ml was taken, 3.0ml of DPPH solution (0.004% concentration) was added thereto, and the reaction was carried out at room temperature for 30 minutes in the absence of light, while the absorbance was measured at a wavelength of 517nm using absolute ethyl alcohol as a blank. The DPPH radical scavenging rate was calculated as follows:
clearance (%) = (absorbance after 1-sample DPPH reaction/blank DPPH absorbance value) ×100%
2.4 whitening test
The test adopts a test method for melanin inhibition of the melanoma cells of the mice, and is used for evaluating the whitening efficacy of the sample.
Preparing a sample solution: the samples of examples 4 to 6 and comparative examples 1 to 6 were prepared as solutions having a volume concentration of 1% using a serum-containing complete culture solution as a diluent, and were sterilized by filtration through a 0.22 μm filter.
Taking melanoma cells of B16 mice in logarithmic growth phase at a ratio of 2×10 4 The cells were inoculated at a density of 100. Mu.L/well into 96-well cell culture plates, and incubated overnight in a carbon dioxide incubator. The old culture solution is discarded, a sample with the volume concentration of 1% is added into the experimental group, the same amount of serum-containing cell culture solution is added into the normal control group, and after the culture is continued for 24 hours, the relative proliferation rate (RGR) of cells is detected by adopting a WST-1 method.
Taking B16 cells in logarithmic growth phase at 2×10 4 Inoculating 3mL of each of the culture plates with the density of 3mL in 6 holes, placing the culture plates in a carbon dioxide incubator for conventional culture for 24 hours, discarding old culture solution, adding 3mL of serum-containing culture solution into a normal control group and a model group, adding 3mL of sample solution into the model group, adding 60 mu L of forskolin solution (2 mM) into each hole of the model group and the model group, stimulating melanin production, and continuously culturing for 72 hours.
Measurement of intracellular melanin content: pancreatin digestion, centrifugation to remove supernatant, adding 1mol/L NaOH solution 500 μL to lyse cells, heating at 80deg.C for 30min, centrifuging at 3000rpm for 10min, adding 96-well plate into the supernatant, 100 μL/well, measuring 450nm absorbance to obtain total melanin, taking the melanin content of model group as 100%, obtaining the relative value and inhibition rate of melanin content of experimental group, and calculating total melanin content inhibition rate by the following method:
inhibition (%) = (1-experimental group absorbance value/model group absorbance value) ×100%
