Abstract
本发明公开了一种用于增强光动治疗的自增敏型纳米组装体及其制备方法与应用,属于药物制剂联合治疗新辅料和新剂型技术领域。所述自增敏型纳米组装体由光敏剂和藤黄酸共组装,共组装后修饰以PEG修饰剂,所述光敏剂与藤黄酸的摩尔比为5:1~1:2,光敏剂和藤黄酸质量之和与PEG修饰剂的质量比为100:0~70:30。本发明的共组装纳米制剂为开发药物的递送提供新的策略和更多的选择,满足临床中对光动治疗策略在制剂上的迫切需求。
Description
技术领域
本发明属于药物制剂联合治疗新辅料和新剂型技术领域,具体涉及一种用于增强光动治疗的自增敏型纳米组装体及其制备方法与应用。
背景技术
虽然化疗是最常用的临床癌症治疗手段,但经常会导致患者出现耐药性和严重的副作用,从而导致临床治疗效果不理想。为了处理无序和难治性肿瘤,光动力疗法(PDT)等新型治疗方式已得到广泛研究。与其他治疗方法不同,PDT作为一种局部治疗方式,由于其高选择性和低毒性而备受关注。因此,各种光敏剂(PSs)被设计成通过将近红外光转化为肿瘤局部活性氧(ROS)来发挥有效的抗肿瘤活性。换言之,PDT已成为癌症治疗方案的新兴选择。
尽管具有独特的优势,但PDT的效率通常受到不利的肿瘤微环境的限制,尤其是细胞内抗氧化系统。人体内有两个独立的抗氧化系统:硫氧还蛋白(Trx) 系统(包括NADPH、硫氧还蛋白还原酶(TrxR)和硫氧还蛋白(Trx))和谷胱甘肽(GSH)系统(包括NADPH、GSH和GSH还原酶(GR))。在这两个系统中,GR和TrxR分别催化电子从NADPH转移到GSSG和Trx。此外,这两个系统还可以参与肿瘤细胞内后续反应以维持氧化还原平衡以及保护肿瘤细胞免受ROS损害的基本解毒机制。因此在PDT期间抑制肿瘤细胞中的抗氧化系统可以显着提高PDT的疗效。因此,将抗氧化阻滞剂和PSs高效同步输送至肿瘤是增强PDT的可靠选择,但也是一项重大挑战。
随着纳米技术和生物医学的快速发展,各种纳米载体被设计用于递送抗癌药物。虽然一些药物可以通过载体的封装成功地递送到肿瘤部位,但这些基于载体材料构建的纳米粒子(NPs)的一些明显缺点也同时暴露出来,例如药物过早泄漏、载药量低和载体相关的毒性等。最近,无载体纳米药物递送系统(CFNDS) 逐渐受到研究人员的关注。在CFNDS中,药物可以在不借助任何载体的情况下形成稳定的纳米组装体,可以是单一药物的自组装,也可以是多种药物的共组装。由于组装过程中不涉及载体材料,大大提高了载药效率和递送效率,同时也避免了载体引起的系统毒性。CFNDS的出现和发展为肿瘤组织的药物递送带来了良好的前景和原理。
近年来,通过纯药物自组装的无载体纳米粒在药物递送中有着广阔前景,特别是对于某些无需载体材料即可自组装成稳定纳米粒的抗癌药物,构建具有多种药物分子的无载体纳米组装体在联合疗法中更具潜力。研究开发一种具有增强光动治疗的自增敏型纳米组装体是当前亟待研究的重要课题。
发明内容
基于现有技术存在的问题,本发明为了进一步消除抗氧化系统对PDT的抑制作用以及其大部分光敏剂水溶性差、组装能力差、包载于聚合物中导致载药量低、药物泄露和辅料相关毒性等问题,我们在此构建了一种双药无载体纳米组装体,以增强肿瘤的PDT功效。在这个组装过程中,焦脱镁叶绿酸a(PPa)和藤黄酸(GA)可以很容易地共同组装成均匀的纳米粒,而无需其他载体或赋形剂。然后,通过一步纳米沉淀法制备非聚乙二醇化纳米粒子(PPa@GA纳米粒)。为了提高PPa@GA纳米粒的稳定性,DSPE-PEG2K(20wt%)被用作稳定化改性剂来构建PEG化的纳米粒(PPa@GA/DSPE-PEG2K纳米粒)。一旦PPa@GA/DSPE-PEG2K纳米粒进入肿瘤细胞,GA可以通过抑制Trx和GSH直接破坏细胞内的抗氧化系统。因此,GA可以显著提高PDT的效果,并在激光照射下PPa具有显着的协同抗肿瘤作用。正如预期的那样,PPa@GA/DSPE-PEG2K纳米粒在携带4T1乳腺肿瘤的BALB/c小鼠中发挥了高效的肿瘤抑制作用。这种双重药物递送策略为提高PDT或其他联合疗法的敏感性提供了一个有前途的纳米平台和可参考的方案。
本发明通过以下技术方案实现上述目的:
本发明提供了一种用于增强光动治疗的自增敏型纳米组装体,所述纳米组装体由光敏剂和藤黄酸通过分子间作用力共组装而成,并修饰以PEG修饰剂,所述光敏剂与藤黄酸的摩尔比为5:1~1:2,优选为3:1;光敏剂和藤黄酸质量之和与PEG修饰剂的质量比为100:0~70:30。
进一步地,所述分子间作用力包括π-π堆积、疏水作用及氢键。
进一步地,所述的光敏剂包括焦脱镁叶绿酸a、叶绿素a、脱镁叶绿酸a、焦脱镁叶绿酸a己醚、二氢卟吩e6中的一种或二种以上。优选为焦脱镁叶绿酸a。
进一步地,所述PEG修饰剂包括PCL-PEG、DSPE-PEG、DSPE-SS-PEG、 PLGA-PEG、PE-PEG中的一种或二种以上,PEG的分子量为200-20000。优选为DSPE-PEG2K。
本发明还提供了一种用于增强光动治疗的自增敏型纳米组装体的制备方法,包括如下步骤:
将光敏剂,藤黄酸和PEG修饰剂分别溶解到有机溶剂中,搅拌下混匀,将混匀后的溶液缓慢滴加到水中,搅拌下自发形成均匀的共组装纳米粒,;除去有机溶剂,即得。
进一步,所述的有机溶剂包括乙醇、四氢呋喃、二甲基亚砜中的一种或任意两种及以上的组合。
进一步地,所述有机溶剂优选为四氢呋喃和乙醇。
进一步,除去有机溶剂的方法包括溶剂蒸发法、超滤法和膜渗透法。
本发明还提供了上述方法制备的光敏剂和藤黄酸共组装形成的自增敏型纳米组装体。
本发明还提供了光敏剂和藤黄酸共组装形成的自增敏型纳米组装体在制备药物递送系统中的应用。
本发明还提供了光敏剂和藤黄酸共组装形成的自增敏型纳米组装体在制备抗肿瘤药物中的应用。
本发明还提供了光敏剂和藤黄酸共组装形成的自增敏型纳米组装体在制备注射给药、口服给药或局部给药系统中的应用。
本发明相对于现有技术具有的有益效果如下:
1.本发明制备的光敏剂(优选为PPa)和藤黄酸共组装形成的自增敏型纳米组装体,可用于肿瘤光动治疗,GA可以通过抑制肿瘤细胞内Trx的表达从而增强光动力治疗,光敏剂在光照条件可以产生大量ROS。
2.本发明的光敏剂(优选为PPa)和藤黄酸共组装形成的自增敏型纳米在组装体实现了载药量高、稳定性好、毒副作用低等技术效果,满足临床中对高效低毒制剂的迫切需求,为光动治疗中光敏剂与其他药物协同联用的组装提供了一个新策略,为开发无载体的杂化纳米组装体以及增强光动治疗提供了一个有效的纳米平台。

具体实施方式
下面结合实施例对本发明进行详细的说明,但本发明的实施方式不限于此,显而易见地,下面描述中的实施例仅是本发明的部分实施例,对于本领域技术人员来讲,在不付出创造性劳动性的前提下,获得其他的类似的实施例均落入本发明的保护范围。
实施例1:PPa与GA协同比例的筛选
使用MTT法检测了PPa和GA在不同比例下(PPa:GA=5:1~1:5)的协同细胞毒性并计算出协同指数。具体操作如下:将处于对数生长期的4T1细胞消化并重新分散,用细胞计数板计算细胞密度后用培养基稀释至1×104细胞/mL,向96孔板的各孔中加入200μL的细胞悬液,调零孔用PBS补齐,置于含5%CO2的37℃培养箱中培养过夜。弃去培养液,分别向各孔内加入含有一系列浓度梯度各组制剂的新鲜培养液,每个浓度平行设置3孔,对照孔加入200μL的空白培养液,调零孔用PBS补齐。将96孔板置于培养箱中培养4小时后取出,对光照组的各孔进行光照(660nm,50mW/cm2,5分钟),光照结束后,将96孔板置于培养箱中继续培养44小时。培养完毕后取出96孔板,每孔加入25μL的MTT 溶液(5mg/mL),37℃保温4小时后弃去板中液体,然后每孔加入200μL DMSO,置于振荡器上振荡10分钟后,利用酶标仪测定490nm下的吸光度值。通过以下公式计算抑制率,并通过Graphpad 8.0软件拟合各组制剂的半数抑制浓度(IC50)。

其中,“A”与“B”分别代表混合溶液中GA和PPa所占摩尔百分比;GAalone和PPaalone分别代表GA或PPa单一溶液剂的IC50值;PPa@GAcombine代表混合溶液的IC50值。当CI<1代表两药协同;CI=1代表联用时的作用是单独使用时的加和;CI>1代表两药拮抗。
检测到的PPa与GA的细胞毒以及在各比例下IC50及CI结果见表1-2。

如表2所示,当PPa与GA的摩尔比为5:1~1:2时,均表现为协同作用。其中,当PPa与GA的摩尔比为3:1时,具有最好的协同效果,CI为0.52,初步优选PPa与GA的比例为3:1。后续实验均在PPa:GA的比例为3:1的条件下进行。
实施例2:PPa@GA/DSPE-PEG2K纳米粒的制备
将不同摩尔比的PPa和GA溶解到四氢呋喃中,得2.5mg/mL(PPa浓度) 含药溶液;将PEG修饰剂溶解到无水乙醇中,并取出25μL加入到200μL 的含药溶液中。搅拌下,将该溶液缓缓滴加到2mL去离子水中,PPa和GA 自发形成均匀的纳米粒,然后采用溶剂蒸发法除去纳米制剂中的有机溶剂,得到不含任何有机溶剂的纳米胶体溶液。
检测所制备的纳米制剂的粒径、粒径分布结果见表3。

如表3所示,纳米粒的粒径都在130-230nm之间。结合协同结果,其中 PPa:GA=3:1时,PPa@GA/DSPE-PEG2K纳米粒的协同效果最好且粒径较小以及分布较均匀。
(1)非PEG化的PPa@GA纳米粒的制备方法:精密称取PPa 0.36mg,和GA 0.14mg,用200μL四氢呋喃将其溶解,搅拌下,将该溶液缓缓滴加到 2mL去离子水中,自发形成均匀的纳米粒,然后经过溶剂蒸发法除去纳米制剂中的有机溶剂,得到不含任何有机溶剂的纳米胶体溶液。
(2)DSPE-PEG2K修饰的PPa@GA/DSPE-PEG2K纳米粒制备方法:精密称取PPa 0.36mg和GA 0.14mg,用200μL四氢呋喃将其溶解,再精密称取不同比例的DSPE-PEG2K,用乙醇溶解,取25μL加入到200μL的含药溶液中;在搅拌下,将三者的混合溶液缓缓滴加到2mL去离子水中,得到均匀 PPa@GA/DSPE-PEG2K纳米粒。然后经过溶剂蒸发法除去纳米制剂中的有机溶剂,得到不含任何有机溶剂的纳米胶体溶液。通过动态光散射法检测所制备的纳米粒的粒径、粒径分布(表4、图1-3)和载药量(表5)。
注:后文PPa@GA/DSPE-PEG2K纳米粒均指代采用PEG修饰的 PPa@GA/DSPE-PEG2K纳米粒,PPa@GA纳米粒均指代未采用PEG修饰的 PPa@GA纳米粒。

从图1,表5和表6可知,质量分数为20%比例下,PPa@GA/DSPE-PEG2K纳米粒和PPa@GA纳米粒的粒径和PDI较好,PEG化的纳米粒,zeta电位为 -30左右,载药量高达80%
通过透射电子显微镜测定实施例1中制备的PPa@GA/DSPE-PEG2K纳米粒和PPa@GA纳米粒的粒径和形态,结果如图4和图5,透射电镜图表明纳米粒为均一的球形,粒径在90nm和120nm左右。
实施例3:纳米粒的胶体稳定性试验
将实施例2中制备的PPa@GA/DSPE-PEG2K纳米粒和PPa@GA纳米粒(0.25 mg/mL)各取出1mL,加入到9mL含有10%FBS的PBS(pH 7.4)中,在37℃的摇床中孵育24小时,并且在预定的时间点(0、1、2、4、8、12和24小时) 通过动态光散射法测定其粒径变化。结果如图6所示,与PPa@GA纳米粒相比, PPa@GA/DSPE-PEG2K纳米粒在含有10%FBS的PBS中胶体稳定性较好,在24 小时内粒径没有发生明显的变化。
实施例4:PPa和GA组装机理分析
通过分子对接模拟实验,初步研究了纳米组装体的组装机理。为了进一步研究组装过程中可能涉及的力,进行了分子间力破坏实验。PPa@GA纳米粒加入到氯化钠(NaCl)、十二烷基硫酸钠(SDS)或尿素(Urea)的溶液中,37℃摇床孵育。在预定的时间点,使用MalvernZetasizer对PPa@GA纳米粒的粒径进行了表征。通过紫外光谱扫描实验,验证了π-π堆积作用力的存在。使用酶标仪测定PPa溶液剂、PPa@GA纳米粒或PPa@GA/DSPE-PEG2K纳米粒(10μg/mL, PPa当量)的紫外吸收光谱。通过荧光光谱扫描实验,验证了聚集诱导淬灭(ACQ) 效应的存在,证明了纳米粒的成功组装。使用酶标仪测定PPa溶液剂、PPa@GA 纳米粒或PPa@GA/DSPE-PEG2K纳米粒(10μg/mL,PPa当量)的荧光激发和发射光谱。
结果表明,如图7所示,组装过程中,参与组装的分子间作用力包括π-π堆积、疏水作用力和氢键。随后,通过分子间力破坏试验继续研究了组装机制,结果在NaCl和尿素溶液中,PPa@GA纳米粒的粒径几乎没有变化,说明静电力和氢键力对组装的影响最小。相比之下,PPa@GA纳米粒在SDS溶液中结构破坏严重,粒径显著增大,说明疏水作用力在PPa@GA纳米粒的组装过程中起关键作用(图8)。PPa@GA纳米粒的紫外吸收光谱与PPa溶液剂相比有明显的红移,这证明了PPa@GA纳米粒中π-π堆积作用力的存在(图9)。因此,PPa@GA纳米粒的组装过程主要由π-π堆积和疏水作用力驱动。荧光光谱结果(图10-11) 显示,PPa@GA纳米粒和PPa@GA/DSPE-PEG2K纳米粒的激发和发射光谱的荧光强度显着降低,这可能归因于聚集引起的猝灭(ACQ)效应,这表明纳米组件的成功形成。总之,这些结果表明,多种力(主要是疏水力)参与了PPa和GA 的组装过程,从而促进了纳米组装体的成功形成。
实施例5:纳米粒的体外单线态氧产生
用SOSG探针测定光诱导的单线态氧(1O2)的生成。简单地说,将PPa溶液剂、GA溶液剂、PPa@GA溶液剂(PPa与GA按摩尔比3:1的混合溶液剂,后文的PPa@GA溶液剂均为该比例)、PPa@GA纳米粒或PPa@GA/DSPE-PEG2K纳米粒(1mM,PPa当量)分别与SOSG(1mM)混合。样品在近红外(660nm, 250mW/cm2,5分钟)光照射下生成的1O2可以与荧光探针结合。用酶标仪测量SOSG结合物的荧光强度。
结果如图12所示,正如预期的那样,含有PPa的激光照射组显示出比没有激光处理的组更高的1O2生成能力,表明PPa驱动和激光敏化的1O2生成能力。此外,与PPa溶液剂和PPa@GA溶液剂相比,PPa@GA纳米粒和 PPa@GA/DSPE-PEG2K纳米粒产生的1O2的量呈现出特定的减少,这可能归因于 ACQ效应。
实施例6:纳米粒的细胞摄取
采用共聚焦显微镜测定实施例2中制备的PPa@GA/DSPE-PEG2K纳米粒在 4T1细胞(小鼠乳腺癌细胞)中的摄取情况。将4T1细胞以1×105cells/well的密度接种到24孔板上,置培养箱中孵育24小时使其贴壁,待细胞贴壁后加入 PPa溶液剂、PPa@GA纳米粒和PPa@GA/DSPE-PEG2K纳米粒,PPa的浓度均为 200nM,在37℃孵化2小时和4小时后,清洗细胞,进行细胞固定,在加入Hoechst 染核,最后用共聚焦显微镜分析细胞对各种制剂的摄取情况。实验结果如图13 所示。通过流式细胞术对2小时和4小时的细胞摄取情况进行定量分析,实验结果如图14所示。
上述实验结果表明,细胞摄取呈现出时间依赖性摄取,且 PPa@GA/DSPE-PEG2K纳米粒处理的细胞比PPa溶液剂处理的细胞具有更高的细胞内荧光强度。因此,制备的PPa@GA/DSPE-PEG2K纳米粒具有比PPa溶液剂更高的细胞摄取效率。
实施例7:纳米粒的细胞毒性
采用MTT法考察了PPa溶液剂、PPa溶液剂+光照、GA溶液剂、PPa@GA 溶液剂、PPa@GA溶液剂+光照、PPa@GA纳米粒、PPa@GA纳米粒+光照、 PPa@GA/DSPE-PEG2K纳米粒和PPa@GA/DSPE-PEG2K纳米粒+光照对小鼠乳腺癌(4T1)细胞的细胞毒性。将状态良好的细胞消化,用培养液稀释至1×104cells/mL的细胞密度,吹匀后于96孔板中每孔加入细胞悬液200μL,调零孔用 PBS补齐,然后置于培养箱中孵育12小时使其贴壁。待细胞贴壁后用含药的培养基培养细胞,每孔200μL。对照组用不含药液的培养基培养,调零孔用PBS 补齐。4小时后,光照组给予激光照射。再继续培养44小时后,将96孔板取出,每孔加入5mg/mL MTT溶液25μL,置培养箱中孵育4小时后甩板,将96孔板倒扣于滤纸上充分吸干残留液体后,每孔加入200μLDMSO于振荡器上振荡10 min以溶解蓝紫色结晶物。使用酶标仪在490nm处测定各孔调零后的吸光度值。
细胞毒性结果如图15所示,没有激光治疗组的PPa溶液剂几乎没有毒性。在激光照射(660nm,50mW/cm2,5分钟)下,PPa@GA溶液剂、PPa@GA纳米粒和PPa@GA/DSPE-PEG2K纳米粒比单独的PPa溶液剂或GA溶液剂表现出更强的细胞毒性。与PPa@GA溶液剂和PPa@GA纳米粒相比, PPa@GA/DSPE-PEG2K纳米粒表现出更有效的抗肿瘤活性,这可归因于合理的剂量比、良好的胶体稳定性和PEG化纳米组件的细胞摄取。
实施例8:纳米粒的活性氧(ROS)的产生
为了评估细胞内ROS水平,将4T1细胞(1×105cells/well)接种到24孔板中,并在37℃下孵育12小时。然后,用含有PPa溶液剂、GA溶液剂、PPa@GA 溶液剂、PPa@GA纳米粒和PPa@GA/DSPE-PEG2K纳米粒的新鲜培养基替换旧培养基,PPa剂量当量为200nM,并孵育4小时(37℃,黑暗)。然后,所有组用冷PBS洗涤,用DCFH-DA(10μM)处理,并孵育30分钟(37℃)。然后,将激光处理组的细胞暴露于激光(660nm,50mW/cm2)下5分钟。最后,用冷 PBS洗涤细胞3次,并使用倒置显微镜记录细胞荧光信号。通过流式细胞术对细胞内产生的ROS进行定量分析。
结果如图16和17所示,GA培养基组除了PPa激光暴露组外还显示出增加细胞内ROS的能力,这表明具有打破氧化还原平衡的能力。与单独的PPa或GA 相比,由于PPa和GA的协同作用,在用PPa@GA溶液剂、PPa@GA纳米粒和 PPa@GA/DSPE-PEG2K纳米粒处理的细胞中观察到更高的ROS水平。正如预期的那样,PPa@GA/DSPE-PEG2K纳米粒表现出与细胞毒性结果一致的最高水平的 ROS积累。
实施例9:肿瘤细胞内GSH浓度和硫氧还蛋白浓度检测。
细胞内GSH浓度采用微量还原型GSH探针检测。将4T1细胞(1×105个/ 孔)接种到24孔板上,在37℃下孵育12小时。12小时后,分别用空白培养基、空白培养基+光照、PPa溶液剂、PPa溶液剂+光照、GA溶液剂、GA溶液剂+光照、PPa@GA溶液剂、PPa@GA溶液剂+光照、PPa@GA纳米粒、PPa@GA纳米粒+光照、PPa@GA/DSPE-PEG2K纳米粒或PPa@GA/DSPE-PEG2K纳米粒+光照,按PPa和GA等量处理细胞,孵育4小时后给予光照组激光(660nM,50mW/cm2, 5分钟)。结束后收集细胞,并用PBS(pH 7.4)清洗1~2次,低速离心收集沉淀细胞,再加入0.3~0.5mL(0.1M,pH 7.4)的PBS缓冲液悬浮细胞,超声破碎细胞待测。在96孔板中进行下一步操作并按制造商的说明加入GSH探针,孵育5 分钟,405nm处酶标仪测定各孔吸光度值。实验结果如图18所示。
使用TXN ELISA试剂盒评估GA对Trx活性的抑制作用。简而言之,将4T1 细胞(6×105/孔)接种到培养皿(100mm)中并孵育12小时。然后,用含有 GA溶液剂(67nM)、PPa溶液剂(200nM)、PPa@GA溶液剂、PPa@GA纳米粒或PPa@GA/DSPE-PEG2K纳米粒(200nM,PPa等量)的新鲜培养基替换旧培养基并孵育48小时。温育后,用冷PBS洗涤细胞3次并离心。弃去上清,将细胞用PBS(pH 7.4)重悬,细胞浓度达到约1×106个细胞/mL。通过反复冻融来破坏细胞并释放其内容物。然后,将混合物离心约20分钟(3000转/分),小心收集上清液。接下来的程序是按照制造商的说明进行的。用酶标仪在450nm处测定吸光度,用标准曲线计算Trx含量,结果如图19所示。
结果表明,在激光照射(660nm,50mW/cm2 5分钟)下,PPa产生的ROS 可以抵消和降低GSH的水平。重要的是,GA和PPa可以在激光照射下协同下调GSH的表达,有利于增加ROS积累和致敏PDT。同时,GA能够抑制Trx表达,协同下调Trx。这些结果表明,GA可以降低GSH和Trx水平以抑制细胞内抗氧化系统,协同放大PDT的抗肿瘤活性。
实施例10:纳米粒的药代动力学研究
取体重在200-250g之间的SD大鼠,随机分组,给药前禁食12小时,自由饮水。分别静脉注射PPa溶液剂以及实施例2制备的PPa@GA纳米粒和 PPa@GA/DSPE-PEG2K纳米粒(均以PPa计2.5mg/kg),于规定的时间点(0.033、 0.083、0.2、0.5、1、2、4、8、12和24小时)眼眶取血,分离获得血浆。之后通过离心和沉淀蛋白法提取PPa,最后用酶标仪(激发415nm,发射675nm) 检测各制剂的药动学行为。实验结果如图20所示,由于半衰期短,PPa溶液剂很快就被代谢清除。相比于PPa溶液剂和PPa@GA纳米粒, PPa@GA/DSPE-PEG2K纳米粒的循环时间明显延长,明显提高了PPa的AUC,为药物在体内肿瘤的蓄积提高了很好基础。
实施例11:纳米粒的组织分布实验
将4T1细胞悬液接种于BALB/c小鼠,当肿瘤体积达到300mm3时,尾静脉注射给药:PPa溶液剂,PPa@GA纳米粒和PPa@GA/DSPE-PEG2K纳米粒(给药剂量均为2.5mg/kg PPa)。于给药后2小时、4小时、8小时和12小时,将小鼠猝死,进行离体组织器官的荧光强度分析并进行定量分析。
结果如图21-23所示,PPa@GA溶液剂、PPa@GA纳米粒和 PPa@GA/DSPE-PEG2K纳米粒成功地积累到肿瘤中。这种蓄积从2小时到12小时呈现先升后降的趋势,并且在纳米组装体注射后4小时左右发现肿瘤中药物蓄积的峰值。值得注意的是,PPa溶液很快被清除,伴随着较低的肿瘤积累。与PPa 不同,两种纳米组件在肿瘤组织中表现出更强的荧光强度,尤其是PPa@GA/DSPE-PEG2K纳米粒,与药代动力学结果一致。
实施例12:纳米粒的体内抗肿瘤实验
在4T1荷瘤小鼠中评估了体内抗肿瘤功效。简而言之,将4T1细胞皮下接种到每只雌性BALB/c小鼠的右侧。当肿瘤体积达到150mm3时,将小鼠随机分为8组(n=6):PBS、GA溶液剂、PPa溶液剂+光照、PPa@GA纳米粒、 PPa@GA/DSPE-PEG2K纳米粒、PPa@GA溶液剂+光照,PPa@GA纳米粒+光照和PPa@GA/DSPE-PEG2K纳米粒+光照。以3.0mg/kg的PPa剂量每隔2天将各种制剂静脉注射到4T1荷瘤雌性BALB/c小鼠体内,一共五次。每次注射后4小时,激光照射组小鼠肿瘤接受激光(5分钟,660nm,50mW/cm2)照射。每天监测小鼠的体重和肿瘤体积。最后一次处理两天后,处死小鼠,取主要器官(心、肝、脾、肺、肾)和肿瘤进行H&E染色。此外,肿瘤组织用于TUNEL染色的组织学评估,收集的血浆用于肝肾功能分析。
结果如图24-27所示,PBS处理的小鼠的肿瘤体积在处理的第10天已经增长到1000mm3。与PBS组相比,GA溶液剂、PPa@GA纳米粒和 PPa@GA/DSPE-PEG2K纳米粒对肿瘤生长表现出适度的抑制。相比之下,激光照射的联合治疗组显示出更有效的肿瘤生长抑制作用。正如预期的那样, PPa@GA/DSPE-PEG2K纳米粒+光照表现出优于PPa溶液剂+光照、PPa@GA+光照和PPa@GA纳米粒+光照的抗肿瘤活性,这主要是由于胶体稳定性好、细胞吸收率高、延长血液循环时间、高肿瘤积累以及GA和PPa协同增强ROS 积累的潜在能力。此外,H&E染色和TUNEL测定的结果也证明了激光处理的PPa@GA/DSPE-PEG2K纳米粒组中的有效肿瘤破坏(图28-30)。
最后应说明的是:以上各实施例仅用以说明本发明的技术方案,而非对其限制;尽管参照前述各实施例对本发明进行了详细的说明,本领域的普通技术人员应当理解:其依然可以对前述各实施例所记载的技术方案进行修改,或者对其中部分或者全部技术特征进行等同替换;而这些修改或者替换,并不使相应技术方案的本质脱离本发明各实施例技术方案的范围。
Claims (7)
1. 一种用于增强光动治疗的自增敏型纳米组装体,其特征在于,所述纳米组装体由光敏剂与藤黄酸通过分子间作用力共组装而成,并修饰以PEG修饰剂,所述光敏剂与藤黄酸的摩尔比为5 : 1 ~ 1 : 2,光敏剂与藤黄酸质量之和与PEG修饰剂的质量比为100 : 0 ~ 70 :30;
所述的光敏剂为焦脱镁叶绿酸a;
所述的PEG修饰剂为PCL-PEG、DSPE-PEG、DSPE-SS-PEG 、PLGA-PEG和PE-PEG中的一种或二种以上,PEG的分子量为200-20000;
所述的用于增强光动治疗的自增敏型纳米组装体的制备方法包括如下步骤:
将光敏剂、藤黄酸及PEG修饰剂分别溶解到有机溶剂中,搅拌下混匀,将混匀后的溶液缓慢滴加到水中,搅拌下自发形成均匀的共组装纳米粒;除去有机溶剂,即得。
2.如权利要求1所述的一种用于增强光动治疗的自增敏型纳米组装体,其特征在于,所述分子间作用力包括π-π堆积、疏水作用及氢键。
3.如权利要求1所述的一种用于增强光动治疗的自增敏型纳米组装体,其特征在于,所述的PEG修饰剂为DSPE-PEG2K。
4.如权利要求1所述的一种用于增强光动治疗的自增敏型纳米组装体,其特征在于,所述的有机溶剂为乙醇、四氢呋喃、二甲基亚砜中的一种或两种以上的组合。
5.权利要求1-4任何一项所述的用于增强光动治疗的自增敏型纳米组装体在制备药物传递系统中的应用。
6.权利要求1-4任何一项所述的用于增强光动治疗的自增敏型纳米组装体在制备抗肿瘤药物中的应用。
7.权利要求1-4任何一项所述的用于增强光动治疗的自增敏型纳米组装体在制备注射给药、口服给药或局部给药系统中的应用。
