技术领域

背景技术
基因疗法是聚焦于将(外来)遗传物质(例如DNA和RNA)治疗性递送到患者细胞中以治疗疾病的医学领域。迄今为止,包括(RPE65突变诱导的失明)和(嵌合抗原受体T细胞疗法)在内的几种基因疗法已经获得了针对多种不同医学病症的监管批准。
基因递送是指用于将遗传物质引入细胞的过程。为了获得成功,遗传物质在运输期间必须保持稳定,并最终被内化到靶细胞中。当遗传物质是DNA时,则其必须内化到靶细胞中并被传递到细胞核中。基因递送需要载体,而合适的载体通常分为两类:病毒载体和非病毒载体。
病毒介导的基因递送利用病毒将其DNA注入到宿主细胞内的能力。遗传物质被包装成复制缺陷型病毒颗粒,以形成病毒载体。病毒方法是高效的,但可能诱导免疫应答。此外,这些方法只能将非常小块的遗传物质递送入细胞,生产病毒载体是劳动密集型的,并且存在随机插入位点、细胞病变效应和诱变的风险。
在结构通用性和可扩展性方面,在基因递送应用方面,合成载体与病毒载体相比具有多个优势;且合成载体可单独且特定设计以实现所希望的目的。可以设计这些物质以将遗传物质包装成纳米颗粒或囊泡,这些纳米颗粒或囊泡被工程化来克服与细胞摄取、运输到细胞溶质以及(如果需要)递送到细胞核相关的生物屏障。一种常见的方法是用材料(例如聚合物、肽或脂质,包含与阴离子核酸缔合的正电荷)将遗传物质包装成多分子组件。正负电荷之间的静电相互作用促使自组装成纳米结构或微粒结构,这些粒子的大小和形状可以由材料类型和冷凝条件控制(Park等人,Adv Drug Del Rev[先进药物输送评论],2006,58(4):467-86)。
例如,将DNA配制成合适的载体(如纳米颗粒)显著提高对DNA的细胞摄取(与对未配制的DNA的摄取相比)。DNA具有净负电荷,并且通常不会自己内化到细胞(其也具有净负电荷)中。未配制的DNA也易引发导致其降解的免疫应答。将DNA配制成合适载体可以中和其负电荷,并且保护其免受在细胞外空间的降解。
为了使载体被有效的内化,必须使其经由称为内吞作用的过程运输到细胞中。在这个过程期间,载体被细胞膜的区域包围,然后其在细胞内出芽形成内体。必须设计载体以允许该过程发生,但随后要降低被溶酶体截留(即隔离到酸性膜结合的溶酶体室中)的可能性。实现这一点的一种方法是确保在溶酶体运输发生前内体破裂。这可以通过载体缓冲内体pH(细胞摄取后内体变得愈发呈酸性)来实现,直到产生的渗透梯度导致内体破裂并将遗传物质释放到可以将其用于进行转录/翻译的细胞质中。在内体缓冲范围(pH 7.4-pH5.0)上具有有效的缓冲能力的合适的载体材料可以通过接受质子来减缓对内体的酸化,其导致更多的质子和反荷离子从细胞溶质中流入。
目前的合成基因递送系统在体内受到遗传物质的低稳定性、高毒性和低效的细胞质进入的限制。将适用于基因递送的多功能材料工程化(在保持高递送效率的同时,实现配制品特性(大小、电荷等)、稳定性、缓冲能力和毒性之间的最佳平衡)已被证明是困难且复杂的,但这样将显著简化最终配制品。
聚赖氨酸(PL)是携带游离胺侧臂的线性多肽,这些侧臂能与带负电荷的核酸相互作用,并经由静电相互作用形成复合物。因此,PL及其共聚物已在实验室中广泛用作非病毒基因递送中的载体。例如,当与氯喹等转染辅助剂偶联时(Yamauchi等人,Biomaterials[生物材料],200324(24):4495-4506)以及在作为嵌段共聚物或混杂系统的重要组分时(Incani等人,ACS Appl.Mater.Interfaces[美国化学会应用材料与界面],2009,1(4):841-848),PL基DNA纳米颗粒已在多种细胞系中被证明为高效转染。PL也是可生物降解的,这有利于体内应用。然而,尽管可促进对DNA的相似细胞摄取水平,但PL转染效率远低于其他阳离子聚合物转染剂(例如聚乙烯亚胺(PEI))。这种低效率的转染与所产生的DNA纳米颗粒无法从内体中逃逸(可能是因为其无法缓冲内体pH)有关(Hwang等人,Biomacro[生物大分子],2014,15(10):2577-86)。

本申请描述了某些可用于形成改性PL的改性赖氨酸,当与未改性PL相比时,改性PL对遗传物质的递送效率提高。这些改性PL是独特的,在于它们具有多功能性,不同于组氨酸改性,只促进了内体缓冲。这些改性PL:i)具有增强的缓冲特性,调整至可在细胞内化和溶酶体运输期间出现的pH转变期间实现质子化,ii)由于通过静电和非静电(例如pi-pi堆叠)相互作用增加核酸结合而更加稳定和/或iii)当使用基于代谢物的核心单元时,具有增加的生物相容性,这些核心单元是天然存在的,并且具有毒性或免疫原性的可能性较低。这些改性PL与质粒DNA形成纳米颗粒(类似于与未改性PL形成的纳米颗粒(约100nm,球体、棒体和超环状体)),并显示在小鼠静脉注射后血清稳定性较高且血液循环延长,无相关毒性。它们通过不易解离来保护被包裹的遗传物质并保持较长循环时间。
本文所述的改性赖氨酸是多功能单元,其可并入载体中并改善合成基因递送系统的转染,同时维持高生物相容性。

实例4
聚(L-赖氨酸)和PEG-聚(L-赖氨酸)多肽
通过使方法1-3中制备的NHS-酯与相应的PLL(MW 5000,阿拉曼达聚合物公司(Alamanda Polymers Inc.),亨茨维尔,亚拉巴马州)或PEG-PLL(MW 13000,阿拉曼达聚合物公司,亨茨维尔,亚拉巴马州)反应来制备改性PEG-PLL和PLL。在PLL的末端羧基端处向所有聚合物提供聚合反应引发剂残基(本文简称为PLL)或MeO-PEG-(CH2)2-NH-基团(本文称为PEG-PLL)。将PLL(20mg,2.5μmol)或PEG-PLL(20mg,1.5μmol)溶解于新鲜制备的0.1M碳酸氢钠(pH 8.0(4mL))中。将40mM的方法1、2或3的化合物在二甲基乙酰胺(DMAC)(1.5摩尔过量)中制备并在搅拌的同时逐滴添加到聚合物溶液中。通过控制NHS-酯的摩尔进料比率,制备了一系列具有不同改性程度的PLL。改性反应在12.5至50摩尔当量NHS酯:聚合物下进行。在室温下将反应搅拌1小时,并且随后使用截留分子量(MWCO)为3.5kDa的透析盒通过在pH7.4的磷酸盐缓冲盐水(PBS),并且然后在水中透析去除未反应基团。对于PLL(TM)和PEG-PLL(TM),通过将冻干产物在30%三氟乙酸/DCM中孵育30分钟,使用现有技术中使用的标准方案去除中间产物(*)上的Boc保护基团。赖氨酸改性的程度由D20中记录的H-NMR光谱,按Lys((CH2)3,δ=1.3-1.9ppm)的β、γ和δ-亚甲基质子的峰强度与来自吗啉(M)(起始材料方法1)和吗啉代-烟酸基团(起始材料方法2)(MN)(针对吗啉和吗啉代-尼克酸((CH2)2,δ=3.86和2.58ppm))和针对硫代吗啉(TM)(起始材料方法3)((CH2)2,δ=3.11和3.56ppm)的吗啉环亚甲基质子的峰值强度之和的比率确定,结果如下表所示。

实例5
含遗传物质的纳米颗粒的制备与表征
通过在中性缓冲液中混合多肽和核酸溶液制备纳米颗粒。使用标准技术(包括动态光散射(DLS)、透射电子显微镜(TEM)和溴化乙锭排除测定)对纳米颗粒进行评估,以确认纳米颗粒的形成。
a)纳米颗粒制备
将DNA(Gwiz荧光素酶;Genlantis公司(Genlantis),圣地亚哥,加利福尼亚州)(66.6μg/mL)和聚合物溶液在pH 7.0的20mM4-(2-羟乙基)-1-哌嗪乙烷磺酸(HEPES)缓冲液中制备。将聚合物溶液用PLL(50单位,阿拉曼达聚合物公司,亨茨维尔,亚拉巴马州),PEG(5K)-PLL(50单位)(阿拉曼达聚合物公司,亨茨维尔,亚拉巴马州)以及“P:PLL(M)29”、“P:PLL(MN)31”、“P:PLL(TM)28”、“P:PEG-PLL(M)33”、“P:PEG-PLL(MN)31”和“P:PEG-PLL(TM)30”制备(均通过类似于实例4的程序制备)。聚合物溶液也在HEPES中制备,因此聚合物中的胺(N)与DNA主链中的磷酸盐(P)的比率(N:P比率)将在0.5至10之间。随后将DNA溶液逐滴添加到聚合物溶液中,同时轻轻旋转以确保颗粒均匀。允许DNA/聚合物纳米颗粒在室温下形成持续30分钟。纳米颗粒溶液中DNA的最终浓度为33.3μg/mL。最初,制备了不同N:P比率(0.5-10)的复合物,且使用DNA凝胶电泳以确定核酸缩合所需的比率。
b)动态光散射(DLS)
使用ZetaSizer Nano ZS仪器(马尔文仪器有限公司(Malvern InstrumentsLtd.),伍斯特郡,英国)用绿色激光(λ=532nm)作为入射光束收集DLS数据。所有测量均在25℃下以173°的检测角进行。将如上所述在20mM HEPES缓冲液(pH 7.4)中以33.3μg的复合物pDNA/mL制备的纳米颗粒样品加载到低容量ZEN2112石英比色皿(12.5μL)中。通过累积量拟合分析推导流体动力学直径和多分散指数(PDI)。所有测量均采用自动衰减器调整和多次扫描(15-20次之间)进行,以确保准确性。所有数据点代表三个或更多个单独制备的样品的平均值。

实验1
a)绥冲
对聚合物溶液进行酸碱滴定以评估其添加酸性溶液后维持pH的缓冲能力(capacity/ability),并进行细胞内溶酶体缓冲研究以进一步评估该材料的缓冲能力。
a)酸碱滴定
将聚合物PEG(5K)-PLL(50单位)(阿拉曼达聚合物公司)、PEI(25K,波利塞斯公司(Polysciences Inc))、“P:PEG-PLL(M)35”(聚合物12)、“P:PEG-PLL(MN)39”(聚合物15)和“P:PEG-PLL(TM)33”(聚合物18)溶液(3mL)在150mM NaCl中以1mg/mL制备,并通过添加1MNaOH滴定至pH为11。搅拌时,用0.1M HCl以5μL增量滴定聚合物溶液至pH为4.5。缓冲能力报告为将聚合物溶液pH改变0.5所需的平均体积(μL)。细胞外中性pH7.4和内体酸性pH 4.5之间质子化程度的变化(Δα7.4-4.5)计算为:所添加的将pH从7.4改变为4.5的HCl摩尔数除以每种溶液中胺的总摩尔数。所有滴定实验一式三份地进行。结果显示于图1A中。
b)溶酶体缓冲
将Gwiz荧光素酶(Genlantis公司,圣地亚哥,加利福尼亚州)用两种荧光团标记(pH不敏感的绿色染料和pH敏感的红色染料),以通过测量在相同像素中共存在的绿色:红色荧光比率来估算pH。首先,使用MFP488核酸试剂盒(Mirus生物有限责任公司(Mirus Bio LLC,Madison WI),威斯康星州麦迪逊)标记DNA,并且然后使用制造商建议的方案用胺核酸试剂盒(Mirus生物有限责任公司,威斯康星州麦迪逊)进行胺功能化。采用乙醇沉淀法去除未反应的染料。随后用NHS酯pHRODO红色(赛默飞世尔公司(ThermoFischer),沃尔瑟姆,马萨诸塞州)标记DNA并经由乙醇沉淀进行纯化。用分光光度法确认和定量具有不同荧光团(每个质粒约40个染料)的DNA的改性。然后使用MFP-488和pHRODO红色双标记DNA如下所述制备纳米颗粒。对于细胞研究,将H1299细胞以每孔10,000个细胞接种在96孔板中,并使其在补充有10%FBS和1%P/S的洛斯维公园纪念研究所(Roswell ParkMemorial Institute,RPMI)培养基中附着24h。接下来,将细胞用100μL PBS洗涤两次,并用100μL OPTI-MEM洗涤一次。如上文所述,用PEI、PEG-PLL、“P:PEG-PLL(M)35”(聚合物12)、“P:PEG-PLL(MN)39”(聚合物15)和“P:PEG-PLL(TM)33”(聚合物18),通过将DNA(66.6μg/mL)和聚合物溶液以比率为5的胺与磷酸盐(最终DNA浓度为33.3μg/mL)混合在20mM HEPES中制备DNA纳米颗粒。然后在OPTI-MEM(1:10稀释)中以每孔0.1μg DNA将NP添加到细胞中。4小时时间段后,用赫斯特染色(5μg/mL储备液于PBS中持续5分钟)处理细胞,并使用荧光显微镜用EVOS细胞成像系统(赛默飞世尔公司,沃尔瑟姆,马萨诸塞州)成像。将纳米颗粒在缓冲液(酸性、中性和碱性缓冲液)中处理后,通过成像固定的透性化细胞产生近似细胞内pH的比例尺。在此测定中,pHRODO红色信号随着pH的降低而显著增加,而MFP488对pH不敏感,因此酸性囊泡呈红色或橙色,中性囊泡呈绿色。结果显示于图1B中。
实验2
纳米颗粒稳定性
针对阴离子解离和活体内药代动力学(PK)研究评估纳米颗粒的稳定性。
a)阴离子解离
如以上实例5中所述,使用PEG-PLL和以N:P为5的聚合物12、15和19与2μg Gwiz荧光素酶DNA(Genlantis公司,圣地亚哥,加利福尼亚州)制备纳米颗粒。将20mM HEPES缓冲液和硫酸葡聚糖(DS)(西格玛奥德里奇公司(Sigma-Aldrich),20mM HEPES缓冲液中5g/mL)溶液添加到每个纳米颗粒样品中,使最终pDNA浓度保持恒定,并且DS浓度在0到200mg/mL之间变化。在37℃下处理30分钟时间段后,将15μL的每种配制品添加到含有溴化乙锭的2%的电泳凝胶(赛默飞世尔科技公司)中,并使用标准E-凝胶方案运行10分钟。结果显示于图2A中。
b)血流中纳米颗粒稳定性
通过多光子共焦荧光显微镜对小鼠耳垂血管成像,以确定血流中纳米颗粒的稳定性。如实例5中所述,用PEG-PLL和聚合物12、15和19,使用Cy5标记的pDNA(20染料/质粒)在5%海藻糖溶液(N:P=5)中以100μg DNA/mL制备纳米颗粒溶液。随后在吸气室中用异氟烷麻醉Balb/c小鼠,随后将该小鼠转移到位于显微镜台的鼻锥上。使用定制的载玻片架和玻璃载玻片将小鼠的耳朵定位并展平,以便使用徕卡SP8 DIVE多光子显微镜进行成像。使用25x1.0NA水浸物镜(具有M32后孔)用于纳米颗粒成像。Cy5染料是用光谱物理X3激光器(Spectra-Physics X3 laser)的1220nM线进行激发的。使用DIVE系统的两个非去扫描探测器(non-descanned detector)进行成像。一个DIVE HyD探测器调整到605-615nM,以用于第二谐波成像。第二探测器调整到635-775nM,以用于Cy5发射检测。第二谐波用于定位静脉和动脉的视野,随后通过尾静脉静脉注射200μL(20μg pDNA)给小鼠静脉内施用纳米颗粒配制品。对小鼠进行成像,直到血管内的信号与周围组织中的信号相等或持续2小时时间段。用ImageJ生成针对每个指定时间点经1s时间段采集的图像的最大强度投影。每种配制品至少在3只小鼠中进行测试。结果显示于图2B中。
实验3
纳米颗粒转染
针对阴离子解离和循环评估纳米颗粒的稳定性。
将H1299和C2C12细胞以10,000个细胞/孔接种于96孔板中的RPMI培养基中;或以20,000个细胞/孔接种于补充有10%胎牛血清(FBS)和1%青霉素链霉素(P/S)的杜尔贝科改良的伊格尔培养基(DMEM)中。在转染前,允许H1299细胞有24h的恢复期,而通过将C2C12成肌细胞在补充有2%的马血清的DMEM中孵育至少7天分化成肌管。随后,将细胞用100μLPBS洗涤两次,并用培养基或改良的伊格尔最低培养基(OPTI-MEM)洗涤一次。如实例5中所述,使用PEI、PLL,“PLL(M)37”(聚合物2)、“PLL(MN)33”(聚合物5)、“PLL(TM)39”(聚合物8)、PEG-PLL、“PEG-PLL(M)35”(聚合物12)、“PEG-PLL(MN)39”(聚合物15)、“PEG-PLL(TM)33”(聚合物18)制备纳米颗粒。未改性的PEI、PLL和PEG-PLL纳米颗粒用1μg DNA以N:P为3制备,并且改性的PLL纳米颗粒以N:P为5制备。复合后,在OPTI-MEM(1∶10稀释)或培养基中以每孔0.1μg DNA将NP添加到细胞中。16小时时间段后,去除补充纳米颗粒的培养基,并添加新鲜的培养基。绿色荧光蛋白(GFP)的表达用Incucyte(埃森生物科学公司(EssenBioScience),安娜堡,密歇根州)成像,并且荧光素酶/活性用ONE-GloTM+Tox荧光素酶报告仪(普洛麦格公司(Promega),麦迪逊,威斯康辛州)和PHERAstarFSX仪器(BMG实验室技术公司(BMG Lab Tech),凯里,北卡罗来纳州)的标准方案定量。结果展示于图3和4中。
4)毒性
使用存活死亡测定评估材料毒性。
将H1299细胞接种于96孔板(10000/孔)中。在24小时恢复期后,用0.1至2μgPEI(25K,波利塞斯公司,费城,宾夕法尼亚州)、PLL(阿拉曼达聚合物公司,亨茨维尔,亚拉巴马州,5kDa),和在OPTI-MEM中制备的P:PLL(M)33、P:PLL(MN)33和P:PLL(TM)33”处理细胞。暴露16小时后,用存活/死亡染色或代谢测定分析细胞。对于存活/死亡测定,将10μL钙黄绿素AM(2mM于DMSO中)和5μL的碘化丙啶(2mM DMSO)储备溶液溶解于5mL PBS中。然后将细胞用PBS洗涤两次,并添加染色溶液(100μL/孔)。在37℃下孵育30分钟时间段后,将细胞使用IncuCyte酶标仪(赛多利斯公司(Sartorius))成像。可替代地,使用制造商标准方案使用发光测定进行代谢测定。使用PHERAStar酶标仪(BMG实验室技术公司)采集发光测量,并通过在Microsoft excel中拟合数据推导IC50剂量。

5)肌内转染
通过监测肌内注射后报告蛋白荧光素酶的表达评估纳米颗粒在体内的转染效率。
将裸(nu/nu)小鼠的每个后肢(n=2/小鼠)肌内注射在50μL的5%的海藻糖溶液中的与PEI、PEG-PLL或P:PEG-PLL(MN)39(聚合物(15))复合的5μg DNA。每天/每周使用IVIS(珀金埃尔默公司)评估表达,接着向小鼠腹膜内(IP)注射施用150μL的RediJect(30mg/mL荧光素-D)。发光采集一式四份,以平均值+/-标准偏差表示。结果显示于图5中。
