本发明公开了一种具有抗抑郁功能的全素食代餐粉,其按照质量比包括以下组分:50g荞麦粉,20g弹性蛋白肽,15g亚麻籽粉,1g鱼油,70mg绿茶粉,0.7g木耳粉,13.4g西蓝花粉,55.3ug维生素A,0.9mg维生素B1,0.9mg维生素B2,0.9mg维生素B6,0.16ug维生素B12,6.67mg维生素C,0.67ug维生素D,26.67ug叶酸。该全素食代餐粉使用15种食材,合理搭配,营养均衡,涵盖人体必需的多种营养素,提供了优质蛋白、优质碳水化合物、优质脂肪以及各种具有抗抑郁功能的活性物质,长期食用可改善抑郁症状。

技术领域
本发明属于功能食品领域,具体地,本申请涉及一种具有抗抑郁功能的全素食代餐粉。
背景技术
抑郁症是一种全球常见病,估计有3.8%的人口受到影响,其中5.0%为成年人,5.7%为60岁以上成年人,世界上大约有2.8亿人患有抑郁症,是世界第四大疾病,但我国对抑郁症的医疗防治还处在识别率低的局面,地级市以上的医院对其识别率不足20%,只有不到10%的患者接受了相关的药物治疗。抑郁症的发病已经开始出现低龄化趋势。抑郁症的病因并不清楚,但肯定是生物、心理与社会环境等诸多方面因素参与了抑郁症的发病过程。生物学因素主要涉及遗传、神经生化、神经内分泌、神经再生等方面。成年期遭遇应激性生活事件,是导致出现具有临床意义的抑郁发作的重要触发条件。抑郁症可以表现为单次或反复多次的抑郁发作,抑郁发作表现为心境低落、思维迟缓、意志活动减退、认知功能损害和躯体症状。
对于抑郁症的治疗目前主要有药物治疗、心理治疗和物理治疗。其中药物治疗是中度以上抑郁发作的主要治疗措施,临床上一线的抗抑郁药主要包括5-羟色胺再摄取抑郁抑制剂、去甲肾上腺素和特异性5-羟色胺能抗抑郁药等,传统的三环类、四环类抗抑郁药和单胺氧化酶抑制剂由于不良反应较大,应用明显减少。物理治疗手段为重复经颅磁刺激治疗,主要适用于轻中度的抑郁发作。传统的治疗手段具有副作用,而近年来国内外研究结果表明饮食结构和食物中营养成分能够影响抑郁症,可以通过控制饮食辅助治疗抑郁症,同时克服药物治疗带来的毒副作用,这是治疗抑郁症的新途径。
发明内容
针对上述问题,本发明以原料种类丰富多样、营养物质覆盖全、纯天然、纯素食、含有多种抗抑郁功能的活性成分等特征和优势弥补了相关专利的空缺。本发明包括荞麦粉、弹性蛋白肽、亚麻籽粉、鱼油、绿茶粉、木耳粉、西蓝花粉、维生素A、维生素B1、维生素B2、维生素B6、维生素B12、维生素C、维生素D、叶酸,集各种人体必需营养素、功能活性成分于一体,是一款天然健康、营养丰富、能抵抗并缓解抑郁程度的代餐佳品。

与现有技术相比,本发明的有益效果是:
1、本发明提供的全素食代餐粉配方,原料种类丰富多样、营养物质覆盖全、纯天然、纯素食、含有多种抗抑郁功能的活性成分。
2、本发明提供的全素食代餐粉配方加入动物饲料中,证明本发明所述配方粉对抑郁程度的抵抗与缓解有明显作用。

具体实施方式
以下实施例便于更好地理解本发明,但不限于此,这些实施例仅用于例证的目的,决不限制本发明的保护范围。
本发明中的维生素A、维生素B1、维生素B2、维生素B6、维生素B12、维生素C、维生素D、叶酸均购自埃法生物公司。
除特殊说明的之外,各实施例中所用的设备和试剂均常规市售可得。
以下实施例中饲喂的配方粉按照以下比例配制:50g荞麦粉,20g弹性蛋白肽,15g亚麻籽粉,1g鱼油,70mg绿茶粉,0.7mg木耳粉,13.4g西蓝花粉,55.3ug维生素A,0.9mg维生素B1,0.9mg维生素B2,0.9mg维生素B6,0.16ug维生素B12,6.67mg维生素C,0.67ug维生素D,26.67ug叶酸。
实施例1、小鼠适应性饲养及慢性不可预见性应激(CUMS)抑郁造模
1、适应性饲养
选取4~5周大的小鼠60只,适应环境一周,期间喂养普通饲料。给60只小鼠打耳号,运用合理的方式进行随机分组,一共分为六大组(普通饲料组:只饲喂普通饲料但是不给予CUMS;实验组一:饲喂50%普通饲料+50%配方粉,给予CUMS;实验组二:饲喂50%高脂高糖饲料+50%配方粉,给予CUMS;实验组三:饲喂50%高脂高糖饲料+50%荞麦粉,给予CUMS;正常饲料组:饲喂普通饲料,给予CUMS;高脂饲料组:饲喂高脂高糖饲料,给予CUMS),称量每个小鼠的体重,给小鼠喂养相应的饲料一周以适应环境。分组如表1

实验中所述高脂高糖饲料由华富康公司提供。
2、抑郁造模:
进行CUMS造模,如下表2。

3.抑郁行为测试
1)旷场实验:旷场实验以“中央区域总路程/总区域路程”及“在中央区域的持续时间”这两个参数为依据进行统计分析。旷场实验中的总区域路程划分为中央区域路程和边缘区域路程,总区域路程反映鼠的活动能力。小鼠在新环境中是由紧张、兴奋、探索再到适应的一系列过程,处于兴奋状态的小鼠表现为行动活跃,四处走动,总路程增加。当小鼠处于抑郁状态时,其运动路程明显缩短,但总路程过长反映鼠对新环境的恐惧。啮齿类动物在空间活动时会本能避开中央区域,沿着边缘区域活动。中央区域停留时间的减少说明动物具有焦虑行为。整个旷场实验中鼠在中央区域运动的总路程以及其所占旷场实验总路程的百分比、中央区域运动时间增加与鼠的探索行为密切相关,中央区域运动总路程百分比降低是鼠焦虑的表现。
将每一只小鼠从中心位置放入,利用软件从小鼠第一次经过中心位置开始时算起,六分钟内小鼠经过中心的停留时长与比例,导出数据至excel表格进行prism统计分析。
2)悬尾实验(Tail suspension test,TST):
悬尾实验原理是利用小鼠悬尾后企图逃脱但又无法逃脱,从而放弃挣扎,进入特有的抑郁不动状态,实验过程中记录动物不动时间来反映抑郁状态,抗抑郁药物、兴奋药物能明显地缩短改变其状态。
将小鼠从尾部1cm处吊起,持续6min。用医用胶带将动物尾巴固定在架子上(距离尾巴远端越1cm)。在6min试验期间,记录最后4min的静止时间。在测试过程中,使用摄像机记录每只小鼠的行为,并使用KEmaze软件(南京卡尔文科技有限公司)分析测量不动时间。
3)强迫游泳测试(Forced swimming test,FST):
强迫游泳实验以不动时长占比作为评价指标。不动状态(immobility)即指动物在水中停止挣扎,呈漂浮状态,仅有轻微的肢体运动以保持头部浮在水面。不动时间占比越高表明抑郁程度越重。强迫游泳实验与悬尾实验虽然在原理和数据分析上比较类似,但不同品系的小鼠在两个实验中可能出现不同的敏感性,从而需要我们利用多个实验进行小鼠抑郁行为的评价。
将每只小鼠放置在一个开放的圆柱(直径10cm,高度30cm)中,容器内有23±1℃、深度为20cm的水,并持续6min。在试验的最后4min内测量不动时间。游泳结束后,立即将小鼠取出,用毛巾擦干,放回笼子。在测试过程中,使用摄像机记录每只小鼠的行为,并使用KEmaze软件(南京卡尔文有限公司)分析测量不动时间。
实施例2、实验结果及分析
1、旷场实验结果分析与结论:
吃普通饲料的小鼠在造模后4周前后差别不明显,而吃50%普通饲料+50%配方粉的小鼠在造模后其中央区域路程/总路程反而比4周前有显著提高,说明配方粉比普通饲料能更好的提高小鼠在旷场中央区行动的比例(图1)。
另外,吃高脂高糖饲料的小鼠在造模后4周其中央区域路程/总路程也发生了上升,但吃50%高脂高糖饲料+50%荞麦粉的小鼠发生了明显下降,而50%高脂高糖饲料+50%配方粉的小鼠依然发生上升,说明配方粉比荞麦粉在混入高脂高糖饲料后,能够更好地提高小鼠在旷场中央区行动的比例(图2)。
从小鼠在中央区域持续时间数据结果分析可见:吃普通饲料的小鼠在造模后4周后发生了明显中央区持续时间的下降,而吃50%普通饲料+50%配方粉的小鼠在造模后其在中央区的持续时间反而发生明显上升;另外,吃高脂高糖饲料的小鼠在造模后4周其中央区持续时间上升了8.25%,而吃50%高脂高糖饲料+50%配方粉的小鼠上升了22.31%,与它们相反的是,吃50%高脂高糖饲料+50%荞麦粉的小鼠发生了明显下降(42.14%)(图3、图4)。
因此,认为吃普通饲料的小鼠在抑郁造模后发生了明显的中央区持续时间的下降,说明抑郁造模导致小鼠发生了一定程度的焦虑行为,但在普通饲料中添加50%的配方粉后,小鼠中央区域路程/总路程以及在中央区的持续时间反而发生明显上升,说明配方粉克服了抑郁造模导致的小鼠焦虑行为。在高脂高糖饲料饲喂小鼠的实验中,高脂高糖饲料具有一定的克服小鼠焦虑行为的作用,但在高脂高糖饲料中加入50%的配方粉能更有效地克服小鼠的焦虑行为,如果单纯加入50%荞麦粉则无法克服,反而使小鼠出现明显焦虑行为。
2、悬尾实验结果分析与结论:
由图5、图6可知,本实验开始前各组悬尾结果差异不明显,从造模前到造模第四周后,50%普通饲料+50%配方粉组小鼠的不动时间比上升很小(5.9%;抑郁造模前:57.9%,抑郁造模后:63.8%),普通饲料组小鼠的不动时间比上升幅度较大(12.7%;抑郁造模前:60.9%,抑郁造模后:73.6%);50%高脂高糖饲料+50%配方粉组小鼠的上升幅度是所有组中最小的(5.02%;抑郁造模前:62.3%,抑郁造模后:67.32%),而50%高脂高糖饲料+50%荞麦粉和高脂高糖组小鼠的不动时长比均显著增加,分别上升13.0%(抑郁造模前:58.6%,抑郁造模后:71.6%)和20.3%(抑郁造模前:52.7%,抑郁造模后:73.0%)。
普通饲料组小鼠在抑郁造模后的不动时间明显上升,表明小鼠经过抑郁造模后发生了明显的抑郁绝望情绪,而在普通饲料里面添加了50%配方粉后,这种情况得到了缓解。另外,高脂高糖饲料会明显加重小鼠在悬尾实验中的不动时间比,达到所有组增幅最高(20.3%),而在高脂高糖饲料中加入50%配方粉后,小鼠的不动时间比增幅只有5.02%,为所有组增幅最低,说明配方粉能够很大程度上缓解高脂高糖饲料组小鼠造模后产生的抑郁绝望情绪。50%荞麦粉在高脂高糖饲料中的添加似乎对小鼠抵抗抑郁方面没有起到缓解作用,这也说明配方粉中各成分的混合效果也远远高于其主要的单一成分荞麦粉。
3、强迫游泳实验结果分析与结论:
从图7、图8可知,从造模前到造模四周后,50%普通饲料+50%配方粉组小鼠的不动时间比不但没有上升,反而发生了下降(8.7%;抑郁造模前43.9%,抑郁造模后35.2%),普通饲料组小鼠的不动时间上升了12.2%(抑郁造模前33.4%,抑郁造模后45.6%);高脂高糖组小鼠的不动时间也上升了6.2%(抑郁造模前15.9%,抑郁造模后22.1%),而在高脂高糖饲料中添加了50%的配方粉后,小鼠的不动时间反而发生下降了2.2%(抑郁造模前19.9%,抑郁造模后17.7%),而只在高脂高糖饲料中添加50%荞麦粉的小鼠的不动时间发生了更加明显的上升(12.7%;抑郁造模前36.3%,抑郁造模后49.0%)。
无论在普通饲料还是高脂高糖饲料中添加50%的配方粉,都能够完全逆转小鼠在抑郁造模后发生游泳不动时间比上升的情况,也就是说,配方粉中的营养成分能够很好的抵抗小鼠在抑郁造模后产生的抑郁绝望情绪。而配方粉中的主要成分荞麦粉无法完成抗抑郁作用。
综上所述,本配方粉无论添加在普通饲料或者高脂高糖饲料中,均能较好地逆转或者缓解小鼠在抑郁造模后产生的焦虑行为和抑郁绝望行为,而只在高脂高糖饲料中添加本配方粉的主要成分荞麦粉没有缓解效果,说明本配方粉各配方成分具有比单一成分更佳的抗抑郁能力。
虽然,上文中已经用一般性说明、具体实施方式及试验,对本发明作了详尽的描述,但是本发明不限于上述实施例,在本发明基础上,可以对之作一些修改或改进,这对本领域技术人员而言是显而易见的。因此,在不偏离本发明精神的基础上所做的这些修改或改进,均属于本发明要求保护的范围。

10.根据权利要求6-9任一项所述的应用、功能食品或保健品,所述功能食品或保健品的形式选自直接食用的和冲调用的粉末或颗粒、面粉、米粉、饼干、糕点。
