Abstract
本发明涉及一种洋葱种子消毒及无菌苗培养方法。本发明首先提供了一种基于PPM的洋葱种子消毒方法,将洋葱种子浸泡在20mL/L的PPM溶液(pH=2)中消毒24h,随后将其直接播种至含0.50mL/L PPM的1/2MS培养基上,可显著提高洋葱种子发芽势、降低污染率,并能够有效进行长期培养而不发生污染。该方法操作及其简单,只需将种子浸泡、点播即可,不需要常规消毒中反复繁琐的冲洗操作,极大的提高了工作效率,降低了二次污染的发生。而且本研究还为种子作物的无菌苗培养、以及开放式组培体系的建立提供了方法支持,进一步为洋葱遗传转化研究奠定了坚实的基础。

洋葱(Allium cepa L.)属于石蒜科葱亚科葱属二年生草本植物,是一种世界性蔬菜,其营养价值和医疗保健价值被世界所公认,素有“蔬菜皇后”的美誉。近年来,随着高通量测序技术的发展,洋葱基因组测序已经完成,众多的功能基因已被预测并注释,然而洋葱遗传转化体系进展缓慢,功能基因的互补验证研究受阻,严重制约了洋葱转基因育种及功能性分子标记辅助育种的应用研究。无菌苗的培养是转基因体系及功能基因验证的关键环节,其培养材料通常来源于种子,因此种子消毒显得尤为重要。
在用种子作为外植体的研究中,其消毒试剂通常采用乙醇、次氯酸钠和升汞,因植物及外植体种类的不同,它们又具有不同的应用范围。已有的研究表明,洋葱种子消毒试剂通常也采用次氯酸钠和升汞,然而其发芽率仅为46.94%-56.00%左右,而污染率却高达25.51%-61.62%,并且次氯酸钠对洋葱种子的萌发具有抑制作用。而且常规的消毒试剂及使用方法通常涉及无菌水反复冲洗,步骤繁琐,极易引入二次污染。
植物组培抗菌剂PPM(Plant Preservative Mixture)是一种广谱抗微生物制剂,能有效的抑制植物组织培养中微生物的污染,而不会影响种子发芽、愈伤组织增殖和再生。并且它还具有极高的热稳定性,能与培养基一起高压灭菌,使用方法非常简便,无需冲洗,极大的降低了试验人员重复繁琐的操作,极大的避免了二次污染引入,应用领域及前景非常广泛,而单独采用PPM对种子消毒的研究却鲜见报道。

针对洋葱种子消毒操作繁琐,且消毒后发芽率低、污染严重的问题,本发明首先探索了PPM(20mL/L)消毒液pH值对洋葱种子消毒效果及发芽势的影响,并明确了pH值为2的PPM消毒液具有最佳的效果。随后进一步优化了消毒时间、PPM维持培养浓度。进而提供了一种简便、高效、低污染的洋葱种子消毒及无菌苗培养方法。该方法有效提高了洋葱种子的发芽势,实现种子消毒效果的同时基本不发生污染,基于该培养方式可以有效缩短培养周期,获得幼嫩组织,为洋葱转基因体系研究奠定了坚实的基础。
植物组培抗菌剂PPM(Plant Preservative Mixture)是一种广谱抗微生物制剂,包含有两种异噻唑啉酮抗菌剂(氯甲基异噻唑啉酮和甲基异噻唑啉酮),能够有效的控制植物组织培养中通过空气、水、人为传播的微生物以及内源性微生物污染,而直接将其用于种子消毒培养无菌苗的研究鲜见报道。本研究预试验结果显示,不同pH值的PPM(20mL/L)对洋葱种子浸泡24h,培养4d污染率无显著差异,延长培养6-10d,未调节pH值(pH=6)的PPM消毒处理,其污染率急剧升高达到25.83%,该值显著高于pH=2的污染率4.17%,其原因可能为PPM的最优杀菌效果需要较低的pH环境,而高的pH环境使得PPM无法杀灭种子内部残留的内生菌(或共生菌),或者酸性环境软化了种子的结构,增加了PPM对洋葱种子的渗透性。为了获得最佳的PPM消毒效果,本发明尝试改良PPM制剂,在含量为20mL/L的PPM工作液中添加1%体积1mol/L的HCl至其pH值为2(PPM=20mL/L;pH=2;下同),通过优化浸泡时间,调整PPM在维持培养基中的浓度达到最佳的种子消毒和无菌苗培养效果。
本发明对上述洋葱种子消毒方法进行了污染率和种子发芽效果的考察,考察结果表明:采用PPM作为种子消毒剂的活性成分,种子消毒效果与PPM浸泡时间密切相关,浸泡时间越长污染率越低,小于12h的浸泡,其污染率超过20.67%,而24h和48h的浸泡其污染率可以降低到与10%NaClO消毒10min无显著差异水平,上述结果表明PPM可以通过长时间浸泡渗入组织内部而杀灭污染微生物,进而有效防治污染。另一方面,本发明还发现20mL/L改良PPM(pH=2)浸泡时间在0-48h以内,不会抑制种子发芽,而NaClO消毒强烈的抑制了洋葱种子10天以内的发芽势,其原因可能为洋葱种子难以抵抗NaClO的次生碱伤害。现有的NaClO消毒方式,在消毒之后往往需要反复冲洗种子才能除尽NaClO残留,上述冲洗过程极大的增加了消毒工作量,并极易引入二次污染,进而造成组培苗污染率的急剧攀升。因此,本发明证实了酸性PPM对洋葱种子的消毒效果具有显著提升,并且消毒后种子的后处理方式更加简便。因此,本发明首先提供一种包含酸性PPM(pH=2)的洋葱种子消毒剂及洋葱种子消毒方法。
植物种子本身含有大量内生和附生微生物菌群,它们的组成和垂直传播或多或少会影响种子发芽、植株生长,并成为下一代宿主植物内生菌的来源。本发明对污染微生物进行了分离鉴定,测序及比对结果显示了造成污染的微生物包括4种真菌(Alternariaalternata、Fusarium proliferatum、Fusarium oxysporum、Stemphylium vesicarium)和2种细菌(Phytobacter diazotrophicus、Atlantibacter hermannii),该结果进一步表明了上述微生物可以通过种子进行垂直传播,针对种传病害具有一定的防治建议。随后对上述微生物进行了抑菌试验,真菌抑菌率大于50%的PPM浓度为0.5mL/L,而对细菌在0.25mL/L即可产生显著的抑菌效果。
基于上述技术效果,本发明首先提供了植物组培抗菌剂PPM或其组合物在制备洋葱种子消毒剂中的应用,其次,本发明还提供了基于PPM的洋葱种子消毒剂及洋葱种子消毒方法。
另外,本发明还针对PPM在洋葱维持培养中的效果进行了验证:
维持培养中适宜的PPM浓度因物种、外植体类型存在明显不同。浓度过高虽然能够抑制污染,但同时也抑制了外植体的生长发育,浓度过低防治污染效果不明显,适宜的浓度需要通过梯度试验进行探索。研究表明甜瓜愈伤组织和体细胞胚诱导培养适宜的PPM浓度为0-5mL/L,烟草花药培养单倍体PPM浓度0-10mL/L均可,矮牵牛由叶片外植体诱导芽分化和幼苗生长PPM浓度为0-2mL/L。Niedz研究报道了PPM对芸香科枸橘(Poncirustrifoliata)和粗皮柠檬(Citrus jambhiri)生长发育的影响,结果显示了2mL/L的PPM对种子发芽、幼苗培养、非胚性愈伤组织的诱导以及下胚轴外植体直接诱导不定芽均无影响。关于葱属的研究结果表明,洋葱幼蕾大孢子培养中合适PPM维持培养浓度1mL/L,而小葱(Allium ascolonicum)茎尖和分生组织培养PPM维持培养浓度在1mL/L以内对叶及根的数量无差异。而本发明对洋葱无菌苗的研究结果表明,PPM浓度为1-9mL/L时可以明显的抑制洋葱子叶和根的生长,PPM浓度越高其抑制程度越强烈,这一现象对根的伸长尤为明显,浓度在9mL/L时其胚根几乎不能伸长,而PPM浓度为0-0.5mL/L时,种子发芽、子叶和根的生长均不受抑制。因此,洋葱种子对于PPM的耐受性明显的低于上述几个物种,且不同组织之间具有明显差异。结合抑菌试验的结果,针对洋葱幼苗培养,将PPM维持培养浓度定为0.5mL/L。
根据本发明的验证结果,采用添加PPM的培养基进行洋葱无菌苗培养在促进洋葱无菌苗生长的同时能够实现杀菌效果,基于上述研究结果,本发明还提供了基于PPM的洋葱无菌苗培养基及洋葱无菌苗的培育方法。

基于上述技术效果的阐述,本发明提供了植物组培抗菌剂PPM在洋葱种子消毒及无菌苗培育方面的应用,将其作为消毒剂进行洋葱种子浸种相比现有消毒方法极其简便,无需冲洗,明显降低了试验人员重复繁琐的操作,极大的避免了二次污染,能够有效的减少种子污染率,提升种子萌发效果,并且在无菌苗培育阶段还体现出显著的促生效果,因此,本发明具体提供以下技术方案:
本发明第一方面,提供植物组培抗菌剂PPM或其组合物在制备洋葱种子消毒剂中的应用。
上述第一方面中,所述植物组培抗菌剂PPM为异噻唑啉酮类抗菌剂,具体包括氯甲基异噻唑啉酮和甲基异噻唑啉酮两种成分。
另外,上述第一方面还涉及PPM的组合物,所述组合物中除包括活性剂量的PPM,还可以包括以下成分中的一种或几种的组合:农药学上可接受的载体、pH调节剂、生长调节剂:
所述农药学上可接受的载体包括但不限于溶剂、增溶剂、悬浮剂、等渗剂、缓冲剂、舒缓剂等,还可以根据需要适当地加入防腐剂、抗氧化剂和其他制剂添加剂;
所述生长调节剂为用于改善洋葱品种、提高产量的活性成分,如乙烯利、复硝酚钠、氯化胆碱、萘乙酸等;

上述洋葱种子消毒剂,其主要的效果为实现种子消毒、杀菌及促进萌发,效果较好的实施方式中,上述洋葱种子消毒剂为酸性PPM溶液。
本发明第二方面,提供一种洋葱种子消毒剂,所述消毒剂中至少包括活性剂量的PPM制剂,并且所述消毒剂的pH为1~3。
优选的,所述PPM制剂的浓度为10~30mL/L,进一步的,为15~25mL/L,具体的实例中,所述消毒剂中PPM制剂的浓度为18mL/L、19mL/L、20mL/L或21mL/L。
优选的,所述消毒剂通过加入无机酸对pH进行调节;进一步的,所述无机酸为盐酸溶液,所述pH为1.5~2.5。
本发明第三方面,提供一种洋葱无菌苗培养基,所述培养基采用1/2MS培养基作为基础培养基,还包括活性剂量的PPM。
优选的,上述基础培养基中,还包括蔗糖添加,所述蔗糖浓度为18~22g/L,pH为5~6;进一步的,所述蔗糖浓度为19~21g/L,pH为5.5~6。
优选的,所述无菌苗培养基为固体培养基,培养基中还包括琼脂,所述琼脂浓度为8~10g/L。
优选的,所述无菌苗培养基中,PPM的浓度为0.5~1.0mL/L;进一步的,所述PPM的浓度为0.5~0.8mL/L,具体的实例如0.5mL/L、0.6mL/L、0.7mL/L,其中以0.5mL/L的效果最好。
本发明第四方面,提供一种洋葱种子消毒方法,所述消毒方法包括采用第二方面所述消毒剂对洋葱种子进行浸泡。
优选的,所述浸泡时间为18~25h;进一步的,所述浸泡时间为22~25h;具体的实例如23h或24h。
本发明第五方面,提供一种洋葱无菌苗的培育方法,所述培育方法包括将第四方面所述方法消毒后的洋葱种子加入第三方面所述无菌苗培养基中进行培养。
优选的,所述培养环境为室温25℃,光照时间为14~18h/d。
上述第五方面所述培育方法,一种效果较好的实施方式如下:将洋葱种子浸泡在消毒剂中消毒24h,随后将洋葱种子播种至无菌苗培养基中;
所述消毒剂中PPM的浓度为20mL/L,消毒剂的pH=2;所述无菌苗培养基中PPM的浓度为0.50mL/L。
为了使得本领域技术人员能够更加清楚地了解本发明的技术方案,以下将结合具体的实施例详细说明本发明的技术方案。
实施例1
1材料与方法
1.1材料
洋葱种子天正201用于方法优化研究,短日照洋葱(YY18)、中日照洋葱(GCH)、长日照洋葱(YOUYUE、JIAMEI、KAMUYI-153)用于方法验证。消毒试剂为PPM(北京酷来搏科技有限公司,PTC1000)和10%NaClO。
1.2培养条件
1/2MS培养基为基本培养基,添加蔗糖20g/L,琼脂9g/L,pH=5.8。培养室昼夜温度均设为25℃,光照时间为16h/d,光照强度为3000lx。
1.3方法
1.3.1酸性PPM消毒制剂及消毒时间对洋葱种子发芽和消毒效果的影响
用1mol/L的盐酸调节PPM(20mL/L)消毒液pH值为1-6,其中pH=6为PPM原液用蒸馏水直接稀释至20mL/L的工作液。随后对洋葱种子进行预实验,浸泡洋葱种子24h,播种至含PPM浓度为1mL/L的1/2MS培养基上。每处理3皿,每皿40粒,统计4-10d种子发芽及污染情况。
采用PPM(20mL/L,pH=2)溶液对洋葱种子进行消毒处理,处理方法为A1-A8共8种方式。其中A1为干种子直接播种;A2为20mL/L PPM表面消毒后播种;A3-A7分别为20mL/LPPM浸泡3h、6h、12h、24h、48h后播种;A8为10.00%NaClO消毒10min。所用培养基为基本培养基添加PPM至其浓度为1mL/L。为统计种子自身的带菌污染情况,以不加PPM的1/2MS为对照,编号为A0。
1.3.2种子污染微生物的分离鉴定与抑菌试验
为了确定培养基中能够抑制微生物生长的PPM浓度,将1.3.1中出现的微生物(包含真菌和细菌),转接继代培养,用于PCR鉴定和抑菌试验。利用优化的碱裂解法提取污染微生物核酸,针对真菌采用ITS1(5′TCCGTAGGTGAACCTGCGG 3′)和ITS4(5′TCCTCCGCTTATTGATATGC 3′)引物进行PCR扩增。细菌鉴定采用27F(5′AGAGTTTGATCMTGGCTCAG 3′)和1492R(5′GGTTACCTTGTTACGACTT 3′)引物,扩增体系总体积为25μL,包含2×ApexHF FS PCR Master Mix 12.5μL,上下游引物均为1μL,模板1μL,用PCR级别的水补足至25μL。反应程序为94℃预变性2min;随后进行10个循环的Touchdown PCR,98℃10s,55℃(-0.9℃/Cycler)10s,72℃30s;接着进行34个循环的普通扩增,98℃10s,45℃10s,72℃30s;循环结束后于72℃延伸10min,10℃保存。PCR产物经1%琼脂糖凝胶电泳检测,随后送至北京擎科生物科技有限公司青岛分公司测序。所得序列利用NCBI的GenBankNT数据库进行BLAST比对,确定其物种分类。
真菌抑菌试验采用生长速率法,用直径5mm的菌碟打孔器打取微生物菌饼,转接至含不同浓度PPM的PDA培养基中,PPM浓度分别为0、0.25、0.50、1.00、2.00、4.00、8.00、16.00mL/L,编号为B1-B8,每个处理重复3次,28℃培养,当对照长至平板2/3时,采用十字划线法测量菌落直径,计算抑菌率大小。抑菌率(%)=(对照菌落直径均值-处理菌落直径)/(对照菌落直径均值-菌饼直径)×100%。
细菌抑菌试验采用滤纸片法,用牙签沾取细菌单菌落至6mL液体LB培养基中,37℃170rpm培养过夜。随后按照1:50接种至新的LB培养基中,继续培养并检测OD值至0.5-0.6时,将上述培养物取100μL利用玻璃珠涂布于LB培养基上。将无菌5mm的滤纸片浸泡于B1-B8的PPM溶液中,随后用镊子取出沥干水分放于上述涂布的LB培养基中,37℃培养24h,采用十字划线法测量菌落直径。
1.3.3植物培养基中PPM浓度对洋葱种子发芽与消毒效果的优化
为了确定对洋葱种子发芽、消毒和无菌苗培养最优效果的PPM维持浓度,结合1.3.1和1.3.2的试验结果,将浸泡处理24h的种子播种于含有不同PPM的1/2MS培养基上。其中PPM设置8个浓度分别为C1-C8(0、0.25、0.50、1.00、2.00、3.00、5.00、7.00、9.00mL/L)。
1.3.4指标测定与统计分析
洋葱种子播种后,每天观察种子生长情况,记录发芽数、污染数。统计分析各处理的发芽率、污染率。SPSS Statistics 19.0用于卡方检验和方差分析,多重比较方法分别为Bonferroni法和Duncan法,结果以平均值±标准误呈现。
2结果与分析
2.1不同pH值PPM消毒制剂的效果
由图1预试验结果可以看出,不同pH值的PPM消毒液对洋葱种子4-10d的发芽势存在差异。卡方检验结果表明,4d发芽势受pH值影响达到显著水平,其中pH为2时发芽势最高为75.83%,pH为3-6时发芽势差异不显著,播种后6-10d发芽势受pH值影响不显著,其数值均超过80%。同时,pH值对4-10d的污染率也存在差异,卡方检验结果表明,播种4d时pH为1-6对污染率的影响无显著差异,随着培养时间延长,高pH值污染率急剧升高;培养至10d时污染率的差异更显著,且pH为2的污染率为4.2%,显著低于单独使用20mL/L PPM(pH=6)的消毒效果。
2.2 PPM消毒时间对洋葱种子发芽和消毒效果的影响
由图2发芽率均值及发芽数卡方检测结果可以看出,与干种子直播(A1)相比PPM(20mL/L,pH=2)浸种0-48h对洋葱种子发芽率无显著影响,播种3-8天发芽率逐渐上升,8天均可达到最大值。而10.00%NaClO(A8)消毒可显著降低洋葱种子发芽率,特别是播种后8天内,其发芽率不及20mL/L PPM浸种消毒的50%,即使考虑到浸种时间的延迟(最长48h),其10天发芽率(A8)仍然明显低于A1-A7的6天发芽率。
图2中,各实验组培养情况如下:A1:干种子直播1/2MS(1mL/L PPM);A2:20mL/LPPM表面消毒后播种至1/2MS(1mL/L PPM);A3-A7:20mL/L PPM分别浸种消毒3h、6h、12h、24h、48h后播种至1/2MS(1mL/L PPM);A8:10%NaClO消毒10min播种至1/2MS;柱形图上方不同字母表示处理间差异显著(p<0.05),横线上方为卡方值和调整p值;下同。
图3展示了洋葱种子播种10天的污染率(对照A0为干种子播种至1/2MS培养基后5天结果,A1-A8编号同图2),方差分析结果表明,PPM能够显著的降低洋葱种子培养过程中的污染率,干种子直播1/2MS培养基(A0),其污染率高达98.67%,而直播至1.00mL/L PPM培养基中(A1),污染率可降低至60.00%。PPM(20mL/L,pH=2)表面消毒(A2)与干种子直播(A1)对种子污染无显著差异,而浸种3-48h后,可显著降低种子污染率,且随着浸种时间延长至24h和48h(A6和A7),污染率可降低至与NaClO标准消毒方法无显著性差异水平。上述结果表明,PPM(20mL/L,pH=2)表面消毒效果较差,需要较长的时间才能渗入组织内部,进而达到消毒和抑制污染的目的。
2.3洋葱种子污染微生物的分离鉴定
针对1.3.2中培养产生的污染微生物,用牙签挑取后转接至新的培养基中进行纯化培养,获得真菌菌株9个,细菌菌株4个。分别对真菌和细菌微生物进行ITS和16S rRNA的PCR扩增(图4)。将测序序列利用BLAST软件比对至NCBI的NT数据库,结果表明9个真菌菌株被鉴定为4个物种,其中F1和F2为Alternaria alternata,F3为Fusarium proliferatum,F4为Fusarium oxysporum,F5-F9为Stemphylium vesicarium。上述四个物种均为洋葱病原真菌,它们可以通过种子进行世代传递。其中Alternaria alternata为链格孢菌属物种,洋葱叶枯病的主要致病菌之一,其多个菌株的基因组序列已被公开。Fusarium proliferatum和Fusarium oxysporum为镰刀菌属物种是洋葱茎腐病和基腐病的主要病原。Stemphyliumvesicarium能够引起葱属作物叶斑病和叶枯病。
4个细菌菌株被鉴定为2个物种,其中B1为Phytobacter diazotrophicus,B2、B3、B4均为Atlantibacter hermannii。Phytobacter diazotrophicus为为固氮内生细菌,生长在种子及根系附近,继代后生长停滞,无法离体培养,其原因可能为该菌是一种植物共生菌,离开寄主代谢产物的支持则无法生长。Atlantibacter hermannii曾在烟草和甘薯中得到分离,但在葱属物种中尚未发现分离该物种的报道,其作用或危害目前尚不明确。
2.4洋葱种子污染微生物抑菌试验
为了明确PPM对真菌的抑制效果,采用生长速率法对4个真菌物种进行了抑菌试验,表1抑菌率均值及多重比较结果表明,PPM对于4个真菌物种具有明显的抑菌效果,即使在极低浓度0.25mL/L下,其最低抑菌率也超过了40.13%,且随着浓度的增加而呈现正相关,抑菌率均大于50%的最低浓度为0.5mL/L。并且PPM对于不同真菌物种的抑菌率具有明显的差异,Stemphylium vesicarium最为敏感,在PPM浓度为0.5mL/L时即可达到65.71%的抑菌率,在4mL/L时其抑菌率超过90%,其他三种真菌对PPM的敏感程度相似。

图5展示了不同浓度的PPM对细菌Atlantibacter hermannii的抑制效果,其抑菌圈的直径随着PPM浓度的增加而增加(表2),与空白对照相比在PPM浓度为0.25mL/L时即可产生显著的抑菌效果,抑菌圈直径为11.12mm。

2.5培养基中PPM维持浓度对于洋葱子叶和根生长的影响
种子浸泡于改良PPM(20mL/L,pH=2)溶液24h后,播种于含不同浓度PPM的1/2MS培养基上,培养6天结果表明,不同PPM浓度对洋葱子叶和根的生长存在明显差异,高浓度(1.00-9.00mL/L)时可以抑制洋葱子叶和根的生长,其抑制程度随着PPM浓度增加而逐渐增强,表现为子叶和胚根短小、胚轴膨大,而在低浓度(0.25-0.50mL/L)时与不添加PPM培养基相比无明显差异(图6)。结合抑菌试验中PPM的抑菌浓度,将洋葱种子萌发培养基中PPM的维持培养浓度设置为0.50mL/L。
2.6洋葱种子消毒及无菌苗培养改良方法
根据洋葱种子萌发、抑菌和维持培养的试验结果,探索开发了一套洋葱种子消毒及无菌苗培养的简单方法:将洋葱种子浸泡在20mL/L的改良PPM溶液(pH=2)中消毒24h,随后将其播种至含0.50mL/L PPM的1/2MS培养基上,洋葱种子8天内即可达到最大萌发率为92.65%,10天污染率仅为3.12%,可维持培养至少20天(图7)。
随后,本实施例又将该方法应用于不同来源的短、中、长日照三种类型的洋葱种子(YY18、GCH、YOUYUE、JIAMEI、KAMUYI-153)进行消毒和无菌苗培养测试,结果表明该方法对三种类型的洋葱种子都具有消毒效果好、稳定和可靠的特性(图8)。该方法操作极其简单方便,无需NaClO和HgCl2,不需无菌水反复冲洗,与NaClO消毒方法相比可显著提高8天内种子萌发势。

1.一种洋葱种子消毒方法,其特征在于,所述消毒方法包括采用消毒剂对洋葱种子进行浸泡;所述浸泡时间为18~25h;
所述消毒剂至少包括活性剂量的植物组培抗菌剂PPM;
所述PPM制剂的浓度为10~30mL/L;
所述消毒剂通过加入盐酸对pH进行调节,pH为1.5~2.5。
2.一种洋葱无菌苗的培育方法,其特征在于,所述培育方法包括将权利要求1所述方法消毒后的洋葱种子加入无菌苗培养基中进行培养;
所述培养基采用1/2MS培养基作为基础培养基,还包括活性剂量的PPM。

4.如权利要求3所述洋葱无菌苗的培育方法,其特征在于,所述培养环境为室温25℃,光照时间为16h/d;
所述培养方法的具体步骤如下:将洋葱种子浸泡在消毒剂中消毒24h,随后将洋葱种子播种至无菌苗培养基中;所述消毒剂中PPM的浓度为20mL/L,消毒剂的pH=2;所述无菌苗培养基中PPM的浓度为0.50mL/L。
