Abstract
本发明公开了一种适用于高尿酸及痛风人群的药食同源酵素及其制备方法,包括包括以下组分:EM菌、乳酸菌、酵母菌、植物乳杆菌、嗜热链球菌、双歧杆菌、鼠李糖乳杆菌、嗜酸乳杆菌、凝结芽孢杆菌、罗伊氏乳杆菌、蜂蜜、珊瑚钙、饮用水。所述药食同源中药材原料,包括以下组分:葵花盘、玉米须、鱼腥草、薏苡仁、葛根、蒲公英、栀子、菊苣、杜仲、芡实、肉桂、淡竹叶、茯苓、马齿苋、百合。本发明利用有益微生物菌群,结合药食同源中药材原料进行发酵,其制取物能够有效降低痛风人群血尿酸含量,缓解疼痛,提高免疫系统,恢复和改善代谢功能,适用性广,无毒无副作用,可日常食用。

技术领域
本发明结合了微生物发酵技术、中药材技术、食品技术领域,尤其涉及一种适用于高尿酸及痛风人群的药食同源酵素及其制备方法。
背景技术
EM菌(Effective Microorganisms)也叫EM益生菌原液,是由日本琉球大学的比嘉照夫教授1982年研究成功,EM技术是目前世界上应用范围最大的一项生物工程技术,和一般生物制剂相比,它具有结构复杂、性能稳定、功能齐全的优势,表现出前所未有的高科技水平。作用机理是形成EM菌群和微生物争夺营养的竞争,进而形成有益的微生物菌的优势群落,利用独特发酵工艺把经过仔细筛选出来的好气性和嫌气性有益微生物混合培养,形成功能强大的EM制剂。EM菌技术已经广泛利用在种植、水产养殖领域,近年来逐步利用于医疗、食品添加领域,其应用于食品添加领域中能产生大量人体需要的有益菌和菌群代谢有益物,并控制有害微生物的繁殖,国内外对EM菌发酵中药材的研究还处于一种真空状态,对采用中药材为原料进行EM菌发酵技术仍是空白。
目前国内受高尿酸人群高达1.8亿人,其中痛风人群超过8000万人,而且正以每年9.7%的年增长率迅速增加,患病年龄也趋于年轻化,且男性高于女性。被称为继“三高”后的第“四高”。
痛风是单钠尿酸盐沉积于关节内、关节周围、肾脏和皮下等部位,引发的急、慢性炎症和组织损伤,与嘌呤代谢紊乱及(或)尿酸排泄减少所致的高尿酸血症直接相关,属于代谢性风湿病范畴。其临床特点是由高尿酸血症、尿酸盐沉积所导致的反复发作的急、慢性关节炎和软组织损伤,尿酸性肾结石所导致的痛风性肾病。高尿酸血症是痛风发生的基础,痛风可并发肾脏病变,严重者可出现关节破坏、肾功能损害,常伴发高脂血症、高血压病、糖尿病、动脉硬化及冠心病等。但是,高尿酸症状和痛风关节炎痛症没有任何一种根治方法,只能缓解急性病发痛苦。
现有技术中:治疗痛风主要有西药、中药、针灸,中药汤剂、中成药、痛风茶等。
西药,痛风人群主要有口服抗生素、秋水仙碱、非布司佐他、别嘌醇等镇痛类药物、消炎类药物和排泄类药物,在治疗痛风过程中疗效确切,短时间之内可取得明显效果,但通常副作用大,常见于胃、肠功能紊乱,肝肾功能受损,皮疹、皮炎等,尤其是停药后,容易复发,故不是根本解决之道。
中药治疗痛风是以清热利湿、活血通络、利尿排酸为指导原则,但辨症施治、辨症用药要求较高、难度大。辨症施治痛风的药方使用各有不同,使用的普遍性较低。同时,中药汤剂在服用过程也存在煎煮麻烦的弊端;中成药虽然使用方便,但由于不能辨症用药,造成最后效果的确定性不强、难以普及的弊病。针灸,技术比较高超的针灸师,可以很好治疗痛风,而且没有副作用,但针灸治疗的技术要求比较高,治疗时间也比较长。
为此,我们提出一种适用于高尿酸及痛风人群的药食同源酵素及其制备方法。
发明内容
为解决背景技术中存在的上述技术问题,本发明结合了微生物发酵、中药材技术、食品技术领域,提供了一种能够降低痛风人群血尿酸含量、缓解疼痛、抑制尿酸增高、提高免疫系统、恢复和改善身体代谢功能以及无毒无副作用的适用于高尿酸及痛风人群的药食同源酵素及其制备方法。

所述有益微生物菌群培养按重量份比算,包括以下组分:EM菌(2~3×108CFU/mL)2-5份、乳酸菌(2~3×108CFU/mL)2-10份、酵母菌(2.5~3×108CFU/mL)2-8份、植物乳杆菌(1.5~2×108CFU/mL)2-6份、嗜热链球菌(2~4×108CFU/mL)2-4份、双歧杆菌(2~4×108CFU/mL)2-6份、鼠李糖乳杆菌(1~2×108CFU/mL)1-4份、嗜酸乳杆菌(2~4×108CFU/mL)2-5份、凝结芽孢杆菌(2~4×108CFU/mL)1-5份、罗伊氏乳杆菌(2~3×108CFU/mL)1-4份、蜂蜜2-10份、珊瑚钙2-5份、饮用水50-100份;
所述药食同源中药材原料按重量份比算,包括以下组分:葵花盘8-15份,玉米须5-12份,鱼腥草4-11份,薏苡仁3-10份,葛根5-12份,蒲公英2-6份,栀子3-8份,菊苣3-9份,杜仲4-8份,芡实2-8份,肉桂5-10份,淡竹叶4-10份、茯苓3-10份,马齿苋3-11份,百合3-10份。
一种适用于高尿酸及痛风人群的药食同源酵素的制备方法,包括以下步骤:
S1:有益微生物菌群培养:将上述EM菌、乳酸菌、酵母菌、植物乳杆菌、嗜热链球菌、双歧杆菌、鼠李糖乳杆菌、嗜酸乳杆菌、凝结芽孢杆菌、罗伊氏乳杆菌、蜂蜜、珊瑚钙和饮用水,均匀混合后置入恒温、带气压表、搅拌的发酵罐进行培养,制得有益微生物菌群及微生物代谢物简称A。
S2:去杂:将葵花盘、玉米须、鱼腥草、薏苡仁、葛根、蒲公英、栀子、菊苣、杜仲、芡实、肉桂、淡竹叶、茯苓、马齿苋、百合,分别去除杂质。
S3:干燥:经过去除杂质的原料分别进行热风干燥,热风干燥分为三级,得到干燥后的原料。
S4:粉碎:将干燥后的原料分别粉碎至200-300目,得到粉碎的后的原料。
S5:混合:将粉碎后的原料均匀混合简称B。
S6:发酵:将A和B均匀混合后置入恒温、带气压表、搅拌的发酵罐进行培养,发酵培养过程为无糖发酵培养,制得药食同源植物酵素简称C。
S7:分离:将C放入固液分离设备,安装300~500目滤布,分离得到药食同源植物酵素液体简称D和发酵后的药食同源中药原料简称E。
S8:二次混合:将80-120份D、赤藓糖醇5-13份、甜菊糖苷1-2份均匀混合简称F。
S9:包装入库:将F放入调配缸升温至78-95℃、过滤设备安装200-500目滤布过滤、灌装、喷淋杀菌、检品、包装、入库,制得无糖植物酵素。
S10:中药原料再利用:将E进行热风干燥,热风干燥分为三级,得到干燥后的原料简称G。
S11:饮片制造:将10-30份G、赤藓糖醇1-10份、甜菊糖苷1-2份、抗性糊精5-10份、硬脂酸镁10-20份、微晶纤维素5-20份、麦芽糊精50-80份均匀混合,造粒、糖衣、检品、包装、入库,制得无糖饮片。
优选的,所述S1中培养,温度:28~45℃,压力:2~15PSI、0.1~1.8kg/㎝2,搅拌:10~20r/min,发酵周期:160~360h,发酵完成后的液体PH:2.5~3.5,Bx:0~1。
优选的,所述S2操作中,鱼腥草烘干后的含水量在3%~10%;葛根需去皮,切成薄片;菊苣选取块根,将菊苣块根置于常温水中浸泡1.5-2h,去除泥土清洗漂净;淡竹叶除杂质残根,切段。
优选的,所述S3中一级干燥温度80~100℃,时间80~120min,搅拌20~50min/次,二级干燥温度:70~95℃,时间:40~80min,搅拌:20~40min/次,三级干燥温度:50~80℃,时间:4~14min,搅拌:20~40min/次,经过三级干燥后的原料水分含量≤10%。
优选的,所述S6中培养温度:28~45℃,压力:2~15PSI、0.1~1.8kg/㎝2,搅拌:10~20r/min,发酵周期:160~360h,发酵完成后的液体PH:2.5~3.5,Bx:0~1。

本发明的有益效果:本发明通过对微生物有益菌群培养,结合选用药食同源中药材原料进行发酵,其制取物能有效控制高尿酸或痛风人群尿酸指标水平,无毒无副作用,可日常食用。
对于药性的论述:
葵花盘:性甘、寒,归肝经,能够平肝止血,清热止痛,可用于治疗高血压、头晕头痛、耳鸣、子宫出血和疮疹等疾病,现代药理研究表明,葵花盘中的生物碱、黄酮、多糖和小分子肽成分同样具有抗炎、消肿、止痛、降低血尿酸、溶解尿酸钠的作用。
玉米须:有利尿、降压、利胆、降血糖。
鱼腥草:清热解毒、消痈排脓、利尿通淋。
薏苡仁:健脾益肾、利水消肿、舒筋除痹。
葛根:生津止渴,升阳止泻,通经活络,解酒毒。
蒲公英:清热解毒,利尿散结。
栀子:泻火除烦、清热利尿、凉血解毒。
菊苣:清肝利胆、健胃消食、利尿消肿。
杜仲:补肝肾、强筋骨。

技术优势:
利用微生物的习性,通过对EM菌、乳酸菌、酵母菌、植物乳杆菌、嗜热链球菌、双歧杆菌、鼠李糖乳杆菌、嗜酸乳杆菌、凝结芽孢杆菌、罗伊氏乳杆菌的共生培养,培养后的有益微生物菌群,结合药食同源中药材原料的萃取发酵,发酵培养过程为无糖发酵培养,发酵过程中可以有效的抑制有害菌产生,得到更安全可靠的原料。
发酵后的原料保留了中药原料的功效,如:清热、消炎、止痛、利尿、护肾等。微生物在代谢过程中产生的氨基酸、黄铜、多酚、维生素、粗多糖等可被人体直接利用吸收;乳酸菌和双歧杆菌代谢产生的有机酸,如:乳酸、醋酸、丙酸、丁酸等,结合膳食纤维,可以直接刺激肠蠕动,调节肠道功能,提高免疫系统,增加NK细胞数量。
