痤疮按轻、中、重程度来分可分为粉刺、丘疹、脓疱、结节、囊肿。重度痤疮一般就医治疗,使用超高浓度酸剥脱,再用温和产品日常护理,而轻中度痤疮一般选择日常护理。日常护理产品通常添加酸类成分,常用酸类成分有水杨酸、壬二酸、果酸(乙醇酸、乳酸、扁桃酸、柠檬酸等),但多数产品都会导致皮肤的接触性刺激,发红刺痛等皮肤问题,长期实验还会出现皮肤变薄,屏障变弱,易激惹易敏感等副作用。
目前,市场摒弃了高浓度单一酸产品,大多选择复合酸产品,降低单一高浓度酸带来的刺激性,且多种酸还能互补,护肤效果加乘。如中国发明专利《祛痘复合酸精华溶液及其制备方法》(申请号:202110529908.1)选择纳米脂质体水杨酸、壬二酸和乳酸复配制得精华液,能在较短时间内实现对毛囊过多角质化的抑制,有效改善闭口类痤疮,但是还加入了乙醇,提高成分溶解性,这对皮肤有一定刺激性,会引起皮肤敏感。中国发明专利《一种祛痘组合物、祛痘啫喱及其制备方法》(申请号:202110841255.0)复合酸选择了甜菜碱水杨酸盐和扁桃酸,搭配了多种舒缓植物活性物,但是化学改性后的水杨酸虽水中溶解度极大提升,刺激性降低,可是功效降低,生物利用率低。化学改性有功效降低的问题,脂质体化、微胶囊化制备工艺又复杂繁琐。中国发明专利《水杨酸增溶缓释组合物及其制备方法和用途》(申请号:201910049611.8)利用超分子化将水杨酸稳定于配方体系中,但使用单一水杨酸,含防腐体系,安全性不高。中国发明专利《一种超分子水杨酸复合凝胶及其制备方法》(申请号:201811325688.5)复合酸选择了水杨酸和扁桃酸,无防腐体系,但是溶剂PPG-26-丁醇聚醚-26添加量达到25%,该原料对皮肤没有明显刺激,但是从成本和利用率上考虑是没有必要的。
基于现有技术较全面分析,本领域迫切需要一种袪痘产品,工艺简单,生物利用率高,高功效,但同时温和低敏,为皮肤提供足够的保湿力,结合目前“精简护肤”的流行趋势,产品成分简单,功效明确,对皮肤无负担。
发明内容
本发明是为解决上述技术问题进行的,提供了一种温和低敏性保湿祛痘凝胶及其制备方法,以期提高治疗痤疮的高效性、全面性,同时提高复合酸在无油产品中的稳定性,以及日常护理对皮肤的温和低敏性。
为了实现该目的,本发明提供的温和低敏性保湿祛痘凝胶包括功效成分、增溶剂以及保湿剂。功效组分用于治疗痤疮,增溶剂实现功效组分在水中稳定,保湿剂同时提供保湿和凝胶形成功能。
为解决上述治疗痤疮的高效性、全面性的问题,本发明功能组分选择以重量百分比计的下述组分复配而成:0.5~2%水杨酸、1~10%壬二酸和0.5~6%聚羟基羧酸。
水杨酸,又名柳酸、β-羟基苯甲酸,可从柳树皮中提取。水杨酸可以使角质形成细胞分化、脱落,有较强的剥落性,有角质调节作用;具有脂溶性性质,可以渗入毛孔以及粉刺,有毛孔疏通、收缩毛孔、溶解粉刺的作用;还可以干扰花生四烯酸(刺激发炎作用)的代谢途径,起到抗炎、止痒的作用;酸性较强,对细菌和真菌都有较强的广谱杀菌作用。
壬二酸,又名杜鹃花酸,可从大麦、小麦这类麦类产品中提取,系天然存在的饱和直链二羧酸。壬二酸对皮肤有抗角质化作用,减少丝状角蛋白的合成,有防止毛囊角化过度作用;能抑制皮脂腺中5α-还原酶的生成,间接抑制雄性激素,使皮肤减少油脂分泌,有控油作用;能够进入细菌细胞质内,降低细胞内pH值,抑制细胞内DNA和蛋白质的合成,对各种需氧菌和厌氧菌均有抑菌和杀菌活性,显著减少皮肤表面和毛囊中葡萄球菌、痤疮丙酸杆菌的数量,有广谱抗菌作用且无耐药性;抑制细胞氧化代谢并清除自由基,有抗炎作用;能够抑制酪氨酸酶,防止炎症后色素沉积,有美白、祛痘印的作用。
聚羟基羧酸,含游离糖、蛋白、氨基酸类等物质,如乳糖酸、葡糖酸。聚羟基羧酸能提高透明质酸合成酶-3(HAS-3)和水通道蛋白AQP-3的表达,改善皮肤水合作用,高保湿作用;是细胞间基质糖胺聚糖的中间体,能促进表面细胞再生,促使伤口愈合;促进与加速皮肤温和微剥落,消除死细胞,促进皮肤更新,低刺激性;能抑制黑色素形成,减少皮肤暗沉,减少痘印的生成。
水杨酸浅层剥脱,侧重预防治疗出油过多、闭口、粉刺(黑、白头),壬二酸抗炎抗菌,侧重预防治疗炎性红肿型痘痘和袪痘印,聚羟基羧酸保湿修护,侧重促进痘痘愈合和皮肤提亮,三者在特定配比下,效果互补,协同增效。
组分含量方面,水杨酸的重量百分比介于0.5~2%之间,壬二酸的重量百分比介于1~10%之间,聚羟基羧酸的重量百分比介于0.5~6%之间。三种组分含量过低难以实现祛痘效果,含量过高反而会对皮肤带来刺激。
为解决上述复合酸在无油产品中的稳定性的问题,增溶剂选自泊洛沙姆,优选泊洛沙姆407,重量百分比为10~20%。
水杨酸和壬二酸都难溶于水,泊洛沙姆407因同时含有EO和PO结构,既亲水又亲油,在水溶液中以分子与分子之间的相互作用超分子化水杨酸和壬二酸,使其在体系中更稳定,最终体系形成凝胶。凝胶具有缓释性,泊洛沙姆形成的超分子结构,使得所有的酸被包裹在超分子中央,以游离酸的形式释放,最大限度地发挥功效,且无需加入强碱中和剂,这都极大程度的降低对皮肤的刺激性。最终形成的凝胶体系pH为2~3,无需再过多加入防腐剂。
为解决上述日常护理对皮肤的无负担性的问题,作为优选,选择甘油、聚乙二醇-8和PPG-9二聚甘油醚为本发明的保湿剂。甘油、聚乙二醇-8和PPG-9二聚甘油醚成分安全,对皮肤没有副作用,同时还有促使凝胶体系更稳定的作用。
进一步,本发明的温和低敏性保湿祛痘凝胶的优选配方如下:由以下质量百分比的组分组成:0.5~2%水杨酸、1~10%壬二酸、0.5~6%聚羟基羧酸、10~20%泊洛沙姆407、5~15%甘油、2~10%聚乙二醇-8、2~10%PPG-9二聚甘油醚,余量为蒸溜水。
本发明的第二方面,提供了上述温和低敏性保湿祛痘凝胶的制备方法,包括下述步骤:
(1)A相:将水与泊洛沙姆407在75℃下搅拌溶解至澄清透明溶液;
(2)将B相组分甘油、壬二酸加入A相在75℃下继续搅拌溶解至澄清透明溶液;
(3)C相:将水杨酸、聚乙二醇-8、PPG-9二聚甘油醚在75℃下搅拌溶解至澄清透明;
(4)将C相加入A相,在75℃下搅拌溶解至澄清透明溶液后,降温至60℃;
(5)将聚羟基羧酸加入A相,搅拌均一后即得温和低敏性保湿祛痘凝胶。
相比现有技术,本发明的技术效果如下:
功能方面,本发明制备的温和低敏性保湿祛痘凝胶中,水杨酸浅层剥脱,侧重预防治疗出油过多、闭口、粉刺(黑、白头);壬二酸抗炎抗菌,侧重预防治疗炎性红肿型痘痘和袪痘印;聚羟基羧酸促进皮肤中糖胺聚糖或者胶原蛋白的生物合成起到保湿舒缓修复作用。三者在特定配比下,效果互补,协同增效,对皮肤温和低敏保湿,同时对不同类型的痤疮都有更高效、更全面的预防治疗作用,具有优异的袪痘效果。
安全性方面,使用泊洛沙姆407作为增溶缓释剂,其形成的超分子结构,使得所有的酸被包裹在超分子中央,以游离酸的形式释放,最大限度地发挥功效,且无需加入强碱中和剂,极大程度的降低对皮肤的刺激性体系稳定,无防腐无酒精溶剂,成分精简,日常护理对皮肤无负担、更温和。

具体实施方式
实施例一、稳定性试验
按照表1中对比例1、2以及实施例1的配方配置祛痘凝胶,在相同条件下放置相同的时间,对稳定性进行考察。

稳定性结果统计:考察不同制备方法下凝胶配方的稳定性,稳定以“√”标识,不稳定以“×”标识,具体结果参见表2。

分析可知,随着泊洛沙姆407含量的提高,在水溶液中以分子与分子之间的相互作用超分子化水杨酸和壬二酸的能力也相应提升,使其在体系中更稳定,最终形成的凝胶体系稳定性也随之上升。
聚乙二醇-8或PPG-9二聚甘油醚的存在改变了泊络沙姆的相变特性,泊络沙姆在该条件下形成的凝胶体系稳定性更佳。
结果显示,本发明制备工艺下所得凝胶配方,在常规的高低温状态下能够保持配方制剂的稳定状态。
实施例二、凝胶制剂的温和低敏性试验
通过人体斑贴实验,对不同凝胶配方的敏感性进行对比。按照表3中对比例3、4以及实施例1的配方配置祛痘凝胶,并将该不同成分配比的凝胶制剂作为待测物。

实验方法:将待测物分别置于斑贴小室,敷贴于受试者的背部,24小时后揭开,观察0.5h,24h和48h的皮肤刺激反应,样本量为35人。
结果统计:分别统计出现刺激反应的例数(代表刺激反应发生频率),结果参见表4。

实施例三、凝胶制剂的祛痘抗炎试验
将本发明实施例1的凝胶制剂与一款市售祛痘产品进行抗痘功效试验对比,所选受试者为18~25岁健康男女30名,诊断为寻常型痤疮,且符合I,II级标准的患者。
痤疮临床评价标准如下:
轻度Ⅰ级:以粉刺为主;可有少量丘疹和脓疱,总病灶数少于30个;
中度Ⅱ级:有粉刺,并有中等数量的丘疹和脓疱,总病灶数在31—50个之间;
中度Ⅲ级:大量丘疹和脓疱,偶见大的炎性皮损,分布广泛,总病灶数在51—100个之间,结节小于3个;
重度Ⅳ级:结节,囊肿性或聚合性痤疮,多数有疼痛并形成囊肿,病灶数在100个以上:结节,囊肿在3个以上。
试验方法:
整个试验4周,于Baseline、3天,1周,2周,4周进行评估,采用半脸对照,医生临床评分参见图1。结果显示,对于Ⅰ、Ⅱ级痤疮,本发明祛痘凝胶具有显著的治疗效果,在D3即出现显著的疗效,使得痤疮减少44%,在持续的使用中持续降低痤疮的数量,且与竞品相比*P<0.05,治疗效果优于竞品。
控油效果评价参见图2,结果显示祛痘凝胶在D7,D14,D28时出现显著的控油效果,且与市售产品相比*P<0.05,控油效果优于市售产品。
上述的对实施例的描述仅用于阐明本发明,以便于本领域技术人员理解和使用本发明,并非对本发明进行限制。本发明不限于上述实施例,本领域技术人员根据本发明在不脱离本发明范畴所做出的改进和修改都在本发明的保护范围之内。
Claims (3)
1.一种温和低敏性保湿祛痘凝胶,其特征在于,由以下质量百分比的组分组成:0.5~2%水杨酸、1~10%壬二酸、0.5~6%聚羟基羧酸、10~20%泊洛沙姆407、5~15%甘油、2~10%聚乙二醇-8、2~10%PPG-9二聚甘油醚,余量为蒸馏水。
2.根据权利要求1所述的温和低敏性保湿祛痘凝胶,其特征在于:
其中,所述聚羟基羧酸选自葡糖酸、乳糖酸、聚乳酸羟基乙酸,聚羟基丁酸,3-羟基丁酸,聚乳酸中的任意一种或多种。
3.权利要求1所述的温和低敏性保湿祛痘凝胶的制备方法,其特征在于,包括下述步骤:
(1)A相:将水与泊洛沙姆407在70℃-75℃下搅拌溶解至澄清透明溶液;
(2)将B相组分甘油、壬二酸加入A相在70℃-75℃下继续搅拌溶解至澄清透明溶液;
(3)C相:将水杨酸、聚乙二醇-8、PPG-9二聚甘油醚在70℃-75℃下搅拌溶解至澄清透明;
(4)将C相加入A相,在75℃下搅拌溶解至澄清透明溶液后,降温至60℃-55℃;
(5)将聚羟基羧酸加入A相,搅拌均一后即得温和低敏性保湿祛痘凝胶。
