Abstract
The invention provides a preparation method of a composition preparation containing tomato water-soluble dietary fiber and lycopene, which comprises the following steps: extracting water-soluble dietary fiber from tomato residue; preparing a lycopene complex; mixing and compounding to obtain the composition preparation containing the water-soluble dietary fiber of the tomatoes and the lycopene. The invention creates a deep utilization approach of lycopene extraction processing by combining beneficial lycopene with a combined preparation of a byproduct of extraction, namely tomato water-soluble fiber. Technical Field

Background
Lycopene, a carotenoid present in plants, has widely known effects of resisting oxidation, protecting heart and cerebral vessels, inhibiting tumors and the like, and is widely used in the fields of health products, food additives and the like.
In the prior art, various methods exist for extracting lycopene, such as organic solvent extraction, alkaline extraction and the like. However, regardless of the method, the tomato residue after extracting lycopene is generally discarded or used as feed and is not processed and utilized, resulting in a great waste of beneficial nutritional components, especially the great amount of water-soluble dietary fiber contained therein.
Water soluble dietary fibers are different from dietary fibers commonly recognized in the art. In the art, dietary fiber includes polysaccharides, oligosaccharides, lignin, etc., such as cellulose, hemicellulose, pectin, etc., which are not normally digested and absorbed by the small intestine, and although not digestible by the gastrointestinal tract, are still beneficial components. Dietary fiber is classified into water-soluble dietary fiber and insoluble dietary fiber according to solubility, and as the name suggests, insoluble dietary fiber refers to fiber insoluble in hot water, such as cellulose, hemicellulose, lignin, etc. The water-soluble dietary fiber is fiber soluble in warm water or hot water, and mainly contains polysaccharide such as glucan derived from pectin. The insoluble dietary fiber has a main effect of improving defecation, and the water-soluble dietary fiber has a remarkable effect on prevention of various diseases besides improving defecation. The current research shows that the water-soluble fiber has the function that the insoluble dietary fiber can not replace in the aspect of preventing gastrointestinal diseases and the like. Nutritionals recommend that the water soluble dietary fiber should not be less than 20% of the total dietary fiber ingested on a daily basis.
In practical applications, carrying cellulose is not only inconvenient but also difficult to swallow in areas where fresh vegetables and fruits are not available in sufficient quantities or in outdoor working environments. Therefore, the development of food rich in water-soluble dietary fiber has important market value.
The tomato yield in China is in the front in the world, and with the continuous expansion of the market demand of lycopene, more and more tomatoes are used for extracting lycopene, so that a large amount of extraction residues are generated, and the comprehensive processing and utilization research based on the tomato residues is less at present.
The residual residue after lycopene extraction contains a large amount of crude fibers such as cellulose, has rough taste, and is not suitable for direct eating even after superfine grinding; however, a large amount of pectin, rhamnose, galacturonic acid and the like contained in the dietary fiber are good water-soluble dietary fiber sources.
Therefore, the development of an improved extraction process of water-soluble dietary fiber based on tomato residues has important economic value.
In addition, lycopene, being oil-like or crystalline, is insoluble in water and is usually consumed in the form of capsules, which severely limits its versatility and is difficult to be used as an additive component in food processing.
As described above, although the prior art provides various lycopene preparation methods, its formulation form is single, and its combined formulation with extraction byproduct, tomato water-soluble fiber, has not been reported.
Disclosure of Invention
As mentioned above, the technical problem to be solved by the present invention is to overcome the defects of single lycopene preparation and extraction residue availability in the prior art, and the present invention aims to provide a preparation containing water-soluble dietary fibers of tomatoes and lycopene and a preparation method thereof.
Specifically, in a first aspect, the invention provides a preparation method of a composition preparation containing water-soluble dietary fibers of tomatoes and lycopene, which comprises the following steps:
s1: extracting water-soluble dietary fiber from tomato residue after separating lycopene;
s2: preparing a lycopene complex;
s3: mixing, compounding and granulating to obtain the composition preparation containing the water-soluble dietary fiber of the tomatoes and the lycopene.
The lycopene is usually oily lycopene oleoresin (rich in other beneficial fat-soluble components), and has relatively complete fiber residue.
In the above preparation method, the step S1 specifically includes the following steps:
1) Drying the tomato residues in an oven, and crushing to a particle size of more than 100 meshes; mixing the crushed powder with water according to a weight ratio of 1 to 4-6, filtering to remove epidermis and seeds, and then grinding for 15-30min to obtain grinding slurry;
2) Diluting the above ground slurry with equal weight of purified water, heating to 50-60 deg.C in a heating kettle, adjusting pH to about 7.5 (preferably 7.5-7.8) with sodium carbonate, and alkalizing under stirring for 3-5 hr to obtain alkalized slurry;
3) Adding 0.3-0.5wt% of pectinase (based on dry weight of residue), performing enzymolysis for 2-3 hr at 48-50 deg.C under stirring, adding 0.5-0.8wt% of cellulase (based on dry weight of residue), adjusting pH to 6.5-7 (preferably acetic acid or citric acid as pH regulator), and performing enzymolysis at 42-45 deg.C for 2-3 hr under stirring;
4) Heating to 55-60 deg.C, adding 0.1-0.3wt% xylanase (preferably the combination enzyme of endonuclease and exonuclease), and continuously stirring for enzymolysis for 3h; boiling the enzymolysis solution for 10-15min to inactivate enzyme, cooling to room temperature, centrifuging, collecting filtrate, and concentrating the filtrate by evaporation (preferably to solid content of above 40 wt%) to obtain water-soluble fiber concentrated solution.
5) Adding the water-soluble fiber concentrated solution into 95% (v/v) ethanol with 5-6 times volume of the water-soluble fiber concentrated solution for alcohol precipitation, standing for precipitation for 1-2h, filtering, separating and precipitating, washing the obtained precipitate with absolute ethanol, drying, and micronizing the dried precipitate to obtain the water-soluble dietary fiber powder with the water holding capacity of 5-6g/g and the yield of more than 8% (based on the dry powder of residues).
In the above preparation method of the present invention, the step S2 specifically includes the following steps:
1) Mixing lycopene oleoresin with corn oil according to the weight part of 1: 2-3, then adding 0.1-0.5 part of beeswax, primarily stirring, and then grinding in a colloid mill until uniform to obtain the lycopene compound.
In the above preparation method of the present invention, the step S3 specifically includes the following steps:
1) Slowly adding the lycopene complex into 35-50wt% cyclodextrin solution at 45-50 deg.C under rapid stirring for coating treatment (by weight ratio, lycopene complex: 10-15) of cyclodextrin solution, stirring for 30-60min, adding dietary fiber or dietary fiber solution (the dosage of the dietary fiber is preferably 2-5 times of the mass of the lycopene complex), continuously stirring until the mixture is uniform, adjusting the water content with proper amount of distilled water, homogenizing in a homogenizer for 1-2 times, and then performing spray drying in a high-speed centrifugal spray dryer to obtain the lycopene-cyclodextrin-water-soluble fiber complex particles.
2) Coating the above compound granules with coating liquid (such as carboxymethyl cellulose solution or heated gelatin swelling liquid) in boiling coating machine to obtain composition preparation containing water soluble dietary fiber of tomato and lycopene, sterilizing, and packaging; wherein, preferably, the carboxymethyl cellulose solution is 1 to 3 weight percent, and the gelatin swelling solution is 5 to 10 weight percent.
Wherein, the lycopene-cyclodextrin-water soluble fiber compound particles obtained by the invention can be dried without adopting a coating and then filled with nitrogen for packaging and storage or capsule storage. But the storage stability of the coating is also suitably enhanced after the spray coating treatment.
Preferably, the particle size of the composition preparation of the present invention is controlled to be 0.1-0.5mm.
In a second aspect of the present invention, there is provided a composition formulation comprising water-soluble dietary fiber of tomato and lycopene prepared by the above preparation method. In a third aspect, the invention also provides the application of the composition preparation, which can be directly eaten as a health-care product and also can be used as an additive for food processing and is used as an additive component of raw materials such as flour and the like.
Preferably, when it is used as an additive, the addition amount thereof is preferably 5 to 50wt%.
The invention has the following beneficial effects:
1) The composition preparation simultaneously considers the coexistence of water-soluble components and oily lycopene, and has the beneficial effects brought by the lycopene besides a large amount of soluble dietary fibers. Meanwhile, through coating treatment and enveloping treatment, the storage stability is obtained, and the food packaging film can be used for food processing.
2) The water-soluble dietary fiber of the tomato is effectively utilized as a byproduct of the extraction of the lycopene, the extraction efficiency of the water-soluble dietary fiber of the tomato can reach more than 80 percent (the residual water-soluble fiber is less than 20 percent) through a specific process procedure, and a deep processing way of the residual residue of the lycopene extract is created.
3) The preparation method is simple, is suitable for large-scale production, and has good application prospect and market value.
Detailed Description
The present invention will be described in detail with reference to specific examples, but the scope of the present invention is not limited to these examples in any way.
Preparation example 1
1) Drying the residual tomato residue after extracting lycopene with chloroform in an oven, crushing and sieving with a 160-mesh sieve; mixing pulverized powder 1kg with purified water 5L, stirring, filtering to remove residue components such as epidermis and seed, and grinding in small grinder for 20min to obtain grinding slurry;
2) Diluting the above ground slurry with 6L of purified water, heating to 60 deg.C in a heating kettle, adjusting pH to about 7.5 with sodium carbonate, and alkalizing for 3 hr under stirring to obtain alkalized slurry;
3) Adding 5g of pectinase (based on dry weight of original tomato residue), performing enzymolysis at 50 deg.C for 3 hr under stirring, adding 6g of cellulase (based on dry weight of residue), adjusting pH to 6.8 with citric acid, and performing enzymolysis at 45 deg.C for 3 hr under stirring;
4) Heating to 58 ℃, adding 3g of xylanase, and continuously stirring for enzymolysis for 3 hours; and boiling the enzymolysis liquid for 15min to inactivate enzyme, cooling to room temperature, performing solid-liquid separation by high-speed centrifugation, collecting filtrate, and performing evaporation concentration on the filtrate to about 20g to obtain a water-soluble fiber concentrated solution.
5) Adding the water-soluble fiber concentrated solution into 110ml of 95% (v/v) ethanol for alcohol precipitation, standing for precipitation for 1h, filtering, separating and precipitating, washing the obtained precipitate with absolute ethanol, drying, and pulverizing to obtain about 8.2g of water-soluble dietary fiber powder with water holding capacity of about 5.8g/g. Correspondingly, when the water-soluble dietary fiber powder is not subjected to alkalization treatment and xylanase, only 6.7g of water-soluble dietary fiber powder is obtained, and the extraction rate is obviously reduced.
Example 1
1) The chloroform-extracted lycopene oleoresin was mixed with corn oil in parts by weight of 1.
2) Slowly adding the lycopene complex into 35wt% of cyclodextrin solution at 50 ℃ under the condition of rapid stirring for coating treatment (410 g of cyclodextrin solution), stirring for 30min, adding 30g of dietary fiber, continuously stirring until the mixture is uniform, adjusting the water content by using distilled water, homogenizing in a homogenizer, and then carrying out spray drying to obtain the lycopene-cyclodextrin-water-soluble fiber complex particles.
3) Coating the composite particles with 2wt% carboxymethyl cellulose solution in a boiling coating machine to obtain a composition preparation containing water-soluble dietary fiber of tomato and lycopene, sterilizing and packaging.
Example 2
1) The chloroform extracted lycopene oleoresin was mixed with corn oil in parts by weight of 1.
2) Slowly adding the lycopene complex into 40wt% of cyclodextrin solution at 50 ℃ under the condition of rapid stirring for coating treatment (550 g of cyclodextrin solution), stirring for 30min, adding 40g of dietary fiber, continuously stirring until the mixture is uniform, adding a proper amount of distilled water into a homogenizer for homogenizing treatment, and then performing spray drying to obtain the lycopene-cyclodextrin-water-soluble fiber complex particles.
3) Spraying the composite particles in a coating machine with 6wt% gelatin swelling solution at 55 deg.C to obtain composition preparation containing water soluble dietary fiber of tomato and lycopene, sterilizing, and packaging.
The above embodiments do not limit the technical solutions of the present invention, and a person skilled in the art may modify the technical solutions described in the above embodiments without departing from the scope of the technical solutions of the present invention.
Claims (10)
1. A composition preparation containing water-soluble dietary fiber of tomato and lycopene is characterized by comprising lycopene complex and water-soluble dietary fiber extracted from tomato residue; the lycopene complex comprises lycopene oleoresin.
2. A preparation method of a composition preparation containing water-soluble dietary fiber of tomatoes and lycopene is characterized by comprising the following steps:
s1: extracting water-soluble dietary fiber from tomato residue after separating lycopene;
s2: preparing a lycopene complex;
s3: mixing, compounding and granulating to obtain the composition preparation containing the water-soluble dietary fiber of the tomatoes and the lycopene.
3. The method according to claim 2, wherein the step S1 comprises the steps of:
1) Drying the tomato residues in an oven, and crushing to obtain powder with the particle size of more than 100 meshes; mixing the crushed powder with water according to a weight ratio of 1-6, filtering out epidermis and seeds, and then grinding for 15-30min to obtain grinding slurry;
2) Diluting the ground slurry with purified water, heating to 50-60 deg.C in a heating kettle, adjusting pH to about 7.5 with sodium carbonate, and alkalizing under stirring for 3-5 hr to obtain alkalized slurry;
3) Firstly, adding 0.3-0.5wt% of pectinase, stirring and performing enzymolysis for 2-3h at 48-50 ℃, then adding 0.5-0.8wt% of cellulase, adjusting the pH value to 6.5-7, and continuously stirring and performing enzymolysis for 2-3h at 42-45 ℃;
4) Heating to 55-60 ℃, adding 0.1-0.3wt% of xylanase, and continuously stirring for enzymolysis for 3h; boiling the enzymolysis solution for 10-15min to inactivate enzyme, cooling to room temperature, centrifuging, collecting filtrate, and concentrating the filtrate by evaporation to obtain water-soluble fiber concentrated solution;
5) Adding the water-soluble fiber concentrated solution into 95% ethanol with volume of 5-6 times of the water-soluble fiber concentrated solution for alcohol precipitation, standing for precipitation for 1-2h, filtering, separating and precipitating, washing the obtained precipitate with absolute ethanol, drying, and micronizing the dried precipitate to obtain the water-soluble dietary fiber powder.
4. The method according to claim 3,
acetic acid or citric acid is used as an acidic pH regulator, and the amount ratio of the enzyme is based on the dry weight of the residue.
5. The preparation method according to claim 2, wherein the step S2 specifically comprises the steps of: mixing lycopene oleoresin with corn oil according to the weight part of 1: 2-3, then adding 0.1-0.5 part of beeswax, primarily stirring, and then grinding in a colloid mill until uniform to obtain the lycopene compound.
6. The preparation method according to claim 2, wherein the step S3 specifically comprises the steps of:
1) Slowly adding the lycopene complex into 35-50wt% of cyclodextrin solution at 45-50 ℃ under the condition of rapid stirring for coating treatment, stirring for 30-60min, adding dietary fiber or dietary fiber solution, continuously stirring to be uniform, adjusting the water content with appropriate amount of distilled water, homogenizing in a homogenizer for 1-2 times, and then carrying out spray drying in a high-speed centrifugal spray dryer to obtain lycopene-cyclodextrin-water-soluble fiber complex particles;
2) Coating the composite particles with coating liquid in a coating machine to obtain a composition preparation containing water-soluble dietary fiber of tomato and lycopene, sterilizing and packaging.
7. The process according to claim 6, wherein the ratio by weight of lycopene complex: cyclodextrin solution = 1; the dosage of the dietary fiber is 2-5 times of the mass of the lycopene complex.
8. The process according to claim 6, wherein the coating solution is selected from the group consisting of a carboxymethyl cellulose solution or a heated gelatin swelling solution; preferably, the carboxymethyl cellulose solution is 1 to 3wt%, and the gelatin swelling solution is 5 to 10wt%.
9. The composition preparation containing the water-soluble dietary fiber of tomatoes and the lycopene prepared by the preparation method of any one of claims 2 to 8.
10. Use of the composition preparation according to claim 9 as an additive for health care products or food processing.
