本发明涉及一种妇炎清洗剂中药组合物的制备方法,属于制药技术领域。
技术背景
湿热下注所致的外阴瘙痒、白带量多是一种常见的妇科疾病。目前治疗妇女湿热下注所致的外阴瘙痒、白带量多的中成药洗剂很多,但大多治疗效果不够理想。现有技术专利检索:1、中国专利公报2005年3月9日公开了名称为“一种治疗妇科炎症的中药外用洗剂”公开号CN1589884A的专利申请,组成此发明原料药的配比为:蛇床子15%-30%、白鲜皮15%-30%、黄柏15%-30%、荆芥6%-10%、防风6%-10%、苦参10%-16%、龙胆6%-10%,工艺为“蛇床子、荆芥、防风水蒸馏提取挥发油,蒸馏后的水溶液与挥发油混合,药渣与其余白鲜皮、黄柏、苦参、龙胆药材加水煎煮三次,滤液浓缩成清膏,加95%乙醇使含醇量达65%,回收乙醇,药液与油水混合液混合,分装,即得洗剂”。在实际应用过程中,我们发现用以上现有技术1这种工艺制成的中成药外用妇科洗剂的效果不够理想;在近十年的时间里,我们在此组方的基础上,通过大量的实验摸索,对我公司产品“妇炎清洗剂”的工艺进行二次开发,意外的发现将工艺中“药渣与其余白鲜皮、黄柏、苦参、龙胆四味药材加水煎煮三次,滤液浓缩成清膏,加95%乙醇使含醇量达65%”改成“药渣与其余白鲜皮、黄柏、苦参、龙胆四味药材加40%乙醇回流提取三次,滤液回收乙醇至无醇味”后,发现本发明工艺制成的中成药,其临床药效学实验效果显著提高,且无任何毒副作用;我们按常规工艺制成妇炎清洗剂。
发明内容
本发明的目的在于:提供疗效更为显著的一种妇炎清洗剂中药组合物的制备方法。其特点是,本发明中药组合物各原料药的用量配比为:
制备方法为:
以上蛇床子、白鲜皮、黄柏、荆芥、防风、苦参、龙胆共七味药材,取荆芥、防风、蛇床子,用水蒸气蒸馏法提取挥发油,蒸馏后的水溶液与挥发油混合,加聚山梨酯807.8ml,使成均匀澄清的药液,备用;药渣与其余黄柏、白鲜皮、苦参、龙胆四味药材加40%乙醇回流提取三次,第一次2小时,第二次1.5小时,第三次1小时,合并煎液,滤过,滤液回收乙醇至无醇味,与上述药液混匀,加苯甲酸钠1.5g,再加水至1000ml,混匀,即得洗剂。
现有技术专利检索:1、中国专利公报2005年3月9日公开了名称为“一种治疗妇科炎症的中药外用洗剂”公开号CN1589884A的专利申请,组成此发明原料药的配比为:蛇床子15%-30%、白鲜皮15%-30%、黄柏15%-30%、荆芥6%-10%、防风6%-10%、苦参10%-16%、龙胆6%-10%,工艺为“蛇床子、荆芥、防风水蒸馏提取挥发油,蒸馏后的水溶液与挥发油混合,药渣与其余白鲜皮、黄柏、苦参、龙胆药材加水煎煮三次,滤液浓缩成清膏,加95%乙醇使含醇量达65%,回收乙醇,药液与油水混合液混合,分装,即得洗剂”。
在实际应用过程中,我们发现用以上现有技术1这种工艺制成的中成药外用妇科洗剂的效果不够理想;在近十年的时间里,我们在此组方的基础上,通过大量的实验摸索,对我公司产品“妇炎清洗剂”的工艺进行二次开发,意外的发现将工艺中“药渣与其余白鲜皮、黄柏、苦参、龙胆四味药材加水煎煮三次,滤液浓缩成清膏,加95%乙醇使含醇量达65%”改成“药渣与其余白鲜皮、黄柏、苦参、龙胆四味药材加40%乙醇回流提取三次,滤液回收乙醇至无醇味”后,发现本发明工艺制成的中成药,其临床药效学实验效果显著提高,且无任何毒副作用;我们按常规工艺制成妇炎清洗剂。

以上蛇床子、白鲜皮、黄柏、荆芥、防风、苦参、龙胆共七味药材,取荆芥、防风、蛇床子,用水蒸气蒸馏法提取挥发油,蒸馏后的水溶液与挥发油混合,加聚山梨酯807.8ml,使成均匀澄清的药液,备用;药渣与其余黄柏、白鲜皮、苦参、龙胆四味药材加40%乙醇回流提取三次,第一次2小时,第二次1.5小时,第三次1小时,合并煎液,滤过,滤液回收乙醇至无醇味,与上述药液混匀,加苯甲酸钠1.5g,再加水至1000ml,混匀,即得洗剂。
本发明妇炎清洗剂具有清热燥湿,杀虫止痒的功效;主要用于湿热下注所致的外阴瘙痒,白带量多;外阴炎、阴道炎见以上证候者。
主要药效学试验
实验药物的制备:
1、试验用药物的制备:
本发明妇炎清洗剂组:按照本发明说明书实施例1的方法制得的洗剂。
a组为:按照对比文件1公开号为CN1589884A的专利申请说明书实施例1的方法制得的外用洗剂,具体处方及工艺如下:
按对比文件1重量配比1组(实施例1):蛇床子22.2%、白鲜皮22.2%、黄柏22.2%、荆芥6.7%、防风6.7%、苦参13.3%、龙胆6.7%配比,a组处方各药材重量配比为:
制备方法为:
(1)取上述药物按《中国药典》2000年版一部各药材项下进行炮制,合格后按下述方法制备;
(2)取蛇床子、荆芥、防风等加10倍量水蒸馏2小时,收集挥发油,蒸馏后的水溶液与挥发油混合,加聚山梨酯-807.8ml,使成均匀澄明药液,备用;
(3)取上述蛇床子、荆芥、防风药渣与白鲜皮、黄柏、苦参、龙胆四味加水煎煮三次,第一次加水10倍量,煎煮2小时,第二次加水8倍量,煎煮1.5小时,第三次加水6倍量,煎煮1小时,滤过,合并滤液,浓缩至相对密度1.1~1.2(70℃),加95%乙醇,使醇含量达65%,静置24小时,滤过,滤液回收乙醇至无醇味,备用;
(4)药液与上述油水混合液混合,加苯甲酸钠1.5g,加蒸馏水调至1000ml,搅匀;
(5)分装,封口,微波灭菌,检验;
(6)外包装,检验合格后入库。
b组为:按照本发明组方配比,工艺为“药渣与其余黄柏、白鲜皮、苦参、龙胆四味药材加30%乙醇提取”,制得的妇科外用洗剂,具体如下:

实验目的:通过对本发明妇炎清洗剂组和a组、b组、c组、d组的抗炎、镇痛、止痒、抗过敏和细胞免疫等药理实验研究,将本发明妇炎清洗剂组与a组、b组、c组、d组进行对比,观察其药理作用的强弱。
试验方法:本发明妇炎清洗剂组和a组、b组、c组、d组对大鼠棉球肉芽肿的影响;对小鼠尾根加压所致疼痛的影响;对磷酸组胺引起豚鼠致痒反应的影响;对二硝基氯苯诱发迟发型皮肤过敏反应的影响;对小鼠细胞免疫功能的影响。
一、对大鼠棉球肉芽肿的影响
实验材料
1、动物:Wistar雌性大鼠,体重180~220g。
2、药物:本发明妇炎清洗剂组和a组、b组、c组、d组。药物在实验前按含生药材量为45%的浓度配制。
实验方法
Wistar雌性大鼠60只,体重180~220g,随机分成6组,每组10只。给大鼠腹腔注射戊巴比妥钠30mg/kg麻醉,将动物固定,下腹部皮肤消毒,行正中切口,向两侧腹股沟皮下各放入1个10mg灭菌棉球,缝合皮肤切口。术后大鼠注射青霉素钾盐每日1次,共2次。各组均于术后次日开始在伤口外涂抹给药,对照组涂抹同体积的生理盐水;本发明妇炎清洗剂组和a组、b组、c组、d组分别涂抹给药1.0g生药/kg。连续15d,每日1次,末次给药后1h,将动物处死,取出棉球肉芽肿,置60℃烘箱中干燥12h后称重,各自减去原棉球重量,即为肉芽肿重量。实验结果:见表1
与对照组相比**P<0.01,*P<0.05;与本发明妇炎清洗剂组比△P<0.05。
结果表明:本发明妇炎清洗剂组和a组、b组、c组、d组可明显抑制大鼠棉球肉芽的增生,本发明妇炎清洗剂组与对照组相比有极显著性差异(P<0.01);a组、b组、c组、d组与对照组相比较有显著性差异(P<0.05);本发明妇炎清洗剂组与a组、b组、c组、d组比较有显著性差异(P<0.05)。可见,本发明妇炎清洗剂组比a组、b组、c组、d组的抗炎作用强。
二、对小鼠尾根加压所致疼痛的影响
实验材料
1、动物:昆明种雌性小鼠,体重23~27g。
2、药物:本发明妇炎清洗剂组和a组、b组、c组、d组。药物在实验前按含生药材量为45%的浓度配制。
实验方法
采用小鼠尾根压痛法,在离小鼠尾根1cm处,作为压痛点,用压痛仪测定痛阈值,以小鼠因尾部受压疼痛嘶叫为准,筛选痛阈值在10~45内的雌性小鼠60只,体重23~27g,随机分为6组,每组10只,测定痛阈两次,以均值作为药前值。各组分别给药,在压痛点处涂药,对照组涂抹同体积的生理盐水;本发明妇炎清洗剂组和a组、b组、c组、d组分别涂抹给药均为2.0g生药/kg,在给药1h后,测定痛阈值。实验结果:见表2
与对照组相比**P<0.01,*P<0.05;与本发明妇炎清洗剂组比△P<0.05。
结果表明:本发明妇炎清洗剂组和a组、b组、c组、d组均对小鼠尾根加压所致疼痛有明显的镇痛作用,可使压痛压力升高,本发明妇炎清洗剂组与对照组相比有极显著性差异(P<0.01);a组、b组、c组、d组与对照组相比较有显著性差异(P<0.05);本发明妇炎清洗剂组与a组、b组、c组、d组比较有显著性差异(P<0.05)。可见,本发明妇炎清洗剂组比a组、b组、c组、d组的镇痛作用强。
三、对磷酸组织胺引起豚鼠致痒反应的影响
实验材料
1、动物:雌性豚鼠,体重250~290g。
2、药物:本发明妇炎清洗剂组和a组、b组、c组、d组。药物在实验前按含生药材量为45%的浓度配制。
实验方法
健康雌性豚鼠60只,体重250~290g,随机分成6组,每组10只。实验前将每只豚鼠右后足背刮毛,在刮毛处各组动物分别涂抹药物,对照组涂抹同体积的生理盐水;本发明妇炎清洗剂组和a组、b组、c组、d组分别涂抹给药均为0.8g生药/kg,每天2次,连续给药5d,末次给药后2h,在每只豚鼠右后足背刮毛处用粗砂纸擦伤脱毛处表皮,面积约1cm2,10min后开始在创面处滴0.01%磷酸组织胺0.05ml/只,此后每隔3min依0.02%、0.03%、0.04%……递增浓度,每次均为0.05ml/只。直至出现豚鼠回头舔擦伤处,豚鼠出现回头舔右后足时所给予的磷酸组织胺总量即为致痒阈。实验结果:见表3
与对照组相比**P<0.01,*P<0.05;与本发明妇炎清洗剂组比△P<0.05。
结果表明:本发明妇炎清洗剂组和a组、b组、c组、d组能明显提高磷酸组织胺引起豚鼠瘙痒的致痒阈,本发明妇炎清洗剂组与对照组相比有极显著性差异(P<0.01);a组、b组、c组、d组与对照组相比较有显著性差异(P<0.05);本发明妇炎清洗剂组与a组、b组、c组、d组比较有显著性差异(P<0.05)。可见,本发明妇炎清洗剂组比a组、b组、c组、d组的止痒作用强。
四、对二硝基氯苯诱发迟发型皮肤过敏反应的影响
实验材料
1、动物:昆明种雌性小鼠,体重23~27g。
2、药物:本发明妇炎清洗剂组和a组、b组、c组、d组。药物在实验前按含生药材量为45%的浓度配制。
实验方法
昆明种雌性小鼠60只,体重23~27g,随机分成6组,每组10只。各组小鼠颈部去毛,用1%的二硝基氯苯丙酮溶液100μl/只均匀涂抹致敏,然后各组小鼠腹部去毛,范围为3cm×3cm,致敏当日开始腹部涂抹给药,对照组涂抹等体积的生理盐水;本发明妇炎清洗剂组和a组、b组、c组、d组分别涂抹给药均为2.0g生药/kg,连续给药12d,每日1次。末次给药后,将1%的二硝基氯苯丙酮溶液10μl/只均匀涂抹于小鼠腹部皮肤进行攻击,攻击24h后,每鼠尾静脉注射1%伊文思蓝10ml/kg,30min后处死小鼠,取下腹部蓝染皮肤,剪碎,置试管中,用1:1丙酮生理盐水混合液5ml浸泡24h,离心取上清液,用721型分光光度计610nm测定吸光度。实验结果:见表4
与对照组相比**P<0.01,*P<0.05;与本发明妇炎清洗剂组比△P<0.05。
结果表明:本发明妇炎清洗剂组和a组、b组、c组、d组可显著增强二硝基氯苯所致小鼠迟发型过敏反应,吸光度明显增加,本发明妇炎清洗剂组与对照组相比有极显著性差异(P<0.01);a组、b组、c组、d组与对照组相比较有显著性差异(P<0.05);本发明妇炎清洗剂组与a组、b组、c组、d组比较有显著性差异(P<0.05)。可见,本发明妇炎清洗剂组比a组、b组、c组、d组抗皮肤过敏的免疫作用强。
五、对小鼠细胞免疫功能的影响
实验材料
1、动物:昆明种雌性小鼠,体重23~27g。
2、药物:本发明妇炎清洗剂组和a组、b组、c组、d组。药物在实验前按含生药材量为45%的浓度配制。
实验方法
昆明种雌性小鼠60只,体重23~27g,随机分成6组,每组10只。每鼠每天肌肉注射植物血凝素10mg/kg,共注射3d,同时实验各组小鼠每日皮下注射给药,对照组注射等体积的生理盐水;本发明妇炎清洗剂组和a组、b组、c组、d组分别注射给药均为2.0g生药/kg,共8d,第8d剪尾,取血推片,瑞氏染色,油镜下计数200个淋巴细胞中过渡性细胞的数量,计算所占百分率。实验结果:见表5
与对照组相比**P<0.01;*P<0.05;与本发明妇炎清洗剂组比△P<0.05。
结果表明:本发明妇炎清洗剂组和a组、b组、c组、d组均可使小鼠对植物血凝素刺激淋巴细胞转化反应增强,过渡性细胞的百分率增大,本发明妇炎清洗剂组与对照组相比有极显著性差异(P<0.01);a组、b组、c组、d组与对照组相比较有显著性差异(P<0.05);本发明妇炎清洗剂组与a组、b组、c组、d组相比较有显著性差异(P<0.05)。可见,本发明妇炎清洗剂组比a组、b组、c组、d组的提高细胞免疫功能强。
实验结果:本发明妇炎清洗剂组和a组、b组、c组、d组可明显抑制大鼠棉球肉芽的增生;对小鼠尾根加压所致疼痛有明显的镇痛作用;能明显提高磷酸组织胺引起豚鼠瘙痒的致痒阈;可显著增强二硝基氯苯所致小鼠迟发型过敏反应,抗皮肤过敏免疫增加;可使小鼠对植物血凝素刺激淋巴细胞转化反应增强,提高细胞免疫功能。
结论:本发明妇炎清洗剂组比a组、b组、c组、d组的抗炎、镇痛、止痒、抗皮肤过敏和提高细胞免疫力等药理作用强,因此,本发明妇炎清洗剂组比a组、b组、c组、d组治疗清热燥湿、杀虫止痒的临床疗效好。
