Abstract
本发明公开了密花荚蒾(Viburnum congestum Rehd.)提取物及以其为活性成分的化妆品和其应用。属于化妆品和医药技术领域。密花荚蒾提取物为密花荚蒾枝叶的95%乙醇提取物,将该提取物分散于水中,乙酸乙酯和水两相等体积萃取得到乙酸乙酯部分和水层部分,该提取物、乙酸乙酯部分、水层部分具有良好的DPPH自由基清除能力以及酪氨酸酶活性抑制效果。在护肤方面,密花荚蒾提取物、乙酸乙酯部分、水层部分拥有很好的抗氧化能力和去黑色素能力,即具备很好的抗皱、防晒、美白或去黑色素(斑)的皮肤化妆品活性应用潜力。本发明有效利用我国的民间药用植物──密花荚蒾,得到具有显著生物活性的密花荚蒾提取物、乙酸乙酯部分、水层部分,该提取物可被用于制作良好的抗氧化剂以及酪氨酸酶活性抑制剂,是良好的天然植物化妆品原料。

技术领域
本发明属于化妆品和医药技术领域,具体涉及从一种我国的民间药用植物密花荚蒾中提取天然活性物质──密花荚蒾提取物、乙酸乙酯部分、水层部分,以其单独或组合为活性成分的化妆品或药物,以及密花荚蒾提取物或乙酸乙酯部分或水层部分在制备保湿、嫩肤、抗皱、防晒、美白等多种皮肤化妆品中的应用,还有在制备治疗或预防皮肤病的药物中的应用。
技术背景
密花荚蒾(Viburnum congestum),又名山连子(会泽)、对节疤(宣威),为忍冬科荚蒾属植物,常绿灌木,叶革质,椭圆状卵形或椭圆形,稀椭圆状矩圆形,聚伞花序小而密,花冠白色,钟状漏斗形,果实圆形,核甚扁。拔1000-2800米的谷或山坡丛林、林缘或灌丛中,产于甘肃西南部、贵州东北部及云南西北部、北部和东南部,资源丰富。
荚蒾属植物作为民间药用植物一直被应用,始载于唐《新修本草》,以其根、茎、叶、枝及果实入药,具有清热解毒、祛风除湿,健脾消食,驱虫,镇咳祛痰,养身抗衰老等作用。《本草拾遗》记载“荚蒾,灌木类。实红味甘,可食。枝、叶皆可疗疾。荚蒾枝甘苦平、无毒。功用,下气、消食、杀三虫、疗六畜疮中蛆。荚蒾叶,性质功用与枝同”。
迄今为止,现有技术中未见密花荚蒾提取物或乙酸乙酯部分或水层部分的报道,也未见其活性作用的报道。
发明内容
本发明目的在于提供密花荚蒾提取物或乙酸乙酯部分或水层部分,以其单独或以其任意组合为活性成分的化妆品,它们的制备方法,及其在制备化妆品中的应用。本发明明确了密花荚蒾提取物或乙酸乙酯部分或水层部分具有良好的护肤效果和安全性,使其成为很好的天然护肤化妆品原料。
为了实现本发明的上述目的,本发明提供了如下的技术方案:
密花荚蒾提取物、乙酸乙酯萃取物、水萃取物,其是由密花荚蒾枝叶经乙醇提取或乙酸乙酯萃取或水萃取而得的。
密花荚蒾提取物、乙酸乙酯萃取物、水萃取物,是由下述方法制备而得的:取干燥密花荚蒾枝叶,研磨粉碎,料液比3:1,95%乙醇体积比冷浸提取3次,每次24h,然后用布氏漏斗过滤,收集滤液,将滤液减压蒸馏,浓缩滤液,低温干燥,得粗提物,将粗提物经过MCI吸附脱色,95%甲醇体积比洗脱后,将洗脱液浓缩蒸干,低温冷冻干燥,得密花荚蒾提取物;
将密花荚蒾提取物溶解分散于水中,采用乙酸乙酯和水两相等体积萃取3次,回收溶剂,低温冷冻干燥,得乙酸乙酯萃取物即乙酸乙酯部分,和水萃取物即水层部分。
所述的密花荚蒾提取物或乙酸乙酯萃取物或水萃取物的制备方法,该方法包括下述步骤:取干燥密花荚蒾枝叶,研磨粉碎,料液比3:1,95%乙醇体积比冷浸提取3次,每次24h,然后用布氏漏斗过滤,收集滤液,将滤液减压蒸馏,浓缩滤液,低温干燥,得粗提物,将粗提物经过MCI吸附脱色,95%甲醇体积比洗脱后,将洗脱液浓缩蒸干,低温冷冻干燥,得密花荚蒾提取物;将密花荚蒾提取物溶解分散于水中,采用乙酸乙酯和水两相等体积萃取3次,回收溶剂,低温冷冻干燥,得乙酸乙酯萃取物即乙酸乙酯部分,和水萃取物即水层部分。

化妆品,由所述的密花荚蒾提取物或乙酸乙酯萃取物或水萃取物单独或其两种或三种的组合,和化妆品常用基质所组成。所述的基质选自一种或多种含水体系,甘油、C1-4醇、脂肪醇、多元醇、脂肪醚、脂肪酯、乙二醇、丙二醇、植物油、矿物油、脂质体、层状类脂材料、硅油、水,或者它们的组合。
药物组合物,由所述的密花荚蒾提取物或/和乙酸乙酯萃取物或/和水萃取物,和药学上可接受的载体所组成。
抗菌剂、抗炎剂、抗氧化剂、增湿剂、防腐剂、芳香剂、润湿剂、皮肤保护剂、防晒剂、表面活性剂,由所述的密花荚蒾提取物或/和乙酸乙酯萃取物或/和水萃取物,和上述制剂常规载体所组成。
本发明的密花荚蒾提取物或乙酸乙酯萃取物或水萃取物作为活性成分,和化妆品上常用基质所组成的化妆品,能抑制皮肤的酪氨酸酶活性,细胞黑色素生成抑制活性,抑制皮肤的氧化;能用于消除细纹和皱纹,减轻皮肤红疹,能用于干性、敏感肌肤,皮肤瘙痒,肤色不均或皮肤黑斑,皮肤发炎和其他与此相关的皮肤不适症。化妆品中含有密花荚蒾提取物、乙酸乙酯部分和水层部分的含量是组合物总重的0.001wt%-20.0wt%。化妆品可以是护肤液、护肤霜或乳剂等。密花荚蒾提取物可以是水溶性提取物,也可以是醇(例如甲醇、30-100%乙醇等)提取物。
本发明含有密花荚蒾提取物或乙酸乙酯萃取物或水萃取物的组合物可用于局部皮肤护理或护发产品。这些产品可以做成乳化剂(例如水包油、油包水、硅油包水、水包硅油,水包油包水,油包水包油,油包水包硅油等)霜剂,护肤液,液体(例如水剂或水-醇溶液),无水基质(例如唇膏或粉剂等),啫喱,油膏,奶液,软膏,喷雾剂,固体剂,眼霜等。
本发明的密花荚蒾提取物或乙酸乙酯萃取物或水萃取物作为活性成分用于制备抗菌剂、抗炎剂、抗氧化剂、增湿剂、防腐剂、芳香剂、润湿剂、皮肤保护剂、防晒剂、表面活性剂,或酪氨酸酶活性抑制剂,在所述的这些应用中,活性成分密花荚蒾提取物或乙酸乙酯萃取物或水萃取物的含量是上述各类制剂总重的0.001wt%-20.0wt%。
本发明中密花荚蒾提取物或乙酸乙酯萃取物或水萃取物和药学上可接受的载体所组成的药物组合物具有抗氧化、抑制酪氨酸酶活性能力。这些组合物可以用于治疗或预防与皮肤氧化、皮肤细胞中酪氨酸酶活性相关的皮肤病。在本发明中,所述药物组合物中密花荚蒾提取物或乙酸乙酯萃取物或水萃取物的质量百分含量优选为0.1~99%,更优选为0.5~90%;所述药物组合物中药用载体和/或赋形剂的总质量百分含量优选为1~99.9%,更优选为10~99.5%。
本发明通过实验提取出密花荚蒾枝叶中的有效成分,并进行细胞毒性及皮肤化妆品活性实验,证明了提取物的安全性,明确了目标提取物优良的化妆品活性,明确了密花荚蒾提取物或乙酸乙酯部分或水层部分不仅拥有良好的安全性还表现出优秀的双重皮肤化妆品活性即良好的DPPH自由基清除效果和良好的酪氨酸酶活性抑制效果,使之成为了良好的皮肤化妆品原材料来源,为密花荚蒾的开发和利用提供了更好的途径,对密花荚蒾的可持续利用具有重大意义。提供了密花荚蒾的一种新的更佳的利用类型,同时拓展了天然护肤品植物原料的来源。

具体实施方式
以下结合附图,用本发明的实施例对本发明进行更详细叙述,但并不限制本发明的范围。
实施例1
密花荚蒾提取物、乙酸乙酯萃取物(乙酸乙酯部分)、水萃取物(水层部分)的制备:
(1)称取密花荚蒾(Viburnum congestum)干燥枝叶50g,将样品研磨粉碎,料液比3:1,95%乙醇(体积比)冷浸提取3次,每次24h,然后用布氏漏斗过滤,收集滤液,将滤液用旋转蒸发仪减压蒸馏,浓缩滤液,低温干燥,得粗提物。将粗提物经过MCI吸附脱色,95%甲醇(体积比)洗脱后,将洗脱液浓缩蒸干,低温冷冻干燥,得密花荚蒾提取物4.5g,提取率为9%;
(2)将上述(1)所得密花荚蒾提取物溶解分散于水中,采用乙酸乙酯和水两相等体积萃取3次,回收溶剂,低温冷冻干燥得到乙酸乙酯萃取物(乙酸乙酯部分)2.0g和水萃取物(水层部分)2.5g。
实施例2
密花荚蒾提取物、乙酸乙酯萃取物(乙酸乙酯部分)、水萃取物(水层部分)的细胞毒性实验:通过MTS比色法,确定样品的CC50(50%cytotoxic concentration),即对50%的细胞产生毒性时的药物浓度,从而确定一下活性实验的安全样品浓度。
实验方法:96孔细胞培养板上,将B16细胞与不同浓度的待测药物混合,设定3个重复孔,同时设置不含药物的空白对照,37℃,5%CO2培养24h,采用MTS比色法检测细胞毒性,酶标仪测定OD值,测定波长490nm。计算得到CC50值。
实验结果:密花荚蒾提取物、乙酸乙酯萃取物(乙酸乙酯部分)、水萃取物(水层部分)的细胞存活率分别为95.4%。可明显看出,在细胞培养24h后,细胞数量几乎没有发生变化,说明密花荚蒾提取物、乙酸乙酯部分、水层部分对细胞没有毒害作用,作为皮肤化妆品的天然原料的安全性是有保证的。
实施例3
DPPH自由基清除实验:将待测药物与DPPH(终浓度为100μM)混合反应,设定3个重复孔,同时设置不含药物的空白对照和Trolox阳性对照,30℃,1h,酶标仪测定OD值,检测波长为515nm。计算得到抗氧化率。
实验结果:在实验中,密花荚蒾提取物体外DPPH自由基清除能力的半数抑制浓度IC50为8.76μg/mL,密花荚蒾乙酸乙酯部分体外DPPH自由基清除能力的半数抑制浓度IC50为7.85μg/mL,密花荚蒾水层部分体外DPPH自由基清除能力的半数抑制浓度IC50为13.14μg/mL。通过数据统计分析,发现密花荚蒾提取物、密花荚蒾乙酸乙酯部分和密花荚蒾水层部分均显示出浓度依赖性的DPPH清除作用,尤其是密花荚蒾乙酸乙酯部分具有较强的DPPH清除作用,其半数抑制浓度IC50为7.85μg/mL。乙酸乙酯萃取物(乙酸乙酯部分)、水萃取物(水层部分)表现出显著的DPPH自由基清除能力,并根据浓度梯度,呈现良好的量效关系(见图1),可被用作具备抗皱或防晒潜力化妆品活性良好天然植被来源。
实施例4
酪氨酸酶抑制实验:将待测药物与L-Dopa(终浓度1.25mM)混合,加入酪氨酸酶(终浓度25U/mL)开始反应,设定3个重复孔,同时设置不含药物的空白对照和Kojic Acid阳性对照,室温,5min,酶标仪测定OD值,检测波长为490nm。计算得到酪氨酸酶活性抑制率。
实验结果:在100μg/mL浓度下,3个样品均有较好的酪氨酸酶抑制活性(>30%)。乙酸乙酯萃取物(乙酸乙酯部分)、水萃取物(水层部分)酪氨酸酶活性抑制率分别为:46.7%、37.8%和32.6%,表现对酪氨酸酶良好的抑制效果(见图2)。说明乙酸乙酯萃取物(乙酸乙酯部分)、水萃取物(水层部分)具美白或去黑色素(斑)的皮肤化妆品活性应用潜力,可被作为美白或黑色素天然化妆品良好的植被原材料。
实施例5
取密花荚蒾提取物、乙酸乙酯萃取物(乙酸乙酯部分)、水萃取物(水层部分),再按下述方法制成片剂:(1)处方:实施例1所得密花荚蒾提取物或乙酸乙酯部分或水层部分单独或其任意组合作为活性成分10mg,淀粉60mg,10%淀粉浆150mg,硬脂酸镁5mg,干淀粉5mg;(2)制备方法:将密花荚蒾提取物或乙酸乙酯部分或水层部分分别单独与淀粉混合,加淀粉浆制成软材,用16目筛制粒后,置70摄氏度干燥后与16目筛整粒,加入干淀粉及硬脂酸镁混合,然后将混合物压片,每片重230mg,活性成分含量10mg。
实施例6
先按实施例1制得密花荚蒾提取物、乙酸乙酯萃取物(乙酸乙酯部分)、水萃取物(水层部分),再按下述方法制成胶囊剂:(1)处方:实施例1所得密花荚蒾提取物或乙酸乙酯部分或水层部分单独或其任意组合作为活性成分5mg,淀粉30mg,10%淀粉浆75mg,硬脂酸镁2.5mg,干淀粉2.5mg;(2)制备方法:将密花荚蒾提取物或乙酸乙酯部分或水层部分分别单独与淀粉混合,加淀粉浆制成软材,用50目筛制粒后,置80℃干燥后与16目筛整粒,加入干淀粉及硬脂酸镁混合,把得到的混合物装入硬明胶胶囊中,每粒胶囊重115mg,活性成分含量5mg。
实施例7
先按实施例1制得乙酸乙酯萃取物(乙酸乙酯部分)、水萃取物(水层部分),再按下述方法制成滴丸:(1)处方:实施例1所得密花荚蒾提取物或乙酸乙酯部分或水层部分单独或其任意组合作为活性成分0.5g,无水乙醇2.0mL,PEG-60004.5g;(2)制备方法:①密花荚蒾提取物、乙酸乙酯部分、水层部分与PEG-6000熔融液的制备:按上述处方量分别称取密花荚蒾提取物、乙酸乙酯部分、水层部分,加入适量无水乙醇,微热溶解后,加入处方量的PEG-6000熔融液中(60℃水浴保温),搅拌混合均匀,直至乙醇挥尽为止,继续静置于60℃水浴中保温30min,待气泡除尽,备用;②安装简易滴丸装置,灌入保温水浴和冷凝水浴;③将上述除尽气泡的混匀熔融液转入贮液筒内,在保温75-85℃的条件下,控制滴速,一滴滴的滴入冷凝液中,待冷凝完全,倾去冷凝液,收集滴丸,沥净并用滤纸除去滴丸上的冷凝液,放置硅胶干燥器中或自然干燥即得。
实施例8
先按实施例1制得密花荚蒾提取物、乙酸乙酯萃取物(乙酸乙酯部分)、水萃取物(水层部分),再按下述方法制成油脂性基质的软膏:(1)处方:实施例1所得密花荚蒾提取物或乙酸乙酯部分或水层部分单独作为活性成分0.5g,液体石蜡2.0g,凡士林加至10.0g;(2)制备方法:取密花荚蒾提取物或乙酸乙酯部分或水层部分分别单独置于研钵中,加入适量液体石蜡研成糊状,分次加入凡士林混合研匀即得。
实施例9
先按实施例1制得密花荚蒾提取物、乙酸乙酯萃取物(乙酸乙酯部分)、水萃取物(水层部分),再按下述方法制成O/W乳剂基质的软膏(雪花膏):(1)处方:实施例1所得密花荚蒾提取物或乙酸乙酯部分或水层部分单独作为活性成分0.5g,白凡士林1.2g,十八醇0.8g,甘油0.7g,单硬脂酸甘油酯0.2g,十二烷基硫酸钠0.1g,对羟基苯甲酸乙酯0.02g,蒸馏水加至10.0g;(2)制备方法:取白凡士林、十八醇和单硬脂酸甘油酯置于烧杯中,水浴加热至70℃~80℃使其熔化,将十二烷基硫酸钠、甘油、对羟基苯甲酸乙酯和计算量的蒸馏水置于另一个烧杯中加热至70℃~80℃使其溶解,在同温下将水液以细流加到油液中,边加边搅拌至冷凝,即得O/W乳剂基质;取密花荚蒾提取物或乙酸乙酯部分或水层部分分别单独置于研钵中,分次加入制得的O/W乳剂基质混合研匀,制成10.0g。
实施例10
先按实施例1制得密花荚蒾提取物、乙酸乙酯萃取物(乙酸乙酯部分)、水萃取物(水层部分),再按下述方法制成W/O乳剂基质的软膏(冷霜):(1)处方:实施例1所得密花荚蒾提取物或乙酸乙酯部分或水层部分单独作为活性成分0.5g,石蜡1.0g,白凡士林0.5g,司盘40 0.05g,乳化剂OP 0.05g,单硬脂酸甘油酯1.0g,液体石蜡5.0g,对羟基苯甲酸乙酯0.01g,蒸馏水10.0mL;(2)制备方法:取锉成细末的石蜡、白凡士林、单硬脂酸甘油酯、液体石蜡、司盘40、乳化剂OP和对羟基苯甲酸乙酯置于烧杯中,水浴上加热熔化并保持80℃,细流加入同温的水,边加边搅拌至冷凝,即得W/O乳剂基质;取密花荚蒾提取物或乙酸乙酯部分或水层部分分别单独置于研钵中,分次加入制得的W/O乳剂基质混合研匀,制成10.0g。
实施例11
先按实施例1制得密花荚蒾提取物、乙酸乙酯萃取物(乙酸乙酯部分)、水萃取物(水层部分),再按下述方法制成水溶性基质的软膏:(1)处方:实施例1所得密花荚蒾提取物或乙酸乙酯部分或水层部分单独作为活性成分0.5g,羧甲基纤维素钠0.6g,甘油1.0g,苯甲酸钠0.1g,蒸馏水加至8.4mL;(2)制备方法:取羧甲基纤维素钠置于研钵中,加入甘油研匀,然后边研边加入溶有苯甲酸钠的水溶液,待溶胀后研匀,即得水溶性基质;取密花荚蒾提取物或乙酸乙酯部分或水层部分分别单独置于研钵中,分次加入制得的水溶性基质混合研匀,制成10.0g。
实施例12
含有密花荚蒾提取物或/和乙酸乙酯萃取物(乙酸乙酯部分)或/和水萃取物(水层部分)(以下统称:活性成分)的美白霜配方(W%):
按常规制作化妆品的方法制得本发明上述配方的化妆品。
实施例13
含有密花荚蒾提取物或/和乙酸乙酯萃取物(乙酸乙酯部分)或/和水萃取物(水层部分)(以下统称:活性成分)的乳剂配方(W%):
按常规制作化妆品的方法制得本发明上述配方的化妆品。
实施例14
化妆水:
水相:聚丙烯酸钠0.35克,甘油4克,1,3-丁二醇2.5克,维生素B5 0.5克,熊果苷0.5克,EDTA-Na2 50mg;去离子水适量,加至100ml。制备方法:将聚丙烯酸钠0.3克溶入70ml水中,搅拌,充分溶胀。缓缓加入处方中其余成分,不停搅拌;然后加入本发明的密花荚蒾提取物或/和乙酸乙酯萃取物(乙酸乙酯部分)或/和水萃取物(水层部分)0.5克,搅拌使其成匀质,去离子水,加至100ml,包装即可。
实施例15:
修复霜:
处方:甘油6克,卡波姆1.5克,三乙醇胺1.5克,丙二醇6克,尼伯金乙酯0.2克,本发明的密花荚蒾提取物或/和乙酸乙酯萃取物(乙酸乙酯部分)或/和水萃取物(水层部分)0.5克,去离子水适量,加至总重量为100克。制备方法:取卡波姆于适量去离子水中,搅匀,静置过夜,使其充分溶胀后,加入甘油后,用三乙胺调节pH值,使凝胶基质粘稠性增加。另取处方量的密花荚蒾提取物或/和乙酸乙酯萃取物(乙酸乙酯部分)或/和水萃取物(水层部分)、丙二醇及去离子水,混合均匀。再将卡波姆凝胶例入,加入0.5%精油,尼伯金乙酯,合并搅拌至均匀细腻,加蒸镏水至足量,研磨均匀,即可。
Claims (11)
1.密花荚蒾提取物、乙酸乙酯萃取物、水萃取物,由密花荚蒾枝叶经乙醇提取或乙酸乙酯萃取或水萃取而得。
2.密花荚蒾提取物、乙酸乙酯萃取物、水萃取物,其特征在于,所述的密花荚蒾提取物、乙酸乙酯萃取物、水萃取物是由下述方法制备而得的:
取干燥密花荚蒾枝叶,研磨粉碎,料液比3:1,95%乙醇体积比冷浸提取3次,每次24 h,然后用布氏漏斗过滤,收集滤液,将滤液减压蒸馏,浓缩滤液,低温干燥,得粗提物,将粗提物经过MCI吸附脱色,95%甲醇体积比洗脱后,将洗脱液浓缩蒸干,低温冷冻干燥,得密花荚蒾提取物;
将密花荚蒾提取物溶解分散于水中,采用乙酸乙酯和水两相等体积萃取3次,回收溶剂,低温冷冻干燥,得乙酸乙酯萃取物即乙酸乙酯部分,和水萃取物即水层部分。
3.权利要求1或2所述的密花荚蒾提取物或乙酸乙酯萃取物或水萃取物的制备方法,其特征在于,该方法包括下述步骤:取干燥密花荚蒾枝叶,研磨粉碎,料液比3:1,95%乙醇体积比冷浸提取3次,每次24 h,然后用布氏漏斗过滤,收集滤液,将滤液减压蒸馏,浓缩滤液,低温干燥,得粗提物,将粗提物经过MCI吸附脱色,95%甲醇体积比洗脱后,将洗脱液浓缩蒸干,低温冷冻干燥,得密花荚蒾提取物;将密花荚蒾提取物溶解分散于水中,采用乙酸乙酯和水两相等体积萃取3次,回收溶剂,低温冷冻干燥,得乙酸乙酯萃取物即乙酸乙酯部分,和水萃取物即水层部分。
4.权利要求1或2所述的密花荚蒾提取物或乙酸乙酯萃取物或水萃取物在制备化妆品中的应用。
5.权利要求1或2所述的密花荚蒾提取物或乙酸乙酯萃取物或水萃取物在制备保湿或嫩肤或抗皱或防晒或美白或抗氧化的化妆品中的应用,以及在制备改善皮肤代谢功能或皮肤障碍功能的化妆品中的应用。
6.权利要求1或2所述的密花荚蒾提取物或乙酸乙酯萃取物或水萃取物在制备抗菌剂、抗炎剂、抗氧化剂、增湿剂、防腐剂、芳香剂、润湿剂、皮肤保护剂、防晒剂、表面活性剂中的应用。
7.权利要求1或2所述的密花荚蒾提取物或乙酸乙酯萃取物或水萃取物在制备酪氨酸酶活性抑制剂中的应用。
8.权利要求1或2所述的密花荚蒾提取物或乙酸乙酯萃取物或水萃取物在制备预防或治疗皮肤病的药物中的应用。
9.化妆品,由权利要求1或2所述的密花荚蒾提取物或乙酸乙酯萃取物或水萃取物单独或其两种或三种的组合,和化妆品常用基质所组成。
10.药物组合物,由权利要求1或2所述的密花荚蒾提取物或/和乙酸乙酯萃取物或/和水萃取物,和药学上可接受的载体所组成。
11.抗菌剂、抗炎剂、抗氧化剂、增湿剂、防腐剂、芳香剂、润湿剂、皮肤保护剂、防晒剂、表面活性剂,由权利要求1或2所述的密花荚蒾提取物或/和乙酸乙酯萃取物或/和水萃取物,和上述制剂常规载体所组成。
