(2) adding ethanol into the thick polygonatum paste to ensure that the ethanol content is 50-90 percent, standing for 2-6 hours, taking supernatant, and concentrating the supernatant into polygonatum alcohol precipitation paste under reduced pressure;
(4) Preparing a nutrient solution:
(1) mixing deer serum concentrate, cornu Cervi Pantotrichum extract concentrate and rhizoma Polygonati ethanol precipitation extract, standing at 5 deg.C, refrigerating for 24 hr, collecting supernatant, centrifuging with 100 mesh sieve, and filtering to obtain supernatant;
(2) adding honey, potassium sorbate and purified water into the centrifuged supernatant, filtering, filling and sterilizing to obtain the finished product of the deer serum rhizoma polygonati nutrient solution.
Preferably, in the step (1), 0.9 mass% NaCl eluent is used for elution, a biuret reagent is used for detecting elution effluent liquid of the collected deer serum, the biuret reagent is added into the sample to show purple color and indicate that protein is eluted, the effluent liquid is continuously collected, the biuret reagent is added into the sample to show colorless color and indicate that no protein is eluted, and the collection is stopped.
Preferably, in step (1) (2), ultrafiltration is carried out by using a molecular membrane filter having a molecular weight of 50000 to 60000D.
Compared with the prior art, the invention has the beneficial effects that:
the invention adopts a chromatographic column purification technology and an ultrasonic cracking technology to purify and produce deer serum, deer antler extract, deer antler insulin growth factor and sealwort extract, and prepares the deer serum and sealwort nutrient solution which takes deer serum as a main component and has multiple nutrition and health care effects by taking the deer serum, the deer antler extract, the deer insulin growth factor and the sealwort extract as main raw materials and reasonably combining and complementing the three main raw materials.
The extracting solution prepared by the preparation method is filtered, precipitated, clarified and concentrated through multiple links, and is added with honey, potassium sorbate and a proper amount of purified water, so that the product has stable quality, no precipitate, safe eating and good taste, and is suitable for people with low immunity and easy fatigue.
Detailed Description
In order to make the technical solution of the present invention clearer, the present invention is further described in detail by the following specific examples.
Example 1:
a preparation method of a deer serum rhizoma polygonati nutrient solution comprises the following steps:
(1) And (3) purifying deer serum:
(1) adding 250 kg of red deer serum into a sephadex G-25 chromatographic column in multiple batches according to the capacity of the chromatographic column, eluting with 0.9% NaCl eluent, and collecting red deer blood washing eluate;

(3) Extracting rhizoma polygonati:
(1) reflux-extracting 120 kg rhizoma Polygonati decoction pieces with water for three times, adding 3.5 times of water for the first time, and extracting at 80 + -2 deg.C for 3 hr, adding 3.5 times of water for the second time, and extracting at 80 + -2 deg.C for 2 hr, adding 2.5 times of water for the third time, and extracting at 80 + -2 deg.C for 1 hr, mixing the three extractive solutions, and concentrating under reduced pressure to obtain rhizoma Polygonati soft extract;
(2) adding ethanol into the rhizoma Polygonati soft extract to make ethanol content 70%, standing for 4 hr, collecting supernatant, and concentrating under reduced pressure to get rhizoma Polygonati ethanol precipitation extract;
(4) Preparing a nutrient solution:
(1) mixing the red deer serum concentrated solution, the red deer antler extract concentrated solution and the sealwort alcohol precipitation paste, standing, refrigerating and precipitating for 24 hours at the temperature of 5 ℃, and performing 100-mesh centrifugal filtration on the supernatant to obtain centrifugal supernatant;
(2) and adding 75 kg of honey and 1 kg of potassium sorbate into the centrifuged supernatant, adding purified water to a constant volume of 1000L, filtering, filling and sterilizing to obtain the finished product of the deer serum rhizoma polygonati nutrient solution.
Example 2:
a preparation method of a deer serum rhizoma polygonati nutrient solution comprises the following steps:
(1) And (3) purifying deer serum:
(1) adding 200 kg of red deer serum into a sephadex G-25 chromatographic column in multiple batches according to the capacity of the chromatographic column, eluting with 0.9% NaCl eluent, and collecting red deer blood washing eluate;
(2) cleaning and removing blood from Cervus Elaphus L, ultrafiltering with 50000D molecular membrane filter to obtain purified Cervus Elaphus L blood concentrated solution;
(2) Extracting pilose antler:
(1) chopping 30 kg of hairy antler and grinding the chopped hairy antler into hairy antler slurry by using a colloid mill;
(2) adding ethanol into an ultrasonic extraction tank according to the mass ratio of the material liquid to the material liquid of 1.8 for extraction, wherein the extraction time is 35 minutes;
(3) starting an ultrasonic generator to perform wall breaking treatment, wherein the ultrasonic frequency is 26MHZ, the ultrasonic intensity is 80W, and the ultrasonic treatment time is 120 minutes;
(4) filtering by hollow fiber ultrafilter with pore diameter of 80nm, and separating to remove solid residue to obtain cornu Cervi Pantotrichum extractive filtrate;
(5) putting the cornu Cervi Pantotrichum leaching filtrate into a reduced pressure distillation tank, recovering extraction medium, and making into cornu Cervi Pantotrichum extract concentrate;
(3) Extracting rhizoma polygonati:
(1) adding water into 100 kg of rhizoma polygonati decoction pieces, carrying out reflux extraction for three times, adding 3 times of water for carrying out reflux extraction for 2.5 hours at 77 +/-2 ℃ for the first time, adding 3 times of water for carrying out reflux extraction for 1.5 hours at 77 +/-2 ℃ for the second time, adding 2 times of water for carrying out reflux extraction for 0.5 hour at 77 +/-2 ℃ for the third time, combining three extracting solutions, and concentrating under reduced pressure to obtain rhizoma polygonati thick paste;
(2) adding ethanol into the rhizoma Polygonati soft extract to make ethanol content 50%, standing for 3 hr, collecting supernatant, and concentrating under reduced pressure to obtain rhizoma Polygonati ethanol precipitation extract;
(4) Preparing a nutrient solution:
(1) mixing the red deer serum concentrated solution, the red deer antler extract concentrated solution and the rhizoma polygonati alcohol precipitation paste, standing, refrigerating and precipitating for 24 hours at the temperature of 5 ℃, and performing 100-mesh centrifugal filtration on supernate to obtain centrifugal supernate;
(2) adding 70 kg of honey and 1 kg of potassium sorbate into the centrifuged supernatant, adding purified water to a constant volume of 1000L, filtering, filling and sterilizing to obtain the finished product of the deer serum rhizoma polygonati nutrient solution.
Example 3:
a preparation method of a deer serum rhizoma polygonati nutrient solution comprises the following steps:
(1) And (3) purifying deer serum:
(1) adding 300 kg of sika deer serum into a sephadex G-25 chromatographic column in multiple batches according to the capacity of the chromatographic column, eluting with a NaCl eluent with the mass concentration of 0.9%, and collecting sika deer serum eluent;
(2) ultrafiltering the sika deer serum eluate with molecular membrane filter with molecular weight of 60000D to obtain purified sika deer serum concentrated solution;
(2) Extracting pilose antler:
(1) chopping 20 kg of spotted deer antler and grinding into spotted deer antler slurry by using a colloid mill;
(2) adding ethanol into an ultrasonic leaching tank according to the mass ratio of the material liquid to the material liquid of 1.7 for leaching for 45 minutes;
(3) starting an ultrasonic generator to perform wall breaking treatment, wherein the ultrasonic frequency is 30MHZ, the ultrasonic intensity is 120W, and the ultrasonic treatment time is 90 minutes;
(4) filtering the wall-broken treatment by hollow fiber ultrafilter with aperture of 30nm, and separating to remove solid residue to obtain filtrate;
(5) putting the extracting filtrate of the spotted deer antler into a reduced pressure distillation tank, and recovering an extracting medium to prepare a concentrated solution of the spotted deer antler extract;
(3) Extracting rhizoma polygonati:
(1) adding 150 kg of sealwort decoction pieces into water for reflux extraction for three times, adding 4 times of water for reflux extraction at 83 +/-2 ℃ for 3.5 hours for the first time, adding 4 times of water for reflux extraction at 83 +/-2 ℃ for 2.5 hours for the second time, adding 3 times of water for reflux extraction at 83 +/-2 ℃ for 1.5 hours for the third time, combining extracting solutions obtained by the three times, and concentrating under reduced pressure to obtain sealwort thick paste;
(2) adding ethanol into the rhizoma Polygonati soft extract to make ethanol content 90%, standing for 5 hr, collecting supernatant, and concentrating under reduced pressure to obtain rhizoma Polygonati ethanol precipitation extract;
(4) Preparing a nutrient solution:
(1) mixing the concentrated solution of Cervus Nippon Temminck blood serum, the concentrated solution of Cervus Nippon Temminck extract and rhizoma Polygonati ethanol precipitation paste, standing at 5 deg.C, refrigerating for precipitation for 24 hr, and centrifuging the supernatant with 100 mesh to obtain centrifugal supernatant;
(2) adding 80 kg of honey and 1 kg of potassium sorbate into the centrifuged supernatant, adding purified water to a constant volume of 1000L, filtering, filling and sterilizing to obtain the finished product of the deer serum rhizoma polygonati nutrient solution.
Claims (6)
1. A deer serum rhizoma polygonati nutrient solution is prepared from the following raw materials in parts by weight: 20 to 30 parts of deer serum, 2 to 3 parts of pilose antler, 10 to 15 parts of rhizoma polygonati, 6 to 9 parts of honey, 0.08 to 0.1 part of potassium sorbate and a proper amount of purified water.

The deer serum rhizoma polygonati nutrient solution according to claim 1, which is characterized in that: the traditional Chinese medicine is prepared from the following raw materials in parts by weight: 22 to 28 parts of deer serum, 2.2 to 2.8 parts of pilose antler, 12 to 13 parts of rhizoma polygonati, 7 to 8 parts of honey, 0.1 part of potassium sorbate and a proper amount of purified water.
3. The deer serum rhizoma polygonati nutrient solution according to claim 2, which is characterized in that: the deer serum is red deer serum or sika deer serum, and the deer antler is red deer antler or sika deer antler.

(4) filtering the obtained product by a hollow fiber ultrafilter with the aperture of 2-100 nm, and separating to remove solid residue to obtain cornu Cervi Pantotrichum extract filtrate;
(5) placing the cornu Cervi Pantotrichum leaching filtrate into a reduced pressure distillation tank, recovering extraction medium to obtain cornu Cervi Pantotrichum extract concentrate;
(3) Extracting rhizoma polygonati:
(1) adding water into rhizoma polygonati decoction pieces for reflux extraction for three times, adding 3-4 times of water for 75-85 ℃ for reflux extraction for 2.5-3.5 hours for the first time, adding 3-4 times of water for reflux extraction for 1.5-2.5 hours for the second time at 75-85 ℃, adding 2-3 times of water for reflux extraction for 0.5-1.5 hours for the third time at 75-85 ℃, combining extracting solutions obtained in three times, and concentrating under reduced pressure to obtain rhizoma polygonati thick paste;
(2) adding ethanol into the thick polygonatum paste to ensure that the ethanol content is 50-90 percent, standing for 2-6 hours, taking supernatant, and concentrating the supernatant into polygonatum alcohol precipitation paste under reduced pressure;
(4) Preparing a nutrient solution:
(1) mixing the deer serum concentrated solution, the pilose antler extract concentrated solution and the sealwort alcohol precipitation paste, standing, refrigerating and precipitating for 24 hours at the temperature of 5 ℃, taking the supernatant, and performing 100-mesh centrifugal filtration to obtain centrifugal supernatant;
(2) adding honey, potassium sorbate and purified water into the centrifuged supernatant, filtering, filling and sterilizing to obtain the finished product of the deer serum rhizoma polygonati nutrient solution.
