Description

Multi-effect oil control composition, essence preparation, and preparation methods and applications thereof
Technical Field
The invention relates to the technical field of A61K8/00, in particular to a multi-effect oil control composition, an essence preparation, a preparation method and application thereof.
Background
With the development of social economy, consumers pay attention to their own mental and external images, so that skin care products with various effects are also required in life. The oil control products are concerned by consumers, and the sebum ingredients in the skin are relatively complex, mainly contain triglyceride and free fatty acid wax, and are mixed with phospholipid and cholesterol ester generated after epithelial cell metabolism. Sebum is oily, has moistening effect on skin, and can prevent skin dryness and chapping; the fatty acid in sebum can kill bacteria and resist mold and virus, and is called as a natural barrier of skin; however, acne, pimple, seborrheic dermatitis, dandruff, etc. may be formed when sebum excessively overflows. In general, facial seborrhea actually means that the skin is subjected to IGF-1 signal increase, sebaceous gland secretion function is high, so that skin microecological confusion and skin internal inflammation outbreak are caused, and further the problems of inflammatory acne, closed mouth, large pores and the like are caused, and severe cases even develop into seborrheic dermatitis.
The Chinese patent with the patent application number of CN201811596142.3 discloses an oil control pore-shrinking essence, which adopts Hamamelis virginiana extract, red clover flower extract and yarrow extract to be matched for solving the problems of controlling oil secretion and shrinking pores, has the function of controlling oil secretion, but has different adopted extracts and the invention conception of solving the problems, and also has no verification of antibacterial effect.
The present invention considers that skin problems such as acne, pore enlargement, seborrheic dermatitis and the like caused by seborrhea are the results of multiple aspects of compound actions, and single-point breakthrough is insufficient to solve the problems. Therefore, providing an oil control product that can combine the mechanism of facial grease spillover and the various skin problems that result, and that is a multi-target combined solution, is a major technical problem that needs to be addressed at present.

When sebum is excessively secreted by a human body, propionibacterium acnes and malassezia are greatly planted in the skin, the skin micro-ecological environment is broken, and skin inflammation is induced; skin diseases that develop inflammatory acne and seborrheic dermatitis further develop after inducing skin inflammation. The inventor found in experiments that when the citrus fruit extract and the olive fruit extract are contained in the raw materials for preparing the multi-effect oil control composition, the weight ratio of the citrus fruit extract to the olive fruit extract is (1-10): 1, can effectively inhibit the excessive proliferation of propionibacterium acnes and can also limit the proliferation of harmful bacteria such as malassezia, probably because the citrus fruit extract contains a certain amount of active ingredients such as flavone and alkaloid, can inhibit TRPV1 activation and reduce calcium ion inflow, has the effects of relieving acne, relieving redness and swelling caused by seborrhea, relieving pain and the like, and the citrus fruit extract and the olive fruit extract are prepared according to the following formula (1-10): after the weight ratio of 1 is matched, the inventor finds that the active ingredients such as oleuropein, acteoside and the like contained in the olive fruit extract can further reduce the oxidation pressure of skin grease, and can achieve the effects of regulating the inflammatory environment in the skin together and reducing inflammatory reaction and allergic discomfort; when the olive fruit extract or the citrus fruit extract is not used in the preparation raw materials, or when the mixing amount of the olive fruit extract or the citrus fruit extract is too large or too small, the effective antibacterial effect cannot be achieved, and particularly the effect of inhibiting propionibacterium acnes is poor.
The second aspect of the invention provides an essence preparation, which comprises the following preparation raw materials, by mass, 0.5-11% of a multi-effect oil control composition, a component A and a component B; the A component comprises a thickener and a humectant, and the B component comprises a skin conditioning agent.

The multi-effect oil control composition contains ionic components, and the vegetable flavone or other active components contained in the used vegetable extract have the problem of poor water solubility. In order to improve the oil control and anti-seborrheic property, antibacterial property and good moisturizing effect of the essence preparation, the inventor generally adds PCA zinc, but the PCA zinc releases zinc ions, so that the conventional thickener can generate destructive effects such as precipitation and flocculation, and serious consequences such as precipitation, composition precipitation and layering can be directly generated on the essence preparation containing the multi-effect oil control composition, so that the normal stability of the essence preparation is affected. The general conventional thickener is used singly and compounded, and can not meet the requirement of the stability of the multi-effect oil control composition, but can not meet the requirement of the essence preparation of the application on the compatibility of PCA zinc, so that the stability of the multi-effect oil control composition in the essence preparation system needs to be met, and the good compatibility of the multi-effect oil control composition on the PCA zinc becomes a technical problem in the industry.
In order to solve the technical problems that the multi-effect oil control composition has good stability in an essence preparation system and provides good compatibility for PCA zinc, in particular to improve the low-temperature stability of an essence preparation, the inventor finds that the stability requirement of the multi-effect oil control composition in the essence preparation system can be improved by singly or compositely adopting carbomer, sodium polyacrylate, acrylic acid (ester) or C10-30 alkanol acrylate crosslinked polymer, hydroxyethyl cellulose and xanthan gum, but the problem that the compatibility of PCA zinc in the essence preparation system is poor can not be solved, and the inventor particularly finds that the combination of the carbomer fermentation product extract, the xanthan gum and the polyacrylate crosslinked polymer-6 is adopted in experiments, wherein the weight ratio of the carbomer fermentation product extract to the xanthan gum to the polyacrylate crosslinked polymer-6 is 1: (1-3): (2-4), in particular, the weight ratio of the Sphingomonas fermentation product extract, the xanthan gum and the polyacrylate crosslinked polymer-6 is 1:2:3.5, the problem that the multi-effect oil control composition has good stability in an essence preparation system and the compatibility of PCA zinc in the essence preparation system is poor can be solved, and particularly the low-temperature stability of the essence preparation can be improved, and the possible reasons are that the polyacrylate crosslinked polymer-6 and xanthan gum adopted in the essence preparation system have a chain segment structure with a certain length, and after the polyacrylate crosslinked polymer-6 and the xanthan gum are matched with a Sphingomonas fermentation product extract, an interactive network structure can be formed, the multi-effect oil control composition can be stabilized in the interactive network structure, and zinc ions released by PCA zinc and PCA zinc can be wrapped, so that the stability of the essence preparation system is enhanced, and the problems of precipitation, composition precipitation and layering can not occur.
In one embodiment of the invention, the weight ratio of the A component to the B component is (18-46): 1.
In one embodiment of the invention, the a component comprises one or more of water, glycerol polyether-26, poloxamer 407, butylene glycol, 1, 2-pentanediol, glycerol, 1, 2-hexanediol, phytosphingosine, polydimethylsiloxane, caprylic/capric triglyceride, sphingomonas fermentation product extract, polyacrylate cross-linked polymer-6, xanthan gum, p-hydroxyacetophenone, sodium citrate, carbomer, sodium polyacrylate, acrylic acid (esters) type/C10-30 alkanol acrylate cross-linked polymer, hydroxyethyl cellulose.
In a preferred embodiment of the present invention, the A-component comprises, in mass percent, one or more of 50.0-86.0% water, 1.0-5.0% glycerol polyether-26, 0.1-1.0% poloxamer 407, 1.0-5.0% butylene glycol, 1.0-3.0%1, 2-pentanediol, 1.0-5.0% glycerol, 0.1-1.0%1, 2-hexanediol, 0.01-0.2% phytosphingosine, 1.0-5.0% polydimethylsiloxane, 1.0-5.0% caprylic/capric triglyceride, 0.01-1.0% Sphingomonas fermentation product extract, 0.1-1.0% polyacrylate crosslinked polymer-6, 0.1-0.5% xanthan gum, 0.1-0.5% p-hydroxyacetophenone, 0.01-0.2% sodium citrate.
In one embodiment of the application, the B component comprises a combination of one or more of zinc PCA, panthenol, acetylglucosamine, bisabolol, eucalyptus globulus oil, allantoin.
In a preferred embodiment of the present application, the B component comprises, in mass percent, one or more of zinc PCA 0.1-1.0%, panthenol 0.1-3.0%, acetylglucosamine 0.1-1%, salicylic acid 0.05-1%, bisabolol 0.1-0.2%, eucalyptus oil 0.01-0.2%, and allantoin 0.1-0.2%.
The inventor finds that precipitation, composition precipitation and layering of the essence preparation can occur at low temperature in experiments, and in order to improve the low-temperature stability of the essence preparation, the inventor finds that the low-temperature stability of the essence preparation can be improved by matching the component A with the component B, wherein the weight ratio of the component A to the component B is (18-46): 1, the low-temperature resistance of a raw material system of the essence preparation is poor when the component A and the component B are not used in a specific dosage, and the precipitation problem can occur at low temperature, and the possible reason is that the raw material system of the essence preparation is changed due to the fact that the different dosage of the component A and the component B is needed in the application, so that the stability of the multi-effect oil control composition in the essence preparation system is affected.
The third aspect of the invention provides a preparation method of an essence preparation, comprising the following steps:

TABLE 2
As can be seen from the test results in table 2, the low temperature stability of the multi-effect oil control composition in the essence formulation can be improved after the component a and the component B are used together according to the limited use ratio.
3. Lipid drop test experiment
Sebaceous cells synthesize large amounts of lipids and secrete them out of the cell in the form of lipid droplets, affecting cellular function.
The test method comprises the following steps: the essence preparation prepared in examples 1-4 was screened out by cytotoxicity test to a safe concentration of 0.000125%, an in vitro oil secretion model was established based on immortalized human sebaceous gland cell SZ95, and after lipid droplets were formed, the sebum secretion inhibition ability of the test compound was evaluated by analyzing the oil secretion amount after administration treatment using oil red O staining.
And (3) result judgment: the oil control effect of the essence preparation is verified by analyzing the significance of the relative IOD value before and after administration. Statistical analysis was performed in t-test, with significance expressed as x, P-value <0.05 as x, and P-value <0.01 as x, for the sample and blank groups. The inhibition ratios of examples 1, 2 and 3 relative to example 4 were evaluated by using example 4 as a standard, and the test results are shown in table 3, fig. 1 and fig. 2.
TABLE 3 Table 3
As can be seen from the results of Table 3 and FIG. 1, the in vitro ability of the present invention to inhibit secretion of oil from sebaceous gland cells is weaker, while the effects of examples 1 to 3 are more remarkable than those of example 4, showing a great difference, and having a good in vitro ability to inhibit secretion of oil from sebaceous gland cells. The results show that the scientific compounding of the citrus fruit extract, the olive fruit extract and the linseed extract in the multi-effect oil control composition can obviously increase the capacity of the lotus seed extract, the lotus extract/the Egypt blue water lily flower extract for inhibiting the secretion of grease by sebaceous gland cells.
As can be seen from fig. 2, examples 1 to 3 are different in the ability to inhibit the secretion of grease by sebaceous gland cells from example 4, and examples 1 and 3 are significantly stronger than example 2 by the difference in the ratio of citrus fruit extract, olive fruit extract, flaxseed extract, and the inhibition ability is significantly different from that of citrus fruit extract, olive fruit extract, flaxseed extract, and the inhibition ability is different from that of the amount of citrus fruit extract, olive fruit extract, flaxseed extract.
4. Propionibacterium acnes inhibition experiments
The test method comprises the following steps: the test uses a minimum inhibitory concentration test, a pre-added bacteria liquid pouring plate method and a punching method to measure the diameter of a bacteriostasis ring on a bacteria plate to evaluate the inhibition of the sample on acne Capacity of propionibacteria. Injecting activated bacterial liquid into solid culture medium cooled at about 60deg.C to obtain final concentration of 0.5-1.0X10 7 CFU/ml, pour the plate after mixing well, horizontally rest. Punching holes on the prepared fungus flat plate by using a stainless steel round tube, 1-4 holes per dish, and carefully picking out small pieces of culture medium in the holes. After 100 μl of the sample to be tested was added to each well, the sample was allowed to diffuse at 4℃for 2 hours, and then anaerobic culture was carried out in an incubator at 37℃for 72 hours, the evaluation criteria are shown in Table 4, and the test results are shown in Table 5.
Test sample: the concentrate formulations prepared in examples 1-4 were tested at 100% concentration with at least 3 duplicate wells per concentrate example.
TABLE 4 Table 4
TABLE 5
From the results in table 5, it can be seen that example 1 and example 2 have inhibitory effects on propionibacterium acnes, both of which are similar in inhibitory ability and are moderately sensitive (++); examples 3 and 4 did not inhibit propionibacterium acnes, demonstrating that none of the essence formulations prepared when the multi-effect oil control composition of the present invention lacked olive fruit extract or citrus fruit extract achieved bacteriostatic effect; meanwhile, the implementation 3 also shows that even though the flaxseed extract has theoretical support for promoting antibacterial peptide, the flaxseed extract has no effect of actually inhibiting propionibacterium acnes; the multiple-effect oil control composition can achieve synergistic effect only when the components of the multiple-effect oil control composition are matched according to a specific proportion, and plays a role in inhibiting Propionibacterium acnes.
5. Test of the subject:
