Description

Composition for treating acne, external gel and preparation method thereof
Technical Field
The invention belongs to the technical field of medicines, relates to a composition for treating acne, and also relates to an external gel of the composition and a preparation method of the external gel.
Background
Acne, commonly known as whelk and comedo, is a common disease in dermatology, and about 85% of people in the adolescence suffer from acne with different degrees, which affects the appearance of patients on one hand and even seriously affects the psychology on the other hand. Modern research has shown that acne is caused by a variety of causes including sex hormone secretion, microbial infection, inflammatory lesions, and immune disorders. The treatment scheme mainly adopts oral administration and local administration, wherein the local administration mainly adopts tretinoin medicaments and antibacterial medicaments, and although the skin injury of a patient is obviously reduced, the problems of skin irritation and drug resistance exist.
The natural components from the traditional Chinese medicine have the dual advantages of both efficacy and safety. The peony seed oil is vegetable oil obtained by processing oil peony seeds, the oil yield is 27-32%, the proportion of unsaturated fatty acid in the total oil is 82.81-93.23%, the content of alpha-linolenic acid (32-67%) is obviously higher than that of other common edible vegetable oil, and in addition, the peony seed oil contains active ingredients such as oleanolic acid, trans-resveratrol, paeoniflorin, vitamin E and the like, is a natural vegetable oil resource and is approved by the state as a new resource food. Modern researches show that the peony seed oil has the effects of sun screening, whitening, radiation resistance, oxidation resistance and inflammation resistance besides unique nutrition and health care functions, and the physiological activity of the peony seed oil provides a basis for the research and development of external preparations of the peony seed oil. The bletilla striata polysaccharide is a water-soluble high-molecular polysaccharide extracted, separated and purified from bletilla striata, has pharmacological activities such as anti-inflammation, tissue repair and the like, is safe and non-toxic, and has good biological adhesiveness. It has been used in the fields of medicine and cosmetics because of its pharmacological activity and adjuvant properties. Aloe polysaccharide is the main active component of aloe, and has physiological activities of resisting bacteria, inflammation, oxidation and radiation, whitening and moisturizing, etc., and has been widely used in the cosmetic industry due to its unique moisturizing property. Based on the research, the peony seed oil, the bletilla striata polysaccharide and the bletilla striata polysaccharide are selected to be compatible for use, and on one hand, the three components have synergistic effect in whitening, anti-inflammation and antioxidation aspects; on the other hand, the three have the property of auxiliary materials in preparation of the ring joint.
The nano-emulsion gel is a transparent and stable colloid with a reticular structure formed by mixing the medicament, proper oil, emulsifier assistant emulsifier and water according to a certain proportion to prepare nano-emulsion and then adding water-soluble high polymer materials such as carbomer and the like. The nano-emulsion and the gel have the advantages of both nano-emulsion and gel, have the characteristics of high transdermal efficiency and delayed release, are suitable for topical treatment of skin, and can be used as a novel transdermal drug delivery carrier of drugs.

step 1, weighing the following components in percentage by mass: 49% of composition for treating acne, 30% of PEG-40 hydrogenated castor oil, 4006% of polyethylene glycol and 15% of carbomer, wherein the sum of the mass percentages of the components is 100%. And (5) standby.
And 2, uniformly mixing the PEG-40 hydrogenated castor oil and the polyethylene glycol 400 in a constant-temperature magnetic stirrer at the rotating speed of 27r/min for 4min, and marking as a material I.
And 3, dropwise adding the peony seed oil into the material I, and recording as a material II when the rotating speed of the constant-temperature magnetic stirrer is 27 r/min.
And 4, dropwise adding the purified water into the material II according to the mass volume ratio of the material II to the purified water of 1:35g/ml, and marking as a material III.
And 5, emulsifying the material III in a high-shear homogenizing and emulsifying machine for 27min at the rotating speed of 9700r/min, and recording as a material IV.
And 6, homogenizing the material IV for 5 times by a high-pressure homogenizer, wherein the homogenizing pressure is 830bar, and marking as a material V.
And 7, placing the material V in ultrasonic treatment for 22min, wherein the ultrasonic power is 270W, and marking as a material VI.
And 8, adding bletilla striata polysaccharide, aloe polysaccharide and carbomer into purified water, heating to 82 ℃ to react for 4.5h, and then adjusting the pH to 7.5 by using 21% triethanolamine to obtain a material VII, wherein the mass volume ratio of the bletilla striata polysaccharide to the aloe polysaccharide to the carbomer to the purified water is 1:95 g/ml.
And 9, slowly dripping the material VI into the material VII, homogenizing for 3 times by a high-pressure homogenizer at the homogenizing pressure of 450bar after the rotating speed of the constant-temperature magnetic stirrer is 27r/min, and obtaining the nanoemulsion gel.
