Abstract
本发明涉及一种宁心安神的中药组合物,它是由以下重量份的原料药制成:合欢花1‑5份、龙眼肉2‑8份、大枣3‑8份、玫瑰花1‑5份、百合1‑8份。本发明还提供了上述中药组合物在制备治疗心神失养型失眠的茶饮、食品、保健品或药物中的应用。该药物由纯中药制成,基于充足的中医及西医理论基础之上,对于心神失养并不仅是针对于症的描述而进行诊疗,更多的是关注整个疾病的发病背景及其病因病机,深入其根本,疗效明显,无毒副作用,易于被患者接受,且原料组分少,丰富易得,价格便宜,制备工艺简单,适合推广使用。

技术领域
本发明涉及中药技术领域,具体地说,是关于一种宁心安神中药茶饮及其应用。
背景技术
心血不足是导致失眠的证型之一。失眠症(insomnia)是以人睡和(或)睡眠维持困难所致的睡眠质量或数量达不到正常生理需求而影响白天社会功能的一种主观体验,是最常见的睡眠障碍性疾患。在社会节奏加快和竞争加剧的今天,失眠是一种十分普遍的现象。失眠症可造成注意力不集中、记忆力减退、判断力和日常工作能力下降,严重者合并焦虑、强迫和抑郁等症。心神失养通常可由心血虚、心阴虚、心气虚导致,病因不同,所表现的症状也有所不同。心主藏神,即在接受外界客观事物的各种刺激时,心有可以作出反应并协调五脏作出意识、思维、情感等活动的机能。人体受到情志所伤时,首伤心神,其次伤及相应脏腑,继而导致脏腑气机紊乱,就会导致心神失养。
1、心血虚:通常由于劳神过度、失血过多、久病等情况导致,也可由脾失健运、肾精亏虚,使肾血不足所致。因心血虚导致的心神失养,患者会出现心悸、失眠多梦、健忘、头晕、眼花,以及面色淡白或萎黄、唇舌色淡、脉细无力等症状;
2、心阴虚:通常由思虑劳神太过而暗耗心阴,或温热火邪灼伤心阴,以及肝肾阴亏无法上济而累及心阴等情况所致。因心阴虚导致的心神失养,患者会出现心悸、心烦、失眠多梦、口燥咽干、形体消瘦、两颧潮红,或手足心热、潮热盗汗、舌红少苔、脉细数等症状;
3、心气虚:通常由素体虚弱、久病失养、劳倦过度,或先天不足、年事高引起气衰等原因所致。因心气虚导致的心神失养,患者会出现心悸怔忡、气短胸闷、精神疲倦、自汗且动则诸症加剧,以及面色淡白、舌淡、脉虚等症状。目前治疗心血不足,心神失养的常用中成药包括,归脾汤、八珍益母颗粒归脾丸、当归补血汤、乌鸡白凤丸等。
西医治疗主要以失眠为主,常用的安眠药是安定类药物,如短效阿普唑仑、中效的安定类和舒乐安定,以及长效的氯硝安定;对于大多数患者来说,西药虽能改善但不能完全控制。对于部分难治性患者疗效有限,停药后易复发。
中国专利文献CN:201410676614.1,公告日2015年1月28日,公开了一种治疗产后抑郁症的中药组合物由以下重量份的原料药组成:广枣8-10份、蓝布正5-10份、龙眼肉3-5份、大枣3-5份、当归4-6份、百合1-5份、合欢花2-3份、钩藤2-3份、玫瑰花1-3份、牛膝2-5份、竹茹2-5份、乳香2-3份、枳实1-3份。结果证实了该中药组合物对患者血虚气弱的症状得到了改善。
中国专利文献CN:201811208182.6,公告日2019年1月11日,公开了一种能够提高睡眠质量的米酒的制备工艺,该中药组合物由以下药物组成:酸枣仁、合欢花、党参、龙眼肉、莲子、茯苓、大枣、车前子、甘草。选择失眠症患者100例,每人每日饮用100ml制备的米酒,连服2周后,观察患者,效果评定标准为:效果显著、睡眠深度、入睡速度明显改善、睡眠时间增加大于 4小时者;有效:睡眠深度、入睡速度有所改善、睡眠时间增加不到4小时。无效:无明显改善。结果显示:58例效果显著,33例有效,9例无效,可见提高睡眠质量的有效率达到91%。
诸如此类宁心安神的中药组合物有很多,但是都有或多或少的缺点,例如,药味过多,取材难,价格昂贵;药效不明显,治疗效果差等等。因此,亟需一种治疗抑郁心神失养效果明显、药味数适中、便于制备的中药组合物。
发明内容
本发明的目的是针对现有技术中的不足,提供一种宁心安神的中药组合物。
本发明的再一的目的是,提供一种宁心安神的中药组合物的用途。
为实现上述目的,本发明采取的技术方案是:
一种宁心安神的中药组合物,它是由下列重量份配比的原料药制成:合欢花1-5份、龙眼肉2-8份、大枣3-8份、玫瑰花1-5份、百合1-8份。
作为一个优选例,所述的中药组合物是由下列重量份配比的原料药制成:合欢花2-4份、龙眼肉4-6份、大枣4-7份、玫瑰花2-4份、百合3-5份。
更优选地,所述的中药组合物是由下列重量份配比的原料药制成:合欢花 2份、龙眼肉5份、大枣5份、玫瑰花3份、百合3份。
为实现上述第二个目的,本发明采取的技术方案是:
如上任一所述的中药组合物在制备治疗心神失养型失眠的茶饮、食品、保健品或药物中的应用。
本发明药物的功效:
合欢花:具有舒郁,理气,安神,活络之效果。治郁结胸闷,失眠,健忘等。《四川中药志》:能合心志,开胃理气,消风明目,解郁。治心虚失眠。现代药理研究表明:合欢花煎剂灌服,能明显减少小鼠的自发活动及被动活动,具有安神助眠效果。
龙眼肉:具有补益心脾,养血安神之效果。用于气血不足,心悸怔忡,健忘失眠,血虚萎黄。
大枣:具有补脾和胃,益气生津,调营卫,解药毒等效果。治胃虚食少,脾弱便溏,气血津液不足,营卫不和,心悸怔忡等。《别录》:补中益气,强力,除烦闷,疗心下悬。
玫瑰花:具有行气解郁之效。现代药理研究发现鲜花含挥发油(玫瑰油) 约0.03%,主要成分为香茅醇、脚6醇、橙花醇、丁香油酚、苯乙醇等。香茅醇含量可达60%,脚6醇含量次于香茅醇,橙花醇为5~10%,丁香油酚和苯乙醇约各为1%,具有宁心安神,改善睡眠效果。
百合:具有养阴润肺,清心安神之效。用于虚烦惊悸,失眠多梦,精神恍惚。现代药理发现百合含有百合苷,有镇静和催眠作用,睡前服用百合汤,有助于提高睡眠质量。
本发明优点在于:
1、本发明中药组合物的组成及其之间的配比是基于大量临床经验得出,治疗效果最为突出。
2、本发明中药组合物无毒副作用,患者接受程度高。
3、本发明中药组合物原料组分少,易获得,制备工艺简单,适合推广使用。

实施例12本发明宁心安神中药组合物的临床实验
1.临床资料
1.1病例来源
筛选研究对象,将时间择取在2020年1月-2020年6月期间,以就诊先后顺序随机择取200例中医辨证为心神失养型失眠患者为本次研究对象,年龄在 35-80岁,经患者同意后,且经医院组委会同意后,采取信封随机分组法的形式将其进行分配为两小组,即对照组、干预组。
1.2诊断标准
①西医诊断标准:参照《中国精神障碍分类与诊断标准》,以睡眠障碍为主要症状,包括难以入睡、睡眠不深、易醒、多梦、早醒或醒后不易再睡,醒后不适、疲乏;上述睡眠障碍每星期至少发生3次,并持续1个月以上;失眠引起显著的情绪变化,或精神活动效率下降,或妨碍社会功能。
②中医医诊断标准:2012年版《中医病证诊断疗效标准》心神失养证中医证候诊断标准,主症:不寐,不易入睡,多梦易醒;次症:头晕目眩,心悸健忘,急躁易怒;舌脉:舌淡苔白,脉弦细。
1.2.1纳入标准
①年龄35-80岁。②中医辨证分型为心神失养型失眠患者。③匹兹堡睡眠质量指数量表(PSQI)评分>7分。④知情同意,签署知情同意书。⑤具有一定读写能力。
1.2.2排除标准
①对研究方案所选用的药物有过敏者。②伴有可能影响效应指标观测、判断的其他生理或病理状况。③严重心、肝、肾损害影响药物代谢。④特征人群 (孕妇、精神病、病情危笃或疾病晚期)。
1.2.3退出标准
①未按规定实施干预措施,无法判定疗效。②资料不全无法判定疗效、安全性。③严重不良反应、并发症,特殊生理变化等,难以继续治疗。(不良反应者纳入不良反映统计)。④退出/脱落病例按不纳入疗效判定。
1.3研究方法
1.3.1分组方法
采用信封随机分组法将其分为对照组和干预组各100例,实验前患者签署知情同意书,由研究人员进行患者基本信息采集,确保两组研究对象基线一致。
1.3.2治疗方法
①对照组:观察期间禁止服用任何中成药及任何中药制剂。服用安慰剂 (水)200mL,早晚温服。
②干预组:在对照组治疗基础上,每日两次服用宁心安神茶。(宁心安神茶:合欢花9g、龙眼肉15g、大枣15g、玫瑰花9g、百合12g。水煎200mL,早晚温服。)
③观察时间:连续观察两组治疗情况12周,若期间患者病情严重变化,需要调整用药,则视为脱落。
1.3.3观察指标
①观察两组心神失养型失眠患者的睡眠质量,所使用评估量表为匹兹堡睡眠质量指数,除记录睡眠时间、入睡时间、催眠药物外,还需对心脾两虚型失眠患者的睡眠效率、睡眠障碍等进行评估,各项均为0~3分。
②不良反应:观察两组患者治疗期间腹泻、心悸、过敏等不良反应的发生情况。
1.3.4疗效判定标准
疗效判定标准:参考《中药新药临床研究指导原则》中的标准,显效表示患者临床各项症状出现明显好转;有效表示患者临床症状有所好转;无效表示患者临床症状无改善甚至情况加重。有效率为(显效+有效)/例数×100%。
1.4统计学方法
统计分析:采用SPSS16.0统计软件,所有数据用SPSS 24.0进行分析。计量资料用(x±s)表示,组间比较采用t检验;计数资料用[n(%)]表示,组间比较采用χ2检验。以P<0.05为差异有统计学意义。
1.5研究结果
干预组心神失养型失眠患者的睡眠质量优于对照组(P<0.05),见表1。说明宁心安神茶在治疗心神失养型失眠患者有一定效果。
实施例13本发明宁心安神中药组合物的临床案例
许某,男,67岁,胃口较差,乏力明显,面色苍白,既往有贫血病史,否认糖尿病,睡眠较差,夜间多梦,心悸不安,苔白苔少,脉细。予以一种宁心安神的中药茶饮,合欢花3g、龙眼肉3g、大枣3g、玫瑰花2g、百合5g,作为一小包,每天早晚用沸水各冲泡一包。服用1月后,患者症状缓解,面色改善红润。
实施例14本发明宁心安神中药组合物的临床案例
张某,女,25岁,节食减肥后乏力明显,面色苍白,查血常规正常,睡眠较差,夜间多梦,心悸不安,苔白苔少,脉细。予以一种宁心安神的中药茶饮,合欢花3g、龙眼肉3g、大枣5g、玫瑰花3g、百合5g,作为一小包,每天早晚用沸水各冲泡一包。服用1月后,患者症状明显,面色红润,夜寐安。
实施例15本发明宁心安神中药组合物的动物实验
1实验材料
1.1动物
昆明小鼠,雄性,体质量18~22g。所有小鼠均自由饮食饮水,适应性喂养1周,称取体质量。
1.2药物与试剂
本发明中药组合物一制备:合欢花2g、龙眼肉5g、大枣5g、玫瑰花3g、百合3g,常规方法泡服。
本发明中药组合物二制备:合欢花4g、龙眼肉2g、大枣8g、玫瑰花3g、百合5g,常规方法泡服。
对照组一制备:合欢花2g、玫瑰花3g、百合3g、远志3g、夜交藤3g,常规方法泡服。
对照组二制备:合欢花2g、龙眼肉5g、大枣5g、酸枣仁3g、远志3g,常规方法泡服。
空白组:生理盐水
主要试剂:环磷酰胺(规格:0.2g/瓶,江苏盛迪医药有限公司),环磷酰胺配制成浓度为10mg/mL的溶液。5-羟色胺标准品,5-羟吲哚乙酸标准品,去甲肾上腺素标准品,多巴胺标准品;荧光剂,瑞士产邻苯二甲醛。
1.3主要仪器
XW-80A型漩涡混合器(海门其林贝尔仪器制造有限公司),JY92-2D型超声波细胞粉碎机(宁波新芝生物科技股份有限公司),Eppendorf5417R型高速冷冻离心机(德国Eppendorf公司),ANTEC电化学检测器(荷兰安泰克公司),Agilent 1100高效液相仪(美国Agilent公司),色谱柱为Diamonsilim C18(150mm×4.6mm,5μm)。
2方法
2.1心神失养小鼠失眠模型的建立
建立心神失养小鼠失眠模型,除空白组小鼠外,其余小鼠均用于模型建立,分别在第1、3、5、7、9天,每只小鼠予眼眶采血0.3~0.5mL;分别第2、4、 6、8天,每只小鼠(注射前需禁食不禁水12h)经腹腔注射环磷酰胺80、40、 40、40mg/kg,空白组腹腔注射相同体积的生理盐水。
2.2试验方案
将50只已造模好的小鼠随机分为本发明组一、本发明组二、对照组一、对照组二、空白组五组,每组10只。通过检测小鼠行为状态、体重变化观察,本发明组一无脱落;本发明组二体重不达标,脱落1例;对照组一行为状态异常,脱落2例;对照组二体重超标,脱落1例;空白组无脱落。
实验期间分别对其给药方案连续10d灌胃,给药结束后,断头处死小鼠, 去颅骨,完全暴露脑组织,在枕骨大孔处迅速剪断延髓,取出脑组织,去大脑、小脑, 称重。参照《脑组织中5-羟色胺、5-羟吲哚乙酸、去甲肾上腺素和多巴胺的同时提取和荧光分光光度测定法》,分别测得本发明组一、本发明组二、对照组一、对照组二,空白组各样本的5-HT、5-HIAA、NA和DA的吸收度,经换算得其含量为μg/g。
2.3统计学方法
全部数据应用进行正态性检验,结果显示,数据呈正态分布,进行方差齐性检验,多巴胺和去甲肾上腺两组指标方差不齐(P<0.05),故进行数值的平方根转换后,方差齐。所有数据以X±S表示,采用Q检验。
2.4结果
经两两比较可知,在给失眠小鼠连续用药后:本发明组一、本发明组二、对照组一、对照组二,其5-HT的含量均升高,但以发明组一、发明组二明显升高, 均与空白组呈显著性差异(P<0.05)。本发明组一、本发明组二5-HIAA含量较空白组明显升高,差异显著(P<0.05),说明本发明组各组可以使5-HT的代谢明显加快。本发明组一、本发明组二、对照组一、对照组二可以明显降低失眠小鼠脑组织内NA的含量(P<0.05)。多巴胺含量的结果显示,本发明组一、本发明组二,均可使DA的含量降低,优于对照组(P<0.05)。见表2

睡眠和觉醒,是生命活动所必须的两个相互转化的生理过程。科研学者认为, 睡眠中枢神经系统内发生的一个主动过程,与脑内神经递质的动态变化密切相关。与睡眠密切相关的中枢神经递质有5-HT、NA和DA。5-HIAA做为5-HT 所生成的无活性的酸性代谢产物而存在。5-HT神经元的胞体主要集中于中缝核群。脑内中缝核端的5-HT神经元对慢波睡眠的发生和维持起着重要作用。神经元的胞体主要集中于延脑、桥脑,蓝斑头部和桥脑的去甲肾上腺素能神经元, 在维持觉醒中起着重要作用,蓝斑中、后主要与快波睡眠有关,被认为是快波睡眠的“执行机制”。本发明对于心神失养型造模小鼠使NA降低,抑制了其维持觉醒的作用,减少了对5-HT的抑制,从而使睡眠改善。
以上所述仅是本发明的优选实施方式,应当指出,对于本技术领域的普通技术人员,在不脱离本发明方法的前提下,还可以做出若干改进和补充,这些改进和补充也应视为本发明的保护范围。
Claims (4)
1.一种宁心安神的中药组合物,其特征在于,它是由下列重量份配比的原料药制成:合欢花1-5份、龙眼肉2-8份、大枣3-8份、玫瑰花1-5份、百合1-8份。
2.根据权利要求1所述的中药组合物,其特征在于,它是由下列重量份配比的原料药制成:合欢花2-4份、龙眼肉4-6份、大枣4-7份、玫瑰花2-4份、百合3-5份。
3.根据权利要求2所述的中药组合物,其特征在于,它是由下列重量份配比的原料药制成:合欢花2份、龙眼肉5份、大枣5份、玫瑰花3份、百合3份。
4.根据权利要求1-3任一所述的中药组合物,在制备治疗心神失养型失眠的茶饮、食品、保健品或药物中的应用。
