Abstract
本发明公开了一种选用血肉动物提取优质蛋白与中草药制备气阳虚康养胶囊食品的方法。该方法,包括如下步骤:(1)将血肉碎块和水依次加入到提取容器中加热提取,当提取液温度达到105℃时,打开冷凝阀,直至表面液体蒸馏完毕为止,过滤取滤液备用;(2)将辅料添加到滤液、搅拌均匀获得血肉动物提取优质蛋白营养液;(3)在步骤(2)获得的营养液中添加中草药复合功能粉,搅拌均匀,喷雾干燥后收集,获得气阳虚康养胶囊食品。本发明提出的制备方法生产的胶囊食品能较好的萃取保留营养成分,特别是针对气阳虚的亚健康和需要康养的人群体质,具有补气温阳、益肾固精,并兼有活血安神的作用,能增强人体免疫力的保健功效。

技术领域
本发明属于功能食品技术领域,具体涉及一种选用血肉动物提取优质蛋白与中草药制备气阳虚康养胶囊食品的方法。
背景技术
根据德勤发布的《2020年健康医疗预测报告》,全球人口正在迅速的走向老龄化阶段,我国人口也进入了老龄化严重阶段,据不完全统计,现阶段我国60岁以上的老年人已经达到了2亿以上。随着人口老龄化进程的不断加剧,随之产生一系列的社会问题。特别是老年人由于生理功能逐步衰退,免疫功能降低,增大了患病的风险。从治未病和康养的角度出发,预防大于治疗,提高老年人的免疫功能成为预防老年人慢性疾病发生的主要方向之一。
据相关研究表明,60以上的老年人大多存在气阳虚体质的情况,长期气阳虚将导致人体免疫力下降,特别是老年群体,近年来60岁以上的人群患病率高达56%。针对气阳虚的现状,开发具有食疗效果的康养健康食品,将满足多数患有气阳虚体质的中老年消费群体的迫切需求。
发明内容
本发明的目的是针对气阳虚的亚健康和需要康养的人群体质,提供一种选用血肉动物提取优质蛋白与中草药制备气阳虚康养胶囊食品的方法。本发明提出的制备方法不仅能很好的萃取保留各组分的营养成分,而且所获得的胶囊食品具有补气温阳,益肾固精,并兼有活血安神的作用,能增强人体免疫力的保健功效。
为实现上述目的,本发明采用的技术方案是:一种选用血肉动物提取优质蛋白与中草药制备气阳虚康养胶囊食品的方法,包括如下步骤:
(1)将血肉家禽碎块和水依次加入到提取容器中加热提取,当提取液温度达到105℃时,打开冷凝阀,直至表面液体蒸馏完毕为止,过滤取滤液备用;
(2)将辅料添加到滤液、搅拌均匀获得血肉家禽动物提取优质蛋白营养液;
(3)在步骤(2)获得的营养液中添加中草药复合功能粉,搅拌均匀,喷雾干燥后收集,经过胶囊填充、包装,获得气阳虚康养胶囊食品。
本发明根据中医理论与现代医学对生物技术领域的应用研究证实,在饮食中补充有一定含量的优质动物蛋白和天然食用植物药结合,可显著增强人体的免疫力。
本发明提出的家禽是选自检疫合格经符合食品安全标准要求的生物实验室或专门生产经营场所的家禽。
优选地,步骤(1)中血肉家禽碎块经如下步骤得到:家禽经屠宰脱毛、去头、去除内脏后,得到血肉家禽,将其清洗干净、切碎得到血肉家禽碎块。
优选地,步骤(1)中所述的家禽包括公鸡、乌鸡、乌龟、鸽子和海参。
本发明中公鸡:入肝、脾、胃经,甘、性温热,具有补虚温中、止血治崩、补虚损、益虚赢、行乳汁的功效;乌鸡:入肝、肾经。性甘,味平。补肝肾、益气血、退虚热;乌龟:入肝、肺、脾;性温味甘酸;除湿痹、补阴虚、滋肾水、止血、解毒;鸽子:入肺;肝;肾经。味咸;性平。滋肾益气、祛风解毒、调经止痛;海参:归肾,肺经。味甘咸,性平。可以补肾益精,养血润燥,止血。
进一步优选,步骤(1)中所述的血肉家禽碎块以质量份数计,包括公鸡12.50~13.33份,乌鸡6.25~6.66份,乌龟3.75~4.00份,鸽子2.50~2.67份和海参1.88~2.00份。进一步优选,步骤(1)中所述的血肉家禽碎块以质量份数计,包括公鸡12.77份、乌鸡6.38份、乌龟3.83份、鸽子2.55份、海参1.91份。
进一步优选,步骤(1)中所述的血肉家禽碎块和水的质量比为26.88:73.12~28.66:71.34。再进一步优选,血肉家禽碎块和水的质量比为27.44:72.56。
优选地,步骤(3)所述的中草药复合功能粉以质量份数计,包括如下组分:扁豆粉40~50份、山药粉20~25份、芡实粉6.0~7.5份、枸杞粉6.0~7.5份、熟地粉6.0~7.5份、五味子粉4~5份、红参粉3.20~4.00份、灵芝粉0.80~1.00份、鹿茸粉0.60~0.75份和藏红花粉0.08~0.10份。
步骤(3)所述的中草药复合功能粉以质量份数计,包括如下组分:扁豆粉46.15份、山药粉23.07份、芡实粉6.92份、枸杞粉6.92份、熟地粉6.92份、五味子粉4.61份、红参粉3.69份、灵芝粉0.92份、鹿茸粉0.69份、藏红花粉0.09份。
本发明中扁豆:入脾、胃经。甘、平。健脾和中、消暑化湿;山药:归脾、肺、肾经;甘、平。补脾养胃、生津益肺、补肾涩精;芡实:入脾、肾经;甘、涩、平。益肾固精、补脾止泻、祛湿止带;枸杞:入肝、肾经;甘、平。滋补肝肾、益精明目;熟地:入肝、肾经。甘,平。滋阴补血、益精填髓;五味子:入肺、心、肾经;味酸;性温。收敛固涩、益气生津、宁心安神;红参:入脾、肺、心经。甘、微苦,温。大补元气、复脉固脱、益气摄血;灵芝:归心、肺、肝、肾经。甘,平。补气安神、止咳平喘;鹿茸:入肾、肝经。甘、咸,温。壮肾阳、益精血、强筋骨、调冲任、托疮毒;藏红花:入心;肝经。甘;平。活血化瘀、散郁开结、凉血解毒。
优选地,营养液和中草药复合功能粉的质量比为75~80:20~25。进一步优选,营养液和中草药复合功能粉的质量比为78.33:21.67。
优选地,中草药复合功能粉中各组分的颗粒粒径为60目。
优选地,步骤(2)所述的辅料为山梨酸钾。山梨酸钾的加入量为滤液质量的0.75%-0.78%。
本发明还保护经上述方法制备得到的气阳虚康养胶囊食品。
与现有技术相比,本发明的有益效果是:
1、本发明提出的气阳虚康养胶囊食品配方组份主要选择由多种血肉家禽动物提取优质蛋白营养液和药食两用中草药复配制成,配方用性温的鹿茸、红参、五味子、熟地和性平的芡实、灵芝、枸杞来补气温阳、益肾安神,味酸的五味子收敛固涩,性平扁豆、山药补脾益气,少佐藏红花可以活血散郁。同时选用的性偏温的公鸡肉和乌龟肉和性平鸽子肉和乌鸡肉,再加一点性温的乌龟肉,配伍一起就可以有补气血,壮肾阳,强筋骨的作用。
2、本发明对于气阳虚体质的人,比如先天不足,禀赋虚弱;房事不节,耗损肾气;饮食不节,损伤脾胃,不能化生精微,气血亏虚;大病之后,失于调养等都可能会导致患者出现疲倦乏力、少气懒言、动则气疑、畏寒喜暖,重者可见形寒肢冷,腰酸腿软,阳痿早泄等症状,均具有明显的改善作用,可作为康养功能食补产品供气阳虚体质人群放心食用。
3、本发明提出的制备方法生产的胶囊食品能较好的萃取保留营养成分,特别是针对气阳虚的亚健康和需要康养的人群体质,具有补气温阳、益肾固精,并兼有活血安神的作用,能增强人体免疫力的保健功效。

实施例1:
一种采用血肉家禽动物提取优质蛋白与中草药制备气阳虚康养胶囊食品的方法,包括如下步骤:
(1)选检疫合格经符合食品安全标准要求的生物实验室或专门生产经营场所进行屠宰脱毛、去头、去除内脏的公鸡、乌鸡、乌龟、鸽子、海参,清洗干净、切碎;
(2)将步骤(1)处理后的血肉家禽,按血肉家禽和清水的总质量分数100%计,12.77%公鸡、6.38%乌鸡、3.83%乌龟、2.55%鸽子、1.91%海参,余量为清水的比例加入到多功能提取罐,0.1 Mpa压力下加热提取;
(3)当提取液温度达到105℃时,打开冷凝阀,直至表面液体蒸馏完毕为止,过滤取滤液备用;
(4)在步骤(3)获得的滤液中缓慢加入滤液质量的0.77%的山梨酸钾,并搅拌均匀,获得血肉家禽动物提取优质蛋白营养液;
(5)在步骤(4)获得的营养液中按78.33:21.67的质量比添加60目的由扁豆粉46.15份、山药粉23.07份、芡实粉6.92份、枸杞粉6.92份、熟地粉6.92份、五味子粉4.61份、红参粉3.69份、灵芝粉0.92份、鹿茸粉0.69份、藏红花粉0.09份组成的中草药复合功能粉,将二者搅拌均匀,喷雾干燥后,按0.5 g/粒进行胶囊填充并灌装,即获得气阳虚康养胶囊食品。
对比例1
胶囊食品的制备方法,包括如下步骤:
(1)选检疫合格经符合食品安全标准要求的生物实验室或专门生产经营场所进行屠宰脱毛、去头、去除内脏的公鸡、乌鸡、乌龟、鸽子、海参,清洗干净、切碎;

(2)将步骤(1)处理后的血肉家禽,按血肉家禽和清水的总质量分数100%计,12.50%公鸡、6.25%乌鸡、3.75%乌龟、2.50%鸽子、1.88%海参,余量为清水的比例加入到多功能提取罐,0.1 Mpa压力下加热提取;
(3)当提取液温度达到105℃时,打开冷凝阀,直至表面液体蒸馏完毕为止,过滤取滤液备用;
(4)在步骤(3)获得的滤液中缓慢加入滤液质量的0.75%的山梨酸钾,并搅拌均匀,获得血肉家禽动物提取优质蛋白营养液;
(5)在步骤(4)获得的营养液中按75:25的质量比添加60目的由扁豆粉40份、山药粉25份、芡实粉6份、枸杞粉6份、熟地粉6份、五味子粉5份、红参粉3.20份、灵芝粉0.80份、鹿茸粉0.60份和藏红花粉0.1份组成的中草药复合功能粉,将二者搅拌均匀,喷雾干燥后,按0.5 g/粒进行胶囊填充并灌装,即获得气阳虚康养胶囊食品。
实施例3
一种采用血肉家禽动物提取优质蛋白与中草药制备气阳虚康养胶囊食品的方法,包括如下步骤:
(1)选检疫合格经符合食品安全标准要求的生物实验室或专门生产经营场所进行屠宰脱毛、去头、去除内脏的公鸡、乌鸡、乌龟、鸽子、海参,清洗干净、切碎;
(2)将步骤(1)处理后的血肉家禽,按血肉家禽和清水的总质量分数100%计,13.33%公鸡、6.66%乌鸡、4.00%乌龟、2.67%鸽子、2.00%海参,余量为清水的比例加入到多功能提取罐,0.1 Mpa压力下加热提取;
(3)当提取液温度达到105℃时,打开冷凝阀,直至表面液体蒸馏完毕为止,过滤取滤液备用;
(4)在步骤(3)获得的滤液中缓慢加入滤液质量的0.78%的山梨酸钾,并搅拌均匀,获得血肉家禽动物提取优质蛋白营养液;
(5)在步骤(4)获得的营养液中按80:20的质量比添加60目的由扁豆粉50份、山药粉20份、芡实粉7.5份、枸杞粉7.5份、熟地粉7.5份、五味子粉4份、红参粉4份、灵芝粉1份、鹿茸粉0.75份、藏红花粉0.08份组成的中草药复合功能粉,将二者搅拌均匀,喷雾干燥后,按0.5 g/粒进行胶囊填充并灌装,即获得气阳虚康养胶囊食品。
实验例1。
一、对实施例1得到的气阳虚康养胶囊进行测试,有效营养成分含量如表3所示:
二、100名60-70岁的气阳虚志愿者在没有其他药物辅助的情况下,每日服用实施例1得到的气阳虚康养胶囊30 g(一日三餐,每次餐后半小时服用20粒),60%的志愿者在食用10天后气阳虚症状有减轻,95%的志愿者在食用15天后气阳虚症状有减轻,98%的志愿者在食用30天后气阳虚症状有显著改善。
三、气阳虚康养胶囊(阳虚胶囊)的急性毒性研究
1、实验分组
采用最大给药量法进行供试品一日内多次给药。将40只ICR小鼠分成不同给药组及一个对照组,每组10只动物。对照组给予与给药组相同体积的蒸馏水。具体实验动物分组和灌胃量情况如表4所示。实施例1得到的阳虚胶囊经水溶解后形成质量浓度为0.3 g/mL的溶液。
2、实验观察分析。
阳虚胶囊经水溶解后(0.3 g/mL),按照表4的灌胃量完成给药,随后连续观察14天,密切观察动物在给药后48小时内的反应。根据《中药、天然药物急性毒性研究技术指导原则》相关内容要求,详细记录动物出现毒性反应情况,中毒症状、中毒症状出现时间、持续时间、恢复时间及动物最短死亡时间、最长死亡时间、平均死亡时间和死亡动物数量,死亡动物及时解剖尸检,其中肉眼观察异常动物进行病理组织学检查,记录但不限于如下内容:。
① 动物外观:皮肤颜色是否有改变,皮肤是否有红肿现象,出现斑点,毛发是否有光泽等
② 神经系统:行为及反应,异常运动,对刺激的反应,神经反射,抽搐,肌张力。
③ 呼吸功能:呼吸深度及频率,鼻分泌物,喘息,是否出现呼吸急促和暂停的情况。
④ 运动功能:观察小鼠步态,运动是否增加或减少,是否有异常运动,俯卧,震颤,肌束震颤等。
⑤ 反射功能:检查角膜眼睑闭合,正位反射,牵张反射,光反射。
⑥ 眼睛指征:如眼球突出,上睑下垂,结膜浑浊,虹膜炎,结膜炎。
⑦ 心血管指征:血管扩张。
⑧ 胃肠指征:粪便性状,腹部形态等。
⑨ 泌尿生殖系统指征:外生殖器情况。
观察期结束时存活动物称体重后处死并进行解剖检查,观察心脏、肝脏、肾、脾、肺及胃肠等脏器的变化,如有明显改变则进一步进行病理检查,统计各脏器重量,记录脏器系数。
3、实验结果
小鼠在接受不同剂量的阳虚胶囊灌胃处理14天后,与对照组相比,其表观正常,无死亡现象发生,小鼠外观形态、动作行为、体姿神态等无明显异常,表明阳虚胶囊的LD50大于小鼠对其的最大耐受剂量,且无明显急性毒性。
小鼠最大给药量换算为15 g/kg体重,约为成人常规服用剂量(1.2 g/70kg)的875倍,显示其良好的安全性。
由于供试品无明显毒性,为使实验结果简明直观,后续仅对正常对照组和阳虚胶囊高剂量实验组进行分析。
(1)实验动物外观观察
动物给药前、给药后均饮食、活动均无明显异常,排泄物外观正常。整个实验过程未发现动物死亡情况。经连续观察,给药前后小鼠外观无明显异常,神志清楚,动作步态稳健,毛色光鲜,无流涎流涕现象(表5和表6)。
(2)给药前后小鼠体重变化记录情况
经不同时间点对小鼠进行称重,发现阳虚胶囊给药小鼠在接受灌胃给药处理后体重变化与对照组变化趋势基本一致,在给药的第11天和14天,阳虚胶囊给药组小鼠体重显著高于对照组(表7)。
(3)灌胃处理后实验动物的脏器变化情况记录
实验结束后,对给药组和对照组动物进行大体解剖,观察到各组小鼠心脏、肺脏、肝脏、脾脏、肾脏和胃肠等器官均未发生明显异常改变(如表8和表9)。对部分脏器如肝、脾、肺、肾和生殖器官进行称重,并计算相关脏器系数(脏器重量/体重),阳虚胶囊给药处理组与对照组相比,主要脏器如心脏、肺、肝脏、肾脏指数均无无显著性差异,同时阳虚胶囊组(阳虚胶囊高剂量组)小鼠脾脏和胸腺指数显著升高,提示阳虚胶囊可能促进小鼠免疫器官生长,提升小鼠免疫力。
(4)小鼠部分脏器进行病理学检查
分别对各组小鼠进行部分脏器取材,做组织切片进行组织病理学检查,各处理组小鼠脏器均未发现有明显异常病变,如图1所示,各脏器质地均匀,核质边缘清晰,形状规则,无组织坏死现象,各组小鼠肝、脾组织形态完整,结构无明显异常。
5、结论
通过一日内三次给小鼠灌胃不同剂量的阳虚胶囊后,进行为期14天的连续观察,在整个观察期过程中未见动物出现任何异常反应,也没有发生动物死亡情况。体重检测结果显示,在给予阳虚胶囊灌胃一周后,小鼠体重显著高于对照组。通过组织观察和组织病理检查,未发现阳虚胶囊对小鼠主要脏器产生明显不良影响,但显著促进脾脏和胸腺质量的增加。
综合以上结果表明,阳虚胶囊对实验小鼠无明显的急性毒性,且对小鼠主要免疫器官具有显著的生长促进作用。
实验例2 阳虚胶囊对肾阳虚小鼠的治疗效果
主要研究方法如下:健康昆明小鼠30只,5-6周龄,适应性饲养一周后,将小鼠随机分为3组,每组10只。具体分组如下:实施例1得到的阳虚胶囊经水溶解后形成质量浓度为0.3 g/mL的溶液。
对照组:腹腔注射生理盐水0.2 mL,1次/天,连续7天。
肾阳虚组:腹腔注射氢化可的松25 mg/(kg体重·天),连续7天;
肾阳虚组治疗组:腹腔注射氢化可的松25mg/(kg·天),口服灌胃阳虚胶囊溶液剂量为10 mL/kg体重,连续7天。
实验期间,小鼠自由饮水饮食,每天观察小鼠外观形态和活动状态并做好相关记录,7天实验处理期结束后,收集小鼠血液进行相关生化和生物学指标检测。
实验结果:
1、形态外观观察:正常组小鼠外观和行为正常,表现为毛色有光泽,行动活跃,饮食正常,粪便湿润呈颗粒状。肾阳虚小鼠则出现毛发蓬松,行为不活跃,精神状态差,大便稀疏,拱背聚集,有怕冷现象。而给以阳虚胶囊灌胃后,小鼠毛色明显改观,小鼠活动量增加,粪便呈颗粒状。
2、生化检测:
通过检测各组小鼠血清中FSH、睾酮和雌激素E2水平发现,肾阳虚小鼠血清中各指标均显著高于正常对照组(P<0.05),经阳虚胶囊治疗后,FSH和E2水平有明显降低,其数值与正常组无显著差异;阳虚胶囊也在一定程度上降低了肾阳虚小鼠血清中睾酮的水平,但仍然高于正常对照小鼠,具体实验结果见表10。
综上结果表明,阳虚胶囊可以显著改善肾阳虚小鼠症状。
3、对小鼠游泳不动时间的影响
该实验方法是一种行为绝望实验法,通过将动物置于一个局限的环境中(如水中),从而提供了一个无可回避的压迫环境,一段时间后,动物即表现出典型的“不动状态”,观察并记录实验动物产生绝望的不动状态过程中的一系列参数,可以用来评价抗抑郁药物的作用效果。
30只小鼠随机分为3组,每组10只,即空白对照组、盐酸氯米帕明组和阳虚胶囊给药组。各给药组按体重0.2 mL/10g体重灌胃给药,对照组给予等量蒸馏水,每天1次,连续12天。第12天给药后1小时内,将小鼠单个地放入盛有水的Pyrex游泳筒中(45 cm × 21cm,水深约20 cm,水温保持在23℃-25℃),待其适应水温后,先进行15 min的游泳预实验应激,24小时后,再次将小鼠放入水筒中,然后测定各给药组和对照组小鼠在5分钟内在水面上保持漂浮不动的时间。小鼠不动的评判标准是小鼠微卷躯体,但保持垂直姿势,漂浮于水中,鼻孔露出水面,四肢保持不动。各组小鼠游泳不动时间如表11所示。
采用SPSS软件分析实验结果,结果以t检验进行两组间差异分析,P<0.05表示有显著性差异。
结果如下表:
由表11结果表明,与对照组相比,盐酸氯米帕明与阳虚胶囊给药组小鼠游泳不动时间均显著减少,这表明阳性胶囊具有一定程度抗小鼠抑郁功能。
4、阳虚胶囊对气阳虚人群的作用效果
参照《中药新药临床研究指导原则(试行)》,结合中医辨证标准对85位候选人进行选择,纳入标准包括符合气阳虚中医辨证标准,年龄40-78岁,入组人员知情同意等。入组后的人员经单盲法进行中医辨证,根据症状严重程度按照无症状(0分)、轻微症状(1分)、较轻症状(2分)、中等症状(3分)、较重症状(4分)以及严重症状(5分)划分为6个等级并计分统计。
入组人员再空腹采血,获取血清样本,进行相关生化指标TNFα、IL-1和IFNγ等细胞因子的检测。入组人员经口服阳虚胶囊(1.2g/kg体重/天)两周后再行上述检测并记录相关数据。
实验结果:
根据入组标准,最终有50人纳入研究队列,其中获取20人的血清样本进行了相关生化因子检测,具体结果见表12。
从表12可以看到,服用阳虚胶囊后,气阳虚人群症状得到显著缓解,主要表现为心悸气短现象减轻,畏寒、肢冷现象明显改善,面色、唇色以及舌表征明显好转。各炎症因子水平均明显降低。
以上结果表明,阳虚胶囊对气阳虚人群各症状具有显著的改善效果。
以上仅是本发明的优选实施方式,应当指出的是,上述优选实施方式不应视为对本发明的限制,本发明的保护范围应当以权利要求所限定的范围为准。对于本技术领域的普通技术人员来说,在不脱离本发明的精神和范围内,还可以做出若干改进和润饰,这些改进和润饰也应视为本发明的保护范围。
Claims (5)
1.一种选用血肉动物提取优质蛋白与中草药制备气阳虚康养胶囊食品的方法,其特征在于,包括如下步骤:
(1)将血肉碎块和水依次加入到提取容器中加热提取,当提取液温度达到105℃时,打开冷凝阀,直至表面液体蒸馏完毕为止,过滤取滤液备用,所述的血肉碎块以质量份数计,包括公鸡12.50~13.33份,乌鸡6.25~6.66份,乌龟3.75~4.00份,鸽子2.50~2.67份和海参1.88~2.00份,所述的血肉碎块和水的质量比为26.88:73.12~28.66:71.34;
(2)将辅料添加到滤液、搅拌均匀获得血肉动物提取优质蛋白营养液;
(3)在步骤(2)获得的营养液中添加中草药复合功能粉,搅拌均匀,喷雾干燥后收集,获得气阳虚康养胶囊食品,所述的中草药复合功能粉以质量份数计,包括如下组分:扁豆粉40~50份、山药粉20~25份、芡实粉6.0~7.5份、枸杞粉6.0~7.5份、熟地粉6.0~7.50份、五味子粉4~5份、红参粉3.20~4.00份、灵芝粉0.80~1.00份、鹿茸粉0.60~0.75份和藏红花粉0.08~0.10份,营养液和中草药复合功能粉的质量比为75~80:20~25。
2.根据权利要求1所述的方法,其特征在于,步骤(1)中血肉碎块经如下步骤得到:动物经屠宰脱毛、去头、去除内脏后,将其清洗干净、切碎得到血肉碎块。
3.根据权利要求1所述的方法,其特征在于,中草药复合功能粉中各组分的颗粒粒径为60目。
4.根据权利要求1所述的方法,其特征在于,步骤(2)所述的辅料为山梨酸钾。
5.权利要求1所述的方法制备得到的气阳虚康养胶囊食品。
