Abstract
本发明公开了一种有效祛除牙斑、牙垢的增白牙膏,其特征在于;其中包含肉桂酸提取物、蛇床子素提取物、唾液乳杆菌发酵液、甘油、山梨醇、丙二醇、二氧化硅、羧甲基纤维素钠、月桂醇硫酸酯钠、氟化钠、氟化钾或单氟磷酸钠、薄荷粉。本发明选用肉桂和蛇床子素提取液、唾液乳杆菌作抑菌、除垢剂以及增白效果、并配合氟类添加剂去除色素沉着和菌斑与牙膏体系结合制备了可以有效祛除牙斑、牙垢的牙膏。

技术领域
本发明属于洗涤用品领域,具体涉及一种牙膏,特别是涉及一种有效祛除牙斑、牙垢的增白牙膏。
背景技术
当今社会倡导天然、健康,产品的安全性、功效性倍受人们关注。口腔护理用品也是如此,植物草本具有副作用小,使用安全性高的优点,已成为市场消费的主流。牙膏的组成部分绝大部分都是摩擦剂,牙膏的作用主要是作为一种中介,作用于牙刷与牙齿的中间,增强牙刷与牙齿之间的摩擦力,促使牙刷清洁干净牙齿。牙斑是口腔里的细菌粘附在牙齿的表面,随着时间的堆积而形成的附着性菌斑膜。牙垢就是我们常说的牙黄,那些附着在牙齿表面脏东西。牙垢,又称牙菌斑,是在牙齿表面逐渐沉积的生物薄膜。牙垢中的细菌主要是正常口腔内存在的链球菌、厌氧菌等。牙垢堆积到一定厚度之后,其内部紧挨牙齿表面的细菌因为与空气隔绝开始转入无氧呼吸。无氧呼吸在此处产生的酸不能及时被唾液冲走,因此会腐蚀珐琅质中的矿物成分,并进一步促进龋齿的形成。在牙根处堆积的牙垢也会刺激牙龈,导致牙周炎等牙周疾病。日常生活中,牙齿上出现黑色或黄色牙斑,多是由于牙齿表面色素沉着造成的,常见于有喝浓茶或咖啡习惯的人群,以及吸烟人群。牙膏起主要作用的是其中的摩擦剂,通过牙刷在牙齿表面进行摩擦来清洁牙齿,但是普通市售牙膏中并不含有能够分解牙齿表面色素的化学成分,很难实现消费者希望通过刷牙彻底去除牙斑的夙愿。另外,有不少人刷牙的时候会有干呕的现象,除了是有咽喉炎症状,其实跟选择的牙膏也有点关系。如刷牙后漱口不彻底,就会摄入牙膏残液,引起局部性胃肠炎,从而困扰人们的生活。
肉桂,是一种樟科属的植物,常见于在热带地区和部分亚热带地区,为常绿乔木,我国的肉桂资源世界第一。并且肉桂作为中药药材已有两千多年历史,据《广西中药志》记载:“肉桂油具有杀灭病菌、病毒的作用,内服可作驱风药和健胃药”。《本草纲目》中记载按照采收的季节不同,肉桂品种分为春桂和秋桂两类,肉桂植物对治疗冬季冻疮、腹痛腹泄、咳嗽呕吐、头晕头痛和体虚胃寒等病有很好的医用价值,并且肉桂中的肉桂醛还具有抗菌防腐的优点。另外,现代药理实验研究表明蛇床子素具有抗高血压、抗心律失常、抗衰老、抗肿瘤、抗骨质疏松症、抗炎及抗变态反应等功效,将含蛇床子素提取物添加到牙膏中,可以起到一定的中药抗炎、抑制牙菌斑的作用,有利于口腔健康。氟是牙齿及骨骼中不可缺少的成份,少量氟可以促进牙齿珐琅质对细菌酸性腐蚀的抵抗力,牙膏中的游离氟与牙齿釉面接触后,生成氟磷石灰会使牙齿变硬,从而起到抗酸溶解的抗龋效果,就是通常所说的防龋作用,含氟牙膏还能抑制口腔细菌产酸,改变口腔内的细菌适于生存的环境,从而起到防龋祛斑的作用。
因此本发明选用肉桂和蛇床子素提取液作抑菌、除垢剂,搭配唾液乳杆菌发酵液,可以起到一定的美白效果。此外再配合氟类添加剂可以更好的去除色素沉着和菌斑,与牙膏体系结合制备了可以有效祛除牙斑、牙垢的增白牙膏。
发明内容
为于克服现有技术的不足,本发明的目的是提供一种牙膏,具有除垢、增白的效果,并且在口腔内具有一定的抑菌效果。并通过以下技术方案来实现:
第一方面,本发明提供了一种有效祛除牙斑、牙垢的组合物,其组成包括:肉桂酸提取物、蛇床子素提取物、唾液乳杆菌发酵液、甘油、山梨醇、丙二醇、二氧化硅、羧甲基纤维素钠、月桂醇硫酸酯钠、氟化钠、氟化钾或单氟磷酸钠、薄荷粉。
第二方面,本发明提供了一种有效去除牙斑、牙垢的增白牙膏的制备方法。其制备方法如下:
S1、采用水蒸气蒸馏提取法制备肉桂提取液:称取晾干的肉桂果200-300g,磨碎后加水没过物料,然后通过超声波处理机进行预处理,超声波预处理有助于肉桂原料细胞壁的破碎,以及有效成分的溶出,再添加3-5L的去离子,然后加热使体系保持沸腾状态2-5h,得到肉桂提取原液。
S2、制备蛇床子素提取液:称取优质伞形科植物蛇床子的果实10-15g磨碎,取蛇床子花3-5g,加入100-200ml水煎煮5次每次30-50min,合并滤液,加入乙醇60ml静置8h,除去沉淀得滤液,然后用旋蒸仪挥去乙醇,加水定容至120mL,取55mL,加丙三醇40mL,连续萃取三次,取丙三醇层溶液备用。
S3、按3%的接种量将唾液乳杆菌种子接种于发酵罐中,37℃恒温培养,收集培养至稳定期的发酵液,发酵液中菌体达到一定的浓度后,将发酵液离心出去菌体,得到唾液乳杆菌发酵液。
S4、液料制备:取S2制备的肉桂提取液80-200ml,取S3制备的蛇床子素提取液50-150ml,取S3制备的唾液乳杆菌发酵液3-5mL,然后将其加入真空制膏机中,再计量称取适量的甘油、山梨醇和丙二醇加入真空制膏机中,混合搅拌20-40min,这些材料的加入可以使膏体保持一定润湿度和柔软流动性。
S5、粉料制备:称取10-15g的二氧化硅、20-25g的羧甲基纤维素钠、8-10g的月桂醇硫酸酯钠,0.3-0.8g的氟化钠、氟化钾或单氟磷酸钠混合均匀。该粉体是膏体的主体成分之一,在刷牙时帮助牙刷清洁牙齿。
S6、液料和粉料混合:将制S4备好的粉料加入真空制膏机中和S3制备的液料混合,搅拌30min后,加入3-5g薄荷粉,同时打开制膏机的真空泵开始抽真空,真空搅拌30-50min后,关闭真空泵和真空制膏机搅拌,将牙膏灌装到软管中,用自制封尾机封尾即可。

优选地:所述S3选用的甘油为30ml、山梨醇为50ml、丙二醇为10ml;

其中本发明涉及的唾液乳杆菌菌株(JYLS-372)均购自陕西飞米生物科技有限公司,唾液乳杆菌为100亿cfu/g,发酵液中的菌体浓度为3.0×109cfu/mL。
1、本发明制备的牙膏无激素,无色素,无合成香料,无防腐剂等有害的化学添加剂,属于纯植物有机牙膏。
2、本发明制备的牙膏选用肉桂和蛇床子素提取液抑菌,并且在此基础上加入唾液乳杆菌发酵液,他们三者在特定的比例下可以是美白效果达到最大,还具有无毒不刺激、杀菌洁齿的优点。
3、本发明制备的牙膏绿色环保,在刷牙时,即使误食,也不会影响身体健康。
4、本发明操作简单,生产的成本较低,有利于批量生产。
5、本发明制备的牙膏膏体细腻、光滑,呈淡黄色半透明状,泡沫细腻丰富,口感较好,气味清凉,粘稠度适中。

具体实施方式
本实施例所用的唾液乳杆菌购自陕西飞米生物科技有限公司,唾液乳杆菌100亿cfu/g。
本实施例所用的青春乳杆菌购自陕西飞米生物科技有限公司,青春乳杆菌100亿cfu/g
实施例1
S1、称取晾干的肉桂果200g,磨碎后加水没过物料,然后通过超声波处理机进行预处理,超声波预处理有助于肉桂原料细胞壁的破碎,以及有效成分的溶出,再添加3L的去离子,然后加热使体系保持沸腾状态2h,得到肉桂提取原液。

对比例1、在S3步骤中除了不加肉桂提取液外,其余参数与实施例1相同。
对比例2、在S3步骤中除了不加蛇床子素提取液外,其余参数与实施例1相同。
本发明试验菌株选择金黄色葡萄球菌(ATCC 6538)、大肠杆菌(ATCC 25922)、白色念珠菌(ATCC 10231)、牙龈卟啉单胞菌(ATCC 33277)、变异链球菌(ATCC 700610)。参照QB/T2738-2012第7.3进行试验,分别测试用去离子水将牙膏稀释3倍、90倍后样品的抑菌率,作用时间5min,试验重复5次,取平均值。

由表1、2、3的结果可知,实施例1、对比例1和对比例2制备的牙膏被稀释4倍和90倍后,对金黄色葡萄球菌、大肠杆菌、白色念珠菌、牙龈卟啉单胞菌、变异链球菌的抑菌作用,表明实施1制备的牙膏抑菌效果最好,并且通过对比例1、2,也说明了肉桂和蛇床子素提取液的具有协同作用作用,两者都加抑菌效果更好。原因可能是肉桂中含有大量的黄酮类、皂苷、多糖等活性成分,此类活性成分本身就具有一定的抗菌作用。而蛇床子素中含有异戊酸龙脑酯等活性成分,此类成分可以和肉桂中的活性成分发生作用,使得肉桂中抗菌作用更强。
实施例2
S1、采用水蒸气蒸馏提取法制备肉桂提取液:称取晾干的肉桂果300g,磨碎后加水没过物料,然后通过超声波处理机进行预处理,超声波预处理有助于肉桂原料细胞壁的破碎,以及有效成分的溶出,再添加5L的去离子,然后加热使体系保持沸腾状态5h,得到肉桂提取原液。
S2、制备蛇床子素提取液:称取优质伞形科植物蛇床子的果实15g磨碎,取蛇床子花5g,加入200ml水煎煮5次每次50min,合并滤液,加入乙醇60ml静置8h,除去沉淀得滤液,然后用旋蒸仪挥去乙醇,加水定容至120mL,取55mL,加丙三醇40mL,连续萃取三次,取丙三醇层溶液备用。
S3、按3%的接种量将唾液乳杆菌种子接种于发酵罐中,37℃恒温培养,收集培养至稳定期的发酵液,发酵液中菌体浓度为3.0×109CFU/mL时,将发酵液离心出去菌体,得到唾液乳杆菌发酵液。

S6、液料和粉料混合:将S5制备好的粉料加入真空制膏机中和S4制备的液料混合,搅拌30min后,加入5g薄荷粉,同时打开制膏机的真空泵开始抽真空,真空搅拌50min后,关闭真空泵和真空制膏机搅拌,将牙膏灌装到软管中,用自制封尾机封尾即可。
对比例3、在S3步骤中除了不加肉桂提取液和蛇床子素提取液外,其余参数与实施例2相同。
参照《消毒技术规范》中关于抑菌环测试方法,进行市售牙膏(1号)、实施例2(2号)和对比例3(3号)制备牙膏的抑菌环测试,实验平行进行5次,取平均值作为结果。图2为本发明中抑菌环实验结果,实施例2、市售牙膏和对比例3制备牙膏的平均抑菌环直径依次为:21mm、18mm、4mm。实施例2抑菌环直径明显大于市售牙膏,表明其具有高于市售牙膏的抑菌活性。对比例3也有4mm的抑菌环,表明步骤S5中添加的薄荷粉液具有一定抑菌作用,可形成较小抑菌环。
实施例3
S1、采用水蒸气蒸馏提取法制备肉桂提取液:称取晾干的肉桂果230g,磨碎后加水没过物料,然后通过超声波处理机进行预处理,超声波预处理有助于肉桂原料细胞壁的破碎,以及有效成分的溶出,再添加4L的去离子,然后加热使体系保持沸腾状态3h,得到肉桂提取原液。
S2、制备蛇床子素提取液:称取优质伞形科植物蛇床子的果实13g磨碎,取蛇床子花4g,加入120ml水煎煮5次每次40min,合并滤液,加入乙醇60ml静置8h,除去沉淀得滤液,然后用旋蒸仪挥去乙醇,加水定容至120mL,取55mL,加丙三醇40mL,连续萃取三次,取丙三醇层溶液备用。
S3、按3%的接种量将唾液乳杆菌种子接种于发酵罐中,37℃恒温培养,收集培养至稳定期的发酵液,发酵液中菌体浓度为3.0×109CFU/mL时,将发酵液离心出去菌体,得到唾液乳杆菌发酵液。
S4、液料制备:取S1制备的肉桂提取液150ml,取S2制备的蛇床子素提取液80ml,S3制备的唾液乳杆菌发酵液5mL,然后将其加入真空制膏机中,再计量称取适量的甘油、山梨醇和丙二醇加入真空制膏机中,混合搅拌30min。
S5、粉料制备:称取13g的二氧化硅、22g的羧甲基纤维素钠、9g的月桂醇硫酸酯钠,0.6g的单氟磷酸钠混合均匀。
S6、液料和粉料混合:将S5制备好的粉料加入真空制膏机中和S4制备的液料混合,搅拌30min后,加入4g薄荷粉,同时打开制膏机的真空泵开始抽真空,真空搅拌38min后,关闭真空泵和真空制膏机搅拌,将牙膏灌装到软管中,用自制封尾机封尾即可。
对比例4、在S4步骤中除了不加羧甲基纤维素钠外,其余参数与实施例3相同。
对比例5、在S4步骤中除了不加月桂醇硫酸酯钠外,其余参数与实施例3相同。
对比例6、在S4步骤中除了不加二氧化硅外,其余参数与实施例3相同。
对比例7、在S4步骤中除了不加单氟磷酸钠外,其余参数与实施例3相同。
流动曲线和粘度曲线反映了牙膏在不同剪切速率下的流动特性,决定了牙膏在制备、灌装以及使用时性能的好坏。对于牙膏要求其具备优良的剪切变稀特性,表现为假塑性流体,假塑性越好,生产过程中更容易输送,稳定性更好,使用更为方便。
图2、3为本发明实施例1-3和对比例1-7制备牙膏的流动曲线与粘度曲线,对比分析可知,所制备牙膏均表现出假塑性流体的流变学特征,粘度随着剪切速率的增长而逐渐减小,即剪切变稀。在剪切作用下,牙膏内部的立体网状结构沿着剪切作用的方向断裂,使原本悬浮在体系中的不规则粒子沿着流动方向排列,链状分子沿驱动力方向解缠绕、拉伸和取向,导致表观粘度降低。
实施例4
S1、采用水蒸气蒸馏提取法制备肉桂提取液:称取晾干的肉桂果250g,磨碎后加水没过物料,然后通过超声波处理机进行预处理,超声波预处理有助于肉桂原料细胞壁的破碎,以及有效成分的溶出,再添加4.5L的去离子,然后加热使体系保持沸腾状态4h,得到肉桂提取原液。
S2、制备蛇床子素提取液:称取优质伞形科植物蛇床子的果实14g磨碎,取蛇床子花4g,加入140ml水煎煮5次每次44min,合并滤液,加入乙醇60ml静置8h,除去沉淀得滤液,然后用旋蒸仪挥去乙醇,加水定容至120mL,取55mL,加丙三醇40mL,连续萃取三次,取丙三醇层溶液备用。
S3、按3%的接种量将唾液乳杆菌种子接种于发酵罐中,37℃恒温培养,收集培养至稳定期的发酵液,发酵液中菌体浓度为3.0×109CFU/mL时,将发酵液离心出去菌体,得到唾液乳杆菌发酵液。
S4、液料制备:取S1制备的肉桂提取液180ml,取S2制备的蛇床子素提取液110ml,S3制备的唾液乳杆菌发酵液5mL,然后将其加入真空制膏机中,再计量称取适量的甘油、山梨醇和丙二醇加入真空制膏机中,混合搅拌34min。
S5、粉料制备:称取14g的二氧化硅、24g的羧甲基纤维素钠、9g的月桂醇硫酸酯钠,0.4g的单氟磷酸钠混合均匀。
S6、液料和粉料混合:将S5制备好的粉料加入真空制膏机中和S4制备的液料混合,搅拌30min后,加入4.4g薄荷粉,同时打开制膏机的真空泵开始抽真空,真空搅拌40min后,关闭真空泵和真空制膏机搅拌,将牙膏灌装到软管中,用自制封尾机封尾即可。
对比例8、在S4步骤中除了不加单氟磷酸钠外,其余参数与实施例4相同。
对比例9、在S4步骤中除了不加入唾液乳杆菌发酵液外,其余参数与实施例1相同。
对比例10、在S4步骤中的唾液乳杆菌发酵液换成青春乳杆菌发酵液,其余参数与实施例1相同。
实验表明,使用含氟牙膏,能降低患口腔疾病的概率。青少年和老年人适当地使用含氟牙膏对牙齿有好处。本发明与普通牙膏的最大不同之处就在于增加了能够杀菌的的肉桂和蛇床子字素提取液,它有杀菌功效,能在一定程度上缓解口腔疾病,微量的氟掺杂结合植物添加剂可以更好的发挥本发明的优势。
本发明中涉及到的可溶氟和游离氟的检测方法及判定依据均为国家标准《牙膏》(GB/8372-2017)中提供的方法进行检测。

美白效果试验
1.试验样品:实施例1-4及对比例1-2及对比例9-10制得的牙膏以及市面上的美白牙膏;
2.实验过程:
2.1试验对象:选取270名年龄范围在30-50岁的牙齿有外源性色斑的志愿者,随机分成9组,每组30人;
2.2试验方法:给予所有志愿者相同的软毛牙刷,分别使用试验样品刷牙,早晚各刷一次,每次刷牙两分钟,并要求志愿者保持平时的其他口腔卫生习惯,日常饮食及吸烟等习惯。临床检查于基线、第四周、第八周进行,并且由同一名检查者完成。数据采用t检验进行统计分析,双侧检验,检验水准α为0.05,结果见表5所示。
2.3评价方法:牙齿色斑采用Lobene色斑指数进行评价,用0~3的数字来评价色斑部位的大小和色斑程度。
Lobene色斑指数分级计分:
0:无色斑存在;
1:色斑不超过牙面的1/3,轻度色斑(黄色或黄褐色);
2:色斑不超过牙面的2/3,中度色斑(中等程度的棕色);
3:色斑大于牙面的2/3,重度色斑(深棕色或黑色)。
3.试验结果:具体的试验结果见表5。

由表5可知,本发明制备的实施例1-4制的的牙膏具有显著的美白牙齿的效果,尤其以实施例1的美白效果最佳,因此,实施例1为本发明的最佳实施例。而对比例1-2,以及对比例9中分别去掉了肉桂提取液、蛇床子提取液、唾液乳杆菌,造成的美白效果显著降低,但是对比例10中,将唾液乳杆菌发酵液换成青春乳杆菌发酵液后,样品的美白效果并没有得到提升,说明并不是所有的益生菌能搭配肉桂和蛇床子提取物达到美白的效果,也说明了三者具有一定的协同作用,可以达到抗菌、除垢、美白的效果。而实施例1中的美白效果最佳,说明是在实施例1的比例下,三者的协同作用达到最大。
以上所述实施例仅表达了本发明的具体实施方式,其描述较为具体和详细,但并不能因此而理解为对本发明保护范围的限制。应当指出的是,对于本领域的普通技术人员来说,在不脱离本发明构思的前提下,其他任何未背离本发明的精神实质与原理下所作的改变、修饰、替代、组合、简化,均应为等效的置换方式,都包含在本发明的保护范围之内。
Claims (8)
1.一种有效祛除牙斑、牙垢的增白牙膏,其特征在于;其中包含肉桂酸提取物、蛇床子素提取物、唾液乳杆菌发酵液、甘油、山梨醇、丙二醇、二氧化硅、羧甲基纤维素钠、月桂醇硫酸酯钠、氟化钠、氟化钾或单氟磷酸钠、薄荷粉。
2.一种有效祛除牙斑、牙垢的增白牙膏的制备方法,其特征在于:S1、称取晾干的肉桂果,磨碎后加水没过物料,然后通过超声波处理机进行预处理后再添加去离子水,然后加热使体系保持沸腾状态,得到肉桂提取原液;S2、称取蛇床子的果实磨碎,取蛇床子花,加入水煎煮5次每次30-50min,合并滤液,加入乙醇静置,除去沉淀得滤液,然后用旋蒸仪挥去乙醇,加水定容至120mL,取55mL,加丙三醇40mL,连续萃取三次,取丙三醇层溶液备用;S3、按3%的接种量将唾液乳杆菌种子液接种于发酵罐中,37℃恒温培养,收集培养至稳定期的发酵液,发酵液中菌体浓度达到一定浓度后,将发酵液离心出去菌体,得到唾液乳杆菌发酵液;S4、取S1制备的肉桂提取液80-200ml,取S2制备的蛇床子提取液50-150ml,S3制备的唾液乳杆菌发酵液3-5mL,然后将其加入真空制膏机中,再计量称取适量的甘油、山梨醇和丙二醇加入真空制膏机中,混合搅拌20-40min;S5、称取10-15g的二氧化硅、20-25g的羧甲基纤维素钠、8-10g的月桂醇硫酸酯钠,0.3-0.8g的氟化钠、氟化钾或单氟磷酸钠混合均匀;S6、将制S5备好的粉料加入真空制膏机中和S4制备的液料混合,搅拌30min后,加入3-5g薄荷粉,同时打开制膏机的真空泵开始抽真空,真空搅拌30-50min后,关闭真空泵和真空制膏机搅拌,将牙膏灌装到软管中,用自制封尾机封尾即可。
3.根据权利要求2所述的一种有效祛除牙斑、牙垢的增白牙膏的制备方法,其特征在于:所述S1称取的肉桂果实为200-300g。
4.根据权利要求2所述的一种有效祛除牙斑、牙垢的增白牙膏的制备方法,其特征在于:所述S2称取的蛇床子的果实为10-15g。
5.根据权利要求2所述的一种有效祛除牙斑、牙垢的增白牙膏的制备方法,其特征在于:所述S2称取的蛇床子花为3-5g。
6.根据权利要求2所述的一种有效祛除牙斑、牙垢的增白牙膏的制备方法,其特征在于:所述S3中菌液的浓度达到2.0×109-3.0×109cfu/mL。
7.根据权利要求2所述的一种有效祛除牙斑、牙垢的增白牙膏的制备方法,其特征在于:所述S4选用的甘油为30-50ml、山梨醇为50-65ml、丙二醇为5-10ml。
8.权利要求2-6任一项所述的制备方法制备得到的牙膏。
