Abstract
The invention relates to A61K8, in particular to a whitening composition with anti-wrinkle and tightening functions and application thereof. Raw materials of the whitening composition with anti-wrinkle and tightening functions comprise an amide compound, a reduced coenzyme and a nutritional agent. The three substances are compounded, so that the generation of nicotinic acid can be obviously reduced, and the stability of nicotinamide is improved; the self structural stability of the reduced coenzyme is improved by adding a water-soluble low-viscosity liquid compound with the viscosity of 170-185 MPa.s and the surface tension of 68-75 mN/m; the ultraviolet light resistance of the system is further enhanced by controlling the composition components of the plant extract mixture in a certain proportion, and the whitening composition is not easy to separate out when being irradiated by long-term ultraviolet light, especially in essence and cream systems, flocculent substances and precipitates are not easy to generate. Through the synergistic effect of all substances in the system, the antioxidant capacity of the essence and the face cream is improved, so that the face cream has the effects of whitening and resisting aging, and can tighten skin and repair skin barriers.

Technical Field
The invention relates to A61K8, in particular to a whitening composition with anti-wrinkle and tightening functions and application thereof.
Background
Since the scholars find that the nicotinamide can whiten skin due to the effect of inhibiting melanin from transferring in 2002, the application of the nicotinamide in skin care products becomes a research hot spot.
The patent No. CN108113920B provides a slow-release whitening composite moisturizing facial cream matrix, a preparation method and application thereof, uses arbutin and nicotinamide to act together with other substances of a system, and adds lecithin to form a water-in-oil structure, so that active ingredients are slowly released, and long-term use is facilitated. The patent No. CN110742822B provides a skin whitening anti-aging composition, whitening anti-aging cream containing the same and a preparation method thereof, and nicotinamide, anthocyanin, ergothioneine and retinol palmitate are mixed, so that the capability of removing free radicals of skin is improved, and the falling of surface cells is accelerated.
Although nicotinamide is not easily decomposed by visible light and has higher light stability than ascorbic acid, it still has many problems, and is easily hydrolyzed in acid and alkali, so that it is limited in cosmetic application. And because nicotinamide has certain irritation, allergic reaction can occur when people with sensitive muscles use the nicotinamide, and the skin problem is aggravated.
Disclosure of Invention
In order to solve the above problems, the first aspect of the present invention provides a whitening composition having an anti-wrinkle, tightening function, the raw materials of which include an amide compound, a reduced coenzyme, and a nutritional agent.
As a preferable technical scheme of the invention, the amide compound is selected from one or more of niacinamide, fatty amide polyoxyethylene ether and asparagine.
Preferably, the amide compound is niacinamide.
The niacinamide not only has the whitening effect, but also can participate in the energy metabolism of the human body, and in the human body, after the niacinamide is converted, the niacinamide becomes Nicotinamide Adenine Dinucleotide (NAD), and then enters mitochondria after participating in the tricarboxylic acid circulation of the human body, so as to promote the energy release.
As a preferred embodiment of the present invention, the reduced coenzyme derivative is selected from one or more of a coenzyme Q10 derivative, a reduced coenzyme II derivative and a reduced coenzyme I derivative.
Preferably, the reduced coenzyme is a reduced coenzyme I derivative (nicotinamide adenine dinucleotide NAD).
Nicotinamide adenine dinucleotide is an indispensable role in cell activities such as energy synthesis and basal metabolism as a reduced coenzyme. At present, biological experiments show that the NAD concentration in a human body has a certain influence on cell aging, the concentration is reduced, and the cell aging process in the human body is accelerated.
As a preferred embodiment of the present invention, the nutrient contains at least one intramolecular hydrogen bond in the molecule.
As a preferred embodiment of the present invention, the nutritional agent is selected from one or more of glycoprotein, nicotinamide mononucleotide, and vitamin E.
Preferably, the nutritional agent is Nicotinamide Mononucleotide (NMN).
Further preferably, the Nicotinamide Mononucleotide (NMN) is a β -nicotinamide mononucleotide.
Nicotinamide Mononucleotide (NMN) is taken as a nutrient and participates in multi-step enzymatic reaction, thereby promoting NAD synthesis and improving metabolic homeostasis, and animal research experiments show that the nicotinamide mononucleotide has the capacity of protecting heart and anti-aging.
As a preferable technical scheme of the invention, the weight ratio of the amide compound, the reduced coenzyme and the nutritional agent is (1-3.5): (0.004-0.25): (0.004-0.25).
When nicotinamide is applied to cosmetics, certain hydrolysis often occurs to generate nicotinic acid, and the hydrolysis reaction not only causes that the nicotinamide has certain instability, but also can negatively influence other components of the system, and in addition, skin allergy can occur to some people when the nicotinamide is used due to the existence of the nicotinic acid. In order to solve the above problems, the inventors have found through a large number of experiments that niacin production can be significantly reduced and stability of niacinamide can be improved when niacinamide, a specific reduced coenzyme and a nutritional agent are compounded. The applicant speculates that this is probably due to the fact that hydrogen on the unsaturated carbon ortho to the amide bond on the pyridine ring on the nicotinamide molecule can form intermolecular hydrogen bonds with hydroxyl groups in the nutritional agent, and at the same time, the intramolecular hydrogen bonds of the nutritional agent and the formed intermolecular hydrogen bonds form a relatively tight connection structure, so that hydrolysis of the nicotinamide molecule is inhibited to a certain extent, and when the added nutritional agent is NMN, since N on the pyridine ring carries a large amount of positive charges, certain electrostatic effect exists between the N and the nicotinamide molecule and the reductive coenzyme, and the self stability of the N is further enhanced. In addition, the inventors have unexpectedly found that adding a specific proportion of a reduced coenzyme I derivative, which has a molecular chain entangled with a certain nicotinamide molecule and a nutrient molecule, improves the stability between the combined molecules and makes it less prone to hydrolysis at high temperatures. The above-mentioned effects are especially obtained in that the weight ratio of the amide compound, the reduced coenzyme and the nutritional agent is (1-3.5): (0.004-0.25): (0.004-0.25), the effect is particularly remarkable.
The second aspect of the invention provides application of a whitening composition with anti-wrinkle and tightening functions, which is applied to skin care products.
As a preferred embodiment of the present invention, the skin care product essence further comprises a low viscosity liquid compound.
Preferably, the low viscosity liquid compound has a viscosity of 170-185 mpa.s at 20 ℃.
Preferably, the low viscosity liquid compound has a surface tension of 68 to 75mN/m at 20 ℃.
mN/m is the liquid tension coefficient and represents the traction force of the adjacent part of the surface unit length. 1 mN/m=10 - 3 N/m, i.e., 0.001N/m.
Further preferably, the low viscosity liquid compound has water solubility.
The self structure of nicotinamide adenine dinucleotide added in the composition is fragile, so that clusters formed by nutrient molecules entangled on a chain and nicotinamide molecules are fragile when the composition is applied. In order to improve such vulnerability, the inventors have conducted a series of exploratory experiments, and finally found that the above problems can be improved by introducing a water-soluble low-viscosity liquid compound of a specific viscosity and a specific tension into the system. When the viscosity of the low-viscosity liquid compound is 170-185 MPa.s, the fluidity of the composition is changed, and the intermolecular spacing of the substances is changed, so that the intermolecular combined agglomeration is affected, and meanwhile, the specific surface tension of the low-viscosity liquid compound plays a role in binding molecules to a certain extent, so that the structural stability of the system is improved.
Preferably, the raw materials of the skin care product essence further comprise water dilution of polyalcohol, plant extract mixture, humectant, emulsifying chelating agent, astringent and preservative.
Preferably, the aqueous diluent of the polyol is an aqueous diluent of glycerol having a water content of 0.2 to 0.8 wt%.
Preferably, the plant extract mixture comprises plant extract and mixture. Wherein, the weight ratio of the plant extract to the mixture is (0.2-0.6): (2-5).
Further preferably, the plant extract is selected from one or more of chamomile extract, chrysanthemum extract.
Still more preferably, the plant extract is a chamomile extract.
Preferably, the particle size of the chamomile extract is 50-90 mesh.
Preferably, the mixture is selected from one or more of 1, 3-butanediol, phenoxyethanol, deionized water.
Further preferred mixtures are 1, 3-butanediol, phenoxyethanol and deionized water. In the mixture, the weight ratio of 1, 3-butanediol, phenoxyethanol and deionized water is (0.8-1.5): (0.4-0.9): (1-2).
In order to further reduce the irritation caused by the hydrolysis of nicotinamide in the whitening composition with anti-wrinkle and tightening functions, the inventor finds that the plant extract mixture added with the specific composition has a certain slow release property and reduces the irritation, and meanwhile, the inventor finds that when the composition components of the plant extract mixture are controlled in a certain proportion, the ultraviolet light resistance of a system can be further enhanced, and the whitening composition is not easy to separate out when being irradiated by long-term ultraviolet light, especially in an essence and cream system, flocculent substances and precipitates are not easy to generate. The triterpene compound contained in the golden chamomile extract in a specific proportion and the 1, 3-butanediol and phenoxyethanol which are diluted by water have a certain effect on the hydrolysis of the whitening composition, so that the irritation is reduced, the system stability is improved, and when the particle size of the golden chamomile extract is 50-90 meshes, the golden chamomile extract can better fill into molecular gaps, and the stability of essence and cream is improved.
Preferably, the humectant is selected from one or more of erythritol, xanthan gum, and sodium hyaluronate.
Further preferably, the humectant is a mixture of erythritol, xanthan gum and sodium hyaluronate, wherein the weight ratio of erythritol, xanthan gum and sodium hyaluronate is (0.4-1): (0.07-0.2): (0.01-0.04).

The preparation method of skin care product essence comprises mixing phase A, stirring at 65-70deg.C for 10-15min, and maintaining at 70-75deg.C for 2-5min to obtain phase A mixture; mixing the phase B, and stirring for 5-8min at 60-65 ℃ to obtain a phase B mixture; mixing the mixture A and the mixture B at 30-35deg.C, and stirring for 2-7min to obtain skin care product essence.
The third aspect of the invention provides application of a whitening composition with anti-wrinkle and tightening functions, which is applied to skin care product face cream.
As a preferred embodiment of the present invention, the skin care product cream further comprises a low viscosity liquid compound.
Preferably, the skin care product cream further comprises an aqueous dilution of polyhydric alcohol, a plant extract mixture, a humectant, an emulsifying chelating agent, an aqueous solution of 10-20wt% conditioning agent, deionized water, an astringent, cetostearyl alcohol, PEG-100 stearate, glyceryl stearate, isohexadecane, cyclopentadimethicone, cyclohexasiloxane, cetostearyl alcohol, cetostearyl glucoside, ethylhexyl palmitate, dimethicone, tocopheryl acetate, a preservative and a fragrance.
Further preferred, the conditioning agent is selected from one or more of polyacrylamide, C13-14 isoparaffin, C9-11 alkanol polyether-6.
Still more preferably, the conditioning agent is a polyacrylamide, a C13-14 isoparaffin, and a C9-11 alkanolamine polyether-6, wherein the weight ratio of polyacrylamide, C13-14 isoparaffin, and C9-11 alkanolamine polyether-6 is (0.1-1): (0.1-1): (0.05-0.4).
The raw materials of the skin care product cream are divided into a C phase and a D phase. The C phase and the D phase respectively comprise the following components in parts by weight:
and C phase: 1.5-5 parts of low-viscosity liquid compound, 1-3 parts of aqueous diluent of polyalcohol, 1-3 parts of whitening composition with anti-wrinkle and tightening functions, 0.8-1.3 parts of plant extract mixture, 0.5-1 part of humectant, 0.2-0.6 part of aqueous solution of conditioner and 72-77 parts of deionized water;
and D phase: 2-5 parts of cetostearyl alcohol, 1-2.5 parts of PEG-100 stearate, 1-2.5 parts of glycerol stearate, 1.5-2.5 parts of isohexadecane, 1-2 parts of cyclopentadimethicone, 1-2 parts of cyclohexasiloxane, 0.3-1.2 parts of cetostearyl alcohol, 0.5-1.2 parts of cetostearyl glucoside, 0.8-1.2 parts of ethylhexyl palmitate, 0.8-1.2 parts of dimethicone, 0.3-0.7 part of isononyl isononanoate, 0.1-0.4 part of phenoxyethanol, 0.1-0.4 part of ethylhexyl glycerol, 0.3-0.5 part of emulsifying chelating agent, 0.03-0.2 part of astringents, 0.1-0.3 part of preservative, 0.1-0.3 part of tocopheryl acetate and 0.005-0.015 part of essence.
The preparation method of skin care product cream comprises mixing phase C at 73-75deg.C, stirring for 10-15min to obtain phase C mixture; mixing the phase D at 58-60deg.C, and stirring for 5-8min to obtain phase D mixture; mixing the mixture of phase C and phase D at 80-85deg.C, stirring for 2-7min, and cooling to obtain skin care product cream.
Compared with the prior art, the invention has the following beneficial effects:
when nicotinamide, NAD and NMN are compounded, the generation of nicotinic acid can be obviously reduced, and the stability of the nicotinamide is improved; the self structural stability of nicotinamide adenine dinucleotide is improved by adding a water-soluble low-viscosity liquid compound with the viscosity of 170-185 MPa.s and the surface tension of 68-75 mN/m; the plant extract mixture comprises golden chamomile extract with the particle size of 50-90 meshes, 1, 3-butanediol, phenoxyethanol and deionized water, the ultraviolet light resistance of the system can be further enhanced, and the whitening composition is not easy to separate out when being irradiated by ultraviolet light for a long time, especially in essence and cream systems. Through the synergistic effect of all substances in the system, the antioxidant capacity of the essence and the face cream is improved, so that the face cream has the effects of whitening and resisting aging, and can tighten skin and repair skin barriers.
Detailed Description
Examples
The compositions of the examples were all prepared from commercially available materials, wherein the niacinamide was purchased from the company of Highway technologies, inc., reduced coenzyme I derivative had a CAS number of 53-84-9, beta-nicotinamide mononucleotide had a CAS number of 1094617, the low viscosity liquid compound was methylpropanediol, the methylpropanediol had a CAS number of 2163-42-0, the viscosity at 20℃of 178 MPa.s, the surface tension at 20℃of 72.2mN/m, the water solubility, the extract of Matricaria chamomilla (Chrysanthellum indicum) was purchased from the company of Volterray Biotechnology, lanzhou, the particle size of 80 mesh, the lubricant was PEG/PPG-15/15 polydimethylsiloxane, the star was model CE-20, the preservative was methylparaben, the solubilizing agent was PEG-40 hydrogenated castor oil, the model was emulsifier HEL-40, and the essence was of the floral type.
Example 1
The whitening composition with the anti-wrinkle and tightening functions comprises an amide compound, a reduced coenzyme and a nutritional agent.
The amide compound is niacinamide. The reduced coenzyme is a reduced coenzyme I derivative (nicotinamide adenine dinucleotide NAD). The nutrient contains 2 intramolecular hydrogen bonds in the molecule. The nutritional agent is Nicotinamide Mononucleotide (NMN). Nicotinamide Mononucleotide (NMN) is a β -nicotinamide mononucleotide.
The weight ratio of the amide compound, the reduced coenzyme and the nutritional agent is 2:0.01:0.01.
the example also provides application of the whitening composition with anti-wrinkle and tightening functions, and the whitening composition with anti-wrinkle and tightening functions is applied to skin care products.
The skin care product essence comprises a phase A and a phase B. The phase A and the phase B respectively comprise the following components in parts by weight:
phase A: 3 parts of low-viscosity liquid compound, 2 parts of aqueous diluent of polyalcohol (specifically, aqueous diluent of glycerin with water content of 0.5 wt%), 2.02 parts of whitening composition with anti-wrinkle and tightening functions, 0.8 part of plant extract mixture, 0.92 part of humectant, 0.6 part of 1,2 hexanediol, 0.3 part of aqueous solution of panthenol with water content of 10wt%, and 89.38 parts of deionized water;
the plant extract mixture comprises plant extract and mixture.
Wherein, the weight ratio of the plant extract to the mixture is 0.5:2.5. the plant extract is flos Matricariae Chamomillae extract. The mixture is 1, 3-butanediol, phenoxyethanol and deionized water. In the mixture, the weight ratio of 1, 3-butanediol, phenoxyethanol and deionized water is 1:0.5:1.
the humectant is a mixture of erythritol, xanthan gum and sodium hyaluronate, wherein the weight ratio of the erythritol, the xanthan gum and the sodium hyaluronate is 0.8:0.1:0.02.
and B phase: 0.3 parts of lubricant, 0.42 parts of emulsifying chelating agent, 0.1 parts of astringents (specifically allantoin), 0.15 parts of preservative, 0.008 parts of solubilizer and 0.002 parts of essence.
The emulsifying chelating agent is acrylic acid (esters) or C10-30 alkanol acrylate cross-linked polymer, triethanolamine and EDTA disodium, wherein the weight ratio of the acrylic acid (esters) or C10-30 alkanol acrylate cross-linked polymer, the triethanolamine and the EDTA disodium is 0.2:0.18:0.04.
the preparation method of skin care product essence comprises mixing phase A, stirring at 68deg.C for 10min, and maintaining at 75deg.C for 5min to obtain phase A mixture; mixing the phase B, and stirring for 5min at 60 ℃ to obtain a phase B mixture; mixing the mixture A and the mixture B at 34 ℃, and stirring for 7min to obtain the skin care product essence.
Example 2
This example provides a whitening composition having anti-wrinkle and tightening functions, as in example 1.
The example also provides application of the whitening composition with the anti-wrinkle and tightening functions, and the whitening composition with the anti-wrinkle and tightening functions is applied to skin care product face cream.
The raw materials of the skin care product cream are divided into a C phase and a D phase. The C phase and the D phase respectively comprise the following components in parts by weight:
and C phase: 3 parts of low-viscosity liquid compound, 2 parts of aqueous diluent of polyalcohol (specifically, aqueous diluent of glycerin with water content of 0.5 wt%), 2.02 parts of whitening composition with anti-wrinkle and tightening functions, 1 part of plant extract mixture, 0.73 part of humectant, 0.4 part of aqueous solution of 15wt% conditioning agent and 74.21 parts of deionized water;
the plant extract mixture comprises plant extract and mixture.
Wherein, the weight ratio of the plant extract to the mixture is 0.5:2.5. the plant extract is flos Matricariae Chamomillae extract. The mixture is 1, 3-butanediol, phenoxyethanol and deionized water. In the mixture, the weight ratio of 1, 3-butanediol, phenoxyethanol and deionized water is 1:0.5:1.
the humectant is a mixture of erythritol, xanthan gum and sodium hyaluronate, wherein the weight ratio of the erythritol, the xanthan gum and the sodium hyaluronate is 0.5:0.2:0.03.
the conditioning agent is polyacrylamide, C13-14 isoparaffin and C9-11 alkanol polyether-6, wherein the weight ratio of the polyacrylamide to the C13-14 isoparaffin to the C9-11 alkanol polyether-6 is 0.15:0.15:0.1.
and D phase: 3 parts of cetostearyl alcohol, 1 part of PEG-100 stearate, 1.5 parts of glycerol stearate, 2 parts of isohexadecane, 1 part of cyclopentadimethicone, 1 part of cyclohexasiloxane, 0.5 part of cetostearyl alcohol, 0.5 part of cetostearyl glucoside, 1 part of ethylhexyl palmitate, 1 part of dimethicone, 0.5 part of isononyl isononanoate, 0.2 part of phenoxyethanol, 0.2 part of ethylhexyl glycerol, 0.42 part of emulsifying chelating agent, 0.11 part of astringents (specifically allantoin), 0.2 part of preservative, 0.2 part of tocopheryl acetate and 0.01 part of essence.
The emulsifying chelating agent is acrylic acid (esters) or C10-30 alkanol acrylate cross-linked polymer, triethanolamine and EDTA disodium, wherein the weight ratio of the acrylic acid (esters) or C10-30 alkanol acrylate cross-linked polymer, the triethanolamine and the EDTA disodium is 0.23:0.15:0.04.
the preparation method of skin care product cream comprises mixing phase C at 75deg.C, stirring for 10min to obtain phase C mixture; mixing the phase D at 60 ℃ and stirring for 5min to obtain a phase D mixture; mixing the mixture of phase C and the mixture of phase D at 85deg.C, stirring for 5min, and cooling to obtain the skin care product cream.
Example 3
The present example provides a whitening composition having anti-wrinkle and tightening functions, unlike example 1, in which the weight ratio of the amide compound, the reduced coenzyme and the nutritional agent is 2:0.5:0.5.

Unlike example 1, the plant extract mixture comprises plant extract and mixture. Wherein, the weight ratio of the plant extract to the mixture is 0.4:4. the mixture is 1, 3-butanediol, phenoxyethanol and deionized water. In the mixture, the weight ratio of 1, 3-butanediol, phenoxyethanol and deionized water is 2:0.5:0.5.
example 6
This example provides a whitening composition having anti-wrinkle and tightening functions, as in example 1.
The example also provides application of the whitening composition with anti-wrinkle and tightening functions, and the whitening composition with anti-wrinkle and tightening functions is applied to skin care products. Unlike example 1, the low-viscosity liquid compound was 1, 2-propanediol, the viscosity at 20℃was 56 MPa.s, and the surface tension at 20℃was 38mN/m.
Performance test:
1. appearance stability test: the appearance of the skin care essences obtained in example 1, example 3, example 5 and example 6 was observed, the appearance of the skin care creams obtained in example 2 and example 4 was observed, and after the above substances were left standing for 28 days in an environment 1 of-5 ℃ and an environment 2 of 40 ℃ respectively, taken out, and after standing again for 12 hours at 25 ℃, the appearance was observed again, and the results are shown in table 1:

Ultraviolet stability test: the skin care product essences obtained in examples 1,3,5 and 6 were irradiated with an ultraviolet lamp tube, and after irradiation time of 12 hours at 150w, the ultraviolet lamp tube was turned off for 12 hours, which is one cycle, and after 20 cycles, the appearance of the skin care product essences was observed, and the results are shown in Table 2:

3. Whitening effect test: 100 persons whose skin was sensitive mixed skin were randomly selected, and were divided into 4 groups of 1 to 4, 25 persons each, using the skin care essence obtained in examples 1,3,5 and 6 at both cheeks of the face, respectively, group 1 using the skin care essence obtained in example 1, group 2 using the skin care essence obtained in example 3, and so on. The preparation is used once in the morning and evening every day for 28 days. After 28 days of continuous use, 100 people are led to score the whitening effect of the essence, the score is fully divided into 10 points, the higher the score is, the more obvious the whitening effect is, the average score of each group is taken, and the result is shown in Table 3:

1. A skin care product essence with anti-wrinkle and tightening functions is characterized in that raw materials of the skin care product essence comprise an amide compound, a reduced coenzyme and a nutritional agent; the amide compound is niacinamide, the reduced coenzyme is a reduced coenzyme I derivative, the nutritional agent is nicotinamide mononucleotide, and the weight ratio of the amide compound to the reduced coenzyme to the nutritional agent is (1-3.5): (0.004-0.25): (0.004-0.25);
the raw materials also comprise a low-viscosity liquid compound, wherein the low-viscosity liquid compound is methyl propylene glycol;
the raw materials also comprise plant extract mixture, wherein the plant extract mixture comprises plant extract and mixture, and the weight ratio of the plant extract to the mixture is (0.2-0.6): (2-5) the plant extract is golden chamomile extract, and the grain size of the golden chamomile extract is 50-90 meshes; in the mixture, the weight ratio of 1, 3-butanediol, phenoxyethanol and deionized water is (0.8-1.5): (0.4-0.9): (1-2);
