本发明公开了一种含岩藻黄质的提取物的制备方法,包括以下步骤:(1)将干海带切成小块并用饮用水清洗后浸润;(2)将饮用水浸润的海带切块加入食用酒精进行浸润;(3)将食用酒精浸润的海带切块加入芝麻油进行浸润;(4)在超声环境下对水、食用酒精、芝麻油浸润的海带切块进行提取;(5)分离提取液中的食用酒精,得到含岩藻黄质的提取物。本发明提供的制备方法,利用了醇水互溶的性质,使食用酒精快速浸润到海带中,再加入芝麻油进行超声提取,使岩藻黄质不断转移到油相中,实现对海带中岩藻黄质成分的快速提取。

技术领域
本发明属于从天然植物中提取有效成分的技术领域,涉及一种岩藻黄质的提取方法。
背景技术
岩藻黄质(fucoxanthin)亦称褐藻黄素,是自可食用褐藻中,如裙带菜(Undariapinnatifida)、海带(Laminaria japonica aresch)、和马尾藻(Sargassum fulvellum)等提取出来的天然类胡萝卜素。
岩藻黄质在干海带中仅占0.05%-0.2%。岩藻黄质是一种脂溶性成分,可捕捉光子参与海带的光合作用,岩藻黄质在常态下遇光、热、氧容易分解,稳定性较差,并且其疏水特性,导致岩藻黄质必需用非极性溶剂提取。
据发明人所知,岩藻黄质的提取工艺有无水乙醇-丙酮-石油醚(4∶1∶1)复合溶剂超声提取、丙酮提取、90%乙醇-丙酮(3∶1)复合溶剂65℃水浴提取、90%乙醇加压110℃加热提取、3%甲醇作为夹带剂超临界萃取等。其中甲醇属于无基因毒性但有动物致癌性的第二类溶剂;丙酮、石油醚中的戊烷属于虽然属于对人体低毒的第三类溶剂,但这些溶剂的长期毒性或致癌性受到消费者广泛关注。
综上目前岩藻黄质的提取工艺均不能从提取溶剂的残留毒性、热不稳定性、提取简便性全面综合地解决岩藻黄质的提取问题。
发明内容
本发明的实施例提供了一种含岩藻黄质的提取物的制备方法,包括以下步骤:
(1)将干海带切成小块并用饮用水清洗后浸润;
(2)将饮用水浸润的海带切块加入食用酒精进行浸润;
(3)将食用酒精浸润的海带切块加入芝麻油进行浸润;
(4)在超声环境下对水、食用酒精、芝麻油浸润的海带切块进行提取;
(5)分离提取液中的食用酒精,得到含岩藻黄质的提取物的油相。
在一些实施例中,所述将干海带切成小块并用饮用水清洗后浸润,包括:将干海带切成2-5cm的小段,用饮用水清洗并进行浸润,使海带吸水量为干海带质量的1.2-1.8倍。
在一些实施例中,所述将饮用水浸润的海带切块加入食用酒精进行浸润,包括:使用干海带质量的3倍体积的食用酒精进行浸润,食用酒精的浓度不低于90%,浸润时间20-40分钟,例如为95%的食用酒精。
在一些实施例中,所述将食用酒精浸润的海带切块加入芝麻油进行浸润,包括使用干海带质量0.5倍的芝麻油进行浸润,浸润的时间为30-50分钟。
在一些实施例中,所述超声环境的工艺条件为超声提取时间为40-80分钟,提取温度为30-50℃。
在一些实施例中,所述分离提取液中的食用酒精,得到含岩藻黄质的提取物,包括采用离心萃取机分离食用酒精和芝麻油,芝麻油相含岩藻黄质的提取物。
在一些实施例中,本发明所提供的提取物的制备方法,所述各步骤均在避光环境操作。
本发明的实施例还提供了一种前述任一实施例的制备方法制备的含岩藻黄质的提取物,所述含岩藻黄质的提取物分布在芝麻油中。
本发明的实施例还提供了前述实施例提供的含岩藻黄质的提取物在制备减肥食品、保健品或药品方面的用途。
具体实施方式
为了充分说明本发明提供的制备方法的具体实施过程、工艺条件等,如下提供了多个实施例对本发明的技术方案进行详细说明。如下所提供的用量、品种、规格、型号等仅为示意性的。
在下述实施例中,所称的饮用水是指满足国家规定的《生活饮用水卫生标准》(GB5749-2022)或其它等同标准的水,例如纯净水、矿泉水等。
在下述实施例中,所称的食用酒精是指满足GB31640-2016《食品安全国家标食用酒精》以谷物、薯类、糖蜜、或其他可食用农作物为主要原料,经发酵、蒸馏、精制而成的供食品工业食用的含水酒精。
在下述实施例中,所称的酒精浓度为体积比浓度。
实施例1
称取干海带100kg,用打碎机切成约2-5cm的小块,饮用水冲洗海带表面盐分及灰尘,再经沥水网带快速沥干水分,此时海带的质量为250kg(即吸水量150kg)。
将润洗后的海带放入2T容量超声提取罐中,喷洒300L食用酒精(浓度95%)搅拌均匀,密封浸润30分钟。
加入50kg芝麻油,搅拌均匀,密封浸润40分钟。
打开超声,利用夹层冷却水控制温度,使料液温度保持在30-50℃之间,超声提取60分钟。将食用酒精及将芝麻油混合液放出,经钛棒过滤,得到含岩藻黄质的提取物的食用酒精和芝麻油混合液。
将前述混合液用离心萃取机分离食用酒精和芝麻油,含岩藻黄质的提取物位于芝麻油相中。
所得油相经高效液相检测,含岩藻黄质0.40mg/g。
实施例2
称取干海带100kg,用打碎机切成约2-5cm的小块,饮用水冲洗海带表面盐分及灰尘,再经沥水网带快速沥干水分,此时海带的质量为230kg(即吸水量130kg)。
将润洗后的海带放入2T容量超声提取罐中,喷洒300L食用酒精(浓度90%)搅拌均匀,密封浸润40分钟。
加入50kg芝麻油,搅拌均匀,密封浸润50分钟。
打开超声,利用夹层冷却水控制温度,使料液温度保持在30-50℃之间,超声提取50分钟。将食用酒精及将芝麻油混合液放出,经钛棒过滤,得到含岩藻黄质的提取物的食用酒精和芝麻油混合液。
将前述混合液用离心萃取机分离食用酒精和芝麻油,含岩藻黄质的提取物位于芝麻油相中。
所得油相经高效液相检测,含岩藻黄质0.39mg/g。
实施例3
减肥乳
制备方法:
(1)取本实施例1所得含岩藻黄质的提取物置鱼干燥乳钵中,加阿拉伯胶粉研磨均匀。
(2)按照油:水:胶(4:2:1)的比例首次加入纯化水6.5ml,迅速沿一个方向研磨,直至产生“噼啪”的乳剂劈裂声,即形成初乳。
(3)用纯化水将初乳分次转移至代刻度的烧杯中,加水至50ml,搅拌均匀即得减肥乳。
实施例4
减肥乳
处方:
岩藻黄质提取物 16ml
聚山梨酯-80 8ml
纯化水 适量
共至制100ml
制备方法:
(1)取本实施例1含岩藻黄质的提取物与聚山梨酯-80共置干燥乳钵中,研磨均匀,加入纯化水11ml研磨,形成初乳。
(2)用纯化水将初乳分次转移至代刻度的烧杯中,加水至100ml,搅拌均匀即得减肥乳。
实施例5
自制凉拌减肥黄瓜
按照凉拌黄瓜的做法,最后加入本实施例1含岩藻黄质的提取物8-10ml,搅拌均匀。
每日食用1次,连用1周以上即有明显的减肥效果。
对比实施例1
取干海带100g,切成约2-5cm的小块,用饮用水清洗海带表面盐分及灰尘,快速取出沥干水分,加入色拉油约400g,将海带浸没,超声提取60分钟,经滤纸过滤,得含岩藻黄质的提取物。
经高效液相检测,所得产物中含岩藻黄质仅为0.003mg/g。
与相关技术中的制备方法相比,本发明提供的制备方法,以含有岩藻黄质的海带为原料,将其用饮用水清洗浸润,除尽外表盐分,加入浓度高于90%的食用酒精中浸润,再加芝麻油浸润,全程避光,再用超声提取得到含岩藻黄质的芝麻油和食用酒精的混合物,用分液装置将食用酒精分离,得到含岩藻黄质的提取物。
所提供的制备工艺至少具有如下技术进步中一个或多个:
1、海带生活在海水中,对芝麻油的浸润性差,但对水的浸润性好,本发明采用水浸润,利用醇水互溶的性质,采用90%以上食用酒精浸润含水海带,使食用酒精快速浸润到海带中,再加入芝麻油进行提取,利用岩藻黄质在芝麻油中溶解度远远大于在食用酒精中的溶解度,将岩藻黄质萃取到芝麻油中。解决了芝麻油对海带难以浸润的问题。
2、本发明使用的饮用水、食用酒精和芝麻油,均属于可食用的无毒溶剂,在原理上保证了提取物的安全性。
3、超声提取,可以在较低温度下,使岩藻黄质快速溶解到芝麻油中,减少热量对岩藻黄质的破坏,提取效率高。
4、芝麻油中氧含量极低,油相可以起到避氧的作用,芝麻油中含芝麻素、芝麻酚、芝麻酚林等有脂溶性抗氧化物质,两者共同作用减少岩藻黄质在提取及贮存中的分解。
5、本含有岩藻黄质的提取物直接作为一种食品与其他食品调配食用,避免了其他提取方法提取后的浓缩、分离、纯化过程,降低了后续处理难度,降低了成本。
6、本含有岩藻黄质的提取物浓度高达0.4mg/g,由于岩藻黄质的活性非常高,人体服用3mg/天,即可达明显的减肥效果,因此每天服用7.5ml含岩藻黄质的提取物即可达到减肥效果。
7、食用酒精和芝麻油混合液中,食用酒精和芝麻油密度相差不大,采用离心萃取机分离提高了分离效果及效率。
8、提取、过滤、罐装过程可以全程避光,可起到避光作用。
Claims (9)
1.一种含岩藻黄质的提取物的制备方法,其特征在于,包括以下步骤:
(1)将干海带切成小块并用饮用水清洗后浸润;
(2)将饮用水浸润的海带切块加入食用酒精进行浸润;
(3)将食用酒精浸润的海带切块加入芝麻油进行浸润;
(4)在超声环境下对水、食用酒精、芝麻油浸润的海带切块进行提取;
(5)分离提取液中的食用酒精,得到含岩藻黄质的提取物。
2.根据权利要求1所述的提取物的制备方法,其特征在于,所述将干海带切成小块并用饮用水清洗后浸润,包括:将海带切成2-5cm的小段,用饮用水清洗并进行浸润,使海带吸水量为干海带质量的1.2-1.8倍。
3.根据权利要求1所述的提取物的制备方法,其特征在于,所述将饮用水浸润的海带切块加入食用酒精进行浸润,包括:使用干海带质量的3倍体积的食用酒精进行浸润,食用酒精的浓度不低于90%,浸润时间20-40分钟。
4.根据权利要求1所述的提取物的制备方法,其特征在于,所述将食用酒精浸润的海带切块加入芝麻油进行浸润,包括使用干海带质量0.5倍的芝麻油进行浸润,浸润的时间为30-50分钟。
5.根据权利要求1所述的提取物的制备方法,其特征在于,所述超声环境的工艺条件为超声提取时间为40-80分钟,提取温度为30-50℃。
6.根据权利要求1所述的提取物的制备方法,其特征在于,所述分离提取液中的食用酒精,得到含岩藻黄质的提取物,包括采用离心萃取机分离食用酒精和芝麻油,芝麻油相含岩藻黄质的提取物。
7.根据权利要求1所述的提取物的制备方法,其特征在于,所述各步骤均在避光环境操作。
8.权利要求1-7任一所述的制备方法制备的含岩藻黄质的提取物,所述含岩藻黄质的提取物分布在芝麻油中。
9.权利要求8所述含岩藻黄质的提取物在制备减肥食品、保健品或药品方面的用途。
