Abstract
本发明属于生物领域,具体涉及一种用于治疗与TGFβ1表达相关的疾病的siRNA。首先公开了一种抑制TGFβ1基因表达的siRNA,所述siRNA包括一条正义链和一条反义链,所述正义链包括与所述反义链互补配对的互补性区域,所述反义链包括与所述正义链互补配对的互补性区域,其中所述正义链选自与SEQID NO:1‑SEQ ID NO:129中任意一条的核苷酸序列区别不多于5个核苷酸的核苷酸序列,所述反义链选自与SEQ ID NO:130‑SEQ ID NO:258中任意一条的核苷酸序列区别不多于5个核苷酸的核苷酸序列。本发明发现了新的靶向于TGFβ1基因的siRNA,可以有效的抑制TGFβ1的表达,高效特异的命中靶标。包含该siRNA的药物组合物可用于治疗增生性瘢痕、纤维化或肿瘤等疾病。

技术领域
本发明属于生物领域,具体涉及一种用于治疗与TGFβ1表达相关的疾病的 siRNA。
技术背景
增生性瘢痕的病理基础是以成纤维细胞为主的细胞成分的过度增殖和以胶原为主的细胞外基质的过度沉积。其特点是损伤后炎症反应延长,导致局部成纤维细胞的血管化、细胞增生和过量胶原沉积。致病因素主要包括:代谢异常、细胞因子、低氧和自由基、基因水平变化。目前国内外治疗瘢痕多采用局部药物封闭,皮肤移植,手术切除植皮,晶体磨疤术,气囊扩张术,抗体注射,放射治疗,压迫疗法,激素疗法,硅酮类等疗法。
肿瘤的治疗方法是很多的,肿瘤分良性肿瘤和恶性肿瘤,良性肿瘤的主要治疗方法是手术治疗,经过手术治疗良性肿瘤是可以彻底治愈的,良性肿瘤也不容易复发转移。恶性肿瘤就是我们平常说的癌症。治疗的主要方法是手术治疗化疗,放疗免疫治疗,靶向药物治疗,生物治疗干细胞移植治疗,pd1治疗等等。配合中医中药治疗,中西医结合治疗效果也是比较好的。
纤维化(fibrosis)是一个医学概念,纤维化可发生于多种器官,主要病理改变为器官组织内纤维结缔组织增多,实质细胞减少,持续进展可致器官结构破坏和功能减退,乃至衰竭,严重威胁人类健康和生命。
然而现有的治疗增生性瘢痕、肿瘤或纤维化的技术在临床上都不是很成熟,疗效有待观察。而且有的所需时间长,痛苦大,容易反复,出现不良后果等,许多都不尽如人意。
发明内容
本发明发现了一种新的方法,该方法以TGFΒ1为靶点,设计小干扰核苷酸(siRNA)双链,并从中选择最有效的siRNA,将这些选定的siRNA进行修饰,或与载体(如分支肽、LNP、Galnac等)制备成纳米颗粒制剂,皮下注射治疗增生性瘢痕、肿瘤或纤维化。
具体的,本发明的技术方案如下:
本发明第一个方面公开了一种抑制TGFβ1基因表达的siRNA,所述siRNA 包括一条正义链和一条反义链,所述反义链包括与所述正义链互补配对的互补性区域,所述正义链包括与所述反义链互补配对的互补性区域,所述正义链选自与SEQ ID NO:1-SEQ ID NO:129中任意一条的核苷酸序列区别不多于5个核苷酸的核苷酸序列,所述反义链选自与SEQID NO:130-SEQ ID NO:258中任意一条的核苷酸序列区别不多于5个核苷酸的核苷酸序列。
本发明第二个方面公开了一种TGFβ1基因沉默试剂盒,所述试剂盒包括上述的siRNA。
本发明第三个方面公开了一种抑制TGFβ1表达的siRNA在制备治疗增生性瘢痕、纤维化或肿瘤的药物组合物中的应用。
本发明第四个方面公开了一种药物组合物,所述药物组合物包含有效量的上述的siRNA。
优选的,所述药物组合物为纳米颗粒制剂。
更优选的,所述纳米颗粒制剂包括siRNA与药物组合物载体。
本发明第五个方面公开了上述的药物组合物在制备用于治疗和/或改善与 TGFβ1表达相关的疾病的药物中的应用。

实施例中所用仪器、设备、试剂均可通过各种渠道获得,例如购买得到,或可以制备而得。
实施例1
本实施例公开了siRNA体外活性检测实验方法,包括以下步骤:
细胞系选用NSFB或A549,对于NSFB细胞铺板细胞数:25000cells/孔,转染前24h铺板,转染siRNA终浓度0.1nM和10nM,所述siRNA的序列如表1 所示。转染试剂RNAiMAX,转染时间48h。48h后提取RNA、反转录(500ng)、稀释10倍后qPCR检测TGFβ1表达水平。细胞系选用A549,对于A549细胞铺板细胞数:50000cells/孔,转染前24h铺板,转染终浓度0.1nM和10nM,转染试剂RNAiMAX,转染时间24h。24h后提取RNA、反转录(500ng)、稀释10 倍后qPCR检测TGFβ1表达水平,qPCR检测时以人PPIB为内参基因,引物序列如下表2所示。

Claims (10)
1.一种用于抑制TGFβ1基因表达的siRNA,其特征在于,所述siRNA包括一条正义链和一条反义链,所述反义链包括与所述正义链互补配对的互补性区域,所述正义链包括与所述反义链互补配对的互补性区域,所述正义链选自与SEQ ID NO:1-SEQ ID NO:129中任意一条的核苷酸序列区别不多于5个核苷酸的核苷酸序列,所述反义链选自与SEQ ID NO:130-SEQ ID NO:258中任意一条的核苷酸序列区别不多于5个核苷酸的核苷酸序列。
2.一种TGFβ1基因沉默试剂盒,其特征在于,所述试剂盒包括权利要求1所述的siRNA。
3.一种抑制TGFβ1基因表达的siRNA在制备治疗增生性瘢痕、纤维化或肿瘤的药物组合物中的应用。
4.一种药物组合物,其特征在于,所述药物组合物包含有效量的权利要求1所述的siRNA。
5.根据权利要求4所述的药物组合物,其特征在于,所述药物组合物包含药学上可接受的赋形剂。
6.根据权利要求5所述的药物组合物,其特征在于,所述药物组合物为纳米颗粒制剂。
7.根据权利要求6所述的药物组合物,其特征在于,所述纳米颗粒制剂包括siRNA与药物组合物载体。
8.根据权利要求5-7任一项所述的药物组合物在制备用于治疗和/或改善与TGFβ1表达相关的疾病的药物中的应用。
9.根据权利要求8所述的应用,其特征在于,所述疾病为增生性瘢痕、纤维化或肿瘤。
10.一种产品,所述产品包括权利要求5-7任一项所述的药物组合物。
