TNF-alpha regulates inflammatory response by activating two inflammatory signal pathways of nuclear transcription factor kB (NF-kB) and activated protein 1(AP-1), is an important inflammatory factor and participates in pathological injury of certain autoimmune diseases.
Bacterial Lipopolysaccharide (LPS) is combined with a macrophage surface antigen recognition receptor to induce macrophages and secrete various cytokines such as TNF-alpha and the like. TNF-alpha can activate three signal paths of Caspase protease, JNK and transcription factor NF-kB, realize biological functions of cytotoxicity, antivirus, immunoregulation, apoptosis and the like, and is one of important inflammatory factors. LPS induces RAW264.7, is a classical cell model for researching inflammatory factors, and evaluates the effect of the test object in inhibiting the secretion of TNF-alpha by comparing the difference of the content of TNF-alpha secreted by RAW264.7 after the negative control and the test object are administrated. The TNF-alpha content is measured by an enzyme-linked immunosorbent assay (ELISA) according to the following specific principle: after the TNF-alpha is specifically combined with the TNF-alpha antibody coated on the enzyme label plate, the TNF-alpha is combined with the anti-TNF-alpha antibody with a substrate label, a colored product is generated after the substrate is catalyzed by enzyme, and the content of the TNF-alpha is in positive correlation with the color depth of the colored product. And measuring an Optical Density (OD) value by using a microplate reader at the wavelength of 450nm, and calculating the content of the TNF-alpha. The rate of TNF-. alpha.inhibition was then calculated according to the following formula.
The inhibition rate (%) - (1-T/C) × 100%, wherein T is the average content of TNF-alpha in the test substance, and C is the average content of TNF-alpha in the negative control under the action of LPS.
The specific test results are shown in table 1.

As can be seen from Table 1, the soothing compositions prepared in the examples of the present invention have a good effect of inhibiting the content of TNF- α.
Example 4
The soothing mask comprises the following components in percentage by mass: 3% of glycerin, 2% of butanediol, 0.5% of betaine, 0.05% of trehalose, 0.05% of xanthan gum, 0.15% of sodium hyaluronate, 0.05% of hydroxyethyl cellulose, 1% of the soothing composition prepared in example 2, 0.5% of hexanediol, 0.5% of p-hydroxyacetophenone, 0.001% of essence and the balance of water.
The preparation method comprises the following steps: putting water and hydroxyethyl cellulose into an emulsifying pot, heating to 85 ℃ under stirring, and homogenizing for 1-3 minutes; uniformly wetting glycerol and butanediol with xanthan gum and sodium hyaluronate in advance, and then putting into an emulsifying pot; putting betaine, trehalose, hexanediol and p-hydroxyacetophenone into an emulsifying pot; starting homogenizing for 1-3 min; stirring at 85 deg.C for 30 min, slowly cooling to 45 deg.C, adding the soothing composition and essence, stirring, and discharging to obtain semi-finished product. And weighing 25g of the semi-finished product, filling the semi-finished product into a mask bag containing mask cloth, and carrying out heat sealing to obtain the allergy-relieving mask.
Test 2 trial test in volunteers
31 volunteers were called out and the soothing mask prepared in example 4 was subjected to a volunteer trial test.
And (3) inclusion standard: the face is red, itching, stabbing and the like.
The using method comprises the following steps: after the face is cleaned by clear water, the mask is covered on the face for 15-30 minutes, the mask is slightly removed, and then the mask is cleaned by clear water, the mask is respectively used for 1 time in the morning and at night, and the observation results are obtained after the mask is continuously used for 1 month, and the statistics are shown in the following table 2.
The criteria for efficacy evaluation are as follows:
the effect is shown: the face is reddish, the itch and the stabbing pain disappear completely;
the method has the following advantages: redness, itching, reduced stinging of the face;
and (4) invalidation: the face was reddened, itchy, and tingling had no detail or was more severe.

As can be seen from table 2, the soothing mask prepared in the examples of the present invention produced good soothing effect.

Claims (7)

1. The soothing composition is characterized by comprising the following components in parts by mass: oat bran extract 2-20 parts, large-leaf gentian root extract 10-50 parts, and selaginella tamariscina extract 1-5 parts.
2. A soothing composition according to claim 1, wherein the soothing composition comprises the following components in parts by weight: 10 parts of oat bran extract, 30 parts of gentiana macrophylla extract and 3 parts of selaginella tamariscina extract.
3. The soothing composition of claim 1, wherein the extract of gentiana macrophylla comprises a combination of extracts of roots, leaves and flowers of gentiana macrophylla.
4. A soothing composition according to any of claims 1-3, wherein the soothing composition is prepared by a process comprising the steps of:
(1) respectively weighing the following raw materials in parts by weight and crushing: oat bran 2-20 parts, large-leaf gentian root 5-30 parts, large-leaf gentian leaf 2-10 parts, large-leaf gentian flower 3-10 parts, and selaginella tamariscina herb 1-5 parts;
(2) soaking oat bran in 90% V/V ethanol water solution, wherein the feed-liquid ratio is 1: 8-1: 15, adjusting the pH value to be weakly acidic, and stirring and extracting to obtain an extracting solution A;
(3) adding water into the extracting solution A to reduce the ethanol concentration to 70% V/V to obtain a diluent B; mixing radix Gentianae Marcrophyllae, folium Gentianae Marcrophyllae, flos Gentianae Marcrophyllae, and herba Selaginellae, adding appropriate amount of diluent B, soaking overnight, and transferring into percolation tank; eluting with the rest diluent B until the eluate is drained to obtain eluate, and concentrating by rotary evaporation to remove ethanol to obtain concentrated solution;
(4) loading the concentrated solution onto AB-8 macroporous resin column, eluting with 80% V/V ethanol water solution, collecting eluate 4-5BV, and concentrating by rotary evaporation to dry to obtain powder.
5. A soothing composition according to claim 4, wherein in step (2) the pH is in the range of 4 to 5; the temperature for stirring and extracting is 55-65 deg.C, and the extraction time is 3-5 hr.
6. A soothing composition according to claim 4, wherein the elution in step (3) is at a flow rate of 1-2 BV/h.
7. A soothing composition according to claim 4, wherein the elution in step (4) is at a flow rate of 1-2 BV/h.
