Abstract
本发明公开了一种放射性核素标记生长抑素受体拮抗剂及其制备方法,所述放射性核素标记的生长抑素受体拮抗剂为64Cu‑NODA‑JR11,制备方法如下:生长抑素受体拮抗剂JR11的NOTA修饰;对NODA‑JR11进行64Cu核素标记,即得反应液;用PD‑10柱对反应液进行纯化,得到64Cu‑NODA‑JR11。本发明所述的一种放射性核素标记生长抑素受体拮抗剂及其制备方法,合成了新型的放射性核素标记生长抑素受体拮抗剂,制备方法简单,能够扩大产量,让更多的患者使用,用于显像时图像的分辨率大大提高,且能够与肿瘤SSTR2抗原特异性结合,对SSTR有更高的亲和力和功能活性,实现肿瘤的靶向分子影像诊断目的,拓宽了以奥曲肽为代表的生长抑素类似物显像剂和治疗药物的应用范围。
Description
技术领域
本发明涉及临床核医学技术领域,特别涉及一种放射性核素标记生长抑素受体拮抗剂及其制备方法。
背景技术
随着核医学的发展,分子影像特别是正电子发射计算机断层显像(PET)的应用在肿瘤诊断和治疗方面显示出了巨大的和潜能优势。神经内分泌瘤是一组起源于肽能神经元和神经内分泌细胞的异质性肿瘤,大多数神经内分泌肿瘤细胞表达生长抑素受体(SSTR),目前正电子放射性核素68Ga标记生长抑素类似物用于神经内分泌肿瘤显像在临床上应用广泛,其有助于肿瘤的早期诊断及预后评估等;生长抑素在中枢内分泌系统和脑外组织分布广泛,是调节组织内外分泌的重要环肽激素家族,通过与靶细胞的SSTR结合发挥作用。肿瘤细胞组织上常有一些SSTR的过度表达,如在神经内分泌肿瘤(胃肠胰神经内分泌肿瘤、垂体瘤、嗜铬细胞瘤等)和神经系统肿瘤(神经母细胞瘤、脑膜瘤等)中都有SSTR的过表达。研究表明生长抑素及其类似物具有抗肿瘤活性,不仅能阻滞肿瘤细胞周期,诱导肿瘤细胞调亡,还能抑制肿瘤血管生成并拮抗其促肿瘤生长的作用。
经典的生长抑素类似物如奥曲肽(Octreotide)等,已广泛用于神经内分泌肿瘤、胃肠道肿瘤等生长抑素受体阳性肿瘤的诊断和治疗。但该类生长抑素受体激动剂在临床应用上也有局限性,放射性核素标记的奥曲肽在显像研究中显示出在肝脏、脾、胰腺有较高摄取,影响对该区域SSTR阳性肿瘤的诊断。故此,我们提出了一种放射性核素标记生长抑素受体拮抗剂及其制备方法。
发明内容
本发明的主要目的在于提供一种放射性核素标记生长抑素受体拮抗剂及其制备方法,可以有效解决背景技术中的问题。
为实现上述目的,本发明采取的技术方案为:
一种放射性核素标记生长抑素受体拮抗剂,所述放射性核素标记的生长抑素受体拮抗剂为64Cu-NODA-JR11。
一种放射性核素标记生长抑素受体拮抗剂的制备方法,包括以下步骤:
S1:生长抑素受体拮抗剂JR11的NOTA修饰;
S2:对NODA-JR11进行64Cu核素标记,即得反应液;
S3、用PD-10柱对反应液进行纯化,得到64Cu-NODA-JR11。
优选的,所述S1具体为:对JR11多肽p-Cl-Phe端的短链伯胺进行蛋白修饰,用1,4,7-叠氮酸钠-N,N',N”-三羧酸修饰后,得到NODA-JR11。
优选的,所述S2具体为:向NODA-JR11中依次加入0.1M NaAc和185MBq64CuCl2进行反应,即得64Cu-NODA-JR11。
优选的,所述NaAc的pH=5.5,反应在37℃下进行30-40min。
优选的,在所述S3中,所述PD-10柱使用前用需要经过去金属离子化的0.01M PBS缓冲液淋洗。
优选的,所述PBS溶液的量浓度为0.03M/L,pH为7.4。
优选的,所述64Cu-NODA-JR11的放射化学纯度大于95%。
与现有技术相比,本发明具有如下有益效果:
1、在本发明的制备方法合成了新型的放射性核素标记生长抑素受体拮抗剂,相比68Ga标记,64Cu标记的优势是能够扩大产量,让更多的患者使用,且用于显像时图像的分辨率大大提高,并能够与肿瘤SSTR2抗原特异性结合,分别通过红外/核医学手段准确定位SSTR2高表达的组织,实现肿瘤的靶向分子影像诊断目的,达到肿瘤早发现、早诊断、早治疗的目标。
2、在本发明合成的分子探针对SSTR有更高的亲和力和功能活性,新型生长抑素拮抗剂的合成及分子影像研究,拓宽了以奥曲肽为代表的生长抑素类似物显像剂和治疗药物的应用范围,经进一步药物使用临床试验评价后,其有望成为具良好前景的生长抑素拮抗剂显像剂。

实施例
一种放射性核素标记生长抑素受体拮抗剂,所述放射性核素标记的生长抑素受体拮抗剂为64Cu-NODA-JR11。
一种放射性核素标记生长抑素受体拮抗剂的制备方法,如图1所述,包括以下步骤:
S1:生长抑素受体拮抗剂JR11的NOTA修饰,具体为:对JR11多肽p-Cl-Phe端的短链伯胺进行蛋白修饰,用1,4,7-叠氮酸钠-N,N',N”-三羧酸修饰后,得到NODA-JR11;
S2:对NODA-JR11进行64Cu核素标记,即得反应液,具体为:向NODA-JR11中依次加入0.1M NaAc和185MBq 64CuCl2进行反应,即得64Cu-NODA-JR11,所述NaAc的pH=5.5,反应在37℃下进行30-40min;
S3、用PD-10柱对反应液进行纯化,得到64Cu-NODA-JR11,所述PD-10柱使用前用需要经过去金属离子化的0.01M PBS缓冲液淋洗,所述PBS溶液的量浓度为0.03M/L,pH为7.4,所述64Cu-NODA-JR11的放射化学纯度大于95%。
在本发明制备方法合成了新型的放射性核素标记生长抑素受体拮抗剂,相比68Ga标记,64Cu标记的优势是能够扩大产量,让更多的患者使用,且用于显像时图像的分辨率大大提高,并能够与肿瘤SSTR2抗原特异性结合,分别通过红外/核医学手段准确定位SSTR2高表达的组织,实现肿瘤的靶向分子影像诊断目的,在同一种分子上进行多种分子影像探针的研究,以达到肿瘤早发现、早诊断、早治疗的目标,本发明合成的分子探针对SSTR有更高的亲和力和功能活性,新型生长抑素拮抗剂的合成及分子影像研究,拓宽了以奥曲肽为代表的生长抑素类似物显像剂和治疗药物的应用范围,经进一步药物使用临床试验评价后,其有望成为具良好前景的生长抑素拮抗剂显像剂。

S1:生长抑素受体拮抗剂JR11的NOTA修饰;
S2:对NODA-JR11进行64Cu核素标记,即得反应液;
S3、用PD-10柱对反应液进行纯化,得到64Cu-NODA-JR11。
3.根据权利要求1所述的一种放射性核素标记生长抑素受体拮抗剂的制备方法,其特征在于:所述S1具体为:对JR11多肽p-Cl-Phe端的短链伯胺进行蛋白修饰,用1,4,7-叠氮酸钠-N,N',N”-三羧酸修饰后,得到NODA-JR11。
