Abstract
本发明的课题在于,提供一种不经由ES细胞或iPS细胞等多能性干细胞而直接(通过直接重编程)得到雪旺氏细胞的方法。作为解决所述的课题的手段,提供一种制备雪旺氏细胞的方法,其包含在哺乳动物的体细胞中导入选自由SOX10基因及KROX20基因构成的组中的至少1种基因或其表达产物的工序。
Description
本发明专利申请是针对申请日为2015年09月11日、申请号为201580049009.4、发明名称为“雪旺氏细胞及其制备方法”的申请提出的分案申请。
技术领域
本发明主要涉及一种雪旺氏细胞及其制备方法,详细而言,涉及一种通过直接重编程来制备雪旺氏细胞的方法。
背景技术
认为雪旺氏细胞在神经的再生中起到决定性的重要作用。与神经缺损或雪旺氏细胞的功能缺陷关联的疾病也多,对于这些疾病,期待如果可以移植自身雪旺氏细胞,则成为理想的再生医疗。实际上,对于外伤或恶性肿瘤切除引起的神经损伤,自身神经移植、或从自身神经中将雪旺氏细胞进行分离、培养而移植的治疗提高效果。但是,神经的采集对患者的侵袭大,无论如何也不能避免二次的神经损伤。另外,可以供给的雪旺氏细胞的数大多不充分。
在非专利文献1及2中,公开有如下方法:将以未分化脂肪干细胞(ADSC)(也称为源自脂肪的间质细胞)为代表的间充质干细胞作为材料,进行雪旺氏细胞样的细胞(dADSC)的分化。但是,在方法的性质上,具有来自外部的感染的危险性,因此,存在品质管理不容易、花费成本和人力的问题。进而也指出:得到的细胞的形质或功能与真的雪旺氏细胞不同。另外,没有报道有用这些方法制作的雪旺氏细胞有髓磷脂化能力,有可能不能贡献于跳跃传导。
最近,显示心肌细胞或肝细胞等可以由成纤维细胞直接诱导(直接重编程或直接转换)。如果可以由能够从患者中以低侵袭采集的成纤维细胞等体细胞直接制造雪旺氏细胞,则侵袭低且癌化的危险性也低,可以期待关系到制造新的移植用自身雪旺氏细胞的技术。
关于在体细胞中导入组织特异性的转录因子的基因组、可以不经过iPS细胞而直接分化诱导为该组织细胞(直接重编程(直接转换)),例如有以下的报道:
小鼠成纤维细胞→软骨细胞(导入SOX9+Klf4+c-Myc基因)
小鼠成纤维细胞→心肌细胞(导入GATA4+Mef2c+Tbx5基因)
小鼠成纤维细胞→肝细胞(Hnf4α+(导入Foxa1或Foxa2或Foxa3)基因)
小鼠成纤维细胞→神经干细胞(导入Sox2+FoxG1基因等)、小鼠、人细胞→造血干细胞等。
但是,没有将体细胞直接转换为雪旺氏细胞的报道。

发明内容
发明所要解决的课题
本发明的目的在于,提供一种可以应用于对与神经缺损或雪旺氏细胞的功能缺陷有关的疾病等的治疗、制备癌化的危险性少的雪旺氏细胞的方法。
用于解决课题的技术方案
本发明发现:通过在哺乳动物的体细胞中组合特定的基因而导入,不经由ES细胞或iPS细胞等多能性干细胞直接(通过直接重编程)可得到雪旺氏细胞。
本发明包含以下的方案。
项1、一种制备雪旺氏细胞的方法,其包含在哺乳动物的体细胞中导入选自由SOX10基因及KROX20基因构成的组中的至少1种基因或其表达产物的工序。
项2、根据项1所述的方法,其中,所述基因为SOX10基因及KROX20基因的组合。
项3、根据项1或2所述的方法,其中,所述体细胞为成纤维细胞、血管内皮细胞或间充质干细胞。
项4、一种雪旺氏细胞,其源自于哺乳动物的体细胞,具有选自由外源性的SOX10基因及KROX20基因构成的组中的至少1种基因或其表达产物。
项5、一种移植材料,其用于治疗基于神经的缺损或者雪旺氏细胞的缺损、不足或功能降低的疾病,包含用项1~3中任一项所述的方法得到的细胞、或项4所述的雪旺氏细胞。
项6、一种用于制备雪旺氏细胞的组合物,其包含选自由SOX10基因及KROX20基因构成的组中的至少1种基因或其表达产物。
发明效果
本发明中,可以通过直接重编程在短期间内由体细胞提供雪旺氏细胞。该雪旺氏细胞可以由进行移植的本人的体细胞容易地诱导,因此,在将得到的雪旺氏细胞进行移植的情况下,也不产生免疫学的排斥应答等问题。另外,可以不经由iPS细胞或ES细胞直接由体细胞诱导雪旺氏细胞,因此,可以避免癌化等起因于多能性干细胞的问题。

具体实施方式
本发明涉及一种雪旺氏细胞的制备方法。本发明的制备方法为不经由ES细胞或iPS细胞等多能性干细胞、制备雪旺氏细胞的方法。
雪旺氏细胞
雪旺氏细胞为末梢神经系中的神经胶质细胞。在生理条件下,进行神经组织的支持、形成髓磷脂(髄鞘)而有助于跳跃传导的实现等。在末梢神经的损伤时,完成神经营养因子的产出及释放、再生轴索的基础、髓磷脂形成等末梢神经再生中的许多重要的作用。
天然的雪旺氏细胞与直接源自外胚层的神经细胞不同,源自神经脊。成熟的雪旺氏细胞经过雪旺氏前体细胞、未成熟雪旺氏细胞而形成。本说明书中,为了简单,这些分化过程的中途的全部的细胞包含在“雪旺氏细胞”中。
另外,在雪旺氏细胞中,有形成髓磷脂的雪旺氏细胞、未形成髓磷脂的游走性的雪旺氏细胞(未分化雪旺氏细胞)等,本说明书中这些全部包含在“雪旺氏细胞”中。
本说明书中,不仅包含与天然的雪旺氏细胞相同的细胞,而且包含细胞的功能的一部分或全部可以视为与天然的雪旺氏细胞相同的细胞(也可以称为“雪旺氏细胞样的细胞”。),均称为“雪旺氏细胞”。
体细胞
体细胞只要是源自哺乳动物即可。特别优选为源自哺乳动物、且并非雪旺氏细胞本身及具有在生物体内分化为雪旺氏细胞的能力的细胞的细胞。将雪旺氏细胞移植到生物体的情况下,为了降低感染或排斥应答等危险,优选使用源自进行移植的被验体的体细胞(自身细胞)。但是,对突然的神经的损伤等进行移植等目的的情况下,可以将从并非自身细胞、而是他人或其它动物的体细胞中预先准备的雪旺氏细胞用于移植。或可以由预先准备的他人或其它动物的体细胞制作雪旺氏细胞,用于移植。即,可以制作雪旺氏细胞库的库(包含雪旺氏细胞前体细胞的库。)并供于移植目的。这种情况下,为了降低排斥应答等危险,可以预先将MHC进行分型。另外,可以预先确认雪旺氏细胞的细胞特性或造肿瘤性等。
本说明书中,作为哺乳动物,可列举小鼠、大鼠、仓鼠、人、狗、猫、猴子、兔、牛、马、猪等,特别可列举人。
作为成为本发明的方法(直接重编程)的对象的体细胞,没有特别限定。
体细胞可以使用能够容易地从生物体中采集的体细胞。可列举例如:成纤维细胞、角化细胞、口腔粘膜上皮细胞、鼻腔粘膜上皮细胞、气管粘膜上皮细胞、胃粘膜上皮细胞、肠道粘膜上皮细胞、血管内皮细胞、平滑肌细胞、脂肪细胞、牙龈细胞(牙龈成纤维细胞、牙龈上皮细胞)、牙髄细胞、牙根膜细胞、骨髄细胞、源自骨髄的间质细胞、白细胞、淋巴细胞、肌细胞、结膜上皮细胞、破骨细胞等,优选列举成纤维细胞、角化细胞、口腔粘膜上皮细胞、牙龈细胞、白细胞、淋巴细胞等。本发明中,优选使用从生物体上采集的上述的细胞。
“生物体”不仅包含胚(胎儿)、幼虫、幼体、成体,而且也包含连结母体和胎儿的胎盘、脐带。脐带血管内皮细胞等源自脐带的细胞或源自胎盘的细胞严格来讲并非体细胞,但这些细胞也包含在本发明的“体细胞”中(此时,将“体细胞”换用另一词句读作“脐带血管内皮细胞”、“源自脐带的细胞”、“源自胎盘的细胞”等)。从采集的容易性的观点出发,这些细胞为优选的体细胞的一例。
另外,也可列举由间充质干细胞(Mesenchymal stem cell:MSC)、神经干细胞(Neural stem cell)、肝干细胞(hepatic stem cell)、肠干细胞、皮肤干细胞、毛包干细胞、色素细胞干细胞等体性干细胞进行分化诱导、或进行脱分化、或进行重编程而制作的体细胞。另外,也可列举由各种各样的体细胞进行分化诱导、或进行脱分化、或进行重编程而诱导为其它的体细胞的细胞。另外,也可列举由生殖系的细胞进行分化诱导、或进行脱分化、或进行重编程而诱导的体细胞。
另外,也可列举由胚胎干细胞(Embryonic stem cell:ES细胞)或人工多能性干细胞(induced pluripotent stem cell:iPS细胞)进行分化诱导、或进行重编程而诱导的体细胞。
另外,除上述的进行了分化的体细胞以外,也可以使用体干细胞。在此,干细胞是指具有自我复制能力及分化为其它种类的细胞的能力的细胞。具体而言,可例示:间充质干细胞(Mesenchymal stem cell:MSC)(例如源自脂肪的间质细胞(ADSC:adipose derivedstromal cell))、神经干细胞(Neural stem cell)、肝干细胞(hepatic stem cell)、肠干细胞、皮肤干细胞、毛包干细胞、色素细胞干细胞等体干细胞。
另外,严格来讲并非体细胞,ES细胞、iPS细胞、或生殖系的细胞也包含在本发明的“体细胞”中(此时,将“体细胞”换用另一词句读作“ES细胞”、“iPS细胞”或“生殖系的细胞”)。
另外,也可列举培养细胞,也可列举由培养细胞进行分化诱导、或进行脱分化、或进行重编程而诱导的体细胞。另外,也可列举由ES细胞、iPS细胞、或生殖系的细胞进行分化诱导、或进行脱分化、或进行重编程而诱导的体细胞。
在本发明的优选的一个方式中,体细胞为成纤维细胞、血管内皮细胞(特别是脐带血管内皮细胞)或间充质干细胞(特别是源自脂肪的间质细胞)。
基因或其表达产物
在本发明的方法中,在体细胞中导入选自由SOX10基因及KROX20基因构成的组中的至少1种基因或其表达产物。在此,作为“表达产物”,可列举SOX10基因和/或KROX20基因的mRNA或蛋白质。
作为可以使用的基因的组合,包含仅SOX10基因、仅KROX20基因、SOX10基因及KROX20基因的组合。从可以得到雪旺氏细胞的效率的观点出发,优选SOX10基因及KROX20基因的组合。
在本发明的方法中,也可以同时使用SOX10基因和/或KROX20基因的1种或2种以外的基因。另外,也可以同时使用microRNA、siRNA、shRNA或表达它们的DNA。另外,也可以同时使用各种蛋白质。另外,也可以与各种其它基因同时导入。从可以得到雪旺氏细胞的效率的观点、及简便性的观点出发,优选使用仅1种或2种、特别是SOX10基因及KROX20基因的两种基因。
SOX10基因编码属于SOX(SRY-related HMG-box)家族的、参与胚胎期的发生中的细胞命运的决定的控制的转录因子。
KROX20基因(别名、EGR2、AT591、CMT1D、CMT4E)编码具有3个C2H2型锌指(zincfingers)的蛋白质。
上述基因均为在脊椎动物中高度保守的基因,本说明书中,只要没有特别示出动物名,则表示包含同族体的基因。另外,还包含含有polymorphism(多态性)、即使为具有变异的基因但与野生型的基因产物具有同等功能的基因。
例如,人(Homo sapiens)及小鼠(Mus musculus)的SOX10基因及KROX20基因的cDNA碱基序列及其所编码的蛋白质的氨基酸序列在美国生物工程信息中心(NCBI;National Center for Biotechnology Information)提供的GenBank中以下述的入藏号(Accession Number)被注册(注册有多个修订本(revision)的情况下,理解为是指最新的修订本。):

具体而言,优选将目的基因插入于1个或多个表达载体,在作为对象的体细胞中导入表达载体,在细胞内进行表达。
作为导入基因的方法,可列举使逆转录病毒载体、腺病毒载体、慢病毒载体、腺随伴病毒载体、疱疹病毒载体、仙台病毒载体等病毒性载体感染的方法。此外,在导入基因和其表达产物的情况下,也可以通过阳离子脂质体、阳离子聚合物、电穿孔法等非病毒载体将质粒载体或附加型载体、基因的表达产物(mRNA、蛋白质)进行转染的方法。另外,也可以导入mRNA。这些用于基因导入的手段全部包括在内,在本说明书中均称为载体。
从导入效率和导入基因的稳定保持的观点出发,优选病毒载体,为了抑制癌化的危险,优选质粒。
另外,通过与目的的基因一起导入成为药剂选择标记物的基因(嘌呤霉素耐性、杀稻瘟菌素S耐性、新霉素耐性、潮霉素耐性等),其后进行药剂选择,可以在选择表达目的基因的细胞后使用。
本发明的基因的导入可以通过质粒进行,也可以使用病毒载体、例如逆转录病毒载体。从导入效率和导入基因的稳定保持的观点出发,优选病毒载体,为了抑制癌化的危险,优选质粒。
被导入于体细胞的基因也可以利用LTR启动子进行转录,也可以由载体内部的其它的启动子进行表达。例如可以利用CMV启动子、EF-1α启动子、CAG启动子等组成型表达启动子、或所期望的诱导性启动子。另外,可以利用将LTR的一部分置换为其它启动子的嵌合启动子。
另外,在导入因子为基因的表达产物(例如蛋白质)的情况下,使被称为ProteinTransduction Domain(PTD)的肽等与作为表达产物的蛋白质结合并添加于培养基,由此可以导入于体细胞内。
在本发明的方法的方式之一中,向体细胞导入基因等之后,可以将被导入的细胞在适于雪旺氏细胞的培养的培养基中进行培养。作为适于雪旺氏细胞的培养的培养基,可以使用公知的培养基。例如可以使用在添加有10%FBS(fetal bovine serum)的DMEM培养基(达尔伯克改良伊格尔培养基(Dulbecco's Modied Eagle's Medium))等通常培养基上含有1~20μM左右(特别是5μM左右)的forskolin;2~50ng/ml左右(特别是10ng/ml左右)的bFGF(碱基性成纤维细胞增殖因子、basic fibroblast growth factor);2~50ng/ml左右(特别是10ng/ml左右)的PDGF(Platelet-Derived Growth Factor);50~1000ng/ml左右(特别是200ng/ml左右)的human neuregulin-β1(别名、heregulin、GGF(Glial growthfactor))等成分的1种或2种以上(优选全部)的培养基(雪旺氏细胞诱导培养基)(以上的浓度均为终浓度)。作为一例,可以使用非专利文献1或2中记载的培养基(可以由未分化脂肪干细胞诱导雪旺氏细胞的培养基)。
培养期间没有特别限定,例如可以设为12小时~1个月左右、1天~3周左右、3天~2周左右。根据需要,可以进行培养基交换。培养条件优选按照常规方法。
制备
这样,由体细胞诱导雪旺氏细胞,制备雪旺氏细胞。
所制备的雪旺氏细胞在某种方式中具有选自由外源性的SOX10基因及KROX20基因构成的组中的至少1种基因或其表达产物。在此,“外源性”是指主要为上述的导入手段的结果所导入的基因或其表达产物的方式、与天然的方式不同的方式。可列举例如受天然的启动子以外的启动子控制表达的基因、天然以外的染色体上的位置、或存在于染色体外的基因的方式等。
得到了雪旺氏细胞这一点,作为雪旺氏细胞的功能的评价等可例示:形态(例如细胞宽度和细胞长度之比。)的观察和评价;S100β、p75NTR、GFAP、Nestin、NG2等雪旺氏细胞特异性标记物的表达的检测(可列举例如利用RT-PCR法等的标记物的基因表达的检测、利用免疫染色等的标记物蛋白质的表达的检测等。);神经营养因子的产生;相对于进行了共培养的神经细胞的神经突起伸长的效果或髓磷脂形成能力等。
雪旺氏细胞典型而言具有比较小型的核的双极、或多极性的细胞形态。
雪旺氏细胞特异性标记物中,p75NTR为未分化雪旺氏细胞的标记物。
形成髓磷脂可以通过Myelin Protein Zero(MPZ、P0)、髓磷脂碱基性蛋白质(MBP:Myelin Basic protein)等髓磷脂细胞的标记物的检测、髓磷脂的形态的观察等来确认。
雪旺氏细胞有时作为与雪旺氏细胞以外的细胞(例如原来的体细胞。)的混合物得到。这种情况下,可以将雪旺氏细胞和雪旺氏细胞以外的细胞根据需要进行分离。用于进行分离的手段没有特别限定。例如,将得到的雪旺氏细胞和作为原来的细胞的成纤维细胞进行分离时,可以基于与相对于基础(例如骨胶原等。)的细胞的粘接性的差异进行分离。一般而言,雪旺氏细胞与成纤维细胞相比,相对于基础的粘附性弱。另外,也可以根据分类将雪旺氏细胞和雪旺氏细胞以外的细胞进行分离。
由本发明制备的雪旺氏细胞例如可以优选用作后述的移植材料。
由本发明制备的雪旺氏细胞另外可以用于使用有雪旺氏细胞的各种各样的研究或技术开发等。例如对雪旺氏细胞的产生、分化、形态形成的机制、相对于这些的力学的应力、营养、激素等影响的分析等基楚研究是有用的。
如果使用由本发明制备的雪旺氏细胞,则可以由具有各种各样的疾病或遗传的背景的人或动物简便、迅速、廉价地建立雪旺氏细胞,因此,可以通过生物化学、分子生物学、免疫学等方法分析与疾病或遗传的背景关联的雪旺氏细胞的异常,由此可以有益于疾病的发症机理的阐明等研究或診断法的开发。另外,如果使用这种雪旺氏细胞进行药剂的开发、药剂的毒性试验等,则可以有益于相对于各种疾病的新型治疗法的开发。
移植材料
由本发明得到的雪旺氏细胞可以用于治疗各种疾病。该情况下,雪旺氏细胞可以移植材料的形态被提供。
移植材料是指为了神经纤维的修复、再建而导入于生物体内的、含有雪旺氏细胞的材料。本发明中得到的雪旺氏细胞可以用于移植材料的制作。雪旺氏细胞自身也成为移植材料。因此,也可以将雪旺氏细胞作为细胞制剂移植于患者,可以与由人工材料构成的基材(支架)一起移植,或与支架一起进行培养后移植。基材(支架)例如作为神经再建起作用。这些情况下,支架可以根据移植目的而制成各种3维的形状。
本发明的移植材料可以通过包含前述的雪旺氏细胞的制备方法作为工序的方法来制造。
作为基材(支架)的具体例,可列举组合有聚乙醇酸(PGA)管、骨胶原管、纤维蛋白胶(Fibrin glue)、聚合物泡沫管、明胶管、聚乙醇酸(PGA)和骨胶原的管等。作为组合有聚乙醇酸(PGA)和骨胶原的管,也可以使用“ナーブブリッジ”(东洋纺株式会社制)等市售品。
移植材料可以按照自身神经移植、或从自身神经将雪旺氏细胞进行分离培养而移植的治疗而使用。这种方法记载于下述的文献:

本发明中得到的雪旺氏细胞另外并不限于疾病的治疗,也可以以美容或功能增强的目的使用。此时,相对于人的处理也在本说明书中方便上称为治疗,“患者”可以换用另一词句读作“健康者”或“人”,“疾病”可以换用另一词句读作“美容”或“功能”。
本发明另外不仅可以用于人的疾病的治疗,而且也可以用于包含狗、猫等赏玩动物或牛、马、猪、绵羊、鸡等家畜的哺乳动物的疾病的治疗。该情况下,将“患者”换用另一词句读作“患畜”或“哺乳动物”。
组合物
如上所述,通过将选自由SOX10基因及KROX20基因构成的组中的至少1种基因或其表达产物导入于体细胞,可以制备雪旺氏细胞。因此,本发明进而提供一种用于制备雪旺氏细胞的组合物,其包含选自由SOX10基因及KROX20基因构成的组中的至少1种基因或其表达产物。用于制备该雪旺氏细胞的组合物包含为了从体细胞中诱导雪旺氏细胞而使用的因子,上述基因或其表达产物优选以可导入于体细胞的形态含有。上述基因作为可导入于体细胞的形态,具体而言,可例示插入有上述基因的载体。在此,上述基因可以分别插入于不同的载体,也可以2种以上的基因同时插入于1个载体。
关于可以使用的载体的种类等,如上所述。
在活体内(in vivo)的直接重编程
如上所述,通过将选自由SOX10基因及KROX20基因构成的组中的至少1种基因或其表达产物导入于体细胞,可以制备雪旺氏细胞。
在此,在神经损伤时,在损伤部聚集成纤维细胞。聚集的成纤维细胞其后形成纤维性的伤疤。
因此,通过应用本发明的制备方法在神经的损伤部的成纤维细胞中导入选自由SOX10基因及KROX20基因构成的组中的至少1种基因或其表达产物,在该损伤部通过直接重编程而诱导雪旺氏细胞,因此,可以有助于神经损伤的治疗或神经的再生。在选自由SOX10基因及KROX20基因构成的组中的至少1种基因或其表达产物的导入中,可以优选使用上述的本发明的组合物。
本发明的制备方法理解为除在活体外(in vitro)的直接重编程之外,也包含如上所述的在活体内(in vivo)的直接重编程。
在活体内(in vivo)的直接重编程除在神经的损伤部的成纤维细胞中导入选自由SOX10基因及KROX20基因构成的组中的至少1种基因或其表达产物以外,例如可以按照下述的文献中记载的在活体内(in vivo)的心肌细胞的直接重编程而进行。

【实施例】
以下,示出实施例,但本发明并不仅限定于该实施例。
实施例中,HDF表示人皮肤成纤维细胞(Human Dermal Fibroblast)。cSC表示从对照的生物体上采集的培养雪旺氏细胞(cultured Schwann cells)。dSC表示通过本发明的方法得到的雪旺氏细胞(directly reprogrammed Schwann cells)。
Cont表示对照(Control)。
实施例1方法的概略(图1)
图1表示用于制备本发明的雪旺氏细胞(图中,“directly reprogrammed Schwanncell:dSC”)的方法的概略。
在逆转录病毒载体质粒pMXs.puro中使用Gene art系统插入SOX10等各种基因的cDNA编码序列。将包装细胞Plat GP细胞悬浮于含有100U/mL青霉素和100μg/ml链霉素的1%NEAA 10%FBS DMEM(通常培养基),在进行了明胶包被的10cm培养盘中以每盘5×106个的浓度播种(第-3天)。24小时培养后,将含有各种基因的pMXs载体以各种组合与pCMV VSV载体同时使用X-tremeGENE 9按以下的比导入。即将导入基因5μg、pCMV.VSV 2.5μg、Opti-MEM 500μl、X-tremeGENE 9 22.5μl的混合液添加于装入10ml的培养基的10cm盘(第-2天)。24小时后,交换成不含有抗生素的通常培养基(第-1天)。同日(第-1天),将人正常皮肤成纤维细胞株(aHDF)(图中,“fibroblast”)以1.5×104~2×104cells/mL播种于培养盘或12孔板。24小时后(第0天),将Plat GP培养上清液通过孔隙的直径为0.45μm的针头过滤器之后,与聚丁烯(最终浓度4μg/mL)进行混合(病毒液)。抽吸除去aHDF的培养上清液之后,快速地加入上述的病毒液1mL,培养24小时(感染;图中、“transfection”)。作为对照组,也准备不进行病毒感染的细胞。1天后(第1天),抽吸除去培养上清液,加入雪旺氏细胞诱导培养基(通常在培养基中加入有5mM forskolin,10ng/ml recombinant human basic fibroblastgrowthfactor(bFGF),5ng/ml recombinant human platelet-derived growth factor(PDGF)and 200ng/ml recombinant human heregulin1-b1(GGF)(均为最终浓度)的培养基),其后,每2天与相同的组成的新的培养液进行交换。第12-22天,对得到的细胞进行作为雪旺氏细胞的代表性的标记物的S100β染色。将不使逆转录病毒载体感染、进行了相同的培养的细胞设为Control。
实施例2从人正常皮肤成纤维细胞向雪旺氏细胞的转换、S100β荧光免疫染色像
(表1)(图2)
将人正常皮肤成纤维细胞(aHDF)在12孔板上进行培养,进行实施例1中记载的操作。第14天,从各孔抽吸除去培养液,用PBS进行1次清洗后,用4%PFA固定。加入封闭溶液,在37℃下静置15分钟。为用抗S100β抗体(一抗)和AlexaFluor566标记抗兔IgG抗体(二抗)进行了染色的细胞。在板的各孔中导入不同的基因的组合,在哪个号码的孔中导入哪个基因的组合记载于表1(表中,在各基因的栏中有记载为“1”的物质,表示使含有该基因的逆转录病毒载体感染,空栏表示不使含有该基因的逆转录病毒载体感染)。作为雪旺氏细胞关联基因的SOX10、Krox20、Oct6的导入的基因候补为作为重编程关联基因的SOX2、C-myc、KLF4、Oct3/4的7因子。例如,表1的No.43为感染含有SOX10、Krox20、Oct6、KLF4的基因的逆转录病毒载体的细胞。
作为代表性的S100β的染色性像,将导入有Sox10和Krox20的2因子的基因的细胞示于图2。图2为与DAPI(4',6-diamidino-2-phenylindole)引起的核染色的共染色。

在与图2相同的实验中,为了确认其它组合的染色性,用荧光显微镜(Olympus制)观察板,以4阶段评价S100β的染色性(从S100β阳性细胞多的物质依次为+++、++、+、-)。
用图3表示各自的评价的典型像。另外,将其评价结果在表1中同时示出。例如可知:No.4的感染含有SOX10和KROX20的2基因的逆转录病毒载体的细胞成为+++的评价,含有最多的S100β阳性细胞。即使SOX10单独、或KROX20单独的导入,也可以以低的效率诱导雪旺氏细胞,但SOX10和KROX20的共导入可以更高效地诱导。另外可知:Oct6在雪旺氏细胞的诱导中几乎没有效果。
实施例4从人正常皮肤成纤维细胞向雪旺氏细胞的转换、细胞形态评价(图4)
在实施例1中,对导入有Sox10和Krox20的2因子的基因的细胞,对于从人正常皮肤成纤维细胞向雪旺氏细胞的转换中的细胞形态变化,以细胞长度/细胞宽度比对雪旺氏细胞特征性的双极型的细胞长度的增加进行评价。
将评价的结果示于图4。通过转换而显示细胞长度/细胞宽度比的增大、即变化为雪旺氏细胞的特征性细胞形态。
实施例5从人正常皮肤成纤维细胞向雪旺氏细胞的转换、使用有其它雪旺氏细胞
关联标记物的免疫细胞荧光染色像(图5)
将人正常皮肤成纤维细胞aHDF在12孔板上进行培养,进行实施例1的操作。示出导入有SOX10及Krox20的2个基因的组的结果。从基因导入12天后,进行免疫染色。
将结果示于图5。抗p75NTR抗体(一抗)、抗GFAP抗体(一抗)、抗NG2抗体(一抗)的宿主为兔,抗Nestin抗体(一抗)的宿主为小鼠。二抗使用AlexaFluor488,566标记抗兔IgG抗体或AlexaFluor488标记抗小鼠IgG抗体。虽然与S100β相比阳性率下降,但是确认各自的表达。
实施例6
A.从人正常皮肤成纤维细胞向雪旺氏细胞的转换、S100β和p75NTR基因的mRNA表达量的测定(图6)
将人正常皮肤成纤维细胞(aHDF)在12孔板上进行培养,进行实施例1的操作。在孔No.1~6的细胞中导入Sox10和Krox20的2因子的基因,孔No.7~12的细胞中没有导入基因,作为对照。基因导入12天后,从各孔用ISOGEN II回收总RNA,使用ReverTra Ace qPCR RTMaster Mix合成cDNA。以定量UCP1和β肌动蛋白基因的mRNA水平的目的将Real-time PCRMaster Mix、Taqman probe、Specific Primer及cDNA进行混合,使用AB7300 Real-timePCR system进行Real-time RT-PCR。计算各细胞的相对于β肌动蛋白mRNA水平的目的基因的mRNA水平的值。
将结果示于图6。图表的纵轴将孔No.1的值作为“1”,将算出了各孔的值的相对值作为相对mRNA水平表示。导入有SOX10和KROX20的2个基因的细胞(No.1~6)与control(No.7~12)相比,看到作为雪旺氏细胞特异性标记物的S100β和p75NTR基因的强表达。
B.RT-PCR产物的使用有琼脂糖凝胶的电泳
对A中得到的Real-time RT-PCR产物进行使用有琼脂糖凝胶的电泳。结果,在导入有SOX10和Krox20的2因子的基因的细胞群中,可确认S100β和p75NTR基因的mRNA的强表达。
C.通过没有因子导入、仅SOX10、仅Krox20、SOX10+Krox20的各自的组合进行的人成纤维细胞向雪旺氏细胞的直接重编程效率的使用有RTPCR的研究。
将作为人正常皮肤成纤维细胞的aHDF在12孔板上进行培养,进行实施例1的操作。用与A同样的方法将没有基因导入的细胞(对照)、仅导入SOX10基因、导入仅Krox20基因、导入有SOX10基因+Krox20基因的细胞各自的S100β和p75NTR的mRNA的表达进行比较。将mRNA的相对值作为相对mRNA水平表示。计算各组的相对于β肌动蛋白mRNA水平的目的基因的mRNA水平的值。将非导入细胞的中的1群的值作为“1”,将算出了各组的值的相对值的平均作为相对mRNA水平表示。结果,导入有SOX10+Krox20的2因子的组中,S100β和p75NTR的mRNA的表达量大大上升。另外,即使仅导入有Sox10的细胞,与导入有Sox10和Krox20这两者的细胞相比时低,但S100β和p75NTR的mRNA表达量上升。另外,即使仅导入有Krox20的细胞,与导入有Sox10和Krox20这两者的细胞相比时低,但S100β的mRNA表达量上升。S100β的相对mRNA水平在非导入细胞中为4.4,在Sox10单独导入细胞中为1712.3,在Krox20单独导入细胞中为25.1,在导入有Sox10+Krox20的细胞中为127615.0。p75NTR的相对mRNA水平在非导入细胞中为3.1,在Sox10单独导入细胞中为74.3,在Krox20单独导入细胞中为0.4,在导入有Sox10+Krox20的细胞中为35397.6。因此,即使SOX10单独或Krox20单独,效率也低,但是有可能产生由人成纤维细胞向雪旺氏细胞的直接重编程。
由以上确认:由人成纤维细胞向雪旺氏细胞的直接重编程在使用SOX10和Krox20的2因子的情况下效率高。另外显示,即使SOX10或Krox20的任意1因子,效率虽然低但可以由人成纤维细胞对雪旺氏细胞的直接重编程。
实施例7 dSC相对于神经细胞的神经突起伸长效果的研究(图7)
将NG108-15培养12小时,其后与HDF、cSC、或dSC(导入Sox10和Krox20的2因子的基因而诱导的dSC)共培养36小时。作为对照,将NG108-15通过仅添加1%FBS的DMEM进行培养。将I.具有神经突起的细胞(neurite-bearing cell)的比例(percent of neurite-bearing-cells)、II.由细胞体直接伸长的一次神经突起数(number of primaryneurites)、III.最长神经突起的长度(Longest neurite length)通过文献:Tomita K,Madura T,Sakai Y,et al:Glialdifferentiation of human adipose-derived stemcells:implications for cell-based transplantation therapy.Neuroscience.2013;236:55-65.的方法进行比较研究。使用抗Tuj-1抗体(一抗)和二抗AlexaFluor566标记抗兔IgG抗体仅对NG108-15进行荧光染色。
图7表示结果。通过与对照组的比较,cSC、dSC这两者在所研究的全部参数(I-III)中均显示高的神经突起伸长促进效果,dSC的成绩与cSC匹敌。由以上确认:dSC培养上清液对于神经细胞促进突起伸长。
实施例8通过无病毒方式Plasmid导入(电穿孔)引起的从人正常皮肤成纤维细胞
向雪旺氏细胞的转换(图8)
将人正常皮肤成纤维细胞aHDF在12孔板上进行培养,进行实施例1的操作。示出导入有SOX10及Krox20的2个基因的组的结果。基因导入14天后,进行免疫染色。
将结果示于图8。抗S100β抗体(一抗)、抗p75NTR抗体(一抗)、抗GAP43抗体(一抗)的宿主为兔,二抗使用AlexaFluor566标记抗兔IgG抗体。虽然与使用有逆转录病毒载体的情况相比阳性率下降,但是,可以确认S100β、p75NTR及GAP43各自的表达,可以确认由成纤维细胞aHDF向雪旺氏细胞的转换。
实施例9从源自人正常脂肪的干细胞向雪旺氏细胞的转换、使用有其它雪旺氏细
胞关联标记物的免疫细胞荧光染色像(图9)
验证了从源自脂肪的间质细胞向dSC的转换。
〈方法〉
将源自人正常脂肪的间质细胞(ADSC:adipose rerived stromal cell)在12孔板上进行培养,进行实施例1的操作。示出导入有SOX10及Krox20的2个基因的组的结果。从基因导入14天后,进行免疫染色。
〈结果〉
将结果示于图9。抗S100β抗体(一抗)、抗p75NTR抗体(一抗)、抗GAP43抗体(一抗)、抗Protein Zero抗体(一抗)的宿主为兔,二抗使用AlexaFluor566标记抗兔IgG抗体。利用S100β的染色中,约40%的细胞显示阳性。关于其它雪旺氏细胞标记物,虽然与S100β相比阳性率下降,但是也可确认各自的表达。作为髓磷脂标记物的Protein Zero也显示阳性。
具体而言,用与成纤维细胞(Fibroblast)同样的方法进行了雪旺氏细胞诱导的结果,细胞的形态由源自人正常脂肪的间质细胞的形态变化成雪旺氏细胞典型性的具有比较小型的核的双极、或多极性的细胞形态。进而,在雪旺氏细胞中变化成典型的细胞形态的细胞为雪旺氏细胞标记物(S100β)阳性(约40-50%)(图9A)。
另外,就诱导后的细胞而言,雪旺氏细胞标记物(S100β、GAP43)、未分化雪旺氏细胞标记物(p75NTR)、髓磷脂标记物(P0)为阳性(图9B)。
实施例10从脐带血管内皮细胞向雪旺氏细胞的转换、使用有其它雪旺氏细胞关联
标记物的免疫细胞荧光染色像(图10)
验证了从血管内皮细胞向dSC的转换。
〈方法〉
将脐带血管内皮细胞(Huvec:Human Umbilical Vein Endothelial Cells)在12孔板上进行培养,进行实施例1的操作。示出导入有SOX10及Krox20的2个基因的组的结果。从基因导入14天后,进行免疫染色。
〈结果〉
将结果示于图10。抗S100β抗体(一抗)、抗p75NTR抗体(一抗)、抗GAP43抗体(一抗)、抗Protein Zero抗体(一抗)的宿主为兔,二抗使用566标记抗兔IgG抗体。利用S100β的染色中,约30%的细胞显示阳性。关于其它雪旺氏细胞标记物,虽然与S100β相比阳性率下降,但是也可确认各自的表达。作为髓磷脂标记物的Protein Zero也显示阳性。
具体而言,用与成纤维细胞(Fibroblast)同样的方法进行了雪旺氏细胞诱导的结果,细胞的形态从脐带血管内皮细胞的形态变化成雪旺氏细胞典型性的具有比较小型的核的双极、或多极性的细胞形态。进而,在雪旺氏细胞中变化成典型的细胞形态的细胞为雪旺氏细胞标记物(S100β)阳性(约50-60%)(图10A)。
另外,就诱导后的细胞而言,雪旺氏细胞标记物(S100β、GAP43)、未分化雪旺氏细胞标记物(p75NTR)为阳性(图10B)。
实施例11用GFP标记的dSC(导入Sox10和Krox20的2因子的基因而诱导的dSC)和
DRGn的共培养引起的dSC的活体外的髓磷脂化形成。使用有雪旺氏细胞关联标记物的免疫
细胞荧光染色像(图11)
评价用GFP标记的dSC和DRGn的共培养引起的dSC的活体外的髓磷脂化能力。
〈方法〉
使用逆转录病毒载体将dSC用GFP进行细胞标记。
具体的方法按照文献:Yoshioka T,Ageyama N,Shibata H,Yasu T,Misawa Y,Takeuchi K,Matsui K,Yamamoto K,Terao K,Shimada K,Ikeda U,Ozawa K,HanazonoY.Repair of infarctedmyocardium mediated by transplanted bone marrow-derivedCD34+ stem cells in anonhuman primate model.Stem Cells.2005 Mar;23(3):355-64.及文献:Hirschmann F,Verhoeyen E,Wirth D,Bauwens S,Hauser H,Rudert M.Vitalmarking of articular chondrocytes by retroviral infection using greenfluorescence protein.Osteoarthritis Cartilage.2002 Feb;10(2):109-18.中记载的方法。
将从生后第5天的小鼠中采集的后根神经节细胞(DRGn:dorsal root ganglionneuron)在12孔板中培养2天,在含有用GFP标记的dSC、神经生长因子、抗坏血酸和cAMP的髓磷脂分化诱导培养基中进行共培养。从共培养基因开始14天后,进行免疫染色。
使用的培养基的组成如下所述:
DMEM containing N2 supplement(Invitrogen),50ng/ml ascorbic acid(Wako,Osaka,Japan),and 50ng/ml recombinant rat b-nerve growth factor(NGF)(R&DSystems,Inc.,Minneapolis,MN,USA),0.5μM cAMP(R&DSystems,Inc.,Minneapolis,MN,USA)。
培养基按照文献:Sango K,Kawakami E,Yanagisawa H,Takaku S,Tsukamoto M,Utsunomiya K,Watabe K.Myelination in coculture of established neuronal andSchwann cell lines.Histochem Cell Biol.2012 Jun;137(6):829-39.的记载而制作。
抗神经丝抗体(一抗)、抗MBP(Myelin Basic Protein)抗体(一抗)的宿主为小鼠,抗Tuj-1抗体(一抗)、抗Protein Zero抗体(一抗)的宿主为兔,二抗使用AlexaFluor566标记抗小鼠和兔的IgG抗体、Cy5标记抗小鼠和兔的IgG抗体。作为髓磷脂标记物抗体的抗MBP抗体和抗Protein Zero抗体用红色标记,作为神经轴索标记物的抗神经丝抗体和抗Tuj-1抗体用灰色标记。
〈结果〉
将结果示于图11。沿DRG的神经突起可看到进行了髓磷脂化的雪旺氏细胞,但一部分髓磷脂化雪旺氏细胞和用GFP标记的dSC细胞发生了重叠。
实施例12相对于坐骨神经损伤模型的用GFP标记的dSC(导入Sox10和Krox20的2因
子的基因而诱导的dSC)移植引起的活体外的髓磷脂化形成。使用有髓磷脂关联标记物的免
疫细胞荧光染色像(图12)
进行了dSC在活体内的髓磷脂化能力的研究。
〈方法〉
使用病毒载体将dSC用GFP进行细胞标记。
使免疫缺陷小鼠的坐骨神经露出,用手术用止血钳将中央部分把持约1分钟,将约5mm的范围内的神经压碎。在其末梢侧注入用GFP标记的dSC约5万个。从移植开始1个月后,采集被压碎的部位的修复神经并进行免疫染色。将方法的概要示于图12。
抗神经丝抗体(一抗)、抗MBP抗体(一抗)的宿主为小鼠,抗Tuj-1抗体(一抗)、抗Protein Zero抗体(一抗)的宿主为兔,二抗使用AlexaFluor566标记抗小鼠和兔的IgG抗体、Cy5标记抗小鼠和兔的IgG抗体。作为髓磷脂标记物抗体的抗MBP抗体和抗Protein Zero抗体用红色标记,作为神经轴索标记物的抗神经丝抗体和抗Tuj-1抗体用灰色标记。
〈结果〉
将结果示于图12。GFP+细胞以沿再生神经的方式排列,这些与髓磷脂标记物+细胞发生重叠(图12B及C)。
不稳定寄生于(erratic parasitism,日语:迷入)再生神经的GFP+细胞也表达髓磷脂标记物(图12B及C、箭头)。
实施例13对免疫缺陷小鼠坐骨缺损(5mm)模型的dSC的移植(图13)
〈方法〉
将培养雪旺氏细胞(SC:比较对照组)从坐骨神经进行分离培养。将这些细胞预先播种于明胶管,在小鼠的坐骨神经干中制作约5mm的缺口。将得到的混合型管移植至神经缺损部。以Sham小鼠及仅含有PBS的管移植组(Cont)为对照,评价神经再生促进效果(图13A)。
再建神经显示宏观像和再生神经组织横轴切片的勒克司坚牢蓝引起的髓磷脂染色像。坐骨神经功能显示移植后6周(6W)和12周(12W)后的结果,支配肌肉的委缩以湿肌肉重量进行评价。
坐骨神经功能索引(SFI:Sciatic functional Index)按照文献:Inserra MM,Bloch DA,Terris DJ.Functional indices for sciatic,peroneal,andposteriortibial nerve lesions in the mouse.Microsurgery.1998;18:119-124.的记载而求出。
支配的肌肉的萎缩及纤维化按照文献:Clavijo-Alvarez JA,Nguyen VT,Santiago LY,Doctor JS,Lee WP,Marra KG.Comparison of biodegradable conduitswithin aged rat sciatic nerve defects.Plast Reconstr Surg.2007;119:1839-1851.的记载进行评价。
〈结果〉
进行了再建的神经的宏观像中,cSC组和dSC的两者均与control相比优异,在两者间没有看到明显的差异(图13B)。
再生神经组织横轴切片的髓磷脂染色像中,dSC组与cSC组匹敌(图13C)。
坐骨神经功能索引(SFI:Sciatic functional Index)中,在12W的时刻,dSC组可看到与cSC匹敌的坐骨神经功能的恢复(图13D)。
关于支配肌肉萎缩及纤维化,在两组中也可看到与对照的显著性差异,但没有看到cSC和dSC间的显著差异(图13E)。
实施例14向雪旺氏细胞的转换(表2)
对表2所示的基因的组合,进行与实施例2同样的实验。
在哪个号码的孔中导入哪个基因的组合记载于表2(表中,在各基因的栏中有记载为“1”的物质表示使含有该基因的逆转录病毒载体感染,空栏表示不使含有该基因的逆转录病毒载体感染)。
与实施例3同样地,用荧光显微镜(Olympus制)观察板,将S100β的染色性以4阶段进行评价(由S100β阳性细胞多的物质依次为+++、++、+、-)。
将评价结果在表2中同时示出。
由该结果可知:已知Oct6为在雪旺氏细胞的分化中完成重要的功能的因子,但几乎不能诱导由体细胞对雪旺氏细胞的直接重编程。另外可知:Oct6不能提高由Sox10单独、Krox20单独、或Sox10+Krox20引起的体细胞对雪旺氏细胞的直接重编程的效率。

实施例15神经营养因子的产生(图14)
将HDF、cSC、及dSC在4×104cells/cm2的条件下播种于细胞培养用盘,确认80%汇合后,在细胞上清液采集用培养基中培养48小时。其后,将各组的培养上清液通入于40-μm过滤器之后,进行采集。对脑源性神经营养因子(Brain-derived Neurotrophic Factor(BDNF))、胶质细胞源性神经营养因子(glial cell line-derived neurotrophic factor(GDNF))、脑神经营养因子(Nerve Growth Factor(NGF)),使用ELISA kits human BDNF、GDNF、NGF(Promega,Madison,WI)测定各自的培养细胞的培养上清液中所含的蛋白量。
其结果是,与对照(HDF)相比,cSC和dSC均较强地产生BDNF、GDNF、NGF的全部。另外,BDNF的产生即使在最高点,cSC和dSC也类似。
Claims (6)
1.一种制备雪旺氏细胞的方法,其包含在哺乳动物的体细胞中导入选自由SOX10基因及KROX20基因构成的组中的至少1种基因或其表达产物的工序。
2.根据权利要求1所述的方法,其中,
所述基因为SOX10基因及KROX20基因的组合。
3.根据权利要求1或2所述的方法,其中,
所述体细胞为成纤维细胞、血管内皮细胞或间充质干细胞。
4.一种雪旺氏细胞,其源自于哺乳动物的体细胞,具有选自由外源性的SOX10基因及KROX20基因构成的组中的至少1种基因或其表达产物。
5.一种移植材料,其用于治疗基于神经的缺损或者雪旺氏细胞的缺损、不足或功能降低的疾病,包含用权利要求1~3中任一项所述的方法得到的细胞、或权利要求4所述的雪旺氏细胞。
6.一种用于制备雪旺氏细胞的组合物,其包含选自由SOX10基因及KROX20基因构成的组中的至少1种基因或其表达产物。
