Description
Technical Field
The invention belongs to the field of aquaculture, relates to the technical field of killing of moss in crayfish and river crab culture ponds, and particularly relates to a preparation method of a biological agent and application of the biological agent in the field of killing of moss.
Background
With the improvement of the living standard of residents in China, the consumption structure is continuously optimized and improved, the specific gravity of aquatic products in the dietary structure is continuously increased, and the total output value of the aquaculture industry is in a stable increasing state. While the aquaculture industry in China gains huge achievements, the problems of pollution around the aquaculture water area, moss prevention and control and the like are also faced. The moss is a product of excessive propagation of algae in the water body, and the growth of a large amount of moss damages the algae phase balance of the water body and seriously damages the water quality, so that the excessive moss in the pond has great harm to aquaculture, and serious economic loss is caused. The currently adopted moss killing technology comprises a physical method, a chemical method, a biological method and the like. The use of physical methods is time-consuming and labor-consuming; the chemical method comprises prometryn powder, copper sulfate and the like, has great environmental pollution, remains and is unsafe; the use of biological methods such as microecologics is costly and has little effect.
Galla chinensis is a general term for insect gall parasitic on Rhus chinensis in Rhus of Anacardiaceae. China is the main producing country and the processing country of gallnuts. Historically, the main function of the development of the gallnut industry in China is medicinal. According to the records of medical history book 'compendium of materia Medica': galla chinensis is sour, astringent and cold, and has effects of stopping nocturnal emission, relieving diarrhea, astringing lung, lowering fire, astringing dampness, astringing intestine, healing sore, and stopping bleeding. Modern medicine proves that the gallnut contains tannic acid, and the gallnut has the effects of local hemostasis, antibiosis, detoxification and the like because the tannic acid can form insoluble compounds with some metals, alkaloids and the like and can denature protein. It is recorded in Kaibao Bencao that it has the actions of astringing lung to reduce pathogenic fire, astringing intestine to stop diarrhea, astringing dampness to heal wound, arresting sweating and stopping bleeding. Can be used for treating chronic cough due to lung deficiency, cough due to lung heat, traumatic hemorrhage, hematochezia, hemorrhoid, skin wet rot, chronic diarrhea, chronic dysentery, spontaneous perspiration, night sweat, skin sore, diabetes, etc. Gallnut has been reported to have an in vitro antibacterial effect on various pathogenic bacteria such as staphylococcus epidermidis, pseudomonas aeruginosa, staphylococcus aureus, candida albicans and the like, and is commonly used for treating anorectal diseases and bacterial cariogenic diseases in clinic at present. Because of the inhibition effect of gallnut on aquatic pathogenic bacteria in various aquaculture, gallnut powder is often used as a pond cleaning medicament in aquaculture.
The dead moss floats on the water surface and is slowly decomposed under the action of microorganisms, so that the inventor is inspired to separate a bacillus from the rotten moss, and the bacterial strain is found to have good growth inhibition effect on the moss through a laboratory culture test. The bacillus used in the technology is bacillus subtilis (China general microbiological culture Collection center, preservation number: CGMCC 1.3358), and in addition, the Chinese gall powder can effectively kill moss according to research findings. Thus, the inventor prepares a preparation capable of effectively killing moss by mixing the Chinese gall powder with the bacterial strain. The main components of the product are plant extract, antagonistic bacteria complex flora and moss lyase. The method can directionally identify and destroy the cell wall structure of the moss, gradually decompose the moss and convert the moss into organic matter components which can be used by the aquatic weeds, eliminate pollution damage of dead moss to water quality, cannot damage aquatic weeds such as hydrilla verticillata and elodea nutans in the aquaculture water body, cannot generate stress to aquaculture animals, does not have drug residues, and has good biological safety.

And (3) sufficiently mixing the gallnut powder with the granularity of 100 meshes obtained by crushing and the bacillus subtilis (the activity is more than or equal to 1 multiplied by 108CFU/g) according to the mass percentage of 1:10 to obtain the biological preparation rich in the plant tannin and the bacillus subtilis. And (3) mixing the prepared biological agent according to a material-liquid ratio of 1: brown sugar water (water: brown sugar 10: 1(kg/kg)) was added to 25(kg/kg) and the activation treatment was carried out at 30 ℃ for 6 hours. In the moss outbreak period, sodium humate is matched, and the full pond sprinkling is carried out after the activation according to the dose of 100g per mu of water surface (the water depth is about 10 meters). Generally, the aquatic weeds turn yellow on the 2 nd day, gradually separate from the aquatic weeds and float upwards along with the increase of time, and the moss dies and decomposes around a week.
Example 4: comparative test without activation
Gallnut powder with the granularity of 100 meshes obtained by crushing is taken and fully mixed with the bacillus subtilis (the activity is more than or equal to 1 multiplied by 108CFU/g) according to the mass percentage of 1:10 to obtain the biological agent rich in the plant tannin and the bacillus subtilis. In the moss outbreak period, sodium humate is matched, and the full pond sprinkling is carried out after the activation according to the dose of 100g per mu of water surface (the water depth is about 10 meters). The moss does not obviously float and die after being used for one week.
Example 5: comparative test with Bacillus alone
Bacillus subtilis (the activity is more than or equal to 1 multiplied by 108CFU/g) is added according to the feed-liquid ratio of 1: brown sugar water (water: brown sugar 10: 1(v/v)) was added at 25(kg/kg) and the activation treatment was carried out at 30 ℃ for 6 hours. In the moss outbreak period, sodium humate is matched, and the full pond sprinkling is carried out after the activation according to the dose of 100g per mu of water surface (the water depth is about 10 meters). The moss does not obviously float and die after being used for one week.
Example 6: laboratory moss cultivation experiment
And (3) sufficiently mixing the gallnut powder with the granularity of 50 meshes obtained by crushing and the bacillus subtilis (the activity is more than or equal to 1 multiplied by 108CFU/g) according to the mass percentage of 1:10 to obtain the biological preparation rich in the plant tannin and the bacillus subtilis. And (3) mixing the prepared biological agent according to a material-liquid ratio of 1: 20(kg/kg) was added with brown sugar water (water: brown sugar 10: 1(kg/kg)) and activated at 30 ℃ for 6 hours to obtain a medicinal solution. Placing 200mL of the above medicinal liquid in 200mL culture flask, adding 2g of moss in vigorous growth stage into 200mL river water of control group, culturing in sunlight, and observing. The result shows that the control solution is green and has light fishy smell after the moss is cultivated for 24 hours, and the medicament group has dark color; after 48 hours, the color deepens and becomes black, and the fish smell is generated. Experiments show that the preparation prepared by the invention has an inhibiting effect on moss.
The above description is only of the preferred embodiments of the present invention, and it should be noted that: it will be apparent to those skilled in the art that various modifications and adaptations can be made without departing from the principles of the invention and these are intended to be within the scope of the invention.
