一种大麻二酚或大麻素酸的稳定方法,属于天然物提纯加工技术领域。本发明的目的是为了解决目前大麻二酚酸不稳定,室温下也会很快的脱羧降解为大麻二酚CDB的问题,所述方法具体为:新鲜的汉麻花叶加入水中,汉麻花叶与水的固液比为1:10,搅拌1~2h,过滤分离,液体中含需要的大麻二酚酸及其它有价值的大麻素酸物质;取beta‑环糊精或其衍生物,加入水中,配制10%的水溶液,在2℃~30℃搅拌条件下,搅拌1‑2h,过滤分离出包合物。beta‑环糊精与大麻二酚酸包合物有效的使大麻二酚酸稳定,方法简便,成本低,并且在各种液体(如,水溶液,护肤液,乳液)中稳定,为大麻二酚酸的应用提供了一个新的方法。

技术领域
本发明属于天然物提纯加工技术领域,具体涉及一种使大麻二酚或大麻素酸稳定的方法。
背景技术
大麻(cannabissatival.)是大麻科大麻属的一年生草本植物,自大麻植物中分离得到的酚类酸性物质,即大麻素酸主要包括大麻萜酚酸(cannabigerolic acid,CBGA)、大麻二酚酸(cannabidiolic acid,CBDA)、大麻环萜酚酸(cannabichromenic acid,CBCA)、四氢大麻酚酸(Tetrahydrocannabinolic acid,THCA)等,其中大麻二酚酸(CBDA),大麻萜酚酸(CBGA)等具有很高的药用价值。汉麻植物中的大麻素主要是以酸的形式存在,包括大麻二酚酸,它比大麻二酚(CBD)更有效。研究发现大麻二酚酸的活性是大麻二酚的几十倍,并且吸收快,目前市场需要大麻二酚酸的产品。但是大麻二酚酸(CBDA)是不稳定的,干燥和加工过程中受热很快的降解成大麻二酚(CBD),因此一直以来在药物开发中是无法使用的。目前市场上的产品都是大麻二酚(CBD),市场需要一种能使大麻二酚酸稳定的方法。
发明内容
本发明的目的是为了解决目前大麻二酚酸不稳定,室温下也会很快的脱羧(CO2)降解为大麻二酚CDB的问题,提供一种简易的大麻二酚或大麻素酸的稳定方法,该方法中大麻二酚酸(CDBA)与beta-cyclodextrin(beta-环糊精)或beta-环糊精的衍生物形成包合物,在中性的条件下,包合物中的大麻二酚酸稳定,70℃可以保持2个星期的降解不到5%,室温可以保持一年以上。
为实现上述目的,本发明采取的技术方案如下:
一种大麻素酸的稳定方法,所述方法具体为:
步骤一:获取汉麻提取液,具体有以下三种方式:
(1)新鲜的汉麻花叶加入水中,汉麻花叶与水的固液比为1:10,搅拌1~2h,过滤分离,液体中含需要的大麻二酚酸(CDBA)及其它有价值的大麻素酸物质;
(2)新鲜的汉麻花叶加入95%乙醇中,汉麻花叶与乙醇的固液比为1:5,搅拌1~2h,过滤分离,液体中含需要的大麻二酚酸(CDBA)及其它有价值的大麻素酸物质;
(3)新鲜的汉麻花叶,利用超临界二氧化碳方式提取,搅拌1~2h,过滤分离,液体中含需要的大麻二酚酸(CDBA)及其它有价值的大麻素酸物质;
步骤二:取beta-环糊精(beta-cyclodextrin)或beta-环糊精的衍生物,加入水中,配制10%的水溶液,在2℃~30℃搅拌条件下,把汉麻提取液加入beta-环糊精液中,开始有beta-环糊精与大麻二酚酸形成包合物,包合物的溶解度很低,会从水中沉淀出来,搅拌1-2h,沉淀物不在增加,过滤分离出包合物。
进一步地,所述方法还包括步骤三:为了进一步提纯大麻二酚酸,去除包合物表面的其它杂质,滤饼可以用水或水和乙醇等有机溶剂的混合物漂洗。
进一步地,所述汉麻提取液与beta-环糊精液的质量比为1:1。
进一步地,包合物可以直接销售或制成不同的剂型。
一种大麻二酚的稳定方法,所述方法为:取100克95%的大麻二酚,加入500克的95%的乙醇,取100克的beta-环糊精,加入500克水中,搅拌,并慢慢加入CBD液体,beta-环糊精与CBD形成包合物,沉淀,搅拌4小时,过滤,用水或水和乙醇的混合物漂洗滤饼,把滤饼上吸附的其它杂质洗掉。
本发明相对于现有技术的有益效果为:beta-环糊精与大麻二酚酸包合物有效的使大麻二酚酸稳定,方法简便,成本低,并且在各种液体(如,水溶液,护肤液,乳液)中稳定,为大麻二酚酸的应用提供了一个新的方法。
同时,由于大麻二酚酸是油状物,基本不溶于水,不利于人体的吸收,溶解性很差,目前主要通过高温雾化方式吸入(电子烟),效率低,损失大。而beta-环糊精及衍生物的水溶性很好,使得包合物的水溶性大大地增加,使大麻二酚更容易吸收,效率高。

下面结合实施例对本发明的技术方案作进一步的说明,但并不局限于此,凡是对本发明技术方案进行修正或等同替换,而不脱离本发明技术方案的精神范围,均应涵盖在本发明的保护范围之中。
本发明用不同的方法提取的新鲜的大麻花叶,为了减少大麻二酚酸的降解,应采用低温提取,减少浓缩温度,得到的含大麻二酚酸的提取物的含量大于99%。
实施例1:
新鲜的大麻花叶,加水,1:10的固液比。搅拌2小时,过滤。滤液在搅拌的条件下,加入1:1花叶干重量的beta-环糊精,搅拌4小时,过滤。用水或水和乙醇的混合物漂洗滤饼,把滤饼上吸附的其它杂质洗掉,得到大麻二酚酸和beta-环糊精的包合物。

新鲜的大麻花叶,加零下20度的95%的乙醇,1:5的固液比。搅拌1小时,过滤。滤液在搅拌的条件下,加一到四倍重量的水,搅拌2小时,过滤。过滤中加入1:1花叶干重量的beta-环糊精,搅拌4小时,过滤。用水或水和乙醇的混合物漂洗滤饼,把滤饼上吸附的其它杂质洗掉,得到大麻二酚酸和beta-环糊精的包合物。
实施例3:
新鲜的大麻花叶,利用超临界二氧化碳提取,过滤。滤液在搅拌的条件下,加一到四倍重量的水,搅拌2小时,过滤分离。加入1:1花叶干重量的beta-环糊精,搅拌4小时,过滤。用水或水和乙醇的混合物漂洗滤饼,把滤饼上吸附的其它杂质洗掉,得到大麻二酚酸和beta-环糊精的包合物。
实施例4:
新鲜的大麻花叶,加零下20度的95%的乙醇,1:5的固液比。用萃取离心机萃取。滤液在搅拌的条件下,加一到四倍重量的水,搅拌2小时,过滤。过滤中加入1:1花叶干重量的羟丙基-beta-环糊精,搅拌4小时,过滤。用水或水和乙醇的混合物漂洗滤饼,把滤饼上吸附的其它杂质洗掉,得到大麻二酚酸和beta-环糊精衍生物的包合物。
将实施例1~4得到的包合物经HPLC分析,含CBDA大于99%,说明在提取包合过程中,大麻二酚酸几乎没有降解。
beta-环糊精与大麻二酚酸包合物的热稳定试验:
把实施例1~4中得到的包合物放70℃烘箱,70%相对湿度,两个星期,少于5%的大麻二酚酸变成大麻二酚。
把实施例1~4包合物放30℃烘箱,70%相对湿度,六个月,少于5%的大麻二酚酸变成大麻二酚。
实施例5:
取100克95%的CBD(大麻二酚),加入500克的95%的乙醇。取100克的beta-环糊精,加入500克水中,搅拌,并慢慢加入CBD液体。beta-环糊精与CBD形成包合物,沉淀,搅拌4小时,过滤。用水或水和乙醇的混合物漂洗滤饼,把滤饼上吸附的其它杂质洗掉。
beta-环糊精与大麻二酚(CBD)包合物的热稳定试验:
把实施例5包合物放70℃烘箱,70%相对湿度,两个星期,大麻二酚含量无变化。
把实施例5包合物放30℃烘箱,70%相对湿度,六个月,少于1%的大麻二酚损失。
实施例6:
新鲜的大麻花叶,加零下20度的95%的乙醇,1:5的固液比。用萃取离心机萃取。滤液在搅拌的条件下,加一到四倍重量的水,搅拌2小时,过滤。过滤中加入1:1花叶干重量的二甲基-beta-环糊精,搅拌4小时,过滤。用水或水和乙醇的混合物漂洗滤饼,把滤饼上吸附的其它杂质洗掉。
Claims (5)
1.一种大麻素酸的稳定方法,其特征在于:所述方法具体为:
步骤一:获取汉麻提取液,具体有以下三种方式:
(1)新鲜的汉麻花叶加入水中,汉麻花叶与水的固液比为1:10,搅拌1~2h,过滤分离;
(2)新鲜的汉麻花叶加入95%乙醇中,汉麻花叶与乙醇的固液比为1:5,搅拌1~2h,过滤分离;
(3)新鲜的汉麻花叶,利用超临界二氧化碳方式提取,搅拌1~2h,过滤分离;
步骤二:取beta-环糊精或beta-环糊精的衍生物,加入水中,配制10%的水溶液,在2℃~30℃搅拌条件下,把汉麻提取液加入beta-环糊精液中,开始有beta-环糊精与大麻二酚酸形成包合物,搅拌1-2h,过滤分离出包合物。
2.根据权利要求1所述的一种大麻素酸的稳定方法,其特征在于:所述方法还包括步骤三:滤饼用水或水和乙醇等有机溶剂的混合物漂洗。
3.根据权利要求1所述的一种大麻素酸的稳定方法,其特征在于:所述汉麻提取液与beta-环糊精液的质量比为1:1。
4.根据权利要求1所述的一种大麻素酸的稳定方法,其特征在于:包合物可以直接销售或制成不同的剂型。
5.一种大麻二酚的稳定方法,其特征在于:所述方法为:取100克95%的大麻二酚,加入500克的95%的乙醇,取100克的beta-环糊精,加入500克水中,搅拌,并慢慢加入CBD液体,beta-环糊精与CBD形成包合物,沉淀,搅拌4小时,过滤,用水或水和乙醇的混合物漂洗滤饼,把滤饼上吸附的其它杂质洗掉。
