本发明涉及渔药领域,更具体涉及基于黄鳝促性腺激素释放激素类似物的黄鳝人工催产方法。本发明的方法是将黄鳝促性腺激素释放激素类似物和地欧酮注射至性腺发育成熟的黄鳝中;本发明提供的修饰设计的黄鳝促性腺激素释放激素类似物MaGnRHa具有显著的促进黄鳝卵细胞成熟和排卵作用。怀卵待产的雌性亲鳝对MaGnRHa催产剂的效应时间短,催产效率高。修饰设计的MaGnRHa是一种高效的、特异针对黄鳝人工催产的活性肽。该催产剂运用黄鳝人工繁育生产具有明显的经济效益。

Description

基于黄鳝促性腺激素释放激素类似物的黄鳝人工催产方法
技术领域
本发明涉及渔药领域,更具体涉及基于黄鳝促性腺激素释放激素类似物的黄鳝人工催产方法。
背景技术
黄鳝(Monopterus albus)隶属于硬骨鱼纲、合鳃目、合鳃科、黄鳝属,是我国重要的名特优养殖鱼类。据2021中国渔业统计年鉴我国黄鳝养殖的年产量达30.7万吨。由于缺乏高效的人工繁育方法,养殖黄鳝的苗种还主要依赖于捕捞自然界的幼鱼,既限制了黄鳝养殖规模,也造成黄鳝种质资源的逐步减少。建立黄鳝人工繁育技术迫切需要高效的催产剂,然而目前市场上还没有针对黄鳝的催产剂。现在市售的常用于鱼类的催产剂主要包括脑垂体(PG)、绒毛膜促性腺激素(HCG)、促黄体生成素释放激素类似物(LRH-A)、地欧酮(DOM)等。脑垂体一般为自制的鲤鱼脑垂体,其原理是利用性成熟鲤鱼脑垂体中含有的促滤泡激素和促黄体素促使精卵细胞进一步发育成熟和诱导雌鱼发情产卵。由于脑垂体来源有限、制备价格高、催产效果不稳定等原因难于大规模运用于鱼类催产。另外,注射脑垂体悬浊液易引入外源病原物导致亲鱼感染死亡。绒毛膜促性腺激素为白色或灰白色粉末易溶于水,遇热易失效。研究表明繁殖季成熟的黄鳝腹腔注射100-4000IU/kg HCG,催产率为11-77%,效应时间长达92-168小时(乔德亮,2006)。陈鹏飞等人比较了雌性亲鳝腹腔注射HCG、LRH-A以及LRH-A+HCG的催产效果,结果表明注射催产剂48小时的亲鳝的排卵率分别为29.41%、35.29%、41.18%。由此可见,LRH-A对黄鳝亲鱼的催产效率略高于HCG。混合注射LRH-A和HCG的催产效果更好,但催产效率仍低于50%(陈鹏飞等,2001)。本发明比较了雌性亲鳝注射不同剂量的地欧酮的催产效果,结果表明单独注射地欧酮不能促使亲鳝排卵。综上所述,目前商业化的催产剂诱导黄鳝亲鱼排卵效应时间长、效率低,不能满足黄鳝人工催产和繁育的需求。
发明内容
本发明的目的在于提供了基于黄鳝促性腺激素释放激素类似物的黄鳝人工催产方法,方法简单,高效。
为了达到上述目的,本发明采取以下技术措施:
基于黄鳝促性腺激素释放激素类似物的黄鳝人工催产方法,其步骤包括将复合催产剂注射至性腺发育成熟的黄鳝中;所述的复合催产剂由黄鳝促性腺激素释放激素类似物和地欧酮组成;黄鳝促性腺激素释放激素类似物和地欧酮的质量比为1:(16.67-300)。
所述的黄鳝促性腺激素释放激素类似物为Pyro-Glu-His-Trp-Ser-Phe-[D-Trp]-Leu-Ser-Pro-Net。
以上所述的方法中,优选的,复合催产剂各组分的注射剂量为黄鳝促性腺激素释放激素类似物MaGnRHa注射量为10-30微克/千克鱼;地欧酮DOM注射注射剂量为0.5-3毫克/千克鱼。
本发明的保护范围还包括,上述催产方法在黄鳝育种或人工养殖中的应用。
本发明与现有技术相比,具有以下优点和效果:
现有的商业化催产剂对黄鳝的催产效果不佳。本发明提供的修饰设计的黄鳝促性腺激素释放激素类似物MaGnRHa具有显著的促进黄鳝卵细胞成熟和排卵作用。怀卵待产的雌性亲鳝对MaGnRHa催产剂的效应时间短,催产效率高。修饰设计的MaGnRHa是一种高效的、特异针对黄鳝人工催产的活性肽。该催产剂运用黄鳝人工繁育生产具有明显的经济效益。

具体实施方式
本发明所述技术方案,如未特别说明,均为本领域的常规方案;所述试剂或材料,如未特别说明,均来源于商业渠道。
实施例1:
黄鳝促性腺激素释放激素类似物的获得:
A、分析黄鳝内源促性腺激素释放激素编码基因结构,获得发挥生物活性的促性腺激素释放激素核心十肽,其氨基酸序列为:Gln-His-Trp-Ser-Phe-Gly-Leu-Ser-Pro-Gly。
B、针对核心十肽的特定位点进行修饰设计:第一位Gln转换为Pyro,第六位Gly转换为D-Trp,第十位Gly转换为Net。
C、人工合成获得修饰设计的MaGnRHa活性肽,其结构式为:Pyro-Glu-His-Trp-Ser-Phe-[D-Trp]-Leu-Ser-Pro-Net。
D、修饰设计的黄鳝促性腺激素释放激素类似物MaGnRHa与地欧酮DOM制备黄鳝特异催产剂。
实施例2:
黄鳝复合催产剂的获得:
将黄鳝促性腺激素释放激素类似物MaGnRHa与地欧酮DOM按照质量比1:(16.67-300)混合,既得。
所述的黄鳝促性腺激素释放激素类似物结构式为:Pyro-Glu-His-Trp-Ser-Phe-[D-Trp]-Leu-Ser-Pro-Net。
在本实施例中,黄鳝促性腺激素释放激素类似物与地欧酮的重量比为1:200。
实施例3:
不同的催产药物用于黄鳝人工催产:
实验过程如下:
以市售的催产剂LHR-A2为对照,对比分析修饰设计的MaGnRHa与LHR-A2促进雌性黄鳝卵细胞成熟和排卵的效果差异。由于鱼类垂体内滤泡刺激素和黄体生成素受多巴胺的抑制作用,采用促性腺激素释放激素类似物催产时通常与地欧酮(DOM)同时注射以消减抑制作用。黄鳝繁殖季节,选取雌性黄鳝亲本40尾,体重约100克/尾。随机分为对照组和实验组,每组20尾黄鳝置于55厘米Х45厘米的水箱中,水深15厘米,水温24℃。
对照组:对照组催产剂为1.5微克LHR-A2和0.3毫克DOM(购自宁波三生生物科技有限公司)溶解于0.5毫升的生理盐水,腹腔注射雌性黄鳝。注射催产剂的黄鳝遮阳网覆盖放置在水箱中,室内保持温度为24℃。
实验组:实验组催产剂为1.5微克MaGnRHa(委托南京金斯瑞生物科技有限公司合成)和0.3毫克DOM溶解于0.5毫升的生理盐水,腹腔注射雌性黄鳝。注射后的黄鳝放置在与对照组相同的环境待产。
结果表明注射含MaGnRHa催产剂后24小时即可检测到雌性黄鳝卵细胞成熟排卵,注射后60小时已有6条黄鳝产卵。而注射对照组催产剂60小时才检测到3条雌鱼排卵。随后,实验组和对照组黄鳝陆续排卵。催产剂注射后108小时后实验组共有17条黄鳝成功排卵,对照组共有13条黄鳝排卵。综上所述,注射含MaGnRHa催产剂的黄鳝排卵效应时间短于对照组,排卵效率高于对照组。MaGnRHa的催产效率显著提高。

1.基于黄鳝促性腺激素释放激素类似物的黄鳝人工催产方法,其步骤包括将复合催产剂注射至性腺发育成熟的黄鳝中;所述的复合催产剂由黄鳝促性腺激素释放激素类似物和地欧酮组成;黄鳝促性腺激素释放激素类似物和地欧酮的质量比为1:(16.67-300);
所述的黄鳝促性腺激素释放激素类似物为Pyro-Glu-His-Trp-Ser-Phe-[D-Trp]-Leu-Ser-Pro-Net。
2.根据权利要求1所述的方法,复合催产剂各组分的注射剂量为黄鳝促性腺激素释放激素类似物MaGnRHa注射量为10-30微克/千克鱼;地欧酮DOM注射注射剂量为0.5-3毫克/千克鱼。
3.权利要求1所述的催产方法在黄鳝育种或人工养殖中的应用。
