Abstract
本发明提供式(1)所示的IL‑15抑制剂Diosgenin,及其筛选方法和应用。本发明筛选方法包括:(1)根据IL‑15/IL‑15Rα复合物的三维晶体结构,确定IL‑15Rα拮抗剂潜在的结合位点;(2)IL‑15Ra蛋白去除所有杂原子和水后,加入氢原子,并合并所有非极性氢,通过AutoDockTools构建的Grid Box,设置网格中心;(3)将备选化合物在Chem3D中转换为3D结构后进行能量最小化,利用分子对接软件Autodock Vina将被选化合物与IL‑15Rα进行对接,并对结合情况打分,筛选出IL‑15抑制剂Diosgenin。本发明提供的IL‑15抑制剂Diosgenin可用于制备肿瘤免疫治疗药物,以及呼吸道系统相关疾病治疗药物。
Description
技术领域
本发明属于医药技术领域,具体涉及IL-15抑制剂Diosgenin,及其筛选方法和应用。
背景技术
IL-15有助于B细胞、T细胞和NK细胞的增殖和分化。通过与三聚复合物结合而发挥其细胞信号传导功能,所述三聚复合物由两个共有受体,即共同γ链(γc;CD132)和IL-2受体B链(IL-2Rβ;CD122),以及IL-15受体α(IL-15Rα;CD215)组成。被认为是肿瘤学中潜在有价值的治疗剂。
IL-15主要呈递是在单核细胞和树突状细胞上与IL-15Rα的膜结合异源二聚体复合物,其作用是通过将IL-15/IL-15Rα复合物反式呈递到例如在NK细胞和CD8+T细胞上发现的中等亲和力受体复合物(即IL-2Rβ/γ复合物)来实现的。
IL-15/IL-15Rα作为经典的免疫活性分子,参与调节多种免疫细胞的存活、增殖与功能,其中包括NK细胞、记忆性CD8+T细胞、NKT细胞等,在自身免疫性疾病、炎症性疾病、肿瘤等疾病的发生发展中发挥着重要的作用。在COVID-19的治疗方面认为IL-15可以作为免疫治疗的明星分子。目前针对IL-15的激动剂和抑制剂药物包括SHR-1501、ALT-803和BNZ-1等用于肿瘤的免疫治疗。然而除已报道的Cef外,目前没有其他商品化的可供临床使用的IL-15抑制剂。
并且免疫分子对神经精神疾病发生发展的调控成为近年来研究的热点,而以IL-15/IL-15Rα系统为靶点的小分子药物在神经系统相关疾病和COVID-19的治疗中鲜有研究。
发明内容
有鉴于此,为了克服现有技术的不足,本发明提供一种IL-15抑制剂Diosgenin,该小分子化合物Diosgenin对IL-15刺激的MO7E细胞增殖具有剂量依赖的显著性抑制作用。

本发明还提供上述IL-15抑制剂Diosgenin的筛选方法,包括步骤:
1)根据IL-15/IL-15Rα复合物的三维晶体结构,观察IL-15与IL-15Rα的结合部位并确定IL-15Rα拮抗剂潜在的结合位点;
3)将备选化合物在Chem3D中转换为3D结构后进行能量最小化,利用分子对接软件Autodock Vina将被选化合物与IL-15Rα进行对接,并对结合情况打分,筛选出IL-15抑制剂Diosgenin。
本发明还提供上述IL-15抑制剂Diosgenin在制备肿瘤免疫治疗药物中的应用。
进一步,所述应用剂量为30μg/mL。
本发明还提供一种肿瘤免疫治疗药物,所述药物成分包括:Diosgenin或其衍生物,以及制药学上可接受的辅助成分。

与现有技术相比,本发明的有益技术效果在于:
1.本研究证明小分子化合物Diosgenin(30μg/mL)对IL-15刺激的MO7E细胞增殖具有剂量依赖的显著性抑制作用。优点:①Diosgenin在30μg/mL处理条件下抑制作用远超过阳性对照Cefazolin(100μg/mL);②除了Diosgenin和已经报道的Cefazolin为IL-15抑制剂以外,目前没有商品化的可供临床使用的IL-15抑制剂。
2.本发明研究表明IL-15抑制剂Diosgenin可用于制备肿瘤免疫治疗药物。
3.本发明研究表明IL-15抑制剂Diosgenin有潜力用于制备呼吸道系统相关疾病治疗药物。
4.本发明研究表明IL-15抑制剂Diosgenin有潜力用于制备神经系统相关疾病治疗药物。

具体实施方式
以下通过具体实施例对本发明进行详细说明,需要理解的是,下述实施例仅作为解释和说明,不以任何形式限制本发明的范围。下述实施方式中,未特别说明的生物化学试剂均为本领域常规试剂,可以按照本领域常规方法配制而得或商购获得,规格为实验室纯级即可。
Diosgenin,中文名:薯蓣皂素,CAS NO:512-04-9,购于普思生物科技。
实施例:Diosgenin的IL-15抑制剂活性
1.分子对接虚拟筛选:
从Protein Data Bank中提取IL-15/IL-15Rα复合物的三维晶体结构(PDB:2Z3Q),通过观察IL-15与IL-15Rα的结合部位(图1)以确定IL-15Rα拮抗剂潜在的结合位点。将IL-15Ra蛋白的所有的杂原子和水去除后,加入所有氢原子,并合并所有非极性氢,随后通过AutoDockTools构建的Grid Box,设置网格中心(x,y,z=52.489,6.812,16.214)。另外,需要筛选的化合物在Chem3D中转换为3D结构,并进行能量最小化。最后利用分子对接软件Autodock Vina对48种中药中的3000多种化合物与IL-15Rα进行对接,并对结合情况打分。
分子对接结果发现化合物Diosgenin(C27H42O3,分子量:414.6,结构式如图2所示):Diosgenin在IL-15Rα与IL-15结合界面中更偏向于结合IL-15Rα的中部和底部,并与残基R35形成氢键作用。另外,IL-15Rα中与Diosgenin相互作用的氨基酸残基R26、K34、R35、S41、P67均参与IL-15与IL-15Rα结合过程。通过Autodock Vina对接打分的Affinity为-6.0kcal/mol。从而推测Diosgenin可能通过图3中的结合方式发挥作用。
2.人巨细胞白血病细胞(MO7E)验证:
MO7E细胞可用于IL-15刺激后的细胞增殖实验,已被R&D公司所证实(https://www.bio-techne.com/cn/)。取对数生长期MO7E细胞,用RPMI 1640完全培养基调整细胞密度至2.15×105个/mL的细胞悬液,接种于96孔培养板中,每孔100μL,置于37℃、5%CO2饱和湿度培养箱中;培养12h后加药,加入100μL含不同浓度Diosgenin(纯度≥98%;厂家:普思生物科技)的RPMI 1640完全培养液,每个浓度设8个复孔,分别设4个浓度梯度,终浓度分别为1、3、10、30μg/mL。空白对照组加入100μL的RPMI 1640完全培养液;IL-15处理组:分别加入含IL-15(30ng/mL)、IL-15+Cefazolin(100μg/mL)、IL-15+Diosgenin(1、3、10、30μg/mL)的RPMI 1640完全培养液;单独Diosgenin处理组:分别加入含Diosgenin(1、3、10、30μg/mL)的RPMI 1640完全培养液。置于IncuCyte S3活细胞分析系统下实时连续监测70h内MO7E细胞的生长增殖情况。
3.统计学方法
采用Graphpad分析软件对实验数据进行处理,计量资料以Mean±SEM表示,采用Two-way AVOVA方法进行数据分析,Tukey’s multiple comparisons test进行组间比较,p<0.05具有显著性差异。
4.研究结果如下:
1.IL-15(30ng/mL)刺激后,可显著提高MO7E细胞的增殖活性(p<0.0001)(图4);
2.Diosgenin(30μg/mL)处理后,对IL-15刺激后的MO7E细胞表现出显著的增殖抑制作用,并呈明显的剂量依赖关系(p<0.0001),阳性药Cefazolin(100μg/mL)处理后出现显著性差异(p<0.01)(图4);
3.单独Diosgenin(1、3、10、30μg/mL)处理对MO7E细胞增殖无抑制作用(p>0.05)(图5)。
