Abstract
本发明涉及一种MazF介导的FTO酶的检测方法及抑制剂筛选方法。RNA去甲基化酶RNA去甲基酶的异常表达与各种人类疾病有关,可作为新的生物标志物和潜在治疗靶点。因此,灵敏的检测RNA去甲基酶并筛选其抑制剂对生物医学研究和临床诊断至关重要。本发明提供了一种利用MazF介导的引物诱导的滚环扩增的无标记方法灵敏检测FTO活性。该方法操作简单,灵敏度高,特异性好,可在单细胞水平检测FTO活性。此外,该方法可用于动力学参数的测定和FTO抑制剂的筛选,有望应用于抗肿瘤活性化合物的开发。

技术领域
本发明属于RNA去甲基酶检测技术领域,具体涉及一种基于MazF介导的滚环扩增(PG-RCA)的无标记荧光方法,及所述检测方法在FTO酶抑制剂筛选领域的应用。
背景技术
公开该背景技术部分的信息仅仅旨在增加对本发明的总体背景的理解,而不必然被视为承认或以任何形式暗示该信息构成已经成为本领域一般技术人员所公知的现有技术。
RNA去甲基酶的异常表达与各种人类疾病有关,例如,ALKBH5与精子发生缺陷和胶质母细胞瘤干细胞样细胞的致瘤性有关,而FTO与代谢和神经退行性疾病、肥胖、糖尿病、癌症和阿尔茨海默病等密切相关。所以,RNA去甲基酶可作为新的生物标志物和潜在治疗靶点。因此,灵敏的检测RNA去甲基酶并筛选其抑制剂对生物医学研究和临床诊断至关重要。
N6甲基腺苷(m6A)修饰是腺嘌呤氮-6基团的转录后甲基化,是真核细胞mRNA中最丰富的内部修饰之一。m6A修饰在mRNA代谢阶段的各个阶段、长链非编码RNA和miRNA的产生以及包括神经元发育、细胞命运转变、免疫反应和肿瘤发生在内的多种生物过程中起着关键作用。N6甲基腺苷(m6A)的水平由RNA甲基转移酶(例如,甲基转移酶-3/甲基转移酶-14复合物)和RNA去甲基化酶(例如,脂肪量和肥胖相关蛋白(fat mass and obesity-associatedprotein,FTO)和AlkB同源物(ALKBH5))动态调节,它们可以对核酸碱基添加和去除“甲基”。FTO和ALKBH5都属于Fe2+和2-氧戊二酸(2OG)依赖的AlkB双加氧酶家族。RNA去甲基酶的异常表达与各种人类疾病有关。例如,ALKBH5与精子发生缺陷和胶质母细胞瘤干细胞样细胞的致瘤性有关,而FTO与代谢和神经退行性疾病(如肥胖、糖尿病、癌症和阿尔茨海默病)密切相关。RNA去甲基酶可作为新的诊断生物标志物和有希望的治疗靶点。因此,灵敏地检测RNA去甲基酶并筛选其抑制剂对生物医学研究和临床诊断至关重要。
RNA去甲基酶活性测定的传统方法包括放射性检测法、高效液相色谱法(HPLC)、质谱法(MS)。然而,放射性分析需要一个特定的放射性同位素标签,该方法成本高且存在放射性污染的危险。高效液相色谱法(HPLC)和质谱法(MS)依靠直接检测甲基化水平的变化检测RNA去甲基化酶活性,但这些方法涉及费力的制备步骤和大量样品。此外,这些方法受到低吞吐量和低灵敏度的限制,无法实现对微量目标物的灵敏检测。针对上述研究背景,发明人认为,提供一种灵敏检测RNA去甲基酶的方法有助于尽早发现机体中RNA去甲基化酶的变化情况,对于若干疾病的早期诊断和相关活性化合物的筛选具有重要意义。
DpnⅡ内切酶(DpnⅡ)是一种甲基化敏感的DNA限制性内切酶,以单链DNA(ssDNA)为底物,用于FTO/ALKBH5活性测定。在单链DNA被FTO和ALKBH5脱甲基后,单链DNA与其互补的DNA序列一起退火,然后在识别位点被DpnⅡ识别和切割。DpnⅡ与聚丙烯酰胺凝胶电泳(PAGE)、HPLC和基于单量子点的荧光共振能量转移(FRET)的结合使得FTO/ALKBH5活性的检测变得非常简单。然而,聚丙烯酰胺凝胶电泳(PAGE)和高效液相色谱法(HPLC)通常需要大量FTO/DNA底物和复杂的样品制备步骤;荧光共振能量转移(FRET)分析需要仔细计算供体和受体之间的距离和角度,以实现高FRET效率。近期,MazF mRNA切割酶(MazF)被引入m6A定位、RNA去甲基化酶分析和抑制剂筛选。大肠杆菌毒素MazF是一种ACA序列特异性内核糖核酸酶,它可以区分A和m6A,并切割RNA的未甲基化5′-ACA-3′序列,但对m6ACA序列没有切割活性。
发明内容
基于本领域对上述内切酶的研究成果,本发明联想到提供一种基于MazF介导的滚环扩增(PG-RCA)的RNA去甲基化酶检测方法,可以实现无标记检测,再引入环形模板进行扩增实现对检测信号的指数放大,从而灵敏地检测RNA去甲基化酶。
为了实现上述技术效果,本发明提供以下技术方案:
本发明第一方面,提供一种检测试剂的组合,所述组合至少包括RNA探针,MazFmRNA切割酶,T4 PNK酶及环形模板;所述RNA探针中具有m6ACA序列,所述环形模板中具有引物结合位点,其扩增链上具有至少一个内切酶识别位点。
本发明设计的检测方法中,上述RNA探针中携带m6ACA,检测环境中的FTO酶能够识别并消除RNA探针中的甲基化,去甲基化的探针被MazF酶识别并且切成为两条短链,A链和B链,所述B链的3’端保留原RNA探针3’的氨基修饰,所述A链的3’端暴露出2′,3′-环磷酸基团,在T4 PNK酶的催化作用下形成3′-羟基末端从而成为环形模板的引物链,所述引物链与环形模板中的识别位点结合引发扩增,扩增形成的扩增链携带内切酶的识别位点从而被剪切成使荧光染料发光的单链DNA。
因此,基于上述检测方法的设计,本发明第一方面提供的RNA探针应当具有一定的长度,从而保证切割后的A链能够特异性的与环形模板结合,所述RNA探针恰当的长度如20~40个碱基,其3’端具有封闭效果的基团进行修饰,如氨基(-NH2)或磷酸基团修饰,由于氨基修饰具有更好的稳定性,本发明提供的RNA探针,其3’端采用氨基修饰。
另外,所述RNA探针的序列中具有m6ACA,并且m6ACA位于RNA探针序列全长的中部,如距离5’端10~30个碱基处;并且m6ACA5’端邻位的碱基具有2′,3′-环磷酸基团修饰。更为具体的方案中,所述RNA探针的长度为28~35个碱基;所述A链的长度为15~20个碱基。
第一方面优选的方案中,所述环形模板在引物结合后,通过DNA聚合酶引发循环扩增,因此所述检测试剂的组合中,还应当具有DNA聚合酶和底物,具体实例如vent(exo-)聚合酶和4种dNTP。
第一方面优选的方案中,所述环形模板扩增后的扩增链中具有一个或多个内切酶的识别位点所述内切酶的一个实例如Nb.BsmI切刻内切酶;所述Nb.BsmI切刻内切酶的识别位点为两个。
能够满足上述优选方案的一种具体实施方式中,所述RNA探针具有如下序列:

上述序列中,加粗字体部分为引物链的结合区域。
上述环形模板的扩增链在内切酶的作用下成指数放大,形成大量单链DNA,此时向检测体系中加入荧光结合染料即可通过检测荧光强度对体系中的FTO酶进行检测。因此,又一种优选的实施方式中,所述检测试剂的组合中还具有荧光DNA结合染料,具体实例如SYBRgold。
本发明第二方面,提供第一方面所述检测试剂的组合在制备RNA去甲基酶检测产品中的应用。
上述第二方面的具体实例中,所述RNA去甲基酶为FTO。
优选的,所述检测产品包括但不限于试剂盒、检测芯片或检测系统中的一种;其中,试剂盒的一种实施方式中,所述试剂盒中包括第一方面所述检测试剂的组合,还包括缓冲体系、防腐剂等;所述缓冲体系包括FTO反应缓冲液、MazF反应缓冲液、T4 PNK反应缓冲液、PG-RCA反应缓冲液:
具体的,所述FTO反应缓冲液为HEPES缓冲液,其中还包括α-酮戊二酸、L-抗坏血酸及(NH4)2Fe(SO4);

本发明第三方面,提供一种MazF介导的FTO酶的检测方法,所述检测方法依靠第一方面所述检测试剂的组合而实现,检测方法包括以下步骤:
将RNA探针加入待测体系中在25~35℃孵育1~2h,将反应体系升温至90~100℃加热4~6min使检测体系中的酶失活,将反应产物加入含有MazF酶的缓冲体系中35~40℃孵育1~2h,再加入T4 PNK在35~40℃下孵育4~6h获得脱磷酸产物,将所述脱磷酸产物与环形模板、DNA聚合酶、底物、内切酶混合启动滚动循环扩增(PG-RCA)反应,反应温度为40~50℃,反应时间为1~3h;反应结束后加入SYBRgold染料并对荧光强度进行检测。
上述荧光强度的检测可采用本领域常规方式,包括采用荧光光谱仪、荧光分光光度计等进行测量,还包括采用凝胶电泳分析方法。
本发明第四方面,提供一种FTO抑制剂的筛选方法,所述筛选方法包括如下步骤:将待测抑制剂与RNA探针、FTO酶混合孵育,采用第三方面所述检测方法对上述孵育后的FTO酶活力进行检测。
以上一个或多个技术方案的有益效果是:
本发明的一个实施方式中提供了一种基于MazF介导的PG-RCA反应在单细胞水平上灵敏检测FTO的无标记荧光方法。利用MazF催化裂解反应的高度特异性和PG-RCA的高扩增效率,该方法灵敏度高,检测限低至7.62×10-6nM和5个数量级的大动态范围(1×10-5nM到1nM),这优于大部分已报道的RNA去甲基酶检测方法。与放射性标记检测法、HPLC和MS等传统方法相比,该方法可避免使用放射性标记底物和复杂设备(如HPLC和MS)以及复杂的样品制备步骤。
与已报道的基于荧光m6A可切换探针的检测方法和基于荧光RNA适配体的检测方法以及基于Dpn II的FRET分析方法相比,该方法非常简单、无标记且成本低益,无需任何荧光修饰和复杂的核酸结构设计。
此外,上述检测方法可以很容易地将癌细胞与正常细胞区分开来,甚至可以在单细胞水平上测量FTO活性。重要的是,该方法可用于筛选FTO抑制剂,在抗癌药物发现方面具有巨大潜力。

正如背景技术所介绍的,RNA去甲基酶与多种人类疾病相关,提供一种早期灵敏检测RNA去甲基酶的方法对于疾病的诊断具有重要意义,另外,上述检测方法还可以应用于RNA去甲基酶抑制剂的筛选,有望作为一种活性药物的筛选方式。
为了实现上述检测,本发明设计了一种含有5′-m6ACA-3′序列的RNA探针为底物。为了防止MazF切割介导的非特异性扩增,本发明采用NH2修饰RNA探针的3′端。本发明设计的环形模板包含一个引物结合位点和两个Nb.BsmI(5′-GAA TGC-3′)的切割位点。RNA去甲基酶测定的原理如图2所示。以FTO作为模型,所述检测方法涉及以下三步:(1)FTO催化的RNA探针去甲基化和随后的MazF介导的去甲基化RNA探针的切割,(2)T4 PNK催化的引物2′,3′-环磷酸端到3′-羟基化端的去磷酸化,(3)PG-RCA反应。当FTO存在时,它从RNA探针中去除甲基,然后去甲基化的RNA探针被识别并被MazF切割成两部分:一部分是一个17碱基的RNA单链,在RNA链的3′端带有2′,3′-环磷酸(A链);另一条为12碱基的RNA链,其3′端为NH2。通过添加T4 PNK(它可以将2′,3′-环磷酸转化为3′-羟基),它催化去除17碱基RNA链3′端的2′,3′-环磷酸基团,以生成具有3′-羟基末端的17碱基RNA链。水解后的17碱基RNA链可以作为引物与环形模板结合,在vent(exo-)聚合酶存在下启动滚动循环扩增(RCA)的级联反应,产生环形模板的重复互补序列的DNA长链。扩增得到的重复序列可被Nb.BsmI切割产生大量的新引物,这些引物可以启动新的RCA反应,以指数放大的方式产生大量的ssDNA。生成的ssDNA以SYBR gold为指示剂,能产生明显的荧光信号。相反,在缺乏FTO的情况下,甲基化RNA探针不能被MazF切割。因此,PG-RCA反应无法启动,且无法观察到荧光增强。
进一步的,本发明还针对上述RNA去甲基化酶活力的检测方法及抑制剂筛选方法的可行性及灵敏度进行验证:
1.可行性验证
本发明进行了非变性PAGE分析,以验证裂解反应(图2A)和PG-RCA反应(图2B)。在没有FTO的情况下,只有一条原始RNA探针带(图2A,lane1),表明既没有发生去甲基化反应,也没有发生裂解反应。但是在FTO存在的情况下,去甲基化RNA探针被MazF切割,产生一条新的17碱基RNA链带(图2A,通道2),表明发生去甲基化和切割反应。PAGE分析也证实了随后的PG-RCA反应(图2B)。在没有FTO的情况下,仅观察到圆形模板(74碱基)和RNA探针(29碱基)的条带(图2B,通道1)。在FTO存在的情况下,检测到PG-RCA反应产物的两个不同条带(27碱基和47碱基)(图2B,通道3)。值得注意的是,在存在FTO但不含T4 PNK的情况下,无法观察到PG-RCA反应产物的条带(27碱基和47碱基)(图2B,通道2),因为MazF对去甲基化RNA探针的切割产生了一条在3′端带有2′,3′-环磷酸的RNA链,该链不能作为诱导PG-RCA反应的引物。
本发明进一步进行了荧光发射光谱测量(图2C)。在缺乏FTO的情况下,MazF无法消化RNA探针,并且由于缺乏PG-RCA反应,因此未检测到明显的荧光信号(图2C)。在FTO存在下,它催化RNA探针的去甲基化,随后MazF识别并切割去甲基化RNA探针的5′-ACA-3′位点以生成引物。由此产生的引物可触发PG-RCA反应,产生丰富的单链DNA,以产生独特的荧光信号(图2C)。此外,本发明还进行了实时荧光测量(图2D)。在没有FTO的情况下,未观察到明显的荧光信号(图2D)。相反,在FTO和Nb.BsmI存在的情况下,荧光强度随反应时间增加,并在2小时内达到平台(图2D),表明PG-RCA反应以指数放大的方式进行。在FTO存在但无Nb.BsmI的情况下,RCA反应以线性方式进行,导致相对较低的荧光强度(图2D)。
2.灵敏度实验
最佳实验条件下,本发明测量了不同浓度FTO产生的荧光信号。如图3A所示,荧光强度随着FTO浓度从1×10-5增加到300nM而增强,并且在1×10-5到1nM的范围内,荧光强度与FTO浓度的对数呈线性关系(R2=0.9966)(图3B),方程为Y=6714.61log10C+1011.08,其中Y为荧光强度,C为FTO浓度(nM)。检测限(LOD)计算为7.62×10-6nM,即空白平均信号加三倍标准差。该方法具有超高灵敏度和极大的动态范围,优于大多数现有分析方法(见ESI,表S2)。值得注意的是,该方法的灵敏度比基于甲基化可切换探针的荧光分析(10nM)高7个数量级,比基于杂交链式反应的荧光分析(2.273nM)高6个数量级,比基于DNAzyme的荧光分析(0.5nM)高5个数量级,比基于单个QD的FRET传感器(7.9×10-14M)高1个数量级。灵敏度的提高可归因于以下三个因素:(1)MazF催化裂解反应的高度特异性使去甲基化底物与甲基化底物得以区分,(2)NH2对RNA探针3′端的修饰有效地防止了非特异性扩增,(3)Nb.BsmI使线性RCA反应转变为PG-RCA反应,显著提高了扩增效率。
3.特异性实验
该方法对FTO具有良好的选择性,对非特异性酶/蛋白质无反应(图4A),可用于准确评估FTO的动力学参数(图4B),测量的最大初始速度(Vmax)和米氏常数(Km)分别为19.97nM/min和670.37nM。
以尿嘧啶-DNA糖基化酶(UDG)、碱性磷酸酶(ALP)、Dam DNA甲基转移酶(DamMTase)、牛血清白蛋白(BSA)和免疫球蛋白G(IgG)为干扰因子,考察该方法的选择性。牛血清白蛋白和IgG是非特异性蛋白。ALP可以催化多种磷酸化底物(如蛋白质和核酸)的去磷酸化。UDG通过催化尿嘧啶与DNA骨架间糖苷键的裂解,特异性地识别并去除尿嘧啶。DamMTase能够以s-腺苷-l-蛋氨酸为甲基供体,催化特定基因组DNA序列中甲基向腺嘌呤残基转移。如图4A所示,没有明显的荧光信号检测的BSA(图4A),IgG(图4A),UDG(图4A),ALP(图4A柱)和Dam(图4A),而FTO生成高荧光信号的存在(图4A),表明该方法对FTO具有良好的选择性。
本实施例中,针对实施例1中检测方法应用于FTO抑制剂的筛选进行了考察,本实施例选择大黄酸和双醋瑞因作为模型抑制剂。如图4所示,相对活性FTO表现出剂量依赖性,分别随双醋瑞因浓度(图4C)和大黄酸浓度(图4D)的增加而降低。计算得出双醋瑞因的半抑制浓度(IC50)为0.96μM,大黄酸为30.91μM,与基于单个量子点的荧光共振能量转移传感器(双醋瑞因为1.51μM)和基于DNA酶的荧光分析方法获得的值(大黄酸为47.2μM)一致,表明该方法用于筛选FTO抑制剂的可行性。
为了使得本领域技术人员能够更加清楚地了解本发明的技术方案,以下将结合具体的实施例详细说明本发明的技术方案。
实施例1一种FTO检测试剂的组合及其检测方法
本实施例中,提供一种RNA去甲基化酶FTO的检测试剂,所述检测试剂包括RNA探针及环形模板,其序列如下表1所示:

在20μL的反应混合物中进行FTO去甲基化酶活性检测,该反应混合物中含有500nMRNA探针、不同浓度的FTO蛋白和1×FTO反应缓冲液(50mM HEPES(pH 7.5)、100μMα-酮戊二酸、100μM L-抗坏血酸和150μM(NH4)2Fe(SO4)2)在30℃下反应90min,随后在95℃下失活5min后终止反应。将10μL反应产物与10U MazF在MazF反应缓冲液(40mM磷酸钠(pH 7.5)、0.01%吐温-20和5mM EDTA)中在37℃孵育90min。然后在30μL含有1μL 10U/μL T4 PNK,3μL10×T4 PNK反应缓冲液(70mM Tris-HCl,10mM MgCl2,5mM DTT,pH 7.6),6μL H2O和20μL消化产物的反应混合物在37℃下反应5h,进行脱磷酸化反应。随后在65℃下孵育20min终止反应。在50μL含有600μM dNTP,60nM圆形模板,0.8U的vent(exo-)DNA聚合酶,15U的Nb.BsmI,30μL脱磷反应产物,和5μL 10×thermopol反应缓冲液(20mM Tris-HCl,10mM(NH4)2SO4,10mM KCl,2mM MgSO4,0.1%Triton X-100,pH 8.8)的反应混合液中进行引物生成滚动循环扩增(PG-RCA)反应,在45℃反应2h。用纯水和1×SYBRgold染料将50μL扩增反应产物稀释至最终体积80μL。使用日立F-7000荧光分光光度计杯测量荧光光谱,激发波长为488nm。在500-700nm波长范围内记录发射光谱,激发和发射的狭缝宽度均为5nm。使用556nm处的荧光强度进行数据分析。
本实施例中,还提供一种凝胶电泳分析检测PG-RCA反应产物的方法:
裂解反应产物用SYBR gold染色,并用12%非变形聚丙烯酰胺凝胶电泳(PAGE)在1×TBE缓冲液(9mM Tris-HCl,pH 8.3,9mM硼酸,0.2mM EDTA)中,110V恒定电压下电泳40min。随后在Bio-Rad ChemiDoc MP成像系统中拍摄照片。PG-RCA反应产物用16%PAGE在1×TBE缓冲液中以150V恒定电压进行电泳60分钟。电泳后,在Bio-Rad ChemiDoc MP成像系统中,使用Epi蓝色(460–490nm激发)光源和516–544nm SYBRgold荧光团滤光片拍摄照片。
实施例2 FTO抑制剂的筛选方法
由于FTO与各种人类疾病相关,FTO抑制剂的筛选可能为抗癌药物的发现提供新的前景,本实施例对上述方案应用于抑制剂筛选的方式进行说明:
1、抑制试验
不同浓度的不同抑制剂与含有500nM RNA探针,0.1nM FTO和1×FTO反应缓冲液的反应混合液在30℃孵育90min。5小时,在95℃下孵育5分钟终止反应。按照上述FTO测定程序进行后续实验。根据方程式1计算FTO的相对活性(RA)。
其中Yt表示存在0.1nM FTO时的荧光强度,Yi表示存在0.1nM FTO和不同抑制剂时的荧光强度。根据线性相关方程获得Ci和Ct(图3B)由曲线拟合方程得到缓蚀剂的IC50值。

2、细胞培养和细胞质提取物的制备
MDA-MB-231细胞在Leibovitz的L-15培养基(美国Gibco)的玻璃底培养皿上生长添加10%胎牛血清和1%青霉素链霉素。MCF-7细胞和HeLa细胞在添加10%胎牛血清和1%青霉素/链霉素的Dulbecco改良Eagle培养基中生长。MCF-10A细胞在MCF-10A完全生长培养基中培养。细胞在37℃下在5%CO2的加湿室内培养。根据制造商的方案,使用核提取试剂盒(Active Motif,Carlsbad,CA,USA)获得细胞质裂解物。所得上清液进行FTO分析。

Claims (8)
1.一种检测试剂的组合,其特征在于,所述检测试剂的组合中,包括RNA探针,MazFmRNA切割酶,T4 PNK酶及环形模板;所述RNA探针中具有m6ACA序列,所述环形模板中具有引物结合位点,其扩增链上具有至少一个内切酶识别位点;
所述RNA探针被检测环境中的FTO酶识别并消除RNA探针中的甲基化,去甲基化的探针被MazF酶识别并且切成为两条短链,A链和B链,所述B链的3’端保留原RNA探针3’的氨基修饰,所述A链的3’端暴露出2′,3′-环磷酸基团,在T4 PNK酶的催化作用下形成3′-羟基末端从而成为环形模板的引物链,所述引物链与环形模板中的识别位点结合引发扩增,扩增形成的扩增链携带内切酶的识别位点从而被剪切成使荧光染料发光的单链DNA;
所述RNA探针的序列如下:

2.如权利要求1所述检测试剂的组合,其特征在于,所述检测试剂的组合中,还具有DNA聚合酶和底物,所述DNA聚合酶为vent(exo-)聚合酶,所述底物为4种dNTP。
3.如权利要求1所述检测试剂的组合,其特征在于,所述环形模板扩增后的扩增链中具有一个或多个内切酶的识别位点的所述内切酶为Nb.BsmI切刻内切酶;所述Nb.BsmI切刻内切酶的识别位点为两个。
4.权利要求1-3任一项所述检测试剂的组合在制备RNA去甲基酶检测产品中的应用,所述RNA去甲基酶为FTO。
5.如权利要求4所述的应用,其特征在于,所述检测产品包括试剂盒、检测芯片或检测系统中的一种;
其中,所述试剂盒中包括权利要求1-3任一项所述检测试剂的组合,还包括缓冲体系、防腐剂。
6.如权利要求5所述的应用,其特征在于,所述缓冲体系选自FTO反应缓冲液、MazF反应缓冲液、PG-RCA反应缓冲液及T4 PNK反应缓冲液:
所述FTO反应缓冲液为HEPES缓冲液,其中还包括α-酮戊二酸、L-抗坏血酸及(NH4)2Fe(SO4);所述MazF反应缓冲液为磷酸钠缓冲液,其中还具有吐温-20和EDTA;
所述T4 PNK反应缓冲液为Tris缓冲液,其中还具有MgCl2及DTT;
所述PG-RCA反应缓冲液为Tris缓冲液,其中还具有(NH4)2SO4、 KCl、MgSO4及Triton X-100。
7.一种非诊断目的的FTO酶的检测方法,其特征在于,所述检测方法依靠权利要求1-3任一项所述检测试剂的组合而实现,检测方法包括以下步骤:
将RNA探针加入待测体系中在25~35℃孵育1~2h,将反应体系升温至90~100℃加热4~6min使检测体系中的酶失活,将反应产物加入含有MazF酶的缓冲体系中35~40℃孵育1~2h,再加入T4 PNK在60~70℃下孵育10~30 min获得脱磷酸产物,将所述脱磷酸产物与环形模板、DNA聚合酶、底物、内切酶混合启动滚动循环扩增反应,反应温度为40~50℃,反应时间为1~3h;反应结束后加入SYBR gold 染料并对荧光强度进行检测;
所述荧光强度的检测包括采用荧光光谱仪和荧光分光光度计进行测量,还包括采用凝胶电泳分析方法。
8.一种FTO抑制剂的筛选方法,其特征在于,所述筛选方法包括如下步骤:将待测抑制剂与RNA探针、FTO酶混合孵育,采用权利要求7所述检测方法对孵育后的FTO酶活力进行检测。
