Abstract

香草酸作为植物源药剂在防治落叶松枯梢病上的应用。本发明涉及香草酸的一种应用,本发明的目的是为了防治落叶松枯梢病,解决化学农药防治落叶松枯梢病容易产生农药残留以及污染环境的问题,本发明将香草酸溶于二甲基亚砜中配制香草酸原液,然后用去离子水稀释配制工作液。香草酸对落叶松枯梢病病原落叶松葡萄座腔菌具有良好的抑菌效果,1~5mg/ml浓度下抑菌率可达30~70%。香草酸的应用能够有效抑制落叶松葡萄座腔菌,且降低了防治过程中对化学药剂的依赖性。本发明应用于植物源抑菌剂领域。

技术领域
本发明涉及香草酸作为植物源药剂在防治落叶松枯梢病上的应用。
背景技术
落叶松生长速度快,具有耐低温、干旱的优良特性,广泛分布于我国东北、华北、西北和西南地区,是我国东北、内蒙古等高山针叶林主要森林组成树种。落叶松具保土、防风功效,可作为防风固沙林、水土保持林和水源涵养林的优良树种。其根系发达,抗风力强,可作为江河流域的高山地区森林更新和荒山造林的主要树种。落叶松木材具有结构致密、材质坚韧、耐腐性强等优良特性,是重要的工业、建筑业用材,具有较高的经济价值。
由落叶松葡萄座腔菌(Botryosphaeria laricin)引起的落叶松枯梢病是落叶松的重要病害之一,该病害危害严重、蔓延迅速,是一种危险性检疫病害。1938年于日本北海道发现落叶松枯梢病,受害落叶松由开始的单株发展为大面积受害。1960~1969年北海道地区因该病害伐除和烧毁的落叶松人工林达9700公顷。落叶松枯梢病于70年代初期在中国发生,十几年后即已遍布东北三省广大林区。根据调查,连续发病9年的落叶松每公顷平均每年损失生长量为2.04立方米,约为该林分9年内每公顷平均生长量的60%。
落叶松枯梢病为害落叶松当年新梢,从幼苗、幼树到成林的当年新梢均能发病,但幼树发病较重。兴安落叶松、长白落叶松、海林落叶松、华北落叶松、朝鲜落叶松和日本落叶松均对其感病,除日本落叶松病情较轻外,其余落叶松感病程度较高。感病落叶松起初在未木质化的新梢嫩茎或嫩轴部褪绿,而后由浅褐色逐渐变为暗褐色、黑色。枝条有树脂溢出,上部弯曲成钩状,针叶枯萎,大部分脱落,只在顶部残留一丛针叶。发病部位以上枝梢枯死,幼苗成为无顶苗。次年春季,由侧芽生小枝代替原来主梢,连年发病,受害严重者,树冠出现五花头和扫帚状枝丛,高生长停止,形成小老树或全株枯死,严重影响落叶松生长。该病害的危害严重、传播迅速,已被列为森林植物检疫对象。目前防治该病害最有效的方式为化学防治,但化学农药的滥用会导致生态环境恶化、生物多样性降低等问题,农药的残留严重威胁人类健康。
发明内容
本发明的目的是为了防治落叶松枯梢病,解决化学农药防治落叶松枯梢病容易产生农药残留以及污染环境的问题,提供了香草酸作为植物源药剂在防治落叶松枯梢病上的应用。

本发明相对于现有技术的优点:
香草酸(4-羟基-3-甲氧基苯甲酸),分子式为C8H8O4。香草酸是胡黄连等抗菌植物的有效成分之一,本发明应用体外抑菌实验验证香草酸对落叶松葡萄座腔菌的抑菌活性,以期为落叶松枯梢病的防治提供一种植物源药剂。
本发明的有益效果体现在,落叶松葡萄座腔菌是一种真菌,能够产生孢子,具有较强的侵染、繁殖、扩散能力,相对细菌具有防治难度。香草酸对落叶松枯梢病病原落叶松葡萄座腔菌具有良好的抑菌效果,1~5mg/ml浓度下抑菌率可达30~70%。香草酸的应用能够有效抑制落叶松葡萄座腔菌,且降低了防治过程中对化学药剂的依赖性。

本实施方式香草酸(4-羟基-3-甲氧基苯甲酸)的相对分子质量为168.15,分子式为:

本实施方式发现香草酸能够抑制落叶松葡萄座腔菌生长。1~5mg/ml浓度下抑菌率可达30~70%。表明香草酸对于防治落叶松葡萄座腔菌具有重要作用,并且具有环境友好的优良特性,可减少化学药剂的使用。
具体实施方式二:本实施方式与具体实施方式一不同的是:香草酸抑制落叶松枯梢病病原落叶松葡萄座腔菌的生长。其它步骤与具体实施方式一相同。
具体实施方式三:本实施方式与具体实施方式一或二之一不同点是:香草酸作为植物源药剂的制备方法是将香草酸溶于二甲基亚砜中配制香草酸原液,然后用去离子水稀释配制工作液。其它步骤与具体实施方式一或二相同。
具体实施方式四:本实施方式与具体实施方式一至三之一不同点:工作液的浓度为1~5mg/mL。其它步骤与具体实施方式一至三相同。
具体实施方式五:本实施方式与具体实施方式一至四之一不同点:香草酸作为植物源药剂还用于抑制对赤松隔尾孢霉菌、松穴褥盘孢菌、罗汉松拟盘多毛孢、胶孢炭疽菌、枯斑盘多毛孢和赤松隔尾孢霉的生长。其它步骤与具体实施方式一至四相同。
采用以下实施例验证本发明的有益效果:
实施例1:
验证由香草酸作为植物源药剂在抑制落叶松葡萄座腔菌生长上的应用效果。
(一)生物材料准备
将落叶松葡萄座腔菌接种至定量马铃薯葡萄糖琼脂(PDA)培养基(8ml/皿),25℃恒温、避光培养,长满板后待用。
(二)药剂溶解与病原菌接种
取200mg香草酸溶解至1ml二甲基亚砜(DMSO),得到香草酸原液。处理组为将1ml香草酸原液加入到99ml溶解的PDA培养基中,混匀,终浓度为2mg/mL;对照组为将1ml DMSO加入到99ml溶解的PDA培养基中,混匀,终浓度为2mg/ml。加药前培养基温度应降到50℃以下。将加药后的培养基定量分装至直径60mm的一次性培养皿(8ml/皿),待培养基冷凝后接种落叶松葡萄座腔菌。
于前期准备的落叶松葡萄座腔菌边缘处取5mm直径的圆形菌苔分别接种至含药剂培养基(处理组)和对照培养基(对照组)。处理组和对照组各设6个重复。25℃恒温、避光培养。
(三)抑菌程度统计
应用十字交叉法测量处理组与对照组的菌苔直径,拍照记录菌苔生长状态。抑菌率按照如下所示公式进行统计。

试验结果显示,处理组菌苔直径显著小于对照组(图1,其中■为对照组,▼为处理组),说明香草酸的处理抑制了该菌的生长。其次,在对照组培养基中落叶松葡萄座腔菌菌丝蓬松,生长状态正常,而在处理组培养基中菌丝生长受到抑制(图2)。经计算,处理9d后,香草酸的抑菌率达54.55%。处理13d后,香草酸的抑菌率达42.13%。
实施例2:
验证由香草酸作为植物源药剂在防治落叶松枯梢病上的应用。
制备落叶松葡萄座腔菌孢子悬浮液。将落叶松苗顶梢用针刺伤。将孢子悬浮液涂抹至落叶松苗伤口,用塑料袋包裹接种部位保湿3d。接种14d后将香草酸药剂(2mg/ml)喷洒至落叶松苗,以相同浓度的DMSO药剂为对照,处理7d后按时观察落叶松苗感病情况,统计感病指数。
病株分级标准如下:0级为健康植株;枝条茎部褪绿,少量针叶脱落的病株为Ⅰ级病株;枝条茎部黄褐色,半数针叶脱落,梢头略微下垂的病株为II级病株;枝条茎部褐色,大部分针叶脱落,梢头下垂的病株为III级病株;枝条茎部呈黑褐色,梢头下垂,除梢头一丛紫灰色枯死针叶外其余针叶全部脱落的病株为IV级病株。

处理香草酸后,病情有所抑制。经计算可知,处理7d后,处理组的病情指数为对照组的55.56%,香草酸的防治效果为44.44%,处理组与对照组具极显著差异(p=0.0003)。
综上所述,本发明提供了一种新的植物源药剂香草酸,可抑制落叶松枯梢病病原落叶松葡萄座腔菌生长,如果大规模推广和投入使用,将大幅减少对化学药剂的依赖,具有安全环保和对环境友好的优良特性。

2.根据权利要求1所述的香草酸的应用,其特征在于香草酸抑制落叶松枯梢病病原落叶松葡萄座腔菌的生长。
3.根据权利要求1所述的香草酸的应用,其特征在于香草酸作为植物源药剂的制备方法是将香草酸溶于二甲基亚砜中配制香草酸原液,然后用去离子水稀释配制工作液。
4.根据权利要求3所述的香草酸作应用,其特征在于工作液的浓度为1~5mg/mL。
5.根据权利要求1所述的香草酸的应用,其特征在于香草酸作为植物源药剂还用于抑制对赤松隔尾孢霉菌、松穴褥盘孢菌、罗汉松拟盘多毛孢、胶孢炭疽菌、枯斑盘多毛孢和赤松隔尾孢霉的生长。
