Abstract
一种清除饲料中多种霉菌毒素复方制剂的生产方法,步骤为:采用半仿生技术将浸泡的甘草加热提取后浓缩,得甘草酸提取液;使用吸附剂对甘草酸提取液进行吸附,得半成品;再将半成品与霉菌毒素降解酶、葡萄糖氧化酶进行混合,制得成品。本发明制得的甘草提取物及复方制剂能快速修复霉菌毒素引起肝肾及其他脏器的损伤,增强肝脏的解毒功能,保肝护肾;霉菌毒素降解酶、葡萄糖氧化酶成分还能全面清除饲料中多种霉菌毒素,有效预防和缓解各类动物因霉菌毒素中毒引起的各种疾病。有效地改善和治疗母猪的假发情、不发情、流产、死胎等病症,提高母猪的繁育能力,提升产仔率,对母猪的健康有很好的保健作用,提升仔猪成活率,提升养殖效益。
Description
技术领域
本发明涉及一种饲料中多种霉菌毒素的清除方法,具体涉及一种清除饲料中多种霉菌毒素复方制剂的生产方法,属于饲料药物添加剂技术领域。
背景技术
由于霉菌毒素的广泛存在,往往1种原料中含有2种或以上的霉菌毒素,而不同霉菌毒素之间往往会产生叠加效应,使得低于国家标准的霉菌毒素剂量就会产生严重的副作用;再加上由于霉菌的大量繁殖所导致的饲料营养不全和某些特殊生理阶段或其他应激因素的存在,饲料中低浓度的霉菌毒素即可对动物造成危害。
因此,某些受霉菌毒素污染的饲料,即使霉菌毒素的检出水平在国家允许的范围内,但仍然会对动物健康和生产性能造成不良影响。
1、影响动物生长发育及生产性能
饲料受到霉菌毒素污染后会导致适口性差、营养物质损失。霉菌毒素通过干扰体内蛋白质、糖类和脂类的代谢、降低能量和营养物质在体内的储存,进而影响生长速度。有些霉菌毒素的毒性具有器官特异性,比如:黄曲霉毒素会侵害动物肝脏,具有严重的肝毒性;赭曲霉毒素和橘青霉毒素具有肾脏毒性;镰孢霉和念珠镰霉等对动物的肺脏有明显的副作用,可引起器官变性坏死。
动物采食受到霉菌毒素污染的饲料后,通常表现为采食量、生产性能和饲料转化率降低等现象,如:猪会表现出厌食、呕吐、拉稀、体重减轻、饲料转化率差等现象;鸡表现为腹泻、糊肛、生长缓慢、饲料转化率降低、产卵率下降、蛋品质降低等现象。
2、影响动物的繁殖机能
各种霉菌毒素因结构不同,作用于动物的靶器官也不同,故会对动物表现出各自特有的危害特点。有些毒素如:玉米赤霉烯酮、F2毒素、黄曲霉毒素和脱氧雪腐镰刀菌烯醇等可引起母畜假发情、受胎率下降、流产、死胎等繁殖机能障碍,还会引起小母猪外阴部红肿、假发情、子宫肿胀及卵巢萎缩等障碍。另外,有些霉菌毒素如玉米赤霉烯酮还能引起公猪睾丸退化、配种能力下降等现象。
3、降低动物的免疫性能
大多数的霉菌毒素都能损坏动物的免疫系统,降低动物的免疫水平,增加动物的易感性,具体表现为免疫器官发育不良、免疫器官指数降低、淋巴细胞数量减少、淋巴细胞活力降低、抗体水平降低等现象。
由于霉菌毒素对免疫性能的影响,使得动物的抵抗力下降、疾病的易感性上升,因此,霉菌毒素中毒后,即使假以药物治疗,防治效果仍然很差。
发明内容
为解决现有技术的不足,本发明的目的在于提供一种清除饲料中多种霉菌毒素复方制剂的生产方法。
为了实现上述目标,本发明采用如下的技术方案:
一种清除饲料中多种霉菌毒素复方制剂的生产方法,包括以下步骤:
S1、浸泡甘草,采用半仿生技术加热提取后浓缩,得甘草酸提取液;
S2、使用吸附剂对甘草酸提取液进行吸附,得半成品;
S3、将半成品与霉菌毒素降解酶、葡萄糖氧化酶进行混合,制得成品。
上述吸附剂包括二氧化硅、硅藻土、沸石、聚酰胺中的任一种。
上述甘草酸提取液与吸附剂的质量比为1:(2-8)。
上述霉菌毒素降解酶、葡萄糖氧化酶的质量比为1:(3-9)。
上述霉菌毒素降解酶与半成品的质量比为1:(2-5)。
上述浸泡使用的浸泡液包括水、水和乙醇和氨水的混合液。
上述浸泡液的用量为甘草体积的5-10倍。
进一步的,上述混合液中乙醇的质量占比为(55-65)%,氨水的质量占比为(1-2)%。
上述半仿生技术提取的工艺条件为:
调节pH为5-10,加热温度至50-100℃,提取时间为1-2h;
再加5-8倍的水,调节pH为2-4,提取时间为1-1.5h;
最后浓缩时间为2-6h。
一种清除饲料中多种霉菌毒素复方制剂,由上述的生产方法制得。
本发明的有益之处在于:
一种清除饲料中多种霉菌毒素复方制剂的生产方法,具有以下优点:
1、效果好成本低:本品由甘草提取物、霉菌毒素降解霉、葡萄糖氧化酶以科学比例复配而成,效果更优、成本更低,每吨饲料添加成本不超过20元。
2、修复肝脏损伤:甘草提取物能快速修复霉菌毒素引起肝肾及其他脏器的损伤,增强肝脏的解毒功能,保肝护肾。
3、全面降解霉菌:霉菌毒素降解酶可直接破坏黄曲霉、呕吐毒素、玉米赤霉烯酮、T~2霉菌毒素、赭曲霉毒素、伏马毒素等多种霉菌的分子结构;在清除霉菌的同时对维生素和微量元素无吸附影响,保证了饲料的营养价值。
4、保护肠道健康:葡萄糖氧化酶除了能抑制多种霉菌毒素,并能保护肠道黏膜完整性,强化肠屏障对霉菌毒素的过滤作用;调节肠道菌群平衡,有效改善动物腹泻。
5、成品复方制剂有效地改善和治疗母猪的假发情、不发情、流产、死胎等病症,提高母猪的繁育能力,提升产仔率,对母猪的健康有很好的保健作用,提升仔猪成活率,提升养殖效益。

具体实施方式
以下结合附图和具体实施例对本发明作具体的介绍。
为便于试验数据的比对,下列实施例中的浸泡液均选用水。
一种清除饲料中多种霉菌毒素复方制剂的生产方法,包括以下步骤:
S1、按体积比,使用5-10倍的水浸泡甘草,浸泡时间为0.5-1h,浸泡温度为25-45℃,采用半仿生技术(SBE)加热提取后浓缩,得甘草提取液;
提取的工艺条件为:调节pH为5-10,加热温度至50-100℃,提取时间为1-2h;再加5-8倍(相对甘草的初始体积)的水,调节pH为2-4,提取时间为1-1.5h;最后浓缩时间为2-6h。
S2、使用二氧化硅作为吸附剂,按质量比,甘草提取液:吸附剂=1:(2-8),对甘草提取液进行吸附,得半成品;
S3、将半成品与霉菌毒素降解酶、葡萄糖氧化酶进行混合,其中,霉菌毒素降解酶、葡萄糖氧化酶的质量比为1:(3-9),霉菌毒素降解酶与半成品的质量比为1:(2-5),制得成品。
实施例1
先称取100kg甘草,加5倍水进行浸泡,调节pH值为8,加热进行提取,提取温度80℃,提取时间1h;再加8倍水,调节pH值为4,提取时间1h,最后浓缩时间2h,制得甘草酸提取液(浸膏)25kg。
加二氧化硅75kg进行吸附;
再加入0.2kg霉菌毒素降解酶、0.4kg葡萄糖氧化酶混合,混合时间15min,制得成品复方制剂。
实施例2
先称取100kg甘草,加8倍水进行浸泡,调节pH值为9,加热进行提取,提取温度90℃,提取时间2h;再加6倍水,调节pH值为3,提取时间1h,最后浓缩时间4h,制得甘草酸提取液(浸膏)20kg。
加二氧化硅60kg进行吸附;
再加入0.4kg霉菌毒素降解酶、0.8kg葡萄糖氧化酶混合,混合时间15min,制得成品复方制剂。
实施例3
先称取100kg甘草,加10倍水进行浸泡,调节pH值10,加热进行提取,提取温度100℃,提取时间2h;再加8倍水,调节pH值为2,提取时间2h,最后浓缩时间6h,制得甘草酸提取液(浸膏)15kg。
加二氧化硅45kg进行吸附;
再加入0.5kg霉菌毒素降解酶、1kg葡萄糖氧化酶混合,混合时间15min,制得成品复方制剂。
对比实验一
实验目的:实施例1-3制得的复方制剂对各种霉菌毒素的吸附和脱除能力
实验过程:取适量发霉的饲料进行检测,分别记录下使用复方制剂前、使用一个月后饲料中各种霉菌毒素的含量,添加量为每吨饲料中添加1-2kg复方制剂。
实验数据:见下表1

对比实验二
实验目的:实施例1-3制得的复方制剂对母猪及仔猪的基础健康状况的作用及影响
实验过程:分别记录下于饲料中添加使用复方制剂前、后母猪和仔猪的健康状况数据,添加量为每吨饲料中添加2kg复方制剂。
实验对象:母猪的总数量为60头,仔猪的总数量为300头,均分为3组实验数据:见下表2

由上表1和2可见,本发明的复方制剂具有较强的吸附力,强劲的脱霉脱菌因子,可以有效清除饲料中的多种有害霉菌毒素;有效地改善和治疗母猪的假发情、不发情、流产、死胎等病症,提高母猪的繁育能力,提升产仔率,对母猪的健康有很好的保健作用,提升仔猪成活率,提升养殖效益。
甘草的成分复杂,主要含有三萜类,黄酮类,香豆精类化合物,本发明通过半仿生技术获取甘草提取液,提高含有的甘草甙、槲皮素的浓度,其中甘草甙是甘草黄酮类化合物中重要的单体活性成分,具有抗氧化、抗促癌、抗致突、抗H IV等多种药理作用,为清除饲料中的有害霉菌毒素提供了积极效用。槲皮素能降低黄曲霉菌内的ROS水平,降低MDA含量,RT-PCR结果证实槲皮素能够激活抗氧化系统转录因子Yap1,导致黄曲霉体内的抗氧化酶系统活性的增加,POD、CAT、SOD都得到了显著的提高,这很可能是槲皮素抑制AFB1产生的关键因素;槲皮素能同时下调AflR与AflS的表达,而AflS能够通过结合AflR调控产毒基因的表达,这很可能是槲皮素抑制AFB1产生的核心分子机制,这种机制也与其激活抗氧化系统缓解菌体内氧化胁迫的作用相对应。以上结果表明槲皮素是一种高效的黄曲霉毒素合成抑制剂,将对提高食品安全保障具有较高的应用价值。

Claims (10)
1.一种清除饲料中多种霉菌毒素复方制剂的生产方法,其特征在于,包括以下步骤:
S1、浸泡甘草,采用半仿生技术加热提取后浓缩,得甘草酸提取液;
S2、使用吸附剂对甘草酸提取液进行吸附,得半成品;
S3、将半成品与霉菌毒素降解酶、葡萄糖氧化酶进行混合,制得成品。
2.根据权利要求1所述的生产方法,其特征在于,所述吸附剂包括二氧化硅、硅藻土、沸石、聚酰胺中的任一种。
3.根据权利要求1所述的生产方法,其特征在于,所述甘草酸提取液与吸附剂的质量比为1:(2-8)。
4.根据权利要求1所述的生产方法,其特征在于,所述霉菌毒素降解酶、葡萄糖氧化酶的质量比为1:(3-9)。
5.根据权利要求1所述的生产方法,其特征在于,所述霉菌毒素降解酶与半成品的质量比为1:(2-5)。
6.根据权利要求1所述的生产方法,其特征在于,所述浸泡使用的浸泡液包括水、水和乙醇和氨水的混合液。
7.根据权利要求1所述的生产方法,其特征在于,所述浸泡液的用量为甘草体积的5-10倍。
8.根据权利要求6所述的生产方法,其特征在于,所述混合液中乙醇的质量占比为(55-65)%,氨水的质量占比为(1-2)%。
9.根据权利要求1所述的生产方法,其特征在于,所述半仿生技术提取的工艺条件为:
调节pH为5-10,加热温度至50-100℃,提取时间为1-2h;
再加5-8倍的水,调节pH为2-4,提取时间为1-1.5h;
最后浓缩时间为2-6h。
10.一种清除饲料中多种霉菌毒素复方制剂,由权利要求1-9任一项所述的生产方法制得。
