技术领域

背景技术
轻度创伤性脑损伤(mild traumatic brain injury,mTBI)是最常见的创伤性脑损伤类型。世界卫生组织(WHO)将mTBI定义为头部受到撞击,导致急性脑功能紊乱,通常导致身体、认知和/或情绪症状。mTBI患者经常报告的长期并发症包括增加的焦虑、易怒、抑郁和情绪不稳定,这已成为一个常见但潜在的临床问题。由于不典型的放射学特征和成像结果、轻微症状以及缺乏更可靠的生物标志物和检测方法,mTBI在早期阶段往往难以被准确识别。这些问题影响司法鉴定的权威性和公正性,以及临床诊断和治疗。因此,一种快速准确地检测mTBI的重要生物标志物的方法是非常关键和不可缺少的。

目前,对S100-β的检测仍依赖于传统的酶联免疫吸附试验(Enzyme-linkedimmunosorbent assay,ELISA)、化学发光免疫分析(chemiluminescence immunoassay,CLIA)、磁珠法等方法。然而,这些检测方法在实际应用中获得结果并不令人十分满意,因为它们存在各种缺点,如假阳性率、仪器成本、复杂操作和检测时间长等问题。
因此,有必要开发一种新型材料能够快速、灵敏检测mTBI生物标志物S100-β。

本发明通过酰胺键在碳化聚合物点表面连接了S100钙结合蛋白,使得复合碳化聚合物点(pep-CPDs)能够快速灵敏检测mTBI生物标志物S100-β,并实现了体外细胞成像。通过乳酸脱氢酶(LDH)试验、MTT试验、细胞成像和小鼠急性毒性试验证明了pep-CPDs的安全性和低细胞毒性。通过检测mTBI患者血液样本中的S100-β,证明了pep-CPDs检测实际样本的可靠性。
根据本发明的一些实施方式,所述碳化聚合物点的平均粒径为2.5~7.2nm。
根据本发明的一些实施方式,以复合碳化聚合物点总质量计算,所述S100钙结合蛋白的含量为95~98%。
本发明的第二方面提供了一种复合碳化聚合物点的制备方法,包括如下步骤:
在N-羟基琥珀酰亚胺和1-乙基-(3-二甲基氨基丙基)碳酰二亚胺的作用下,将含有羧基的碳化聚合物点与S100钙结合蛋白在室温下混合反应。
本发明关于复合碳化聚合物点的制备方法的技术方案中的一个技术方案,至少具有以下有益效果:
本发明的氨基修饰的S100钙结合蛋白通过氨基-羧基反应连接到含有羧基的碳化聚合物点表面,制备方法简单,原料易得,制备得到的复合碳化聚合物点对目标S100-β保持高的亲和力。
根据本发明的一些实施方式,所述含有羧基的碳化聚合物点通过如下方法制备:
S1.将邻苯二胺与硝酸混合,形成混合反应液;
S2.将步骤S1得到的混合反应液在温度200~250℃,密封条件下水热反应5~12小时,得到溶液;
S3.将步骤S2得到的溶液用聚醚砜膜过滤,再经过透析、干燥得到所述含有羧基的碳化聚合物点。
根据本发明的一些实施方式,所述含有羧基的碳化聚合物点的浓度为0.5~2mg/mL。由此,
根据本发明的一些实施方式,所述S100钙结合蛋白的浓度为0.1~5mg/mL。
根据本发明的一些实施方式,所述邻苯二胺与硝酸的摩尔比为(62.6~68.9):1。

根据本发明的一些实施方式,一种检测S100钙结合蛋白浓度的方法,包括如下步骤:
(1)将不同浓度的S100钙结合蛋白与复合碳化聚合物点混合,进行荧光检测,获取复合碳化聚合物点与S100钙结合蛋白浓度的荧光强度关系;
(2)加入待测液S100钙结合蛋白,进行荧光检测,得到荧光强度,根据步骤(1)所述的S100钙结合蛋白荧光强度与浓度的关系计算得到待测品中S100钙结合蛋白的浓度;所述复合碳化聚合物点为上述所述的复合碳化聚合物点。
在本发明中“室温”是指温度为15~30℃。

具体实施方式
下面将对本发明实施例中的技术方案进行清楚、完整地描述,但本发明的实施方式不限于此。
本发明所采用的试剂、方法和设备,如无特殊说明,均为本技术领域常规试剂、方法和设备。
以下实施例及对比例中采用的原料如下:
N-羟基琥珀酰亚胺(NHS,99%)、1-乙基-(3-二甲基氨基丙基)碳酰二亚胺(EDC,99%)、邻苯二胺(o-PD,99%)、HNO3(GC,70%)和牛血清白蛋白均购自生工(中国上海)。S100钙结合蛋白序列:NH2-TRTKIDWNKILS由GL生化公司(中国上海)合成和纯化。S100-β、血清淀粉样蛋白A(SAA)、乙二胺(AR97%)、赖氨酸(BC级)、柠檬酸(试剂级|纯度≥99.5%)和葡萄糖(生物技术级|纯度≥99.5%)均来自生工(上海,中国)。
Hela细胞:购自Beyotime(C6330)。
血清样本:从样本库中登记的平均年龄为27岁(23-30岁)的患者中收集血清样本,所有这些患者在样本采集前没有潜在的严重脑出血。凝血后,通过在4℃下以4000×g离心15分钟的方式分离血清样品,将血清分离成等份并储存在-80℃下。采用标准添加法检测血清中的S100-β。
6周龄雄性/雌性C57小鼠购自中南大学实验动物部。
使用F-4700荧光分光光度计(日本东京日立公司)来记录发射光谱。激发和发射的狭缝宽度分别为5nm和10nm。紫外可见分光光度计(岛津,UV-2450)记录紫外-可见光谱。使用FEI Tecnai G2 60-300显微镜(Hillsboro,USA)进行透射电子显微镜的拍摄。Zeta电位由Zeta-sizer Nano ZS仪器(Malvern Inc.,UK)测定。用傅里叶变换红外光谱仪FTIR-850(天津港东科技有限公司,中国)检测表面的化学结构和官能团。细胞成像是使用激光扫描共聚焦显微镜(ZEISSCelldiscoverer 7with LSM 900,德国)进行。碳化聚合物点的量子产率(QY)测量是通过爱丁堡FS920P荧光光谱仪进行测试。
实施例1
实施例1提供一种复合碳化聚合物点,制备方法如下:
S1.将0.054g邻苯二胺溶解在10mL的去离子水中,然后加入50μL(0.725mM)HNO3,在室温下搅拌5分钟后,形成混合反应液;
S2.将混合反应液转移到一个有聚四氟乙烯内衬的反应釜中,并在200℃的烤箱中加热过夜,反应混合物被冷却到室温。
S3.将步骤S2得到的溶液用0.22μm的聚醚砜膜过滤去除大颗粒后,在500D透析袋中对去离子水进行透析,得到碳化聚合物点溶液,将碳化聚合物点溶液冻干保存。
将3.5mg EDC和5mg NHS加入到1mg/mL的碳化聚合物点溶液中,然后搅拌30分钟。然后将100μL的1mg/mL S100-β肽加入到混合物中,在室温下反应30分钟,得到复合碳化聚合物点(pep-CPDs)。
实施例2
实施例2提供一种复合碳化聚合物点,制备方法和原料同实施例1,其区别在于,S100-β肽的浓度为0.1mg/mL。
实施例3
实施例3提供一种复合碳化聚合物点,制备方法和原料同实施例1,其区别在于,S100-β肽的浓度为0.5mg/mL。
实施例4
实施例4提供一种复合碳化聚合物点,制备方法和原料同实施例1,其区别在于,S100-β肽的浓度为0.8mg/mL。
实施例5
实施例5提供一种复合碳化聚合物点,制备方法和原料同实施例1,其区别在于,S100-β肽的浓度为2mg/mL。
实施例6
实施例6提供一种复合碳化聚合物点,制备方法和原料同实施例1,其区别在于,S100-β肽的浓度为5mg/mL。
应用例1
将100ng/mL的S100-β加入到实施例1制备的pep-CPDs中反应10min并检测荧光强度。
应用例2
将100ng/mL的S100-β加入到实施例1制备的pep-CPDs中反应1min并检测荧光强度。
应用例3
将100ng/mL的S100-β加入到实施例1制备的pep-CPDs中反应5min并检测荧光强度。
应用例4
将100ng/mL的S100-β加入到实施例1制备的pep-CPDs中反应20min并检测荧光强度。
应用例5
将100ng/mL的S100-β加入到实施例1制备的pep-CPDs中反应40min并检测荧光强度。
应用例6
将100ng/mL的S100-β加入到实施例1制备的pep-CPDs中反应60min并检测荧光强度。
性能检测
将碳化聚合物点(CPDs)进行了透射电子显微镜和红外光谱测试。如图1和图2所示。透射电子显微镜(TEM)用于观察所制备的CPDs的结构特征,平均尺寸为5.1nm。从图2观察到,CPDs的FTIR光谱在2965cm-1附近有一个明显的峰值,表明存在-OH基团,在1407cm-1处有一个峰值,表明存在C=O基团。1640cm-1处的峰值对应于c=c的伸缩振动。这些数据表明,合成的CPDs富含羧基。CPDs的水溶液通过Zeta电位测量,检测结果如图3显示为负电荷(-13.4mV)。与S100-β肽结合后,Zeta电位从-13.4mV变为-3.2mV,表明肽在CPDs表面成功结合。与肽结合的CPDs仍然呈现出负的整体Zeta电位,避免了在生物实验中CPDs从血液循环中快速移除。
将CPDs进行了紫外-可见吸收和荧光发射光谱,如图4所示,其中A显示pep-CPDs和CPDs的紫外-可见吸收光谱显示在450和630nm之间有多个吸收带,当添加目标S100-β后,与pep-CPDs相比,其吸光度值降低。此外,在B中观察到CPDs的荧光发射光谱在640纳米处呈现出一个荧光强度峰值,激发波长为520nm。当加入目标S100-β后,引起pep-CPD的荧光猝灭。经检测计算,所制备的CPDs的量子产率约为4.1%。这些结果证明,所制备的pep-CPDs具有良好的光学性能,可实现S100-β的荧光检测。
对实施例1~6制备的pep-CPDs进行了荧光检测,如图5所示,随着当S100-β肽的浓度从0.1mg/mL增加到1mg/mL时,相对荧光变化增加,并在1mg/mL开始保持平稳。
应用例1~6的荧光强度见图6所示,加入100ng/mL的S100-β后,pep-CPD的荧光在10分钟内逐渐减弱。基于pep-CPDs的体系在20分钟内完成了对S100-β的检测,而之前报道的传统检测S100-β的方法通常需要0.5~6小时。因此,本发明检测S100-β的时间短。
pep-CPDs的特异性检测:如图7中的A所示,S100-β和干扰物(BSA蛋白、SAA蛋白、乙二胺、赖氨酸、柠檬酸和葡萄糖)的浓度均为103ng/mL,观察到干扰蛋白(BSA和SAA蛋白)表现出较小的荧光变化,并且没有观察到明显的变化。而在使用乙二胺、赖氨酸、柠檬酸和葡萄糖后,也没有明显的变化。而加入S100-β后,实施例1的pep-CPDs的相对荧光变化是这些分子中最高的。因此,pep-CPDs被验证可用于S100-β的特异性检测。
如图7中的B所示,pep-CPDs与目标S100-β浓度(0到106ng/mL)的荧光猝灭图,图7中的C显示了荧光响应随S100-β浓度变化的对应曲线图。它在S100-β浓度从1到106ng/mL的范围内显示了良好的线性关系(F0和F1代表目标S100-β不存在和存在时纳米探针的荧光强度)。S100-β的线性回归方程被描述为Y=-33.04*X+226.0(R2=0.9848)。根据S/N=3计算,检测极限(LOD)为0.1ng/mL。并且可以在20分钟内完成整个检测过程,大大提升了在临床上的快速检测。
细胞毒性评价和细胞荧光成像:通过LDH和MTT试验对不同浓度的实施例1的pep-CPDs评估了pep-CPDs对Hela细胞的影响。数据以平均值±标准差表示(n=6)。如图8中的A所示,LDH试验被用来评估pep-CPDs对Hela细胞的毒性,结果表明,即使高达1000ng/mL的pep-CPDs,其毒性也较低。如图8中的B显示,在培养的4小时内,细胞在pep-CPDs的存在下表现出很好的活力,在高达1000ng/mL的pep-CPDs浓度下,细胞存活率>85%。图8中C显示了分别在明场和480纳米激发下的加入pep-CPDs培养Hela细胞的荧光图像。与没有S100-β时观察到的红色荧光相比,加入S100-β时红色荧光较弱,表明S100-β的存在导致了细胞中pep-CPD的荧光猝灭。因此,pep-CPD可以作为细胞成像中S100-β检测的一种有价值的探针。
活体C57小鼠荧光成像:首先,建立脑损伤动物模型,我们利用改进的重物撞击器来诱发mTBI(图9中的A)。通过神经系统严重程度评分(NSS)和平衡木测试来测量神经严重程度评分(表1)。运动测试(正常=0;mTBI的范围为1-2)和平衡木测试(正常=0;mTBI的范围为2-3)来表征mTBI。经评估,39只小鼠符合mTBI的纳入标准。mTBI小鼠的相关评分见表2。此外,考虑到小鼠最合适的性别、年龄和体重,我们从25只mTBI小鼠中随机选择了16只雄性小鼠(6W至8W大,体重约25.1±2.0克),通过CCI机器建模进行活体成像。在内眦部注射150μL实施例1的pep-CPD或正常生理盐水后,用二乙醚对小鼠进行麻醉,以进行后续的成像实验。C57小鼠脑部有三组接受以下处理:(A)对照组(无损伤)注射pep-CPDs;(B)mTBI组注射生理盐水;(C)mTBI组注射pep-CPDs。如图9中的B所示,只注射正常生理盐水的mTBI组头部没有发现明显的荧光发射,表明背景荧光可以忽略不计。与对照组表现出的强荧光信号相比(图9中的D),用pep-CPD处理的mTBI组表现出较弱的荧光信号(图9中的C),这表明结合目标生物标志物S100-β的探针pep-CPDs可以区分mTBI组和对照组。半定量分析结果显示对照组和mTBI组的平均荧光强度乘以面积的比率为6.118,差异明显。因此,可以通过活体成像来观察S100-β蛋白的动态水平。

急性毒性测试试验:开展了急性毒性研究来评价pep-CPDs在证据权重分析中的影响。通过连续监测对照组(注射150μL正常生理盐水,n=10)和实验组(分别注射150μL1000μg/mL、500μg/mL和100μg/mL实施例1的pep-CPDs,n=10)在相同培养条件下记录28天的小鼠体重的长期变化。每四天对各组小鼠的体重进行一次称重。在实验过程中,对照组和实验组在最高剂量下均未见死亡(图10)。此外,实验组的体重与对照组保持相同的增长趋势。综上结果表明,pep-CPDs在体内并没有引起一定的毒性效应。
实际样品分析:用于检测血清样本中mTBI生物标志物S100-β,将实施例1制备的pep-CPDs对血清中的S100-β的进行荧光测试,从图11中的A和B上看,荧光强度信号和目标S100-β浓度显示出良好的线性关系,图11中的C为pep-CPDs的相对荧光强度与血清中不同浓度的S100-β之间的线性关系图(F0和F1代表加入S100-β和不加入S100-β时的pep-CPDs的荧光强度)。基于信噪比(S/N=3)的LOD为0.3ng/mL。如表3所示,pep-CPDs纳米探针对目标S100-β的检测分析性能与传统的检测方法进行了比较。此外,将不同量的S100-β加入到人血清样品中,然后进行分析,该方法分析的LOD为0.3ng/mL,检测范围为1~500ng/mL。人血清样品的检测回收率在96-104%之间,相对标准偏差(RSD)为1.0%-1.5%(表4),这证实了该方法对S100-β的测定具有精确性和良好的重现性。

比较了ELISA分析法和本发明的方法,以评估检测mTBI患者和健康对照组血清中S100-β水平的效率。对于mTBI组,基于传统的ELISA分析方法和新型传感方法检测血清样本中的S100-β水平,两者具有相似的趋势和一致性(表5)。此外,与ELISA的结果相比,我们的传感方法也能灵敏地检测到对照组的S100-β水平(0.9±1.9ng/mL),其中的S100-β的水平处于平均值范围内。该实验进一步证明了S100-β作为mTBI患者生物标志物的有用性。因此,这种基于pep-CPDs的新方法在灵敏度、特异性、准确性和实用性方面具有很多优点,比在法医和临床实验室诊断中的ELISA和其他传统检测mTBI的方法更具优势。

1.一种复合碳化聚合物点,其特征在于,包括碳化聚合物点和S100钙结合蛋白,所述碳化聚合物点和所述S100钙结合蛋白通过酰胺键连接。
2.根据权利要求1所述的复合碳化聚合物点,其特征在于,所述碳化聚合物点的平均粒径为2.5~7.2nm。
3.根据权利要求1所述的复合碳化聚合物点,其特征在于,以复合碳化聚合物点总质量计算,所述S100钙结合蛋白的含量为95~98%。
4.一种制备如权利要求1~3任一项所述复合碳化聚合物点的方法,其特征在于,包括如下步骤:
在N-羟基琥珀酰亚胺和1-乙基-(3-二甲基氨基丙基)碳酰二亚胺的作用下,将含有羧基的碳化聚合物点与S100钙结合蛋白在室温下混合反应。
5.根据权利要求4所述的复合碳化聚合物点的制备方法,其特征在于,所述含有羧基的碳化聚合物点通过如下方法制备:
S1.将邻苯二胺与硝酸混合,形成混合反应液;
S2.将步骤S1得到的混合反应液在温度200~250℃,密封条件下水热反应5~12小时,得到溶液;
S3.将步骤S2得到的溶液用聚醚砜膜过滤,再经过透析、干燥得到所述含有羧基的碳化聚合物点。
6.根据权利要求4所述的复合碳化聚合物点的制备方法,其特征在于,所述含有羧基的碳化聚合物点的浓度为0.5~2mg/mL。
7.根据权利要求4所述的复合碳化聚合物点的制备方法,其特征在于,所述S100钙结合蛋白的浓度为0.1~5mg/mL。
8.一种荧光探针,其特征在于,包括权利要求1~3任一项所述的复合碳化聚合物点。
9.根据权利要求1~3任一项所述的复合碳化聚合物点或权利要求4~7任一项所述的制备方法制备得到的复合碳化聚合物点在荧光检测、活细胞成像试剂制备中的应用。
10.一种检测S100钙结合蛋白浓度的方法,其特征在于,包括如下步骤:
(1)将不同浓度的S100钙结合蛋白与复合碳化聚合物点混合,进行荧光检测,获取复合碳化聚合物点与S100钙结合蛋白浓度的荧光强度关系;
(2)加入待测液S100钙结合蛋白,进行荧光检测,得到荧光强度,根据步骤(1)所述的S100钙结合蛋白荧光强度与浓度的关系计算得到待测品中S100钙结合蛋白的浓度;所述复合碳化聚合物点为权利要求1~3任一项所述的复合碳化聚合物点。
