Abstract
The invention provides a composition for improving the motility and quality of human sperms, which comprises the following components in percentage by weight: 100-200 parts of liquid phase substances of acetic acid brewed by traditional Chinese medicines; 10-20 parts of solid phase substances of acetic acid brewed by traditional Chinese medicines; 80-100 parts of pollen pini; 10-20 parts of chitosan; 3-5 parts of menthol and/or tea tree oil; 1-2 parts of chlorhexidine acetate; wherein, the Chinese medicine brewed acetic acid is brewed by taking corn starch and Chinese medicinal materials for promoting blood circulation as raw materials and fermenting through acetic acid bacillus. The invention adopts the technical scheme of combining traditional Chinese medicines and modern medicines, starts with the more neglected factors of the male scrotum health in the prior art, such as scrotum temperature and humidity, microbial flora, varicocele and the like, treats oligospermia or asthenospermia by means of external application, can obviously improve the activity and quality of male sperms, and further improves the male fertility.
Description
Technical Field
The invention belongs to the field of medicines, and particularly relates to a composition for improving the motility and quality of human sperms and a preparation method and a use method thereof.
Background
Currently, the decline and aging of the population is becoming a serious problem worldwide, especially for the developed economies, and it is imperative in many countries to encourage fertility to improve population architecture. However, under the influence of a plurality of complex factors such as environmental pollution, food quality, living habits, psychological stress and the like, male fertility has a general decline trend in recent years, which is mainly represented by the reduction of sperm quantity, sperm motility and sperm quality. From the viewpoint of sperm count, the standards of male sperm quantity and sperm concentration reach 2 milliliters and 2000 ten thousand per milliliter respectively in 1999 according to the standards of the world health organization, the standards are reduced to 1.5 milliliters and 1500 ten thousand per milliliter in 2010, and the latest standards to 2021 are 1.4 milliliters and 1600 ten thousand per milliliter. From the viewpoint of sperm motility and sperm quality, the standards for the total sperm motility rate and the normal sperm morphology rate in 1999 were 50% and 15%, respectively, according to the world health organization standard, which decreased to 40% and 4% by 2010, and the latest standard to 2021 was 42% and 4%. Therefore, the improvement of the number, the vitality and the quality of the sperms of the male is important for solving the population problems and improving the population structure and the social structure.
In the prior art, most of the medicines for treating male oligospermia or asthenospermia are oral medicines, and the generation of sperms is promoted by supplementing trace elements such as protein, vitamins, zinc, magnesium and the like; or treating oligospermia or asthenospermia caused by different reasons by means of regulating endocrine, treating inflammation and the like. It can be seen that the prior art neglects to start with the health of the male scrotum, such as scrotum temperature and humidity, microbial flora, varicocele and other factors, and treats oligospermia or asthenospermia by external application so as to improve the activity and quality of the male sperms.
Disclosure of Invention
In view of the above-mentioned defects of the prior art, the present invention aims to provide a composition for improving human sperm motility and quality and a preparation and use method thereof, and adopts the technical scheme of combining traditional Chinese medicines and modern medicines, starting from the more neglected factors of male scrotum health in the prior art, such as scrotum temperature and humidity, microbial flora, varicocele and the like, and treating oligospermia or asthenospermia by a mode of external application, so that the male sperm motility and quality can be significantly improved, and the male fertility can be further improved.
In order to achieve the above objects, in a first aspect, the present invention provides a composition for improving motility and quality of human sperm, comprising the following components by weight:
100-200 parts of liquid phase (containing water) of acetic acid brewed by traditional Chinese medicines; 10-20 parts of solid phase substances of acetic acid brewed by traditional Chinese medicines; 80-100 parts of pollen pini; 10-20 parts of chitosan; 3-5 parts of menthol and/or tea tree oil; 1-2 parts of chlorhexidine acetate; wherein the content of the first and second substances,
the Chinese medicine brewed acetic acid is brewed by taking corn starch and Chinese medicinal materials for promoting blood circulation as raw materials and fermenting with acetobacter.
Further, the pH value of the liquid phase material of acetic acid brewed by the traditional Chinese medicines is 3-3.5.
Further, the pine pollen is wall-broken pine pollen, the grain diameter of which is less than 100 meshes, namely the pine pollen can pass through a 100-mesh sieve.
Furthermore, the molecular weight of the chitosan is 5000-20000, and the deacetylation degree is higher than 90%.
Furthermore, in the raw materials for brewing acetic acid by the traditional Chinese medicines, the weight ratio of the corn starch to the traditional Chinese medicines for activating blood circulation is 4: 6-7: 3. More preferably, the weight ratio of the corn starch to the Chinese medicinal materials for activating blood circulation is 5: 5-6: 4.
Further, the traditional Chinese medicinal materials for promoting blood circulation comprise, by weight, 10-30% of traditional Chinese medicinal materials for dispelling cold and removing blood stasis, 10-30% of traditional Chinese medicinal materials for eliminating dampness and removing blood stasis, 10-30% of traditional Chinese medicinal materials for clearing heat and removing blood stasis, 10-30% of traditional Chinese medicinal materials for regulating qi and removing blood stasis, and 10-30% of traditional Chinese medicinal materials for enriching blood, nourishing yin and removing blood stasis, wherein the total amount is 100%. Wherein the Chinese medicinal materials for dispelling cold and removing blood stasis are selected from one of herba Epimedii, Eucommiae cortex, rhizoma Ligustici Chuanxiong, radix Angelicae sinensis, and Carthami flos; the Chinese medicinal materials with effects of eliminating dampness and removing blood stasis are selected from one of Poria, semen plantaginis, folium Bambusae, radix Stephaniae Tetrandrae, and Alismatis rhizoma; the Chinese medicinal materials with effects of clearing heat and removing blood stasis are selected from one of cornu Bubali, radix Paeoniae Rubra, cortex moutan, radix Arnebiae, radix rehmanniae, radix scrophulariae, and folium Isatidis; the Chinese medicinal materials with effects of regulating qi-flowing and removing blood stasis are selected from one of bupleuri radix, fructus Toosendan, radix Curcumae, fructus Citri Sarcodactylis, pericarpium Citri Reticulatae viride, fructus Aurantii, rhizoma Cyperi, and fructus Citri; the Chinese medicinal materials with effects of replenishing blood, nourishing yin and removing blood stasis are selected from one of radix rehmanniae Preparata, colla Corii Asini, Polygoni Multiflori radix, fructus Lycii and arillus longan.
Further, the acetobacter is selected from one or more of acetobacter striatum, acetobacter oxydans, acetobacter pasteurianus, acetobacter schwanii and acetomonas oxydans.
In a second aspect, the present invention provides a method for preparing the above composition for improving motility and quality of human sperm, comprising the steps of:
s1: pulverizing the Chinese medicinal materials with blood circulation promoting effect into powder of 100 mesh or finer, mixing with corn starch powder of 100 mesh or finer, decocting in water, cooling to room temperature, adding Acetobacter, mixing, and fermenting for a certain period to obtain Chinese medicinal brewed acetic acid;
s2: sterilizing the acetic acid brewed by the traditional Chinese medicines at high temperature, and filtering by using a screen mesh with 1000-2000 meshes, wherein the filtrate is a liquid phase substance of the acetic acid brewed by the traditional Chinese medicines, and the filter cake is a solid phase substance of the acetic acid brewed by the traditional Chinese medicines; adjusting the pH value of a liquid phase substance of acetic acid brewed by traditional Chinese medicines to 3-3.5 by using alkali liquor and/or acid liquor;
s3: adding chitosan into the liquid phase material of acetic acid brewed by the traditional Chinese medicines, fully stirring to dissolve the chitosan, and filtering out insoluble substances by using a screen mesh with 1000-2000 meshes; adding solid phase substances of acetic acid brewed by traditional Chinese medicines, chlorhexidine acetate, menthol and/or tea tree oil into the filtrate, fully stirring, dropwise adding alkali liquor while stirring to adjust the pH value to 6.5-7, and standing for a period of time to obtain a gel substance;
s4: freeze drying the gel material in a freeze dryer to remove water to obtain gel solid; putting the gel solid into a pulverizer to pulverize to reach the particle size of 100 meshes or finer, so as to obtain gel powder;
s5: mixing the gel powder with 100 mesh or finer pollen Pini completely to obtain composition for improving sperm motility and quality, which is generally yellow brown or dark brown powder, and has color changed according to different Chinese medicinal materials.
Further, in step S1, the weight of the added water is 5 to 10 times of the weight of the powder, the cooking temperature is 100 (normal pressure) to 105 ℃ (high pressure), and the cooking time is 0.5 to 2 hours.
Further, the acetobacter aceti in step S1 may be a bacterial powder, or a bacterial liquid cultured by a culture medium; the fermentation temperature is 28-32 ℃, and the fermentation time is 3-5 days.
Further, the temperature for high-temperature sterilization in the step S2 is 100 ℃, and the time is 5-30 minutes.
Further, the alkali solution in steps S2 and S3 is sodium hydroxide, potassium hydroxide or ammonia water; the acid solution in step S2 is hydrochloric acid or acetic acid.
In a third aspect, the present invention provides a method of using the above composition for improving sperm motility and quality in humans, wherein the following two administration modes are available:
the method of use of the composition of the invention may resemble a talcum powder: dipping powder of a proper amount of the composition into a puff or a sponge, and uniformly coating the powder on the surfaces of the scrotum and the spermatic cord 2-3 times a day;
the method of use of the composition of the invention may also be similar to an ointment: taking a proper amount of powder of the composition, adding a proper amount of clear water, uniformly stirring the mixture by using a cotton swab to form a paste, and uniformly coating the paste on the surfaces of scrotum and spermatic cord 2-3 times a day.
The beneficial technical effects of the invention are at least reflected in the following aspects:
(1) the invention adopts the technical scheme of combining traditional Chinese medicines and modern medicines, starts with the more neglected factors of the male scrotum health in the prior art, such as scrotum temperature and humidity, microbial flora, varicocele and the like, treats oligospermia or asthenospermia by means of external application, can obviously improve the activity and quality of male sperms, and further improves the male fertility.
(2) In the formula of the invention, the chitosan and the chlorhexidine acetate have the functions of antibiosis and bacteriostasis, can inhibit the growth of harmful microbial flora near the scrotum of a male, and ensures the sanitation and health of the scrotum and perineum. In addition, chitosan also serves as a film forming agent and can form a film on the surfaces of scrotum and spermatic cord so as to maintain the drug effect for a long time.
(3) In the formula, the traditional Chinese medicinal materials for promoting blood circulation comprise various traditional Chinese medicinal materials with different effects of promoting blood circulation and removing blood stasis, such as traditional Chinese medicinal materials for dispelling cold and removing blood stasis, traditional Chinese medicinal materials for dispelling dampness and removing blood stasis, traditional Chinese medicinal materials for clearing heat and removing blood stasis, traditional Chinese medicinal materials for regulating qi and removing blood stasis, traditional Chinese medicinal materials for enriching blood, nourishing yin and removing blood stasis and the like, and can promote blood circulation in spermatic veins, thereby being beneficial to promoting the generation of sperms and improving the activity and quality of sperms.
(4) In the formula, the pine pollen and the traditional Chinese medicinal materials for eliminating dampness and removing blood stasis have the effects of eliminating dampness and healing sores, and the menthol and/or the tea tree oil have the effects of dispelling wind and dissipating heat, have the effects of calming and astringing scrotum, and are beneficial to adjusting the temperature and the humidity of the scrotum, so that the generation of sperms is promoted, and the vitality and the quality of the sperms are improved. Particularly for patients with varicocele, the scrotum needs to be supported by the scrotum support to prevent the feeling of tenesmus, but the scrotum support and tight-fitting underpants are easy to cause the scrotum to be more moist and hot, and the use of the composition of the invention can enable the patients to use the scrotum support and tight-fitting underpants while still keeping the scrotum fresh and dry.
(5) The Chinese medicinal materials for activating blood circulation are crushed and fermented by the acetobacter, so that the full release of effective substances in the Chinese medicinal materials is facilitated; the invention adopts a freeze drying mode to process the gel-like substance formed by chitosan, can lock the effective substances in the combined group and prevent organic substances (such as menthol and tea tree oil) in the combined group from volatilizing.
Detailed Description
The following examples are given to illustrate the present invention in detail, and the following examples are given to illustrate the detailed embodiments and specific procedures of the present invention, but the scope of the present invention is not limited to the following examples.
Example 1
In a preferred embodiment, the composition for improving motility and quality of human sperm of the invention is prepared from the following raw materials by weight:
liquid phase material (containing water) of acetic acid brewed by traditional Chinese medicines is 1000 g;
solid phase substances of acetic acid brewed by traditional Chinese medicines are 100 g;
800g of broken pine pollen below 100 meshes;
100g of chitosan with the average molecular weight of 10000 and the deacetylation degree of 90 percent;
20g of menthol;
20g of tea tree oil;
chlorhexidine acetate 10 g; wherein the content of the first and second substances,
the Chinese medicine brewed acetic acid is prepared by fermenting corn starch 500g below 100 meshes, herba Epimedii 100g, Poria 100g, cornu Bubali 100g, bupleuri radix 100g, and fructus Lycii 100g with Acetobacter pasteurianus.
The method of preparing the composition of example 1 to improve sperm motility and quality in humans comprises the steps of:
s1: mixing herba Epimedii 100g, Poria 100g, cornu Bubali 100g, bupleuri radix 100g and fructus Lycii 100g, pulverizing into powder of 100 mesh or finer, mixing with corn starch powder 500g below 100 mesh, adding water 5kg, stirring, steaming at 100 deg.C for 1 hr, cooling to room temperature, adding pasteurella acetobacter liquid pre-cultured by culture medium, mixing, and fermenting at 30 deg.C for 5 days to obtain Chinese medicinal brewed acetic acid;
s2: sterilizing the Chinese medicinal brewed acetic acid at 100 deg.C for 10 min, filtering with 1000 mesh sieve to obtain filtrate as liquid phase of Chinese medicinal brewed acetic acid, and filter cake as solid phase of Chinese medicinal brewed acetic acid; adjusting the pH value of a liquid phase substance of acetic acid brewed by traditional Chinese medicines to 3 by using 1M sodium hydroxide solution and 1M hydrochloric acid solution;
s3: adding 100g of chitosan into 1000g of liquid phase material of acetic acid brewed from traditional Chinese medicines, fully stirring to dissolve the chitosan, and filtering insoluble substances by using a 1000-mesh sieve; adding 100g of solid phase substance of acetic acid brewed by traditional Chinese medicines, 10g of chlorhexidine acetate, 20g of menthol and 20g of tea tree oil into the filtrate, fully stirring, dropwise adding 1M sodium hydroxide solution while stirring to adjust the pH value to 7, and standing for a period of time to obtain a gel substance;
s4: freeze drying the gel material in a freeze dryer to remove water to obtain gel solid; putting the gel solid into a grinder to grind the gel solid to a particle size of less than 100 meshes to obtain gel powder;
s5: and (3) fully mixing the gel powder with 800g of broken pine pollen with the particle size of less than 100 meshes to obtain the composition for improving the motility and the quality of the human sperms in the embodiment 1, wherein the composition is tawny powder.
The composition of example 1 that improves sperm motility and quality in humans can be administered in two modes: (1) similar to toilet powder, a powder puff or a sponge is used for dipping powder of a proper amount of the composition and is uniformly smeared on the surfaces of scrotum and spermatic cord 2-3 times a day; (2) also similar to ointments: taking a proper amount of powder of the composition, adding a proper amount of clear water, uniformly stirring the mixture by using a cotton swab to form a paste, and uniformly coating the paste on the surfaces of scrotum and spermatic cord 2-3 times a day.
Example 2
The composition of example 2, which improves sperm motility and quality in humans, was prepared in essentially the same manner as in example 1, except that the raw material formulation was varied, as shown in the following table:
liquid phase material (containing water) of acetic acid brewed by traditional Chinese medicines is 2000 g;
solid phase substances of acetic acid brewed by traditional Chinese medicines are 200 g;
1000g of broken pine pollen with the particle size of below 100 meshes;
200g of chitosan with the average molecular weight of 10000 and the deacetylation degree of 90 percent;
30g of menthol;
20g of tea tree oil;
20g of chlorhexidine acetate; wherein the content of the first and second substances,
the Chinese medicine brewed acetic acid is prepared by fermenting corn starch 500g below 100 meshes, rhizoma Ligustici Chuanxiong 100g, Alismatis rhizoma 100g, radix scrophulariae 100g, fructus Toosendan 100g, and radix rehmanniae Preparata 100g with Acetobacter striatum.
Example 3
The composition of example 3, which improves human sperm motility and quality, was prepared essentially the same as in example 1, except for the raw material formulation, as shown in the following table:
liquid phase material (containing water) of acetic acid brewed by traditional Chinese medicines is 1500 g;
solid phase substances of acetic acid brewed by traditional Chinese medicines are 150 g;
900g of broken pine pollen with the particle size of less than 100 meshes;
150g of chitosan with the average molecular weight of 10000 and the deacetylation degree of 90 percent;
50g of menthol;
chlorhexidine acetate 15 g; wherein the content of the first and second substances,
the Chinese medicine brewed acetic acid is prepared by fermenting corn starch 500g below 100 meshes, safflower 100g, lophatherum gracile 100g, lithospermum 100g, curcuma aromatica 100g and fleece-flower root 100g with acetobacter schdonii.
To verify the efficacy of the composition for improving sperm motility and quality in humans according to the present invention, the applicant summoned 30 patients with oligozoospermia or asthenospermia on the pregnancy preparation for drug trials, 15 of which had marked symptoms of varicocele and the other 15 of which had no marked symptoms of varicocele. The 30 subjects were divided into three groups of 10 persons each and 5 of them had marked varicocele symptoms and were administered the compositions of examples 1-3 separately by the following administration: taking appropriate amount of the powder of the composition, adding appropriate amount of clear water, stirring with cotton swab to paste, and applying on the surface of scrotum and spermatic cord uniformly for 2 times a day for 6 months. The sperm quality of the subjects before and after administration was measured every two months according to the World Health Organization (WHO) fifth edition standard, and the results are shown in the following table (mean values for each group):
from the above results, it can be seen that the compositions of examples 1-3 of the present invention are all capable of significantly improving sperm motility and quality in men, especially in patients with symptoms of varicocele. From the average value, the sperm motility and quality of the subjects had fully reached the WHO specified normal standards over 6 months of treatment. Also, during the 6 month treatment period, 19 subjects had successfully allowed the partner to conceive.
The foregoing detailed description of the preferred embodiments of the invention has been presented. It should be understood that numerous modifications and variations could be devised by those skilled in the art in light of the present teachings without departing from the inventive concepts. Therefore, the technical solutions available to those skilled in the art through logic analysis, reasoning and limited experiments based on the prior art according to the concept of the present invention should be within the scope of protection defined by the claims.
Claims (10)
1. A composition for improving sperm motility and quality in humans, comprising by weight:
100-200 parts of liquid phase substances of acetic acid brewed by traditional Chinese medicines; 10-20 parts of solid phase substances of acetic acid brewed by traditional Chinese medicines; 80-100 parts of pollen pini; 10-20 parts of chitosan; 3-5 parts of menthol and/or tea tree oil; 1-2 parts of chlorhexidine acetate; wherein the content of the first and second substances,
the Chinese medicine brewed acetic acid is brewed by taking corn starch and Chinese medicinal materials for activating blood as raw materials and fermenting through acetic acid bacillus.

The composition for improving human sperm motility and quality as claimed in claim 1, wherein said liquid phase of acetic acid brewed from traditional Chinese medicine has a pH value of 3-3.5; the pine pollen is wall-broken pine pollen, and the grain diameter of the pine pollen is smaller than 100 meshes; the molecular weight of the chitosan is 5000-20000, and the deacetylation degree is higher than 90%.
3. The composition for improving human sperm motility and quality as claimed in claim 1, wherein in the raw material for brewing acetic acid by traditional Chinese medicine, the weight ratio of corn starch to the traditional Chinese medicine for promoting blood circulation is 4: 6-7: 3.

The composition for improving sperm motility and quality as claimed in claim 3, wherein said blood-activating Chinese medicinal materials comprise 10-30% by weight of cold-dispelling and stasis-resolving Chinese medicinal materials, 10-30% by weight of dampness-eliminating and stasis-resolving Chinese medicinal materials, 10-30% by weight of heat-clearing and stasis-resolving Chinese medicinal materials, 10-30% by weight of qi-regulating and stasis-resolving Chinese medicinal materials, and 10-30% by weight of blood-enriching, yin-nourishing and stasis-resolving Chinese medicinal materials, the total being 100%; wherein the content of the first and second substances,
the traditional Chinese medicinal materials for dispelling cold and removing blood stasis are selected from one of epimedium, eucommia bark, ligusticum wallichii, angelica sinensis and safflower;
the traditional Chinese medicinal materials with effects of eliminating dampness and removing blood stasis are selected from one of Poria cocos, semen plantaginis, herba Lophatheri, Stephania tetrandra and rhizoma Alismatis;
the traditional Chinese medicinal materials for clearing heat and removing blood stasis are selected from one of buffalo horn, red paeony root, tree peony bark, sinkiang arnebia root, rehmannia root, figwort root and folium isatidis;
the Chinese medicinal materials with effects of regulating qi-flowing and removing blood stasis are selected from one of bupleuri radix, fructus Toosendan, radix Curcumae, fructus Citri Sarcodactylis, pericarpium Citri Reticulatae viride, fructus Aurantii, rhizoma Cyperi, and fructus Citri;
the Chinese medicinal materials with effects of replenishing blood, nourishing yin and removing blood stasis are selected from one of radix rehmanniae Preparata, colla Corii Asini, Polygoni Multiflori radix, fructus Lycii and arillus longan.
5. The composition for improving sperm motility and quality as described in claim 1, wherein said acetobacter is selected from one or more of acetobacter veillanthi, acetobacter oxyacantha, acetobacter pasteurianus, acetobacter schwanii, and acetomonas oxydans.
6. A method of preparing a composition for improving sperm motility and quality as described in any one of claims 1 to 5, wherein said method of preparation comprises the steps of:
s1: pulverizing the Chinese medicinal materials with blood circulation promoting effect into powder of 100 mesh or finer, mixing with corn starch powder of 100 mesh or finer, decocting in water, cooling to room temperature, adding Acetobacter, mixing, and fermenting for a certain period to obtain Chinese medicinal brewed acetic acid;
s2: sterilizing the acetic acid brewed by the traditional Chinese medicines at high temperature, and filtering by using a screen mesh with 1000-2000 meshes, wherein the filtrate is a liquid phase substance of the acetic acid brewed by the traditional Chinese medicines, and the filter cake is a solid phase substance of the acetic acid brewed by the traditional Chinese medicines; adjusting the pH value of a liquid phase substance of acetic acid brewed by traditional Chinese medicines to 3-3.5 by using alkali liquor and/or acid liquor;
s3: adding chitosan into the liquid phase material of acetic acid brewed by the traditional Chinese medicines, fully stirring to dissolve the chitosan, and filtering out insoluble substances by using a screen mesh with 1000-2000 meshes; adding solid phase substances of acetic acid brewed by traditional Chinese medicines, chlorhexidine acetate, menthol and/or tea tree oil into the filtrate, fully stirring, dropwise adding alkali liquor while stirring to adjust the pH value to 6.5-7, and standing for a period of time to obtain a gel substance;
s4: freeze drying the gel material in a freeze dryer to remove water to obtain gel solid; putting the gel solid into a pulverizer to pulverize to reach the particle size of 100 meshes or finer, so as to obtain gel powder;
s5: and (3) fully mixing the gel powder with pine pollen of 100 meshes or finer to obtain the composition for improving the motility and the quality of human sperms.

The method of claim 6, wherein the amount of water added in step S1 is 5-10 times the weight of the powder, the cooking temperature is 100-105 ℃, and the cooking time is 0.5-2 hours; the acetobacter can be bacterium powder or bacterium liquid cultured by a culture medium; the fermentation temperature is 28-32 ℃, and the fermentation time is 3-5 days.
8. The method of claim 6, wherein the temperature for sterilization at high temperature in step S2 is 100 ℃ for 5-30 minutes.
9. The method of claim 6, wherein the alkaline solution in steps S2 and S3 is sodium hydroxide, potassium hydroxide or ammonia; the acid solution in step S2 is hydrochloric acid or acetic acid.
10. A method of using a composition for improving sperm motility and quality as described in any one of claims 1 to 5, wherein there are two possible methods of use:
