Abstract

本发明提供一种木薯FRK1类似基因及其应用,所述木薯FRK1类似基因,在植物受到病原体侵染时,能够响应侵染信号并进行表达,可作为响应病原体侵染信号的标记基因,用于检测病原体对木薯的侵染情况,及时发现病害,采取相应的防治措施,对木薯抗病品种和抗病分子机理研究起到了重要的推动作用。

一种木薯FRK1类似基因及其应用
技术领域
本发明涉及植物基因工程技术领域,特别涉及一种木薯FRK1类似基因及其应用。
背景技术
木薯(Manihot esculenta Crantz)是热带地区重要的粮食作物,同时它也是一种重要的、很有发展潜力的再生能源作物。病害一直是影响木薯产业健康发展的重要制约因素之一。然而,木薯抗病分子机理还不清楚,抗病相关基因资源的报道缺乏,防控技术研究基础薄弱,已不能满足不断扩大的木薯产业的发展需求。因此,对木薯抗病机理及防控技术进行研究,进而采取相应的措施以减少病害对木薯产业的影响,稳步提高木薯产量,已成为木薯产业持续健康发展亟待解决的现实问题。
病原相关分子模式(PAMP)激发的植物先天免疫反应(PTI)是植物抵御病原菌的第一道防线,是植物界中更加广泛地存在着的植物防卫机制,表现为植物对绝大多数病原微生物种类普遍存在的、高效的和持续的抗病能力。因此植物先天免疫反应成为近年来植物抗病领域的研究热点。现有研究成果主要集中在模式植物拟南芥上,热带作物广谱抗性的研究,相对于植物抗病研究中的其它领域来说,还非常薄弱。因此,开展热带作物-木薯的抗病分子机理研究有望为我国热带作物广谱抗性育种提供基因资源,相关研究成果也可供热带作物抗病分子机理的研究参考。
发明内容
鉴以此,本发明提出一种木薯FRK1类似基因及其在检测病原体侵染情况中应用。
本发明的技术方案是这样实现的:
一种木薯FRK1类似基因为MeFRK1基因。
优选的,所述MeFRK1基因的核苷酸序列如SEQ ID NO.1所示。
优选的,所述MeFRK1基因编码的蛋白的氨基酸序列如SEQ ID NO.9所示。
另一方面的,本发明还提供了所述木薯FRK1类似基因在检测病原体侵染情况中应用。
优选的,所述应用为FRK1类似基因在响应病原体侵染信号中的应用。
优选的,所述FRK1类似基因可作为响应病原体侵染信号的标记基因的标记基因。
优选的,所述检测病原体侵染情况的方法为检测样品中所述FRK1类似基因表达情况。

本发明提供了一种木薯FRK1类似基因及其应用,本发明所述木薯FRK1类似基因,在植物受到病原体侵染时,能够响应侵染信号并进行表达,可作为响应病原体侵染信号的标记基因,用于检测病原体对木薯的侵染情况,及时发现病害,采取相应的防治措施,对木薯抗病品种和抗病分子机理研究起到了重要的推动作用。

具体实施方式
为了更好理解本发明技术内容,下面提供具体实施例,对本发明做进一步的说明。
实施例1木薯FRK1类似基因的筛选
首先,在Phytozome(https://phytozome.jgi.doe.gov/)网站上下载木薯全基因组数据,运用BLASTP软件,以来自NCBI(https://www.ncbi.nlm.nih.gov/protrin/)数据库的拟南芥FRK1基因的氨基酸蛋白序列作为输入序列,以木薯全基因组氨基酸蛋白序列为数据库,e值设置为-10,对木薯FRK基因家族全部成员进行鉴定,选取e值为0的8条木薯FRK基因,按照比对结果顺序命名为MeFRK1~MeFRK8,核苷酸序列见SEQ ID NO.1~8,氨基酸序列见SEQ ID NO.9~16,具体信息见表1。

筛选得到的8个FRK基因氨基酸长度均不相同,最长的是MeFRK8,有921个氨基酸残基,最短的是MeFRK3,有804个氨基酸残基,蛋白质序列平均长度为863aa。8个木薯FRK家族成员在木薯染色体上不均匀分布,其中,MeFRK1位于4号染色体上,MeFRK4、MeFRK5、MeFRK6、MeFRK7、MeFRK8都位于11号染色体,而MeFRK2和MeFRK3位于核骨架Scaffold上。
实施例2木薯FRK1类似基因的生物信息学分析
2.1木薯FRK1类似基因的结构分析
在Phytozome(https://phytozome.jgi.doe.gov/)网站下载木薯FRK1基因的内含子和外显子信息,使用在线网站GSDS(http://gsds.cbi.pku.edu.cn/)绘制基因模式图(图1)。
根据基因结构分析结果,8个候选基因聚为3大类。其中,MeFRK2和MeFRK6为一大类,MeFRK4单独为一大类,MeFRK1、MeFRK5、MeFRK7、MeFRK3和MeFRK8为一大类。
2.2木薯FRK1类似基因的保守基序分析
使用MEME(http://meme.nbcr.net/meme/cgi-bin/meme.cgi)预测木薯FRK1类似基因的保守基序,设置查找基序重复的数量为“any”,基序长度为50~250aa,预测基序的数量为3个,结果见图2和表2;

表中,Motif1为酪氨酸蛋白激酶(Protein tyrosine kinase,PTK)结构域,能催化多种底物蛋白质酪氨酸残基磷酸化,在细胞生长、增殖、分化中具有重要作用。Motif2为内质网的碳水化合物结合蛋白(Carbohydrate-binding protein of the ER)结构域,Motif3为富含亮氨酸重复(Leucine Rich Repea,LRR)结构域,参与蛋白质的相互作用,是已知抗病蛋白的重要结构。
2.3木薯FRK1类似基因染色体定位分析
运用在Phytozome(https://phytozome.jgi.doe.gov/)网站上下载的木薯FRK基因家族信息,来制作染色体定位图(图3)。
从图3中可以看出,8个木薯FRK1类似基因在木薯染色体上不均匀分布,MeFRK1位于4号染色体上,MeFRK4、MeFRK5、MeFRK6、MeFRK7、MeFRK8这五个基因都位于11号染色体,图中表示位置的数字单位为Mb。
综合结构分析、保守基序分析和染色体定位等结果,确定MeFRK1基因为木薯基因组中先天免疫下游的标记基因。
实施例3木薯FRK1类似基因上游基因共表达分析
使用在线网站STRING(http://www.string-db.org/)的数据库,预测木薯FRK基因家族有类似的互作关系,预测得到的木薯FRK1类似基因蛋白互作网络如图4所示,互作蛋白的基因信息见表3。

从蛋白互作网络中可以看出,木薯FRK1类似基因的互作蛋白大部分为2C类蛋白磷酸酶,表明木薯FRK1类似基因可能也参与了胁迫信号、激素信号转导等途径,为研究木薯FRK1类似基因在木薯中的作用提供了新的方向。
实施例4木薯FRK1类似基因响应病原体侵染的表达值热图
在NCBI的SRA数据库中下载来自木薯KU50品种的不同生长发育阶段的块根和叶的4个转录组数据包(SRR1299001,SRR1299002,SRR1299003,SRR1299000),Xam细菌侵染木薯所得的3个转录组数据包(SRR1050891,SRR1050892,SRR1050893),粉蚧虫侵染木薯所得的3个转录组原始数据(SRR2041628,SRR2043186,SRR2043192)。用Peiner et al.(2003)中的方法计算MeERF基因在组织特异性及各种逆境处理下的RPKM值。用Mev(MultiExperimentViewe)软件对数据进行处理,内置HCL程序进行层次聚类,结果用Heatmap图示(图5~6)。
结果显示,MeFRK1基因在Xam细菌和粉蚧虫侵染木薯后,均上调表达,说明MeFRK1基因能够响应不同病原体侵染信号,可作为木薯病害侵染情况检测的标记基因。
实施例5木薯FRK1类似基因的实时定量分析
使用Oligo7软件,为FRK家族8个基因设计实时定量引物,进行普通PCR实验检验其扩增特异性。

以木薯KU50品种的根部组织(生长期分别是73天、116天、143天)以及用激素水杨酸(SA)、茉莉酸(JA)、乙烯(Ethylene,ETH)处理的木薯SC8品种(华南八号)的叶片为样品,参照植物总RNA提取试剂(TIANGEN)说明书进行总RNA的分离,cDNA第一链的合成参照RevertAid First Stand cDNA synthesis(Thermo)说明书进行。Real-time PCR所用仪器为Rotor-Gene Q(QIAGEN),试剂为SYBR Premix Ex TaqTM(TaKaRa)。根据引物Tm值按照TaKaRa试剂说明书,Real-time PCR扩增程序为:95℃30s;“95℃5s,55℃30s,72℃30s,40个循环”;72℃延伸10min。
结果采用2-△△Ct的计算方法进行相对定量,以木薯EF1a基因作为内标校正模板量,每个样品进行3次独立的生物学重复。各激素处理时间点MeFRK1基因的表达水平均与该处理0min(处理前)相比。使用SigmaPLot软件处理数据并作柱形图(图7~10)。
从图7中可以看出,在JA诱导下MeFRK1基因的表达量显著上调,说明该基因参与了的JA信号应答过程。
从图8中可以看出,在乙烯诱导下该基因在30min后的表达量显著上调,说明该基因能够响应乙烯信号。
从图9中可以看出该基因能响应SA的刺激,但是响应强度比JA低。
从图10中可以看出,FRK1图表示该基因在病原菌Xam刺激后能显著增强表达,也进一步表明该基因可以作为木薯应答病原菌尤其是是JA信号相关的转导过程的标记基因。

1.一种木薯FRK1类似基因在检测病原体侵染情况中应用,所述木薯FRK1类似基因的核苷酸序列如SEQ ID NO.1所示,所述病原体为地毯草黄单胞木薯萎蔫致病变种(Xanthomonas axonopodis pv.manihotis,Xam)和/或粉蚧虫。
2.根据权利要求1所述的应用,其特征在于,所述木薯FRK1类似基因在响应病原体侵染信号中的应用。
3.根据权利要求2所述的应用,其特征在于,所述木薯FRK1类似基因为响应病原体侵染信号的标记基因。
4.根据权利要求3所述的应用,其特征在于,检测病原体侵染情况的方法为检测样品中所述FRK1类似基因表达情况。
