The test principle is as follows: the erythrocyte hemolysis experiment is based on the principle that lipid and protein on the surface of human skin are similar to cell membranes of erythrocytes, and the irritation degree of cosmetics is evaluated by detecting the influence of the cosmetics on hemoglobin release and hemoglobin denaturation of erythrocytes.
Preparation of sample suspension
Physiological saline or PBS is generally used as a solvent, and DMSO or ethanol is used as a solvent for a sample which is not dissolved in the physiological saline or PBS. It must be ensured that each diluted sample suspension is well mixed. Sample concentration range detection experiment: preparing a series of gradient suspensions of 0.001%, 0.01%, 0.1%, 1%, 10%, 20%, 30%, 40% and the like of a test object by using a solvent, mixing the suspensions with the erythrocyte suspension in an equal volume ratio, culturing at 37 ℃ for 3h, centrifuging to take supernatant, measuring absorbance at 410nm, 540nm and 575nm of an enzyme labeling instrument, and comparing with the absorbance values of a negative control and a positive control. According to the results, the concentration range of the sample is selected from the upper and lower intervals covering 50% of hemolysis rate.
And selecting proper test concentration according to the detection experiment result to prepare the concentration of the sample series.
Hemolysis and protein denaturation assays
To a 2mL centrifuge tube, 500. mu.L of the test substance and 500. mu.L of the 2% sheep red blood cell suspension (mixed in equal volumes) were added at different concentrations and mixed well. Each concentration was set in 3 replicates. Solvents (saline or PBS) were used as negative controls (i.e. no hemolysis at all) and 0.1% SDS solution was used as positive control (i.e. complete hemolysis). Incubating in a constant temperature incubator at 37 deg.C for 3h, centrifuging at 2000rpm/min for 3min, collecting supernatant 10 μ L, adding into an ELISA plate (adding 90 μ L physiological saline or PBS per well), mixing, and measuring absorbance at 410nm, 540nm, and 575nm with an ELISA reader (A410nm, A540nm, A575 nm).
The hemolysis rate (%) was calculated according to the following equation:
hemolysis ratio (%) - (a-C)/(B-C) × 100;
wherein A is the average absorbance of the sample at 410nm, B is the average absorbance of the positive control at 410nm, and C is the average absorbance of the negative control at 410 nm.
The concentration of the test substance HC50(mg/L) at which the test substance caused 50% hemolysis of red blood cells was determined according to the regression equation.
The protein denaturation index DI (%) was calculated according to the following formula:
DI (%) - (test substance a)575nm/A540nm) /(Positive control A)575nm/A540nm)×100。
Calculating the L/D value of the sample from the HC50 value and the DI value: HC50/DI, results are shown in Table 2; grading the irritation degree of the cosmetics according to RBC experiment grading standard of European Alternatives validation center (ECVAM), wherein the grading method comprises the following steps:
L/D >100 was rated as non-irritating;
the micro-stimulation is evaluated as the L/D is less than 10 and less than or equal to 100;
1< L/D < 10 is evaluated as mild irritability;
0.1< L/D.ltoreq.1 is evaluated as moderate irritability;
the L/D is less than or equal to 0.1, and the evaluation is the severe irritation.
TABLE 2
The results show that: the L/D values obtained by testing the makeup removing water provided by the application examples 1-5 are very high, which shows that the irritation is very small, and the L/D values obtained by testing the makeup removing water provided by the application examples 1-8 are lower and higher than that of the application example 1, which shows that the combination of the reed extract, the peony root extract, the honeysuckle flower extract and the philippine violet herb extract has a more remarkable effect on reducing the irritation of the raw materials of the cosmetics than the combination of any three or any single component, and the synergistic effect of the four components is proved.
Test example 4
Cleansing Water cleansing force test
140 women aged 18 to 35 years were selected and randomly divided into 14 groups of 10 persons each, and the cleansing water provided in examples 1 to 5 and comparative examples 1 to 8 and the commercially available cleansing water were used. Respectively using the same water-based eyebrow pencil, oil-based eyebrow pencil, water-based eyeliner, oil-based eyeliner, eye shadow, water-based mascara, oil-based mascara, concealer, blush and lipstick to equally coat the hand backs of cleaned testees (each testee needs to test all 10 color cosmetic products), after completely drying, using the same cosmetic cotton to dip the same amount of makeup removing water to cover the positions, on which the cosmetics are coated, of the hand backs for 5s, then wiping the hand backs in the same direction, replacing the cosmetic cotton, repeating the wiping step until no color cosmetic product remains on the hand backs, and recording the wiping times, wherein the average result of each group is shown in table 3.
TABLE 3
The results show that: the cleansing water prepared by the application examples has stronger cleaning capability on various water-based or oil-based color cosmetic products, wherein the cleansing capability of the cleansing water prepared by the application examples 4 and 5 is poorer than that of the application example 1, which shows that the cleansing water has excellent cleaning capability due to the combination of two surfactants, namely PEG-6 caprylic acid/capric acid glyceride and caprylyl/capric acid aminopropyl betaine, which are matched with each other and synergically. In addition, the cleaning ability of the cleansing water of application example 1 is not inferior to that of each comparative application example, which shows that the cleansing water provided by the invention has stronger cleaning ability and simultaneously achieves the purpose of remarkably reducing the irritation of cosmetic raw materials.
