本发明公开了一株链霉菌JXJA01及生物药剂、应用,该链霉菌JXJA01名称为Streptomyces europaeiscabiei JXJA01,已于2021年7月13日保藏于广东省微生物菌种保藏中心,地址为广州市先烈中路100号大院59号楼5楼,保藏编号为GDMCC NO:61805。该菌株和/或其发酵液能够用于制备防治根结线虫的生物药剂。本发明发现的链霉菌JXJA01菌株对根结线虫二龄幼虫具有更强的致死活性,且对根结线虫卵囊的孵化具有较强的抑制作用,对于作物根结线虫病的防治具有重要的应用价值,能够广泛应用于根结线虫病的防治生物药剂的制备当中。

背景技术
根结线虫(Root-knot nematodes,RKN),是一种高度专化型的杂食性植物病原线虫,寄生范围非常广泛,能侵染几乎所有的维管植物,包括蔬菜、果树、粮食作物和观赏花卉在内的3000多种植物,且生命力顽强、传播途径广泛、抗药性强,导致防治十分困难。
化学防治虽然可在一定程度上减轻根结线虫病的危害,但农户往往存在严重的滥用农药现象,长期大量使用会引发严重的农药残留、环境污染和抗药性等问题,对人类和环境产生危害。生物防治是一种重要的替代性控制策略,具有无毒、无残留、不破坏生态平衡、可利用资源丰富等优点,且有利于延缓病虫害抗药性的发生与发展,符合农业可持续发展的要求。因此,利用拮抗性微生物防治植物根结线虫病越来越受到人们的广泛关注。

本发明的目的是解决现有技术的不足,发明人发现了一株链霉菌JXJA01菌株,该菌株相比于现有的其它菌株,对根结线虫二龄幼虫具有更强的致死活性,且对根结线虫卵囊的孵化具有更强的抑制作用,对于作物根结线虫病的防治具有重要的应用价值。该链霉菌JXJA01菌株名称为欧洲疮痂链霉菌(Streptomyces europaeiscabiei)JXJA01,发明人已于2021年7月13日保藏于广东省微生物菌种保藏中心,地址为广州市先烈中路100号大院59号楼5楼,保藏编号为GDMCC NO:61805。该菌株的16S rDNA序列如SEQ ID No:1所示:

链霉菌JXJA01菌株和/或其发酵液可用于制备防治根结线虫的生物药剂中。发酵液的制备过程为:将菌落于挑至高氏一号培养基中200rpm培养96h,然后1000rpm离心10min,上清液用0.22μm细菌过滤膜过滤得到。其中,高氏一号培养基包括:可溶性淀粉20g,KNO3 1g,NaCl 0.5g,K2PO4·3H2O 0.5g,MgSO4·7H2O 0.5g,FeSO4·7H2O 0.01g,琼脂15-20g,蒸馏水1000mL,调节pH值至7.2-7.4。
本发明的有益效果为:本发明发现的链霉菌JXJA01菌株对根结线虫二龄幼虫具有更强的致死活性,且对根结线虫卵囊的孵化具有更强的抑制作用,对于作物根结线虫病的防治具有重要的应用价值,能够广泛应用于根结线虫病的防治生物药剂的制备当中。

具体实施方式
以下将结合实施例和附图对本发明的构思及产生的技术效果进行清楚、完整的描述,以充分地理解本发明的目的、方案和效果。
实施例1:
欧洲疮痂链霉菌(Streptomyces europaeiscabiei)JXJA01的分离与筛选:
发明人于2020年11月采集江西省吉安市泰和县竹篙薯种植区0~20cm深度土壤,做好标记后放入自封袋中,带回实验室4℃保存备用。采用稀释平板涂布法,称取新鲜保存的土样10g,倒入含90mL无菌水和10粒直径5mm玻璃珠的250mL锥形瓶中,放入28℃摇床中震荡30min混匀,使微生物细胞分散,静置20-30s,即为10-1稀释液,然后取0.1mL锥形瓶中混匀的土壤悬浊液加入到含有0.9mL无菌水的2mL离心管中,吹吸混匀,得到10-2土壤稀释液,以此类推,连续稀释得到10-3和10-4土壤悬浊稀释液。取10-4土壤悬浊稀释液进行涂板,所用固体培养基为高氏一号培养基(可溶性淀粉20g,KNO3 1g,NaCl 0.5g,K2PO4·3H2O 0.5g,MgSO4·7H2O 0.5g,FeSO4·7H2O 0.01g,琼脂15-20g,蒸馏水1000mL,调节pH值至7.2-7.4),将涂好的平板放置于桌面上20-30min,使菌液渗入培养基,然后将平板倒置于28℃恒温培养箱中培养。待长出放线菌菌落后,在无菌环境下,将平板上长出的放线菌单菌落用灭菌的移植钩转入新的培养基平板进行继代培养2-3代,直到菌株纯化为止。利用上述方法,通过筛选获得菌株编号为JXJA01的欧洲疮痂链霉菌(Streptomyces europaeiscabiei)(见图1)。发明人已于2021年7月13日保藏于广东省微生物菌种保藏中心,保藏编号为GDMCCNo.61805,保藏地址为广州市先烈中路100号大院59号楼5楼,广东省科学院微生物研究所。
实施例2:
欧洲疮痂链霉菌(Streptomyces europaeiscabiei)JXJA01的鉴定:
(1)形态学特征:用插片法培养5、7、10天后观察:JXJA01菌株的孢子丝弯曲或略弯曲;在酵母精麦芽精琼脂培养基(ISP2)、燕麦粉琼脂培养基(ISP3)、无机盐淀粉琼脂培养基(ISP4)和甘油天冬素琼脂培养基(ISP5)上培养,气生菌丝体均呈灰色,基质菌丝体黄色至黄褐色,在ISP2和ISP4培养基上分别可产生黄色和褐色的可溶性色素,而在ISP3和ISP5培养基上均无可溶性色素产生。JXJA01菌株在光学显微镜下的照片如图2所示。

(2)生理特性:对菌株JXJA01的碳源进行筛选,结果如表2所示:该菌株对葡萄糖、棉籽糖、阿拉伯糖、蔗糖和甘露醇均能利用;不能够使明胶液化,能使牛奶凝固和胨化,亦可还原硝酸盐和水解淀粉,能产生黑色素和硫化氢。

(3)分子生物学特性:将得到的菌株JXJA01在高氏一号固体培养基上培养7d后,在超净工作台中用无菌枪头挑取代表菌株菌团块至2mL无菌离心管中,利用植物DNA提取试剂盒(通用型)提取其DNA。分别利用27F(AGTTTGATCMTGGCTCAG),1492R(GGTTACCTTGTTACGACTT)和rpo B-5':ATCAACATC-CGGCCGG TGGT-3',rpo B-3':GGTTTCGATCGGGCA-CATCC-5'为引物,试验菌株为模板,扩增其16S rRNA基因和rpo B基因并纯化测序。并采用MEGA软件,基于16S rDNA序列,邻接法构建系统发育树,1000次重复计算,节点显示Bootstrap值大于50%数值,上标“T”表示模式菌株。
欧洲疮痂链霉菌(Streptomyces europaeiscabiei)JXJA01的16S rDNA的序列如SEQ ID No:1所示;rpoB基因测序序列如SEQ ID No:2所示。
菌株JXJA01测序得到的16S rRNA序列为1347bp,用Blast程序在Genebank与已登录的序列进行核酸同源性比较,该菌的16S rRNA序列与Streptomyces europaeiscabiei的同源性达99.93%,与Streptomyces scabiei(疮痂病链霉菌)的同源性均达99.85%;测序得到的rpo B基因序列为682bp,用Blast程序在Genebank与已登录的序列进行核酸同源性比较,该菌的rpo B基因序列与Streptomyces europaeiscabiei的同源性达98.53%;系统进化树的分析结果也表明该菌与Streptomyces europaeiscabiei遗传距离更接近(如图3所示)。
结合菌落形态学特征、生理生化特征及分子生物学特性,明确菌株JXJA01为欧洲疮痂链霉菌(Streptomyces europaeiscabiei)JXJA01。
实施例3:
欧洲疮痂链霉菌(Streptomyces europaeiscabiei)JXJA01发酵液的制备:
将菌株JXJA01于无菌条件下接种到实施例1中的9cm高氏一号固体培养基上,25℃黑暗培养4d左右即可长出使用。挑取平板上生长良好的单菌落于100mL改良的高氏一号液体培养基中,200rpm摇床培养96h后制成放线菌菌悬液,10000rpm离心10min,取上清液用0.22μm细菌过滤膜过滤即制成菌株JXJA01的发酵液。同时摇瓶空白高氏一号液体培养基(无菌株)、离心、过0.22μm滤膜后作为对照,4℃保存备用。
实施例4:
欧洲疮痂链霉菌(Streptomyces europaeiscabiei)JXJA01致病性的测定:
萝卜幼苗检测法:将萝卜籽先用0.26%NaClO溶液浸泡约10min,用无菌水冲洗3遍,晾干,置于无菌滤纸保湿的培养皿中催芽24h,选择萌发一致的籽粒放入装有5mL 1%水琼脂培养基的试管中,每管2粒。分别接种200μL JXJA01菌悬液、200μL JXJA01菌株代谢产物,以接种200μL无菌水和200μL空白高氏一号液体液体培养基(G-CK)为对照,于24℃光照培养箱中培养,光照时间为16h/d,分别于接种1d、2d、3d、5d后测量幼苗长度,以各处理的抑制率作为致病性指标。抑制率=(1-接种幼苗长度的平均值/对照幼苗长度平均值)×100%。
结果:萝卜幼苗检测法结果表现为接种菌株JXJA01菌悬液和发酵液的胡萝卜幼苗长势与对照相比无显著差异(见图4),由表3也可看出,菌株JXJA01菌悬液和发酵液对幼苗抑制作用不明显,菌株JXJA01发酵液甚至可略微促进胡萝卜幼苗的生长。由此表明筛选到的欧洲疮痂链霉菌(Streptomyces europaeiscabiei)JXJA01无致病性。

实施例5:
欧洲疮痂链霉菌(Streptomyces europaeiscabiei)JXJA01发酵液杀根结线虫的作用测定:
(1)根结线虫的培养
收集受南方根结线虫侵染的黄瓜根系,用水轻轻冲洗,小心取下卵囊。放入1%次氯酸钠溶液中消毒3min,再用无菌水冲洗3次,放入内有少量无菌水的培养皿里,置于25℃恒温中培养,每隔24h收集一次新孵化的南方根结线虫2龄幼虫悬浮液,置于4℃冰箱保存备用。
(2)对比菌株
淡紫褐链霉菌(Streptomyces enissocaesilis):购买自中国普通微生物菌种保藏管理中心的根结线虫生防菌株,保藏号为CGMCC 4.1586。
JZ-1菌株和JZ-2菌株:发明人在筛选欧洲疮痂链霉菌(Streptomyceseuropaeiscabiei)JXJA01菌株时,一同发现的其它链霉菌属菌株。
(3)杀线虫作用测定
将实施例3中的菌株JXJA01发酵液500μL分别加入到24孔细胞培养板中,并加入500μL南方根结线虫悬液(30-100条),进行毒力测定。以目前现有的根结线虫生防菌株淡紫褐链霉菌和相同方法筛选到的JZ-1菌株和JZ-2菌株为对比菌株,以无菌水和高氏一号空白液体培养基(G-CK)为对照,每组处理4次重复。放入25℃培养箱中,24h后记录线虫死亡情况,针触法判断线虫死活,计算校正死亡率。线虫致死率=死亡线虫/供试线虫总数×100%;校正死亡率(%)=(处理线虫死亡率-对照线虫死亡率)/(1-对照线虫致死率)×100%,死亡线虫是指在物理刺激条件下不运动的线虫,每个时间点进行4个生物学重复。
结果:如表4所示,菌株JXJA01发酵液处理24h后,线虫基本均呈僵直死亡状态,针刺不动,校正死亡率达94.63%,而对比菌株JZ-1菌株、JZ-2菌株和淡紫褐链霉菌的校正死亡率分别仅为16.13%、15.16%和36.26%。由此表明,菌株JXJA01发酵液对南方根结线虫二龄幼虫的毒杀作用明显优于对比菌株。

实施例6:
欧洲疮痂链霉菌(Streptomyces europaeiscabiei)JXJA01发酵液对根结线虫卵囊孵化的作用测定:
将实施例3中的菌株JXJA01发酵液2.5mL加入到无菌培养皿(直径6cm)中,并加入2.5mL无菌水,使菌悬液的稀释倍数为2×,再每皿加入20个颜色相近、大小相同消毒后的卵囊,以等体积淡紫褐链霉菌、JZ-1菌株和JZ-2菌株为对比菌株,以等体积空白高氏一号液体培养基和无菌水为对照,每个处理重复3次,放入28℃培养箱于24h后统计卵囊孵化线虫数,计算卵囊孵化的相对抑制率。相对抑制率(%)=(对照卵囊孵化线虫数-处理卵囊孵化线虫数)/对照卵囊孵化线虫数×100。
结果如表5所示:菌株JXJA01发酵液2倍稀释液对根结线虫的卵囊孵化相对抑制率达87.10%,对比菌株JZ-1菌株、JZ-2菌株和淡紫褐链霉菌对根结线虫的卵囊孵化相对抑制率仅为27.24%、22.65%和46.85%。由此表明,菌株JXJA01发酵液对对根结线虫卵囊的孵化具有更强的抑制作用,且抑制作用显著优于对比菌株JZ-1菌株、JZ-2菌株和淡紫褐链霉菌。

综上所述,欧洲疮痂链霉菌(Streptomyces europaeiscabiei)JXJA01发酵液不仅对根结线虫二龄幼虫具有强烈的毒杀作用,也可在很大程度上抑制卵囊的孵化率,对作物根结线虫病的防治具有重要的应用价值。
