2 method
The method referred to in "technical Specification for health food inspection and evaluation (2003 edition)" is used for establishing a constipation model of mice by using loperamide hydrochloride. The experiment comprises a normal control group, a model control group, a low-dose test group (0.5g/kg), a medium-dose test group (1.0g/kg) and a high-dose test group (3.0g/kg), wherein each group contains 10 animals, the animals are administrated by gastric gavage for 10 days continuously for 1 time every day, and fresh feces are collected and are conveniently stored for later use at-80 ℃. 3 results
Constipation mice intestinal flora was detected by 16S rRNA gene sequencing method, Alpha diversity analysis showed: compared with a normal control, the number of the flora at the genus level of the model control group is obviously reduced by P <0.05, while the number of the flora at the genus level of each test group has no statistical difference, namely, the intestinal flora of the mouse at the model group is obviously reduced, and the test group can improve the reduction condition of the flora. The effect on the diversity of the intestinal flora is shown in FIG. 1.
Fourth, conclusion
