Abstract
The invention belongs to the technical field of aquaculture, and discloses a digestion-aiding fish liquid feed additive, a preparation method and an application thereof. According to the invention, four dominant bacterial strains are added to compete with harmful bacteria for oxygen attachment sites and nutrients, so that the growth of the harmful bacteria is inhibited, and the flora in intestinal tracts of fishes is regulated to be normalized; the raw materials with reasonable proportion are added to promote the proliferation of beneficial bacteria in fish intestinal tracts and improve the feed conversion rate, thereby balancing the nutritional ingredients in the fish feed, helping the fish digestion and meeting the requirements of the fish in each growth period.
Description
Technical Field
The invention belongs to the technical field of aquaculture, and relates to a feed additive and a preparation method thereof, in particular to a digestion-aiding fish liquid feed additive and a preparation method and application thereof.
Background
The ornamental fish is fish with ornamental value, bright color or peculiar shape, has various varieties, beautiful body, bright color and elegant swimming appearance, is deeply favored by the majority of fish enthusiasts, and more people for breeding the ornamental fish. In the process of feeding ornamental fishes, requirements on fish feed are strict in addition to requirements on external factors such as water quality, fish tanks, oxygen and the like. In the current market, the ornamental fish feed has a plurality of defects, the feed has single components, incomplete nutritional ingredients and poor palatability, and dyspepsia of the ornamental fish is easily caused, even the ornamental fish is eaten by overeating and dies; if the fed fish feed is not eaten in time, the fish feed floats on the water surface to pollute the water quality, so that the aquarium fish is diseased and died. In order to solve the problem of insufficient nutritional ingredients of the aquarium fish feed, most of the aquarium fish feed sold in the market is prepared by mixing raw materials, puffing, granulating, drying and forming and other processes, and high-temperature drying inevitably loses part of nutritional ingredients such as vitamins and proteins, so that comprehensive nutritional ingredients cannot be provided for the aquarium fish.
Disclosure of Invention
The invention aims to provide a digestion-aiding fish liquid feed additive to balance nutrient components in fish feed, help fish digestion and meet the requirements of fish in each growth period;
the invention also aims to provide a preparation method of the digestion-aiding fish liquid feed additive;
still another object of the present invention is to provide an application of the liquid feed additive for fish for aiding digestion.
In order to achieve the purpose, the invention adopts the technical scheme that:
a digestion-aiding fish liquid feed additive contains four probiotics, namely bacillus licheniformis, enterococcus faecium, lactobacillus acidophilus and bifidobacterium animalis;
the raw materials for preparing the effective components of the digestion-aiding fish liquid feed additive in every 1000 volume parts comprise:
main materials: 3-15 parts of compound amino acid, 0.2-0.6 part of saccharicterpenin, 0.3-0.8 part of galactooligosaccharide, 0.2-0.9 part of mannooligosaccharide, 0.3-1.5 parts of cellulase, 0.3-1.6 parts of yeast cell wall powder and 2-8 parts of compound vitamin;
auxiliary materials: 0.2-0.6 part of antioxidant and 0.1-0.3 part of complexing agent;
supplementing to 1000 parts by volume with sterile water;
wherein, the proportion relation of the parts by weight to the parts by volume is 1 kg: 1L;
bacillus licheniformis and enterococcus faecium were purchased from Oucaceae Behcer Biotech Ltd;
lactobacillus acidophilus and Bifidobacterium animalis were purchased from Jiayi bioengineering, Inc., of middle-school of Shandong;
the cellulase (with enzyme activity of 10000U/g) is purchased from Shandong Su Koehan bioengineering GmbH;
in the present invention, these probiotics and cellulases are used, but not limited to the probiotics and cellulases of the above companies, and it is to be understood that the probiotics to be referred to in the technical solution claimed in the present invention are all probiotics of the same genus and the cellulases to be referred to are all cellulases having the same performance.
As a limitation, the raw materials for preparing the effective components of the digestion-aiding fish liquid feed additive per 1000 parts by volume further comprise: 3-10 parts of bacillus licheniformis powder, 2-8 parts of enterococcus faecium powder, 1-6 parts of lactobacillus acidophilus powder and 1-5 parts of bifidobacterium animalis powder.
As another limitation, in the digestion-aiding fish liquid feed additive, the content of the bacillus licheniformis in the digestion-aiding fish liquid feed additive is more than or equal to 3.0 multiplied by 105CFU/mL, enterococcus faecium content greater than or equal to 2.0 × 105CFU/mL, Lactobacillus acidophilus content not less than 1.0 × 105CFU/mL and the content of animal bifidobacterium is more than or equal to 1.0 multiplied by 104CFU/mL。
As a third limitation, the raw materials for preparing the effective components of the compound amino acid comprise, by weight: 2.5-4.5 parts of lysine hydrochloride, 2-4 parts of arginine hydrochloride and 0.1-0.7 part of tyrosine.
As a fourth definition, the antioxidant is sodium bisulfite, anhydrous sodium sulfite, sodium metabisulfite, thiourea, or dibutylhydroxytoluene;
the complexing agent is disodium ethylene diamine tetraacetate or citric acid.
The invention also provides a preparation method of the digestion-aiding fish liquid feed additive, which comprises the steps of adding the main material and the auxiliary material into sterile water, dissolving, filtering and sterilizing, then adding the four probiotics, uniformly mixing and fixing the volume to obtain the digestion-aiding fish liquid feed additive.
As a limitation, both the dissolving and the blending are performed at room temperature.
As another limitation, the filter sterilization is performed using a microporous filter membrane.
As a third limitation, the temperature for filtration sterilization is 4-8 ℃;
the digestion-aiding fish liquid feed additive also needs to be filled at room temperature.
The invention also provides an application of the digestion-aiding fish liquid feed additive, and the digestion-aiding fish liquid feed additive is added into fish feed for feeding fish so as to promote the digestion of the fish.
Due to the adoption of the technical scheme, compared with the prior art, the invention has the technical progress that:
according to the invention, beneficial bacteria such as lactobacillus and bacillus can be used as members of inherent flora in fish bodies, and can be planted in fish digestive tracts, antagonize potential pathogenic bacteria, maintain intestinal microecological balance, compete for oxygen attachment sites and nutrients with harmful bacteria, competitively inhibit the growth of the harmful bacteria, regulate the intestinal flora of fish to be normalized and enhance the disease resistance; the four probiotics are metabolized to generate organic acid, so that the pH value in the intestinal tract can be reduced, and harmful bacteria which are not acid-resistant can be killed;
the bacillus licheniformis can generate various digestive enzymes, is beneficial to decomposing feed nutrients, synthesizing nutrient substances such as growth promoting factors and the like, and stimulating intestinal immune cells, so that local immune antibodies are increased, and the disease resistance of an organism is enhanced;
enterococcus faecium can be quickly adhered to the intestinal mucosa, and pathogenic bacteria are inhibited from adhering to the intestinal mucosa through exclusion effect, so that an intestinal barrier is formed, the intestinal health is protected, and the micro-ecological balance is maintained. Enterococcus faecium can also produce substances such as lactic acid, bacteriocin, hydrogen peroxide and the like in the metabolic process, so that the pH value of the intestinal tract is reduced, the reproduction of pathogenic bacteria in the intestinal tract is inhibited, the microecological balance of the intestinal tract is maintained and adjusted, the contents of endotoxin and urease in the intestinal tract are reduced, the contents of toxin and ammonia in blood are reduced, the organ maturation of fishes is promoted, and the physiological state of the fishes is improved. Enterococcus faecium can also produce various nutrient substances (such as vitamins, amino acids, growth promotion factors and the like), and the nutrient substances can participate in fish metabolism and promote fish growth. The enterococcus faecium can also induce organisms to generate cytokines, interferon, interleukin and the like, enhance the nonspecific immunity of the organisms, improve the disease resistance, reduce the inflammatory reaction and promote the intestinal health;
the lactobacillus acidophilus can secrete antibiotic substances, such as acidophilic lactobacillus (acidophilolin), acidophilic lactobacillus (acidophilophilin), lactobacillin (1 aerocidon) and the like, and has antagonistic action on intestinal pathogenic bacteria, so that the effects of regulating the balance of intestinal flora and inhibiting the proliferation of harmful bacteria in the intestinal tract are achieved;

the compound amino acid contains lysine, arginine and tyrosine;
lysine is a limiting amino acid, the balance degree of the amino acid can be improved, the amino acid and the protein in the feed have the highest nutritional value, and the digestion utilization rate of the protein in the feed is improved;
the addition of a proper amount of arginine can promote the growth of the fish and also can improve the intestinal tract morphological structure of the fish;
tyrosine is an important nutritional essential amino acid and plays an important role in the metabolism, growth and development of fishes;
after the saccharicterpenin is fed, the immunity and disease resistance of fish organisms can be obviously enhanced, the growth is promoted, the daily gain and the feed conversion rate are improved, the anti-stress and anti-oxidation effects are achieved, and meanwhile, the saccharicterpenin has a strong prevention effect on intestinal bacterial diseases;
the galacto-oligosaccharide can selectively promote the proliferation of beneficial bacteria in the intestinal tracts of the aquatic animals, and the beneficial bacteria utilize oligosaccharide fermentation to generate short-chain fatty acid, reduce the pH value of the intestinal tracts, inhibit pathogenic bacteria from consuming nutrients in vivo and reduce the generation of toxic and pathogenic metabolites, thereby maintaining and improving the health of the aquatic animals; the mannan oligosaccharide can improve the immune response capability of organisms to drugs and antigens and enhance the immune capability of aquatic animals;
the cellulase can promote the digestion and absorption of feed and promote the ingestion and growth of aquatic animals; the cellulase has effects of improving digestive system function, increasing feed effect, and reducing content of nutrients in excrement.
The yeast cell wall powder has the functions of stimulating and enhancing immunity, and can enhance the resistance to diseases and environmental changes and improve the survival rate in the face of the stress of water environment changes;
the compound vitamin comprises vitamin D, vitamin E, and vitamin B1. The digestion-aiding fish liquid feed additive is used for being added into fish feed to feed fishes, and is particularly suitable for feeding ornamental fishes.
Detailed Description
The present invention is further illustrated by the following specific examples, which are to be construed as merely illustrative, and not limitative of the remainder of the disclosure.
Example 1 preparation of a digestive liquid feed additive for fish
The preparation method of the 1000L digestion-aiding fish liquid feed additive comprises the following steps in sequence:
1) 2.8kg of lysine hydrochloride, 2.5kg of arginine hydrochloride and 0.4kg of tyrosine are taken and mixed evenly to obtain the compound amino acid (marked as FA 1).
2) Collecting 1.5kg vitamin D, 1kg vitamin E and 0.8kg vitamin B1And mixing uniformly to obtain the compound vitamin (marked as W1).
3) Adding 5.7kg of compound amino acid A1, 0.2kg of saccharicterpenin, 0.4kg of galacto-oligosaccharide, 0.3kg of mannooligosaccharide, 0.8kg of cellulase (enzyme activity is 10000U/g), 0.6kg of yeast cell wall powder, 4kg of compound vitamin W1, 0.35kg of sodium bisulfite and 0.15kg of disodium ethylene diamine tetraacetate into 700L of sterile water (recorded as sterile water for dissolution), stirring and dissolving at room temperature, cooling to 5 ℃, filtering and sterilizing by a microporous filter membrane sterilizing filter with the diameter of 0.22 mu m, and then adding 4kg of bacillus licheniformis powder (the content of bacillus licheniformis is 1.03 multiplied by 10)8CFU/g), 3kg enterococcus faecium powder (enterococcus faecium content is 1.02 × 10)8CFU/g), 2kg Lactobacillus acidophilus powder (Lactobacillus acidophilus content is 1.01 × 10)8CFU/g) and 2kg Bifidobacterium animalis powder (Bifidobacterium animalis content of 1.05 × 10)7And (3) stirring and mixing the CFU/g uniformly at room temperature, and then adding sterile water to a constant volume of 1000L to obtain the digestion-aiding fish liquid feed additive.
Aseptically packaging the digestion-promoting fish liquid feed additive at room temperature to obtain commercially available digestion-promoting fish liquid feed additive (labeled as N1) containing 4.12 × 105CFU/mL Bacillus licheniformis, 3.06X 105CFU/mL enterococcus faecium, 2.02X 105CFU/mL Lactobacillus acidophilus and 2.1X 104CFU/mL Bifidobacterium animalis.
The digestion-aiding fish liquid feed additive prepared in the embodiment is used for being added into fish feed to feed fishes, and is particularly suitable for feeding ornamental fishes.
Example 2-6 preparation method of digestion-promoting fish liquid feed additive
Examples 2 to 6 are methods for preparing a digestion-aiding liquid fish feed additive, which have substantially the same steps as in example 1, but differ only in the amount of raw materials and process parameters, as detailed in table 1:

EXAMPLE 7 use of a liquid Fish feed additive to aid digestion
700 peacock fishes (half of male and half of female) with 7 days old and good health condition are selected and randomly divided into 7 groups, each group comprises 100 peacock (half of male and half of female), a control group and A1-A6 groups, wherein the control group is fed with common commercial fish feed (Italian small fish feed), the A1-A6 group is also fed with common commercial fish feed (Italian small fish feed), meanwhile, the A1-A6 group is also fed with liquid feed additives N1-N6 respectively, the addition amount is 0.5 percent of the fish feed amount, the feeding is carried out for 1 time every day, and the test period is 90 days according to the standard feeding method of the peacock fishes under other conditions of illumination, oxygen, water changing and the like. The health condition, fish body performance, and the like of the sparrow fish are observed every day during the test period, statistics is carried out, and the specific results are shown in the following table:

As can be seen from table 2, the average body weight of peacock fish increases with the day-old peacock fish, but the average body weight of peacock fish after 90 days in the a1-a6 groups was 1.9g or more higher than that of the control group, the survival rate of the a1-a6 groups was 13% higher than that of the control group, and the fish body appearance with bright color and uniform body color was exhibited after 90 days in the a1-a6 groups, while the fish body appearance with dark color was exhibited in the control group. The experimental results prove that the digestion-aiding fish liquid feed additive prepared by the invention is beneficial to balancing the nutrient components in fish feed, improving the feed conversion rate, accelerating the growth and development of fish, enhancing the immunity and disease resistance of fish, improving the survival rate, promoting the healthy growth of ornamental fish and forming bright body color.
Claims (10)
1. A fish liquid feed additive for aiding digestion is characterized in that,
the digestion-aiding fish liquid feed additive contains four probiotics, namely bacillus licheniformis, enterococcus faecium, lactobacillus acidophilus and bifidobacterium animalis;
the raw materials for preparing the effective components of the digestion-aiding fish liquid feed additive in every 1000 volume parts comprise:
main materials: 3-15 parts of compound amino acid, 0.2-0.6 part of saccharicterpenin, 0.3-0.8 part of galactooligosaccharide, 0.2-0.9 part of mannooligosaccharide, 0.3-1.5 parts of cellulase, 0.3-1.6 parts of yeast cell wall powder and 2-8 parts of compound vitamin;
auxiliary materials: 0.2-0.6 part of antioxidant and 0.1-0.3 part of complexing agent;
supplementing 1000 parts by volume of water;

7. The method for preparing a liquid feed additive for fish according to claim 6, wherein the dissolving and the mixing are performed at room temperature.
8. The method for preparing a liquid feed additive for digestive fishes according to claim 6 or 7, wherein the filtration sterilization is performed by using a microporous membrane.
9. The method for preparing the digestion-aiding fish liquid feed additive according to claim 6 or 7, wherein the temperature for filtration sterilization is 4-8 ℃.
10. Use of the digestive fish liquid feed additive according to any one of claims 1 to 5, wherein the digestive fish liquid feed additive is added to fish feed for feeding fish to promote fish digestion.
