本发明提供了一种人工合成多肽药物制剂的制备方法。将溶剂溶解多肽,与载体均匀混合,将混合液平铺于玻璃皿上为一薄层,置于干燥箱中,在其下放干燥剂,在0~40℃条件下抽真空干燥,直到恒重,其中溶剂采用甲醇、乙醇、丙酮,载体采用寡糖类或氨基酸或空微囊类或与多肽复配的药物。方法简单,除去了冻干设备和运转费用,可大大降低成本。

人工合成多肽药物制剂的制备方法
技术领域:
本发明涉及的是一种非冷冻干燥人工合成多肽药物制剂的制备方法。
背景技术:
自然界中有各种各样的活性多肽具有特有的生物功能。从化学结构上看都是由氨基酸按特定顺序组成。多才多肽在高温、高温强光下,易发生构象改变,肽键断裂、氧化、水解等化学变化,导致失活,所以其制剂都是冷冻干燥,属生化制剂。已有的制备这种制剂的方法是采用真空冷冻干燥,可精确分装,可保证多肽药物不被失活。但由于真空冷冻干燥需真空冷冻干燥机,投资大,运转费用高,使制剂成本高。
发明内容:
鉴于以上原因,本发明的目的是为了克服以上不足,提供一种可大大降低成本的人工合成多肽药物制剂的制备方法。
本发明人工合成多肽药物制剂的制备方法是将溶剂溶解多肽,与载体均匀混合,将混合液平铺于玻璃皿上为一薄层,置于干燥箱中,在其下放干燥剂,在0~40℃条件下抽真空干燥,防止多肽降解失活,直到恒重,其中溶剂采用甲醇、乙醇、丙酮,载体采用寡糖类或氨基酸或空微囊类或与多肽复配的药物,干燥剂可采用P2O5、CaO、硅胶等。
上述的寡糖类采用左旋糖菇或甘露醇。
上述的复配药物为与多肽复配的多潘立酮、孕酮、克罗米芬等等。
本发明方法是通过低沸点极性溶剂溶解多肽,以可溶于水并对多肽有支承作用的寡糖类或氨基酸类或空微囊类或可与多肽复配的药物为载体,抽真空干燥得粉剂。其制剂不需冻干机,大大降低了成本。
本发明方法简单,除去了冻干设备和运转费用,可大大降低制剂的成本。
具体实施方式:
实施例1:
本实施例1制剂的方法是将九肽、十肽化合物促性腺素释放素GnRH-A溶于甲醇中,与载体——多潘立酮均匀混合(GnRH-A与多潘立酮按配方比混合,如1∶1)将混合液平铺于玻璃皿上为一薄层,置于干燥箱中,其下放干燥剂,抽真空干燥直到恒重得非冷冻干燥人工合成多肽药物复配制剂。将制剂注射麦端加拉鲮催产,产卵率>90%。
实施例2:
将促性腺激素释放素GnRH-A溶解于甲醇中,加载体左旋糖苷、甘露醇(以GnRH-A20~100μg、左旋糖苷3-10mg、甘露醇3-10mg配制)均匀混合,将混合液平铺于玻璃皿上为一薄层,置于干燥箱中,其下放置干燥剂,抽真空干燥直到恒重。将制剂注射革胡子鲶,产卵率>90%。
实施例3:
将促进腺激素释放素GnRH-A溶解于甲醇,加入载体氨基酸(赖氨酸、甘氨酸、谷氨酸)(以GnRH-A500~1000μg,氨基酸10~20mg配制),将混合液置于玻璃皿上为一薄层,置于干燥箱中,其下放置干燥剂,抽真空干燥直到恒重。将此制剂注射于牛,引起发情、排卵。

1、人工合成多肽药物制剂的制备方法,其特征在于将溶剂溶解多肽,与载体均匀混合,将混合液平铺于玻璃皿上为一薄层,置于干燥箱中,在其下放干燥剂,在0~40℃条件下抽真空干燥,直到恒重,其中溶剂采用甲醇、乙醇、丙酮,载体采用寡糖类或氨基酸或空微囊类或与多肽复配的药物。
2、如权利要求1所述的人工合成多肽药物制剂的制备方法,其特征在于寡糖类采用左旋糖菇或甘露醇。
3、如权利要求1所述的人工合成多肽药物制剂的制备方法,其特征在于药物为与多肽复配的多潘立酮、孕酮、克罗米芬。
