Technical Field
The invention relates to a plant extract and application of the plant extract in preventing and treating cow mastitis.
Background
Oxidative stress is liable to cause mastitis, ketosis, etc. in dairy cows. The mastitis of the dairy cow is one of the most common and important diseases for restricting the development of the dairy cow breeding industry, not only affects the reduction of milk yield and the reduction of milk quality, but also even leads the dairy cow to lose production performance and causes serious economic loss, and in addition, the mastitis is treated, and the problems of medicament residue and the like are caused by using a large amount of antibiotics, thereby seriously harming human health. It is reported that about 2.2 million cows are stocked globally and about 1/3 cows have each type of mastitis, causing economic losses of up to $ 350 million; more than 1000 million cows are kept in a stall in China, the incidence rate of clinical mastitis is about 30 percent, and the incidence rate of recessive mastitis reaches 70 percent.
Antibiotics are listed as the first choice drugs for treating mastitis all over the world for decades, however, due to long-term repeated use, pathogens generate drug resistance to drugs, the curative effect is reduced, and the residue of antibiotics in milk is increased, so that the development and utilization of antibiotics substitutes becomes a current research hotspot.
The technology for preventing and treating mastitis by adding the plant extract into daily ration of the dairy cow is a hot point of scientific research in recent years, the plant extract is a natural plant component, has no toxic action on animals, and has a very wide application prospect, so that the screening of the plant extract suitable for preventing and treating the mastitis of the dairy cow has important significance.

The arabidopsis thaliana, also known as goat arabidopsis thaliana, radishes, sauerkraut, sandcap and Mongolian name 'Menglason-Lao pump', is a big annual or biennial herb of brassicaceae and arabidopsis, and the plant height is 0.5-2 m. The arabidopsis thaliana is a special plant of Mongolian plateau desert and sand land, and is grown on the sand land or a semi-fixed and mobile sand dune in a grassland area. The arabidopsis thaliana is mainly distributed in inner Mongolia, Shaanxi, Ningxia, Gansu and other places, has higher nutritional value and health care value, and is in good standing of desert ginseng. The wasabi has fresh and unique mustard flavor, high nutrition and health care value, and is rich in various vitamins, trace elements and crude fibers. The leaf of Thellungiella salsuginea has strong antibacterial effect, and the active ingredients of the leaf mainly comprise polyphenol, flavone, alkaloid and the like.
The invention is based on that the Arabidopsis contains a plurality of active ingredients and has strong bacteriostatic and antioxidant effects, so the research and establishment of the extraction process conditions of the Arabidopsis active ingredients provide a specific technical scheme for preventing and controlling the cow mastitis.
Therefore, the present disclosure describes specific technical solutions adopted to implement the above inventive concepts in combination with specific embodiments.
The first embodiment is as follows: ethanol reflux extraction of Thellungiella salsuginea
Drying caulis et folium Brassicae Junceae, pulverizing, mixing with ethanol, heating the mixture to 75 deg.C, refluxing for 120 min for extraction, recovering ethanol, and drying the extractive solution by freeze-drying machine to obtain powder.

TABLE 2 ultrasonic extraction of Arabidopsis ethanol Experimental data
Experimental example: bacteriostasis test
The experimental example disclosed herein adopts an oxford cup method for carrying out a bacteriostatic test, and the specific experimental method is as follows:
80g of the ethanol extract powder of the Arabidopsis thaliana obtained in the first and second examples are respectively weighed and dissolved in ultrapure water to prepare a 1g/mL solution of the Arabidopsis thaliana extract, and the solution is sterilized at 121 ℃ for 20min under ultra-high pressure and then placed at 4 ℃ for later use.
The strain placed at-80 ℃ was thawed and cultured in nutrient broth with shaking at 37 ℃ for 18 h. And (4) marking the activated bacterial liquid on a nutrient agar culture medium, and standing and culturing at 37 ℃ for 24 h. And selecting the purified single strain, inoculating the single strain into a nutrient broth culture medium, and performing shaking culture at 37 ℃ for 18h to obtain a raw bacterium solution. The concentration of staphylococcus aureus liquid is adjusted to 1 × 106CFU/mL by using sterilized normal saline, and the staphylococcus aureus liquid is used as test liquid for standby.
Transferring 0.1mL of prepared test bacterial liquid of staphylococcus aureus to a prepared nutrient agar plate, uniformly coating the bacterial liquid by using an aseptic coating rod, drying the bacterial liquid, placing sterilized oxford cups in the plate by using a pair of tweezers, slightly pressurizing the plate to enable the plate to be in seamless contact with a culture medium, placing 4 oxford cups in each plate, adding the same abrotanum extract into 3 oxford cups to serve as a test group, adding the nutrient broth with the same volume in the other oxford cup to serve as a negative control, and simultaneously using norfloxacin as a positive control. And (3) placing the flat plate in an incubator at 37 ℃ for static culture for 24h, measuring the bacteriostatic diameter of the flat plate by using a vernier caliper according to a cross method, and judging the bacteriostatic effect.
The judgment standard of the antibacterial effect is according to the standard of traditional Chinese medicine pharmacology, namely: the bacteriostatic diameter is more than or equal to 20mm, the hypersensitivity is 15-19mm, the medium hypersensitivity is 10-14mm, and the hypohypersensitivity is below 10 mm.

Note: indicates high sensitivity (15-19mm), medium sensitivity (10-14mm), low sensitivity (less than 10mm), no inhibitory effect
As can be seen from tables 1, 2, 3 and figures 1-8, the four ultrasonic extraction methods of the arabidopsis thaliana have an inhibiting effect on staphylococcus aureus, and only the first two extraction methods of the reflux extraction method have an antibacterial effect, which indicates that the ultrasonic extraction method is superior to the reflux extraction method; and the reflux extraction methods SJ-1 and SJ-2 and the ultrasonic extraction methods SJ-5 and SJ-7 are superior to other methods.
The above-mentioned embodiments are merely embodiments for expressing the invention, and the description is specific and detailed, but not construed as limiting the scope of the invention. It should be noted that, for those skilled in the art, various changes, substitutions of equivalents, improvements and the like can be made without departing from the spirit of the invention, and these are all within the scope of the invention. Therefore, the protection scope of the present patent shall be subject to the appended claims.
