Abstract
本申请公开了一种具有解酒保肝功能的组合物及其制备方法和应用,具体涉及药品、保健品和食品技术领域。具有解酒保肝功能的组合物,以重量计包括枳椇子300‑800份、葛根100‑500份、人参50‑150份、高良姜5‑20份、玉米低聚肽20‑70份、姜黄10‑50份、维生素C1‑20份。同时本申请还进一步提供了组合物的制备方法,包括以下步骤:将葛根、枳椇子、人参、高良姜用水提取,得提取液;将所述提取液真空浓缩后,得浓缩液;将所述浓缩液减压干燥后,得干浸膏;再进行粉碎,过筛。本申请提供的组合物可通过加快酒精的代谢达到醒酒和解酒的效果,从而预防或减少酒精引起的肝损伤。
Description
技术领域
本申请涉及药品、保健品和食品技术领域,具体而言,涉及一种具有解酒保肝功能的组合物及其制备方法和应用。
背景技术
饮酒在我国有悠久的历史,酒文化已渗透到人们的生活和工作中。比烟的关注度与需求度还要高;自古就有无酒不成席,酒的种类很多,饮酒是一种文化,更是一种交流与沟通;在市场竞争激烈的环境下,喝酒是很多商务人士必选的交流手段之一。然而,长期饮酒,尤其是白酒,会造成身体各内脏器官的损害,加重肝损伤,因为酒精或可使肝脏积累越来越多的脂肪,这些脂肪会慢慢导致炎症。当饮酒者出现肝损害症状时,往往超过3/4的肝细胞已失去了正常功能,严重的会引起肝纤维化、肝硬化甚至原发性肝癌。近年来,我国酒精消费群体日益庞大,由此引起的酒精性肝病(ALD)亦成为社会关注的焦点。我国人口众多。因此,研制醒酒、解酒,减轻和预防酒精造成的肝损伤的并具有高安全性的产品,具有重要的意义。
发明内容
本申请目的之一在于提供一种具有解酒保肝功能的组合物,该组合物所用的原料均为传统医学领域的药食同源原料,加上现代理论研究的原料或其它形态等,所述组合物按照重量份计算,包括:枳椇子300-800份、葛根100-500份、人参50-150份、高良姜5-20份、玉米低聚肽20-70份、姜黄10-50份、维生素C1-20份。
本申请另一实施方式中,所述组合物按照重量份计算,包括:枳椇子500-700份、葛根200-400份、人参70-120份、高良姜5-15份、玉米低聚肽30-50份、姜黄15-40份、维生素C5-15份。
本申请另一实施方式中,所述组合物按照重量份计算,包括:枳椇子600份、葛根300份、人参90份、高良姜10份、玉米低聚肽45份、姜黄30份、维生素C12份。
本申请还提出了如前任一项组合物在制备具有解酒保肝功能的药物、保健品或食品中的应用。所述解酒保肝功能例如,可以是预防或减少酒精引起的肝损伤。
本申请还提出了一种含有如前任一项具有解酒保肝功能的组合物的药物、保健品或食品。
本申请另一具体实施方式中,该药物、保健品或食品选自压片糖果、片剂、硬胶囊、软胶囊剂、颗粒剂、糖浆、酒剂、口服液。
本申请另一具体实施方式中,该药物、保健品或食品包含药学或食品上可接受的辅料或添加剂;例如可以是,赤藓糖醇、山梨糖醇、微晶纤维素、草莓果粉、麦芽糊精、甜菊糖苷、硬脂酸镁。
本申请还提出了一种药物、保健品或食品的制备方法,所述药物、保健品或食品选自压片糖果时,其特征在于,所述制备方法包括以下步骤:
称量前述比例的葛根、枳椇子、人参、高良姜,加10倍水,提取(可选地提取时间为1h,提取次数为2次)后60~70℃浓缩(至密度为1.1~1.2g/mL),干燥,粉碎后过筛;
将过筛后的物料与玉米低聚肽、山梨糖醇、姜黄、维生素C混合均匀,混合时间可选为20min;
将混合粉搅拌后,用酒精进行湿法制粒后干燥;干燥温度可选为60℃,干燥时间为50~70分钟,干燥至湿颗粒水分在5%以下;
整粒和总混:将干燥后的颗粒整粒,将硬脂酸镁与整粒颗粒混合,混合时间可选为25~30分钟;
压片,获得压片糖果。
目前市面上少有解酒保肝脏作用的药食同源中药加现代原料的组方。而本申请提出了一种具有解酒保肝功能的组合物,其中的主要原料中,葛根,辛,凉,解肌退热,生津,透疹,升阳止泻,葛根解酒毒,止渴止呕,为君药;枳椇子能解酒毒,润五脏,为臣药,葛根与枳椇子相互配伍,消湿热,清升浊降,解酒毒,生津解渴、利尿除湿,促进肝脏排毒,促进体内的酒精的排泄和代谢,消除体内残余的酒精,解酒毒,从而达到醒酒的目的。高良姜温胃散寒,止呕吐,人参大补元气,补脾益肺,生津,调中,止消渴,通血脉,破坚积,泻心火,高良姜和人参两味药材为佐药,共同加速血液中酒精代谢,达到快速醒酒目的。另外,玉米低聚肽作为营养强化剂能缩短醉酒时间,降低血液中乙醇和乙醛含量,食品添加剂姜黄素可明显改善小鼠醉酒后的行为学指标,一定程度上降低血中乙醇和乙醛浓度。玉米低聚肽和姜黄素均可增强肝组织乙醇脱氢酶ADH和乙醛脱氢酶ALDH活性,提高细胞色素CYP450、过氧化氢酶CAT和超氧化物歧化酶SOD含量,降低黄嘌呤氧化酶XOD活性。玉米低聚肽和姜黄素联合应用时对醉酒小鼠的活性强于单独使用姜黄素,但与单独使用玉米低聚肽组无显著差异.在该实验条件下,玉米低聚肽和姜黄素对醉酒小鼠具有明显的醒酒活性。维生素C给药组小鼠血清中谷草转氨酶AST,谷丙转氨酶ALT水平下降,肝脏丙二醛MDA水平下降,SOD活性上升,肝脂肪变性减轻。因此辅以姜黄、玉米低聚肽、维生素C,降低酒精对肝脏的损伤,对酒精性肝损伤有明显的保护作用。
本申请所提供的解酒护肝的压片糖果,能通过加快酒精的代谢达到醒酒和解酒的效果,且其还能改善酒精导致的新陈代谢的异常、调节人体机能,具有清热解毒、生津止渴和健脾胃护肝的效果,从而降低了酒精性肝损伤的发生。

制备例
提取:葛根300份、枳椇子600份、人参90份、高良姜10份,加10倍量水,100±5℃条件下提取2次,每次1小时,获提取液;
浓缩:所述提取液采用真空浓缩至60℃下相对密度为1.10-1.20,过滤,得浓缩液备用;
干燥:将所述浸膏平铺于托盘中,70℃下采用减压干燥的方式干燥,得到干浸膏;
粉碎:采用万能粉碎机粉碎,过80目,得干膏粉备用;
混合:将所述干膏粉、玉米低聚肽、山梨糖醇、姜黄、维生素C混合均匀,得混匀粉末备用;
制粒:所述混匀粉末中加入95%乙醇制备软材,用20目筛进行制粒,60℃烘干,用20目筛进行整粒,得颗粒;
压片:将所得颗粒加入硬脂酸镁混合,进行压片,得片剂。
本申请组合物的药效考察
1.材料
山东大学实验动物中心提供的健康SPF级昆明种小鼠,体重18-22g,实验动物生产许可证号为SCXK(鲁)20130009;实验动物使用合格证号SYXK(鲁)20150015号,实验动物饲料生产许可证SCXK(京)2014-0008。
丙二醛(MDA)试剂盒、还原型谷胱甘肽(GSH)试剂盒购自南京建成生物工程研究所;无水乙醇(分析纯),国药集团化学试剂有限公司生产;甘油三酯(TG)试剂盒由中生北控生物科技股份有限公司提供。上海垒固JA5003A分析天平,上海精天JT1001N电子天平,OLYMPUS BX53倒置显微镜,Tecan 200多功能酶标仪,HM525 Thermo冰冻切片机。
2.解酒保肝实验
雄性昆明小鼠50只,适应性喂养3天后,随机分为五组,每组10只。按每人每日药物摄入量(64mg/kg.bw)的5倍、10倍、30倍分别设低、中、高三个剂量组,即320mg/kg.bw、640mg/kg.bw、1920mg/kg.bw,同时设阴性对照组及乙醇阳性对照组。分别称取制备好的组合物样品1.6g、3.2g、9.6g,用蒸馏水溶解并定容至100ml。每日一次经口灌胃给予各剂量组动物,灌胃量0.2ml/l0g.bw,阴性对照组及阳性对照组给予等体积蒸馏水。连续灌胃30天,每4天称1次体重,按体重变化调整给药量。第30天三个剂量组及阳性对照组小鼠均灌胃50%(V/V)乙醇0.14ml/10g.bw,禁食18小时后处死动物,取适量肝组织制成10%(W/V)的肝匀浆,用试剂盒测定MDA(过氧化物酶)、GSH(谷胱甘肽)和TG(甘油三酯)的含量。同时从肝脏左叶中部做横切面取材,制作冰冻切片,苏丹Ⅲ染色,苏木素复染细胞核,封片,观察脂滴在肝脏的分布范围和面积。
3.实验结果
3.1组合物对小鼠体重的影响
如表1所示,各组动物试验期内体重持续增长,各组之间无统计学差异(P>0.05)。
3.1组合物对小鼠肝脏中丙二醛(MDA)含量的影响
由表2结果可见,阳性对照组与阴性对照组相比,肝组织中MDA含量明显升高,有统计学差异(P<0.05);与阳性对照组相比,高剂量组MDA含量显著降低并具有统计学差异(P<0.05)。

3.3组合物对小鼠肝脏谷胱甘肽(GSH)含量的影响
由表3结果可见,阳性对照组与阴性对照组相比,肝组织中GSH含量明显降低,有统计学差异(P<0.05);与阳性对照组相比,高剂量组GSH含量明显升高,有统计学差异(P<0.05)。

3.4组合物对小鼠肝脏中甘油三酯(TG)含量的影响
由表4结果可见,阳性对照组与阴性对照组比较,肝脏TG含量明显升高,有统计学差异(P<0.05)。与阳性对照组相比,高剂量组TG含量均显著降低并具有统计学差异(P<0.05)。

3.5肝脏病理组织学检查
从肝脏左叶中部做横切面取材,制作冰冻切片,苏丹Ⅲ染色,苏木素复染细胞核,甘油明胶封片,观察脂滴在肝脏的分布范围和面积。①阴性对照组:肝脏基本正常;②阳性对照组:全部动物出现肝细胞脂肪变性,与阴性对照组比较,有统计学差异(P<0.05);③高剂量组与阳性对照组相比,肝细胞脂肪变性程度明显减轻,脂肪变性评分明显减少,有统计学差异(P<0.05)。

肝细胞脂肪变性评分标准:肝细胞内脂滴散在,稀少。0分
含脂滴的肝细胞不超过1/4。1分

4小结
组合物高剂量组的肝组织MDA含量低于阳性对照组(P<0.05),高剂量组TG含量低于阳性对照组(P<0.05),高剂量组GSH含量高于阳性对照组(P<0.05);病理检查显示高剂量组肝细胞脂肪变性比阳性对照组明显减轻(P<0.05)。根据《保健食品检验与评价技术规范》判定组合物对化学性肝损伤有辅助保护功能。

1.一种具有解酒保肝功能的组合物,其特征在于,以重量计,所述组合物包括枳椇子300-800份、葛根100-500份、人参50-150份、高良姜5-20份、玉米低聚肽20-70份、姜黄10-50份、维生素C 1-20份。
2.根据权利要求1所述的组合物,其特征在于,所述组合物包括枳椇子500-700份、葛根200-400份、人参70-120份、高良姜5-15份、玉米低聚肽30-50份、姜黄15-40份、维生素C 5-15份。
3.根据权利要求1所述的组合物,其特征在于,所述组合物包括枳椇子600份、葛根300份、人参90份、高良姜10份、玉米低聚肽45份、姜黄30份、维生素C 12份。
4.权利要求1-3任一项组合物在制备具有解酒保肝功能的药物、保健品或食品中的应用。
5.一种含有权利要求1-3任一项具有解酒保肝功能的组合物的药物、保健品或食品。
6.根据权利要求5所述的药物、保健品或食品,其特征在于,该药物、保健品或食品选自口服、注射或外用剂型。
7.根据权利要求6所述的药物、保健品或食品,其特征在于,该药物、保健品或食品选自压片糖果、片剂、硬胶囊、软胶囊剂、颗粒剂、糖浆、酒剂、口服液。
8.一种如权利要求1-3任一项所述的组合物的制备方法,其特征在于,包括以下步骤:
提取:葛根、枳椇子、人参、高良姜用水提取,得提取液;
浓缩:所述提取液真空浓缩后,得浓缩液;
干燥:所述浓缩液减压干燥后,得中间体。
9.一种如权利要求8所述的制备方法,其特征在于,包括以下步骤:
提取:葛根、枳椇子、人参、高良姜加8-12倍水,100±5℃条件下提取1-3次得提取液;
浓缩:所述提取液采用真空浓缩至60-70℃下相对密度为1.10-1.20,过滤,得浓缩液;
干燥:将所述浓缩液减压干燥,得到干浸膏;
粉碎:粉碎,过筛。
10.一种具有解酒保肝功能的药物、保健品或食品的制备方法,其特征在于,包括以下步骤:
提取:葛根、枳椇子、人参、高良姜,加10倍量水提取2次,每次1小时,获提取液;
浓缩:所述提取液采用真空浓缩至60℃下相对密度为1.10-1.20,过滤,得浓缩液备用;
