Abstract
The invention discloses a compound Tibetan schizonepeta composition with skin brightening, anti-allergy and relieving effects and a preparation method thereof, wherein the compound Tibetan schizonepeta composition is prepared from the following raw materials in parts by weight: 10-50 parts of Tibetan schizonepeta, 10-50 parts of corydalis tuber, 5-30 parts of tribulus fruit and 5-30 parts of myrobalan. The invention also comprises a preparation method and application of the composition. The composition disclosed by the invention has the functions of brightening the skin and resisting allergy and relieving when being used in a skin care product, and has milder skin effect, no irritation and higher safety.
Description
Technical Field
The invention relates to the technical field of fine chemical and daily necessities, in particular to a compound Tibetan schizonepeta composition with skin brightening, anti-allergy and relieving effects and a preparation method thereof.
Background
Skin color, which is the color of the skin, is the color of the epidermis of the human skin as reflected by pigmentation of the epidermis of the human skin, such as melanin, protopanarubicin, and phyllanthin. Skin color is distributed differently in different areas and populations. Skin color is related to the content and distribution (granular or dispersed) of melanin in the skin. Melanin is concentrated in and among the cells of the germinal layer of the epidermis. In order to achieve the purpose, the technical scheme adopted by the invention for solving the technical problems is as follows: the compound Tibetan schizonepeta composition with the effects of brightening skin and resisting allergy and relieving is provided, and is prepared from the following raw materials in parts by weight: 10-50 parts of Tibetan schizonepeta, 10-50 parts of corydalis tuber, 5-30 parts of tribulus fruit and 5-30 parts of myrobalan.
Further, the compound Tibetan schizonepeta composition with the effects of brightening skin and resisting allergy and relieving allergy is prepared from the following raw materials in parts by weight: 20-40 parts of Tibetan schizonepeta, 20-40 parts of corydalis tuber, 10-20 parts of tribulus fruit and 10-20 parts of myrobalan.
Further, the compound Tibetan schizonepeta composition with the effects of brightening skin and resisting allergy and relieving allergy is prepared from the following raw materials in parts by weight: 40 parts of Tibetan schizonepeta, 30 parts of corydalis tuber, 15 parts of tribulus fruit and 15 parts of myrobalan.
The herba Schizonepetae is whole plant of Nepeta angustifolia C.Y.Wu of Labiatae. Has effects in calming endogenous wind, relieving spasm, relieving swelling, and relieving pain, and can be used for treating epilepsy, wound swelling and pain, apoplexy, hemorrhagic cerebral infarction, wound, and pain, and contains nepetalactone, ursolic acid, oleanolic acid, and betulinic acid.
Rhizoma Corydalis is dried tuber of Corydalis yanhusuo W.T.Wang of Papaveraceae. Has warm nature and pungent and bitter taste, has effects of promoting blood circulation, activating qi-flowing, and relieving pain, and can be used for treating chest and hypochondrium pain, abdominal pain, thoracic obstruction, cardialgia, amenorrhea, dysmenorrhea, puerperal blood stasis, and traumatic injury with swelling and pain, and contains alkaloids such as corydaline and tetrahydropalmatine.
The fructus Tribuli fruit is dried mature fruit of Tribuli terrestris L. Has mild warm nature and pungent and bitter taste, has effects of suppressing hyperactive liver, resolving stagnation, promoting blood circulation, dispelling pathogenic wind, improving eyesight, and relieving itching, and can be used for treating headache, giddiness, chest and hypochondrium distending pain, breast closure, acute mastitis, conjunctival congestion, nebula, rubella, pruritus, and saponin components such as dioscin and lucidus stage.
The fructus Chebulae is dry mature fruit of Terminalia chebula Retz of Combretaceae or Terminalia villosa Retz. var. tomentella Kurt. Neutral in nature, bitter, sour and astringent in taste, has the effects of astringing intestines to stop diarrhea, astringing lung to relieve cough, lowering fire and relieving sore throat, is used for treating chronic diarrhea and dysentery, hematochezia and rectocele, lung deficiency and cough with dyspnea, chronic cough, pharyngalgia and hoarseness, and contains polyphenols such as myrobalic acid, chebulagic acid, gallic acid and the like.
The preparation method of the compound Tibetan schizonepeta composition with the effects of brightening skin and resisting allergy and relieving allergy comprises the following steps:
(1) pulverizing dried Tibetan herba Schizonepetae and rhizoma corydalis, sieving with 20 mesh sieve, mixing, adding 5-15 times of 30-80 vt% ethanol according to volume and mass ratio, reflux extracting for 1-3 times, each time for 1-3 hr, filtering, mixing filtrates, and concentrating under reduced pressure at 55-75 deg.C to obtain fluid extract; then adding water into the clear paste to disperse to 0.05-0.5g/mL, eluting by macroporous adsorption resin, collecting eluent, and drying under reduced pressure to obtain a composition A;
(2) pulverizing dried fructus Tribuli and fructus Chebulae, sieving with 20 mesh sieve, mixing, adding 10-30 times of water according to volume-to-mass ratio, reflux extracting for 1-3 times (1-3 hr each time), filtering, mixing filtrates, and concentrating to obtain fluid extract; then adding water into the clear paste to disperse to 0.05-0.5g/mL, eluting by macroporous adsorption resin, collecting eluent, and drying under reduced pressure to obtain a composition B;
(3) and (3) uniformly mixing the composition A obtained in the step (1) and the composition B obtained in the step (2) to obtain the compound Tibetan schizonepeta composition with the effects of brightening the skin, resisting allergy and relieving.
Further, in the steps (1) and (2), when the macroporous adsorption resin is eluted, the ratio of the diameter to the height of the resin is 1:3-8, the loading amount is 0.1-1g/mL calculated by the amount of the whole formula crude drug, the adsorption flow rate is 1-4BV/h, the resin is eluted by water for removing impurities by 2-10 times of the volume of the resin, the resin is eluted by 30-80% of ethanol for 2-8 times of the volume of the resin, and the elution flow rate is 2-8 BV/h.
Further, in the steps (1) and (2), the macroporous adsorption resin is AB-8, D101, HPD-100, HPD-300 or HPD-5000.
The compound Tibetan schizonepeta composition with the skin brightening, anti-allergy and relieving effects is applied to preparing skin care products with the skin brightening, anti-allergy and relieving effects.
Furthermore, the compound Tibetan herba schizonepetae composition with the effects of brightening skin and resisting allergy and relieving is added in an amount of 0.1-10%.
Furthermore, the compound Tibetan herba schizonepetae composition with the effects of brightening skin and resisting allergy and relieving is added in an amount of 1-8%.
Further, the skin care product is a face wash, a face cream, a soap, a cream, an emulsion, water, a mask or a gel.

Example 1
A compound Tibetan herba schizonepetae composition with skin brightening, anti-allergy and relieving effects is prepared from the following raw materials in parts by weight: 40 parts of Tibetan schizonepeta, 30 parts of corydalis tuber, 15 parts of tribulus fruit and 15 parts of myrobalan.
The preparation method of the compound Tibetan schizonepeta composition with the effects of brightening skin and resisting allergy and relieving allergy comprises the following steps:
(1) pulverizing dried Tibetan herba Schizonepetae and rhizoma corydalis, sieving with 20 mesh sieve, mixing, adding 10 times of 50 vt% ethanol according to volume and mass ratio, reflux extracting for 2 times (2 hr each time), filtering, mixing filtrates, and concentrating under reduced pressure at 75 deg.C to obtain fluid extract; then adding water into the clear paste to disperse to 0.2g/mL, eluting by macroporous adsorption resin, collecting eluent, and drying under reduced pressure to obtain a composition A;
(2) pulverizing dried fructus Tribuli and fructus Chebulae, sieving with 20 mesh sieve, mixing, adding 15 times of water according to volume-mass ratio, reflux-extracting for 2 times (2 hr each time), filtering, mixing filtrates, and concentrating to obtain fluid extract; then adding water into the clear paste to disperse to 0.5g/mL, eluting by macroporous adsorption resin, collecting eluent, and drying under reduced pressure to obtain a composition B;
(3) and (3) uniformly mixing the composition A obtained in the step (1) and the composition B obtained in the step (2) to obtain the compound Tibetan schizonepeta composition with the effects of brightening the skin, resisting allergy and relieving.
In the steps (1) and (2), when the macroporous adsorption resin is eluted, HPD-100 type macroporous adsorption resin is adopted, the diameter-height ratio of the resin is 1:6, the loading amount is 0.5g/mL based on the amount of the whole formula crude drug, the adsorption flow rate is 2BV/h, the resin is eluted by water by 8 times of the volume of the resin to remove impurities, 50% ethanol is used for eluting by 5 times of the volume of the resin, and the elution flow rate is 2 BV/h.
Example 2
A compound Tibetan herba schizonepetae composition with skin brightening, anti-allergy and relieving effects is prepared from the following raw materials in parts by weight: 30 parts of Tibetan schizonepeta, 30 parts of corydalis tuber, 20 parts of tribulus fruit and 20 parts of myrobalan.
The preparation method of the compound Tibetan schizonepeta composition with the effects of brightening skin and resisting allergy and relieving allergy comprises the following steps:
(1) pulverizing dried Tibetan herba Schizonepetae and rhizoma corydalis, sieving with 20 mesh sieve, mixing, adding 5 times of 80 vt% ethanol according to volume and mass ratio, reflux extracting for 3 times, each time for 1 hr, filtering, mixing filtrates, and concentrating under reduced pressure at 65 deg.C to obtain fluid extract; then adding water into the clear paste to disperse to 0.5g/mL, eluting by macroporous adsorption resin, collecting eluent, and drying under reduced pressure to obtain a composition A;
(2) pulverizing dried fructus Tribuli and fructus Chebulae, sieving with 20 mesh sieve, mixing, adding 30 times of water according to volume-mass ratio, reflux extracting for 1 time, each time for 3 hr, filtering, mixing filtrates, and concentrating to obtain fluid extract; then adding water into the clear paste to disperse to 0.1g/mL, eluting by macroporous adsorption resin, collecting eluent, and drying under reduced pressure to obtain a composition B;
(3) and (3) uniformly mixing the composition A obtained in the step (1) and the composition B obtained in the step (2) to obtain the compound Tibetan schizonepeta composition with the effects of brightening the skin, resisting allergy and relieving.
In the steps (1) and (2), D-101 type macroporous adsorption resin is adopted when the macroporous adsorption resin is eluted, the ratio of the diameter to the height of the resin is 1:8, the loading amount is 1g/mL based on the amount of the whole formula crude drugs, the adsorption flow rate is 4BV/h, the resin is eluted by water by 5 times of the volume of the resin to remove impurities, the resin is eluted by 380 percent ethanol by 8 times of the volume of the resin, and the elution flow rate is 8 BV/h.
Example 3
A compound Tibetan herba schizonepetae composition with skin brightening, anti-allergy and relieving effects is prepared from the following raw materials in parts by weight: 40 parts of Tibetan schizonepeta, 40 parts of corydalis tuber, 10 parts of tribulus fruit and 10 parts of myrobalan.

(3) and (3) uniformly mixing the composition A obtained in the step (1) and the composition B obtained in the step (2) to obtain the compound Tibetan schizonepeta composition with the effects of brightening the skin, resisting allergy and relieving.
In the steps (1) and (2), when the macroporous adsorption resin is eluted, HPD-300 type macroporous adsorption resin is adopted, the diameter-height ratio of the resin is 1:3, the loading amount is 0.1g/mL based on the amount of the whole formula crude drug, the adsorption flow rate is 3BV/h, 6 times of the volume of the resin is eluted by water to remove impurities, 8 times of the volume of the resin is eluted by 30 percent ethanol, and the elution flow rate is 5 BV/h.

The preparation method of the compound Tibetan schizonepeta composition with the effects of brightening skin and resisting allergy and relieving allergy comprises the following steps:

(3) and (3) uniformly mixing the composition A obtained in the step (1) and the composition B obtained in the step (2) to obtain the compound Tibetan schizonepeta composition with the effects of brightening the skin, resisting allergy and relieving.

(3) and (3) uniformly mixing the composition A obtained in the step (1) and the composition B obtained in the step (2) to obtain the compound Tibetan schizonepeta composition with the effects of brightening the skin, resisting allergy and relieving.
In the steps (1) and (2), when the macroporous adsorption resin is eluted, AB-8 type macroporous adsorption resin is adopted, the ratio of the diameter to the height of the resin is 1:6, the loading amount is 0.8g/mL based on the amount of the whole formula crude drugs, the adsorption flow rate is 4BV/h, the resin is eluted by 7 times of the volume of water to remove impurities, the resin is eluted by 6 times of the volume of 40% ethanol, and the elution flow rate is 3 BV/h.

Examples of the experiments
The compositions obtained in example 1 and comparative examples 1 to 6 were used to prepare corresponding skin creams according to the formulation shown in Table 1.

1. Comparative experiment for skin color brightening effect
(1) A subject: selecting 140 subjects, female, 20-45 years old and indoor workers, wherein the exclusion conditions of the subjects meet the inclusion and exclusion standards of the cosmetic contact dermatitis diagnosis standard and treatment principle; no serious systemic disease, no immunodeficiency or autoimmune disease; an inactive allergic disease; no allergic history to skin care cosmetics; hormone medicine and immunosuppressant have not been used systemically in 1 month, and are not in gestation or lactation period; no ethical contraindication; skin color was tested between II-IV using healthy subjects according to Chardon's grouping. The subjects were randomly divided into 14 groups of 10 persons each, and skin color lightening effect comparison experiments were performed using the skin cream prepared by the composition of example 1 and the above comparative example.
(2) Testing indexes are as follows: skin whiteness (brightness) (L)*Value): l is*The value is white balance, and the color is more white when the value is larger, and is more black when the value is smaller.
Skin brightness (dark yellowness) (ITA ° value): ITA DEG value is the skin individual type angle, is equal to L*Value b and*value-dependent values characterizing skin brightness. The greater the ITA value, the brighter the skin, whereas the darker the skin.
The test method comprises the following steps: selecting the inner sides of the left arm crank and the right arm crank of the testee as tested areas (with the same area), randomly marking, setting blank areas (without using any sample), smearing experimental samples on the tested areas for 1 time in the morning and at night every day, and smearing the experimental samples according to personal use habits. The use period was 8 weeks. The subjects did not change the brand and habit of daily use of the cleaning product during the test. After 8 weeks of use of the test product, data were collected from the test site using a test instrument (Mexameter MX 18 skin melanin and heme tester, MPA9 multifunctional skin tester and colorimetric test probe, CK company, germany).
And (3) data analysis: if the data are normally distributed, the variation of the data before and after the use of the sample and the difference between the test area and the matrix area are analyzed by using the t-test. If the data are distributed in a non-normal way, corresponding statistical analysis is carried out by applying rank sum test, and a conclusion whether the data are effective or not is obtained according to a calculation result. The data calculation formulas are shown in formula (1) and formula (2).

the results in table 3 show that after 8 weeks using the product of example 1 according to the invention, the value of Δ ITA ° was also significantly higher for the product of example 1 according to the invention than for the other comparative products (P <0.01) in comparison with the Δ ITA ° values of the blank zone of the same subjects. The formula of the invention has obvious synergistic effect of increasing skin brightness, and compared with a comparative product containing only one, two or three raw materials, the skin color brightening effect is obvious, and the formula has definite synergistic effect.
2. Anti-allergic and antipruritic test
After 1 week of acclimatization of 160 mice, they were randomly divided into 16 groups: blank control group, histamine group, inventive example 1, comparative examples 1-13, 10 per group. In the experiment, 1% sodium sulfide is used for removing the hair of the mouse back, the area of the skin area for removing the hair is 2cm multiplied by 3cm, and the mouse is used for the experiment after 2 days. In the experiment, the back hairless area of each group of animals is locally coated with corresponding samples (0.2 ml/mouse) for 5 days continuously, and the neck and the back of each group of mice are injected with 50 mu L/mouse intracutaneously after 1h of the last administration except for a blank control group. After histamine administration, mice were observed for itching and the number of scratching was recorded within 30 min. The test data were statistically analyzed using SPSS 22.0 statistical software, and the statistics were expressed as mean. + -. standard deviation, and if the data were normally distributed, the variation of the data before and after the use of the sample and the difference between the test area and the matrix area were analyzed using the t-test method. If the data are distributed in a non-normal way, corresponding statistical analysis is carried out by applying rank sum test, and a conclusion whether the data are effective or not is obtained according to a calculation result. The results of the anti-allergic and antipruritic tests are shown in Table 4.

The formula of the invention has obvious synergistic anti-allergy and relieving effects, and compared with a comparative product containing only one, two or three raw materials, the formula of the invention has obvious anti-allergy and itching relieving effects and has definite synergistic effect.

Claims (10)
1. The compound Tibetan schizonepeta composition with the effects of brightening skin and resisting allergy and relieving is characterized by being prepared from the following raw materials in parts by weight: 10-50 parts of Tibetan schizonepeta, 10-50 parts of corydalis tuber, 5-30 parts of tribulus fruit and 5-30 parts of myrobalan.
2. The compound Tibetan catnip composition with skin lightening and anti-allergy relieving effects as claimed in claim 1, which is prepared from the following raw materials in parts by weight: 20-40 parts of Tibetan schizonepeta, 20-40 parts of corydalis tuber, 10-20 parts of tribulus fruit and 10-20 parts of myrobalan.
3. The compound Tibetan catnip composition with skin lightening and anti-allergy relieving effects as claimed in claim 1, which is prepared from the following raw materials in parts by weight: 40 parts of Tibetan schizonepeta, 30 parts of corydalis tuber, 15 parts of tribulus fruit and 15 parts of myrobalan.
4. A process for preparing a compound nepeta cataria composition for skin lightening and anti-allergy relief as claimed in any one of claims 1-3, comprising the steps of:
(1) pulverizing dried Tibetan herba Schizonepetae and rhizoma corydalis, sieving with 20 mesh sieve, mixing, adding 5-15 times of 30-80 vt% ethanol according to volume and mass ratio, reflux extracting for 1-3 times, each time for 1-3 hr, filtering, mixing filtrates, and concentrating under reduced pressure at 55-75 deg.C to obtain fluid extract; then adding water into the clear paste to disperse to 0.05-0.5g/mL, eluting by macroporous adsorption resin, collecting eluent, and drying under reduced pressure to obtain a composition A;
(2) pulverizing dried fructus Tribuli and fructus Chebulae, sieving with 20 mesh sieve, mixing, adding 10-30 times of water according to volume-to-mass ratio, reflux extracting for 1-3 times (1-3 hr each time), filtering, mixing filtrates, and concentrating to obtain fluid extract; then adding water into the clear paste to disperse to 0.05-0.5g/mL, eluting by macroporous adsorption resin, collecting eluent, and drying under reduced pressure to obtain a composition B;
(3) and (3) uniformly mixing the composition A obtained in the step (1) and the composition B obtained in the step (2) to obtain the compound Tibetan schizonepeta composition with the effects of brightening the skin, resisting allergy and relieving.
5. The method for preparing the compound Tibetan herba schizonepetae composition with the effects of brightening skin and resisting allergy and relieving pain as claimed in claim 4, wherein in the steps (1) and (2), when macroporous adsorption resin is eluted, the diameter-height ratio of the resin is 1:3-8, the loading amount is 0.1-1g/mL calculated by the amount of the whole formula crude drugs, the adsorption flow rate is 1-4BV/h, the resin volume which is 2-10 times of that of water elution is used for removing impurities, the resin volume which is 2-8 times of that of 30-80% ethanol elution is used for removing impurities, and the elution flow rate is 2-8 BV/h.
6. The method of claim 4, wherein the macroporous adsorbent resin of steps (1) and (2) is AB-8, D101, HPD-100, HPD-300, or HPD-5000.
7. Use of the compound Tibetan catnip composition having skin lightening and anti-allergy soothing effects as claimed in any one of claims 1 to 3 in the preparation of a skin lightening and anti-allergy soothing skin care product.
8. The use of claim 7, wherein the compound nepeta cataria composition is added in an amount of 0.1% to 10% for skin lightening and anti-allergy relief.
9. The use of claim 7, wherein the compound nepeta cataria composition is added in an amount of 1-8% for skin lightening and anti-allergy relief.
10. The use of claim 7, wherein the skin care product is a facial cleanser, facial cream, soap, cream, lotion, water, mask, or gel.
