Abstract
本发明提供一种重组溶瘤基因腺病毒及其构建方法与应用。所述的重组溶瘤基因腺病毒的启动子包括肝癌特异性启动子,所述的重组溶瘤基因腺病毒包括IL‑24的编码核苷酸序列。本发明提供一种重组溶瘤基因腺病毒具有高效的癌症特异性,用其构建获得的重组溶瘤腺病毒对正常细胞的毒性低、杀伤癌细胞效果好,且与BRD4抑制剂联用具有协同作用,抗癌效果优于一般的重组溶瘤腺病毒。

技术领域
本发明属于重组溶瘤基因-腺病毒技术领域,具体涉及一种重组溶瘤基因腺病毒及其构建方法与应用。
背景技术
癌症严重影响人类的健康和发展,受医疗和环境条件的影响,中国癌症死亡率高于全球平均水平。肿瘤的传统疗法存在疗效差,死亡率高及预后复发率高等缺点,因此人们开始对新兴的肿瘤治疗方法寄予厚望。新兴的治疗肿瘤的策略也在不断的发展之中,目前比较被认同的策略包括免疫疗法,基因疗法,溶瘤病毒疗法等。目前的各种基因载体并不能够在人体内特异性选择肿瘤细胞,也不能在肿瘤细胞中高效表达抗癌基因,从而导致基因治疗的效果较弱和误伤人体正常细胞的可能性。仅仅使用基因治疗或者特异性靶向肿瘤的病毒治疗癌症,其效果都十分有限。
因此,有必要提供改进的技术方案以克服现有技术中存在的技术问题。
发明内容
为解决现有技术存在的问题,本发明提供一种重组溶瘤基因腺病毒及其构建方法与应用,所述的重组溶瘤基因腺病毒具有高效的癌症特异性,用其构建获得的重组溶瘤腺病毒对正常细胞的毒性低、杀伤癌细胞效果好,且与BRD4抑制剂联用具有协同作用,抗癌效果优于一般的重组溶瘤腺病毒。
本发明第一方面提供一种重组溶瘤基因腺病毒,所述的重组溶瘤基因腺病毒的启动子包括肝癌特异性启动子,所述的重组溶瘤基因腺病毒包括IL-24的编码核苷酸序列。通过该技术步骤,得到的重组溶瘤基因腺病毒对正常细胞的毒性低、杀伤肝癌细胞效果好,靶向抗癌效果优于一般的重组溶瘤腺病毒。
优选地,所述的重组溶瘤基因腺病毒的启动子包括P28GANK启动子,所述的重组溶瘤基因腺病毒的基因组敲除了E1B基因区域。通过该技术步骤的处理,增强了重组溶瘤基因腺病毒的肝癌靶向性,提高了重组溶瘤基因腺病毒的外源基因装载容量。
本发明第二方面提供一种重组溶瘤基因腺病毒的构建方法,所述的构建方法包括以下步骤:
步骤一、重组小质粒的构建:以含IL-24表达框的质粒为模板进行PCR,克隆得到IL-24的表达框;再以限制性内切酶单酶切pShuttle-P28-E1A-ΔE1B小质粒并进行去磷处理,得到线性化并去磷的pShuttle-P28-E1A-ΔE1B小质粒,连接所述IL-24的表达框和线性化并去磷的pShuttle-P28-E1A-ΔE1B小质粒,得到重组小质粒pShuttle-P28-E1A-ΔE1B-IL-24;
步骤二、重组大质粒的构建:将步骤一构建的所述pShuttle-P28-E1A-ΔE1B-IL-24小质粒用限制性内切酶线性化,去磷后转入带有腺病毒骨架质粒的大肠杆菌感受态中,使二者发生同源重组,筛选获得阳性克隆,得到重组大质粒;
步骤三、重组溶瘤腺病毒的包装:将步骤二得到的重组大质粒使用限制性内切酶线性化,转染细胞,转染时间不少于3天,收取病毒液。
发明人通过上述技术步骤,构建了包括IL-24的编码核苷酸序列的重组溶瘤基因腺病毒,可实现对肝癌细胞的特异性杀伤,靶向抗癌效果优于一般的重组溶瘤腺病毒。
优选地,在所述的构建方法中,步骤三所述的细胞为HEK293细胞,转染时间不少于5天,所述的收取病毒液的方法包括:将步骤三转染后的细胞及其培养液反复冻融,离心得到的上清液,即为病毒液。该技术步骤的参数设置通过HEK293细胞收取的包括IL-24的编码核苷酸序列的重组溶瘤基因腺病毒病毒含量提高了至少100%,病变时间缩短了至少20%,实现重组溶瘤基因腺病毒的快速高效包装。
本发明第三方面提供一种重组溶瘤基因腺病毒在癌症治疗药物中的应用,所述的重组溶瘤基因腺病毒为OncoAd-P28-E1A-ΔE1B-IL-24。
优选地,在所述的应用中,所述的重组溶瘤基因腺病毒还包括OncoAd-P28-E1A-ΔE1B。
优选地,在所述的应用中,所述的癌症治疗药物还包括BRD4特异性的抑制剂。该技术步骤,将BRD4抑制剂作为一种联用药物,与重组溶瘤基因腺病毒联用具有协同抗癌作用,抗癌效果显著优于重组溶瘤基因腺病毒或者BRD4抑制剂的单用药。
优选地,在所述的应用中,所述的重组溶瘤基因腺病毒的启动子包括肝癌特异性启动子,所述的重组溶瘤基因腺病毒包括IL-24的编码核苷酸序列。通过该技术步骤,可实现技术效果为:重组溶瘤基因腺病毒对正常细胞的毒性低、杀伤肝癌细胞效果好,靶向抗癌效果优于一般的重组溶瘤腺病毒。

在本发明中,在肿瘤特异性溶瘤病毒上插入治疗基因,让溶瘤病毒治疗与基因治疗协同作战,即同时发挥多种生物治疗策略的多重治疗效果,可大大提高溶瘤病毒治疗的安全性并增强基因治疗的疗效。靶向基因-病毒治疗中的病毒载体本身会显著性地发挥靶向杀伤肿瘤的作用,再者,靶向基因-病毒治疗中的治疗基因的拷贝数会随着病毒复制增殖而链式增长,同时治疗基因的表达量也会极大地提高。总而言之,靶向基因-病毒治疗策略所具备的靶向性、安全性、基因表达量极高等独特优势一定会极大地推动靶向基因-病毒治疗的长足发展,为人类最终战胜癌症发挥重要的作用。
在本发明中,本发明所提到的IL-24基因作为目前被发现的唯一的既抑制肿瘤细胞生长、转移和血管形成,诱导肿瘤细胞凋亡,又刺激表达次级细胞因子免疫调理而日益受到关注。正常表达的IL-24通过配体-受体介导的JAK/STAT通路在免疫应答的调控中起着重要作用。IL-24因其抗肿瘤谱较广、抗肿瘤作用较强,发明人经研究发现,IL-24在较广谱的人肿瘤细胞系中具有生长抑制作用,而对正常细胞没有影响,在体内研究中利用腺病毒或溶瘤腺病毒表达IL-24基因都能取得较好的抗肿瘤效果,其抗肿瘤主要通过诱导凋亡等作用。因此,我们将这样一个具有高效抗肿瘤能力的基因插入靶向肝癌的溶瘤腺病毒内,可以增强特异性抗肝癌作用。发明人经研究发现ROS水平高的多药耐药性癌症细胞比ROS水平低的细胞对Ad.IL-24诱导的细胞死亡更敏感。在恶性胶质瘤中,IL-24能诱导自我吞噬。以重组IL-24蛋白处理人外周血单核细胞能诱导产生IL-6、IFNγ、TNFα、以及粒细胞/巨噬细胞集落刺激因子等免疫调节因子。Ad.IL-24与其他药物的联合使用能增强肿瘤杀伤效果,McKenzie等研究了在Her-2/neu超表达的乳腺癌细胞中Ad.IL-24与herceptin(小鼠抗p185ErbB2单克隆抗体)联合使用效果。在MCF-7-Her-18细胞中,联合使用引起了β-catenin、Akt和磷酸化Akt水平较单独使用时的降低,在MCF-7-Her-18成瘤的裸鼠中,联合使用也引起了肿瘤体积明显的减小,IL-24的相关临床试验也表现出了良好的治疗效果。
在本发明中,本发明所提到的BRD4是溴结构域和超末端结构(bromodomain andextraterminal domain,BET)家族蛋白成员之一,在哺乳动物中广泛存在,初次报道是附着在有丝分裂染色体上,参与细胞周期的调控。BRD4通过招募不同的转录调节因子,如Mediator、正性转录延伸因子P-TEFb来介导靶基因的表达,在调控细胞周期、胚胎发育、基因转录、炎症反应等生物过程中发挥着重要作用。近年来的研究也发现BRD4与恶性中线癌、急性骨髓性白血病、乳腺癌、黑色素瘤、伯基特淋巴癌、结肠癌等癌症的发生有关,BRD4shRNA或BET抑制剂可以诱导上述癌症发生细胞周期阻滞、凋亡及细胞分化,表现出强大的抗癌活性。JQ-1、PFI-1与MS645均为BRD4特异性的抑制剂。
实施例1. 重组腺病毒OncoAd-P28-E1A-ΔE1B-IL-24的构建
本实施例主要描述了一种重组溶瘤基因腺病毒OncoAd-P28-E1A-ΔE1B-IL-24的构建方法。
为了提高腺病毒的肿瘤特异性,将控制腺病毒复制的重要元件 E1A 区的野生型启动子替换为肝癌特异性启动子 P28GANK(P28),同时删除了腺病毒的 E1B 区,并且插入IL-24表达框来提高病毒对肿瘤细胞的杀伤效果。重组溶瘤腺病毒OncoAd-P28-E1A-ΔE1B-IL-24及相应空载病毒的结构示意图见图1。
(1)重组小质粒pShuttle-P28-E1A-ΔE1B-IL-24的构建:以pZD55-IL-24质粒(参见中国专利ZL 200510026151.1)为模板进行PCR,克隆得到IL-24的表达框。再以BglⅡ单酶切pShuttle-P28-E1A-ΔE1B小质粒(参见中国专利CN201810097442.0),得到线性化的pShuttle-P28-E1A-ΔE1B小质粒,经去磷处理后再用汉恒一步克隆试剂盒连接IL-24表达框和线性化的pShuttle-P28-E1A-ΔE1B小质粒,得到重组小质粒pShuttle-P28-E1A-ΔE1B-IL-24。图2A显示了克隆的IL-24表达框,图2B显示了线性化的pShuttle-P28-E1A-ΔE1B小质粒。
(2)重组大质粒的构建:将上步构建的pShuttle-P28-E1A-ΔE1B-IL-24小质粒用限制性内切酶PmeI线性化,去磷后转入带有腺病毒骨架质粒pAdEasy-E3的BJ5183感受态(参见中国专利CN201810097442.0)中,使二者发生同源重组,筛选获得阳性克隆,即重组大质粒。正确的重组大质粒使用限制性内切酶MluI酶切后可得到大小分别约为24 kb、5.8kb、4.3 kb、1.8 kb、1.1 kb的五条带,重组大质粒酶切鉴定结果见图3。
(3)重组溶瘤腺病毒OncoAd-P28-E1A-ΔE1B-IL-24的包装:将上步重组好的大质粒使用限制性内切酶PacI线性化,转染HEK-293细胞,8-10天后,细胞出现病变,收取病毒液即为重组溶瘤腺病毒OncoAd-P28-E1A-ΔE1B-IL-24。
(4)重组溶瘤腺病毒OncoAd-P28-E1A-ΔE1B-IL-24的鉴定:考虑到HEK-293细胞基因组整合有腺病毒的E1区,无法直接用HEK-293扩增的重组腺病毒来鉴定是否含有野生型腺病毒,故再用A549细胞进行重组溶瘤腺病毒的扩增,抽取病毒的基因组进行鉴定。首先鉴定IL-24基因及P28启动子是否存在。使用特异性的引物,以抽提的病毒基因组为模板进行PCR后跑琼脂糖凝胶电泳,鉴定结果如图4所示,说明IL-24基因及P28启动子均在重组病毒的基因组上。
然后鉴定重组溶瘤腺病毒OncoAd-P28-E1A-ΔE1B-IL-24中是否含有野生型腺病毒。分别以野生型腺病毒基因组和抽提的重组溶瘤腺病毒基因组为模板,以针对野生型腺病毒E1A启动子的引物进行PCR。同时以针对野生型腺病毒E1B区的引物进行PCR,检测是否含有E1A野生型启动子或E1B区的非目标腺病毒。鉴定结果如图5所示,说明重组溶瘤腺病毒OncoAd-P28-E1A-ΔE1B-IL-24中无非目标腺病毒。
进一步地,为了检测重组溶瘤腺病毒OncoAd-P28-E1A-ΔE1B-IL-24中所携带的治疗基因是否能正常表达,使用WB方法从蛋白质水平上分析IL-24的表达情况。用重组溶瘤腺病毒OncoAd-P28-E1A-ΔE1B和OncoAd-P28-E1A-ΔE1B-IL-24分别感染肝正常细胞QSG-7701和肝癌细胞Huh-7、Hep3B,在加入病毒42小时后,收取全细胞蛋白质进行WB检测。实验结果如图6所示,在溶瘤腺病毒OncoAd-P28-E1A-ΔE1B-IL-24感染42小时后,肝正常细胞QSG-7701和肝癌细胞Huh-7、Hep3B中都能检测到IL-24蛋白的表达。但相应空载溶瘤腺病毒OncoAd-P28-E1A-ΔE1B感染和不做处理的空白组中则没有检测到IL-24蛋白的表达,这说明重组溶瘤腺病毒OncoAd-P28-E1A-ΔE1B-IL-24中所携带的治疗基因能够正常表达。
实施例2. 重组溶瘤腺病毒OncoAd-P28-E1A-ΔE1B-IL-24对肝癌细胞的靶向杀伤
为了检测重组溶瘤腺病毒OncoAd-P28-E1A-ΔE1B-IL-24是否能够靶向杀伤肝癌细胞,选取了肝正常细胞QSG-7701、肝癌细胞Huh-7和Hep3B进行体外杀伤测试。上述细胞按照每孔1×105个细胞的密度铺24孔板,待细胞贴壁后,分别以不同MOI的OncoAd-P28-E1A-ΔE1B和OncoAd-P28-E1A-ΔE1B-IL-24感染细胞,感染72小时后用结晶紫染色。实验结果如图7所示,重组溶瘤腺病毒 OncoAd-P28-E1A-ΔE1B和OncoAd-P28-E1A-ΔE1B-IL-24对肝正常细胞QSG-7701几乎都没有杀伤能力,只有当病毒滴度高达100 MOI时才产生明显杀伤作用,表现出良好的安全性。而OncoAd-P28-E1A-ΔE1B和OncoAd-P28-E1A-ΔE1B-IL-24对肝癌细胞Huh-7和Hep3B却有着明显的杀伤,在1 MOI时就对肝癌细胞有极明显的杀伤作用,当MOI升高到10时,能将所有肝癌细胞全部杀死。这表明构建的以P28肝癌特异性启动子调控的重组溶瘤腺病毒对肝癌细胞具有特异性的杀伤作用。
为进一步确认上述结晶紫染色实验的结果,我们采用CCK8法验证该结果。同样选用肝正常细胞QSG-7701作为对照,选用肝癌细胞Hep3B和Huh-7作为实验组,分别用不同MOI的病毒去感染这三种细胞。72小时后,进行CCK8试验检测不同条件下的细胞存活率,结果如图8所示,重组溶瘤腺病毒 OncoAd-P28-E1A-ΔE1B和OncoAd-P28-E1A-ΔE1B-IL-24在10MOI及以下时几乎不杀伤肝正常细胞QSG-7701,即使MOI高达100也只是有少量杀伤,但其对肝癌细胞Hep3B和Huh-7具有强大的杀伤作用,与上述结晶紫染色实验的结果保持一致。
实施例3. 重组溶瘤腺病毒OncoAd-P28-E1A-ΔE1B-IL-24与JQ1联用具有协同作用
BRD4是溴结构域和超末端结构(bromodomain and extraterminal domain,BET)家族蛋白成员之一,在哺乳动物中广泛存在,参与细胞周期的调控。近年来的研究发现BRD4与急性骨髓性白血病、乳腺癌、黑色素瘤、伯基特淋巴癌、结肠癌等癌症的发生有关,BRD4shRNA或BET抑制剂可以诱导上述癌症发生细胞周期阻滞、凋亡及细胞分化,表现出强大的抗癌活性。而根据我们之前的研究进展,溶瘤腺病毒直接杀伤肿瘤的主要方式之一就是引起凋亡,因而可尝试将BRD4抑制剂与溶瘤腺病毒联用,可能会有意想不到的药效。
JQ-1为BRD4特异性的抑制剂,我们首次尝试将JQ1和重组溶瘤腺病毒联合使用处理肝癌细胞。测试的肝癌细胞为Hep3B和Huh-7,分别单独用JQ1抑制剂、空载对照OncoAd-P28-E1A-ΔE1B、重组溶瘤腺病毒OncoAd-P28-E1A-ΔE1B-IL-24以及JQ1抑制剂与重组溶瘤腺病毒OncoAd-P28-E1A-ΔE1B-IL-24联合使用处理,然后在处理24、48、72、96小时后使用CCK8法检测不同条件下的细胞存活率。结果如图9所示,单独使用BRD4抑制剂JQ1时,癌细胞的存活率与空白组没有显著差异。但当JQ1和OncoAd-P28-E1A-ΔE1B-IL-24联合使用时,对Hep3B细胞在处理48小时后,可以看到细胞活性已经降到30%左右,显著低于单独重组溶瘤腺病毒处理组OncoAd-P28-E1A-ΔE1B和OncoAd-P28-E1A-ΔE1B- IL-24。由于Hep3B细胞比较敏感,处理48小时后细胞死亡率过高,后续联用组与重组溶瘤腺病毒组无明显差异。对于Huh-7细胞,JQ1和OncoAd-P28-E1A-ΔE1B-IL-24联合使用在处理72小时后细胞活性降到30%左右,同样显著低于单独重组溶瘤腺病毒处理组OncoAd-P28-E1A-ΔE1B和OncoAd-P28-E1A-ΔE1B- IL-24,继续处理至96小时得到了同样的结果。综上,BRD4抑制剂JQ1和重组溶瘤腺病毒联合使用对肝癌细胞的杀伤效果要显著好于两者单用,表现出明显的协同杀伤作用。
为进一步确认抑制剂JQ1与重组溶瘤腺病毒OncoAd-P28-E1A-ΔE1B-IL-24具有协同杀伤作用,再次分别单独用JQ1抑制剂、空载病毒OncoAd-P28-E1A-ΔE1B、重组病毒OncoAd-P28-E1A-ΔE1B-IL-24以及JQ1抑制剂与重组病毒重组OncoAd-P28-E1A-ΔE1B-IL-24联合使用处理肝癌细胞Huh-7和Hep3B,处理24小时后进行Hoechst33342染色,在目镜10×和物镜20×的荧光倒置显微镜下观察。染色结果如图10 A所示,统计结果如图10 B所示。与空白对照相比,单独JQ1抑制剂、OncoAd-P28-E1A-ΔE1B、OncoAd-P28-E1A-ΔE1B-IL-24处理都可以引起部分肝癌细胞凋亡。JQ1和OncoAd-P28-E1A-ΔE1B-IL-24联合处理组引起的肝癌细胞凋亡显著高于单独JQ1或OncoAd-P28-E1A-ΔE1B-IL-24处理,与之前CCK-8结果保持一致。
为更进一步确认抑制剂JQ1与重组溶瘤腺病毒OncoAd-P28-E1A-ΔE1B-IL-24具有协同杀伤作用,再次分别单独用JQ1抑制剂、空载病毒OncoAd-P28-E1A-ΔE1B、重组病毒OncoAd-P28-E1A-ΔE1B-IL-24以及JQ1抑制剂与重组病毒重组OncoAd-P28-E1A-ΔE1B-IL-24联合使用处理肝癌细胞Huh-7和Hep3B,分别处理48、24小时后,将上清中的细胞和贴壁的细胞一起收集起来用于流式细胞术检测。流式结果如图11 A所示,统计结果如图11 B所示。与空白对照mock组相比,单独JQ1抑制剂、OncoAd-P28-E1A-ΔE1B、OncoAd-P28-E1A-ΔE1B-IL-24处理都可以引起部分肝癌细胞凋亡。JQ1和OncoAd-P28-E1A-ΔE1B-IL-24联合处理组引起的肝癌细胞凋亡显著高于单独JQ1或OncoAd-P28-E1A-ΔE1B-IL-24处理,与之前CCK-8、Hoechst33342染色结果保持一致。
实施例4. OncoAd-P28-E1A-ΔE1B-IL-24与BRD4抑制剂PFI-1、MS645联用都具有协同作用
PFI-1、MS645均为BRD4特异性的抑制剂,分别单独用BRD4抑制剂、重组溶瘤腺病毒OncoAd-P28-E1A-ΔE1B-IL-24以及BRD4抑制剂与重组溶瘤腺病毒OncoAd-P28-E1A-ΔE1B-IL-24联合使用处理Hep3B和Huh-7细胞,然后在处理48或72小时后使用CCK8法检测不同条件下的细胞存活率。结果如图12所示,在Hep3B细胞中,单独使用BRD4抑制剂PFI-1(1 uM)或MS645(0.1 uM)处理48小时,癌细胞的存活率与空白组没有显著差异;单独使用0.5 MOI的重组溶瘤腺病毒OncoAd-P28-E1A-ΔE1B- IL-24感染48小时,癌细胞的存活率与空白对照组亦没有显著性差异。但当抑制剂PFI-1或MS645和OncoAd-P28-E1A-ΔE1B-IL-24联合使用48小时后,Hep3B细胞活性出现了显著性下降,显著低于两组单用药组,表现出协同效应。在Huh-7细胞中也得到了类似的结果。综上,BRD4抑制剂PFI-1、MS645和重组溶瘤腺病毒OncoAd-P28-E1A-ΔE1B-IL-24联合使用均表现出协同作用。
从上述结果可知,BRD4抑制剂和重组溶瘤腺病毒OncoAd-P28-E1A-ΔE1B-IL-24联合使用具有明显的协同杀伤作用。下面以抑制剂JQ-1为例,探索协同杀伤的机理。首先检测了抑制剂JQ1对重组溶瘤腺病毒OncoAd-P28-E1A-ΔE1B-IL-24复制能力的影响,因为影响重组溶瘤腺病毒直接杀伤能力的主要是病毒复制能力。将肝癌细胞Hep3B以每孔3×105个细胞的密度铺六孔板,待细胞完全贴壁后分别用OncoAd-P28-E1A-ΔE1B-IL-24或者OncoAd-P28-E1A-ΔE1B- IL-24与JQ1联合处理细胞,处理48小时收集上清和所有的细胞,反复冻融三次,进行病毒滴度的测定。测得结果显示OncoAd-P28-E1A-ΔE1B-IL-24单独处理的病毒滴度为5.4×107 IU/ml,OncoAd-P28-E1A-ΔE1B-IL-24与JQ1联合处理后的病毒滴度为4.2×107 IU/ml,两者并无显著性差异,说明JQ1并不影响重组溶瘤腺病毒OncoAd-P28-E1A-ΔE1B-IL-24的复制。
为了探究JQ1和重组溶瘤腺病毒OncoAd-P28-E1A-ΔE1B-IL-24具有协同杀伤作用的原因,我们选取肝癌细胞Huh-7和Hep3B均以每孔3×105个细胞的密度铺六孔板,等细胞完全贴壁后分别用OncoAd-P28-E1A-ΔE1B-IL-24单独处理、JQ1单独处理、OncoAd-P28-E1A-ΔE1B- IL-24与JQ1联合处理,在处理48小时后收集全细胞蛋白质进行WB检测。结果出乎意料,如图13所示,在肝癌细胞Huh-7和Hep3B中,OncoAd-P28-E1A-ΔE1B-IL-24和JQ1联合处理后,外源基因IL-24的表达量显著高于单独OncoAd-p28-E1A-ΔE1B- IL-24处理,说明JQ1能够显著提升重组溶瘤腺病毒OncoAd-P28-E1A-ΔE1B-IL-24所携带的IL-24外源基因表达。

Claims (5)
1.一种重组溶瘤基因腺病毒在制备癌症治疗药物中的应用,其特征在于,所述的重组溶瘤基因腺病毒为OncoAd-P28-E1A-ΔE1B-IL-24;
所述的癌症治疗药物还包括BRD4特异性的抑制剂;
其中,所述的癌症为肝癌。
2.根据权利要求1所述的应用,其特征在于,所述的重组溶瘤基因腺病毒还包括OncoAd-P28-E1A-ΔE1B;
其中,所述重组溶瘤基因腺病毒的构建方法,包括以下步骤:
步骤一、重组小质粒的构建:以含IL-24表达框的质粒为模板进行PCR,克隆得到IL-24的表达框;再以限制性内切酶单酶切pShuttle-P28-E1A-ΔE1B小质粒并进行去磷处理,得到线性化并去磷的pShuttle-P28-E1A-ΔE1B小质粒,连接所述IL-24的表达框和线性化并去磷的pShuttle-P28-E1A-ΔE1B小质粒,得到重组小质粒pShuttle-P28-E1A-ΔE1B-IL-24;
步骤二、重组大质粒的构建:将步骤一构建的所述pShuttle-P28-E1A-ΔE1B-IL-24小质粒用限制性内切酶线性化,去磷后转入带有腺病毒骨架质粒的大肠杆菌感受态中,使二者发生同源重组,筛选获得阳性克隆,得到重组大质粒;
步骤三、重组溶瘤腺病毒的包装:将步骤二得到的重组大质粒使用限制性内切酶线性化,转染细胞,转染时间不少于3天,收取病毒液。
3.根据权利要求2所述的应用,其特征在于,所述的重组溶瘤基因腺病毒的启动子包括肝癌特异性启动子,所述的重组溶瘤基因腺病毒包括IL-24的编码核苷酸序列;
其中,步骤三所述的细胞为HEK293细胞,转染时间不少于5天,所述的收取病毒液的方法包括:将步骤三转染后的细胞及其培养液反复冻融,离心得到的上清液,即为病毒液。
4.根据权利要求1所述的应用,其特征在于,所述的重组溶瘤基因腺病毒的启动子包括P28GANK启动子,所述的重组溶瘤基因腺病毒的基因组敲除了E1B基因区域。
5.根据权利要求1所述的应用,其特征在于,所述的BRD4特异性的抑制剂包括JQ-1、PFI-1、MS645中的一种或几种。
