Abstract
本发明提供一种阴道微生物荧光染色液,由2.5%‑5%的抗氧化剂、0.005%‑0.01%的稳定剂、30%‑40%的缓冲剂、3%的荧光染色剂、30%的辅助染色剂、10%‑20%的促溶剂、0.01%‑0.02%的抑菌剂组成,余量为纯化水;其中抗氧化剂为没食子酸丙脂,稳定剂为柠檬酸钾。本发明以没食子酸丙酯为主抗氧化剂的荧光染色液的抗氧化效果优于以维生素C为主抗氧化剂的荧光染色液,且不易淬灭,较为稳定。
Description
技术领域
本发明涉及荧光染色液技术领域,尤其是一种阴道微生物荧光染色液。
背景技术
阴道炎是妇产科门诊常见疾病之一,大多是由各类病原体感染引起。阴道微生物检测是快速辨别感染情况,使患者得到更及时、更准确治疗的主要途径之一。
目前,市面上一些免疫荧光染色试剂存在易被氧化,易淬灭的染色不稳定现象,导致染色效果不佳的问题;与此同时,利用免疫荧光特异性标记微生物,无法避免因透光率太强或者对比度不明显而导致的漏检。
发明内容
针对现有的荧光染色液在阴道微生物检测中存在的不足之处,本发明提供一种稳定且不易氧化的阴道微生物荧光染色液。
一种阴道微生物荧光染色液,由2.5%-5%的抗氧化剂、0.005%-0.01%的稳定剂、30%-40%的缓冲剂、3%的荧光染色剂、30%的辅助染色剂、10%-20%的促溶剂、0.01%-0.02%的抑菌剂组成,余量为纯化水;其中抗氧化剂为没食子酸丙脂,稳定剂为柠檬酸钾。
进一步的,缓冲剂为无水磷酸二氢钠和十二水合磷酸氢二钠;荧光染色剂为吖啶橙,辅助染色剂为甲基绿;促溶剂为二甲基亚砜;抑菌剂为PC300。
本发明以没食子酸丙酯为主抗氧化剂的荧光染色液的抗氧化效果优于以维生素C为主抗氧化剂的荧光染色液,且不易淬灭,较为稳定;提供的阴道微生物荧光染色液将脱落细胞染出绿色,细菌染成红色,使得线索细胞能够呈现更加明显的颜色反差,辨别更容易、更准确,保证检测结果的准确性;在镜检时染色效果更稳定,清晰度及对比度更佳;免疫荧光特异性标记微生物,背景差异化明显,能够解决因透光率太强或者对比度不明显而导致的漏检问题;荧光染色液在镜检金标准上的技术升级,提高了湿片镜检的检出率、准确率,避免可能存在的漏检和误检,提高了镜检的效率,对临床诊断应用具有重大提升意义。

具体实施方式
下面结合附图和具体实施例对本发明实施例中的技术方案进行清楚、完整地描述,显然,所描述的实施例仅仅是本发明一部分实施例,而不是全部的实施例。基于本发明中的实施例,本领域普通技术人员所获得的所有其他实施例,都属于本发明保护的范围。
实施例1
一种阴道微生物荧光染色液,由2.5%的没食子酸丙脂(抗氧化剂)、0.005%的柠檬酸钾(稳定剂)、30%的无水磷酸二氢钠和十二水合磷酸氢二钠(缓冲剂)、3%的吖啶橙(荧光染色剂)、30%的甲基绿(辅助染色剂)、10%的促溶剂(二甲基亚砜)、0.01%的PC300(抑菌剂)组成,余量为纯化水。
本发明公开的荧光染色液配方中的抗氧化主体是没食子酸丙酯和柠檬酸钾。没食子酸丙酯具有抗氧化及防淬灭作用;柠檬酸钾具有耐氧化作用,同时也是荧光染色液中的稳定剂。两者在荧光染色液配方中协同作用,提升荧光染色液的抗氧化效果。
目前,市场上荧光染色液基本上以维生素C作为抗氧化剂。现以本发明公开的以没食子酸丙酯作为主抗氧化剂,与以维生素C作为主抗氧化剂进行试验分析对比。
按照实施例1公开的组分配制阴道微生物荧光染色液S1;其他组分及含量不变,将没食子酸丙酯替换为维生素C,配制阴道微生物荧光染色液D1。将两份荧光染色液置入80℃水浴锅中,进行14天加速稳定性试验。通过观察两份荧光染色液的颜色变化以及主要荧光染色剂吖啶橙含量的测定。
经过14天的加速稳定性试验,荧光染色液D1的颜色由墨绿色变为红棕色,而荧光染色液S1由墨绿色变为草绿色,由此可见,荧光染色液D1的氧化程度更严重。
经过14天的加速稳定性试验,荧光染色液D1的镜检结果如图1所示,而荧光染色液S1的镜检结果如图2所示。对比而言,经过14天的加速稳定性试验后,荧光染色液D1的染色背景较为模糊,对比度差。
基于本发明提供的荧光染色液进行阴道微生物检测,具有较高的准确率,操作步骤简单,实际成本适中,能够区分混合感染,同一视野全标记,成功克服了现有检测存在的容易漏检的缺点。
吖啶橙含量测定结果如图3所示,数据显示以没食子酸丙酯为主抗氧化剂的荧光染色液的抗氧化效果优于以维生素C为主抗氧化剂的荧光染色液,且不易淬灭,较为稳定。
实施例2
一种阴道微生物荧光染色液,由5%的没食子酸丙脂(抗氧化剂)、0.01%的柠檬酸钾(稳定剂)、40%的无水磷酸二氢钠和十二水合磷酸氢二钠(缓冲剂)、3%的吖啶橙(荧光染色剂)、30%的甲基绿(辅助染色剂)、20%的促溶剂(二甲基亚砜)、0.02%的PC300(抑菌剂)组成,余量为纯化水。
按照实施例2公开的组分配制阴道微生物荧光染色液S2;其他组分及含量不变,将没食子酸丙酯替换为维生素C,配制阴道微生物荧光染色液D2。将两份荧光染色液置入80℃水浴锅中,进行14天加速稳定性试验。通过观察两份荧光染色液的颜色变化以及主要荧光染色剂吖啶橙含量的测定。
经过14天的加速稳定性试验,两份荧光染色液的颜色变化和镜检结果与实施例1基本一致,吖啶橙含量测定结果如图4所示,数据依旧显示以没食子酸丙酯为主抗氧化剂的荧光染色液的抗氧化效果优于以维生素C为主抗氧化剂的荧光染色液,且不易淬灭,较为稳定。
实施例3
一种阴道微生物荧光染色液,由4%的没食子酸丙脂(抗氧化剂)、0.008%的柠檬酸钾(稳定剂)、35%的无水磷酸二氢钠和十二水合磷酸氢二钠(缓冲剂)、3%的吖啶橙(荧光染色剂)、30%的甲基绿(辅助染色剂)、15%的促溶剂(二甲基亚砜)、0.015%的PC300(抑菌剂)组成,余量为纯化水。
按照实施例4公开的组分配制阴道微生物荧光染色液S3;其他组分及含量不变,将没食子酸丙酯替换为维生素C,配制阴道微生物荧光染色液D3。将两份荧光染色液置入80℃水浴锅中,进行14天加速稳定性试验。通过观察两份荧光染色液的颜色变化以及主要荧光染色剂吖啶橙含量的测定。
经过14天的加速稳定性试验,两份荧光染色液的颜色变化和镜检结果与实施例1基本一致,吖啶橙含量测定结果如图5所示,数据依旧显示以没食子酸丙酯为主抗氧化剂的荧光染色液的抗氧化效果优于以维生素C为主抗氧化剂的荧光染色液,且不易淬灭,较为稳定。
最后,还需要注意的是,以上列举仅是本发明一个具体实施例。显然,本发明不限于以上实施例,还可以有许多变形。本领域的普通技术人员能从本发明公开的内容直接导出或联想到的所有变形,均应认为是本发明的保护范围。
Claims (1)
1.一种阴道微生物荧光染色液,其特征在于,由2.5%-5%的抗氧化剂、0.005%-0.01%的稳定剂、30%-40%的缓冲剂、3%的荧光染色剂、30%的辅助染色剂、10%-20%的促溶剂、0.01%-0.02%的抑菌剂组成,余量为纯化水;其中抗氧化剂为没食子酸丙脂,稳定剂为柠檬酸钾;
缓冲剂为无水磷酸二氢钠和十二水合磷酸氢二钠,荧光染色剂为吖啶橙,辅助染色剂为甲基绿,促溶剂为二甲基亚砜,抑菌剂为PC300。
