Animal experiments show that the composition can obviously prolong the sleeping time from the sodium pentobarbital to the mice, effectively improve the sleeping incidence of the mice with the sub-threshold dosage of the sodium pentobarbital, and has obvious effect of improving sleeping.
Meanwhile, the composition is prepared from pure natural plants, is green and safe, and has no toxic or side effect after long-term administration.
Detailed Description
In order that the invention may be readily understood, a more complete description of the invention will be rendered by reference to the embodiments that are illustrated below. This invention may, however, be embodied in many different forms and should not be construed as limited to the embodiments set forth herein. Rather, these embodiments are provided so that this disclosure will be thorough and complete.
Unless defined otherwise, all technical and scientific terms used herein have the same meaning as commonly understood by one of ordinary skill in the art to which this invention belongs. The terminology used herein in the description of the invention is for the purpose of describing particular embodiments only and is not intended to be limiting of the invention. The term "and/or" as used herein includes any and all combinations of one or more of the associated listed items.
The raw materials used in the following examples, unless otherwise specified, are all commercially available; the methods used in the examples below, unless otherwise specified, are all conventional.
Example 1
A composition for improving sleep, which consists of the following raw materials in parts by weight.
40 parts of cordyceps militaris, 40 parts of ground tip melon, 10 parts of eupatorium fortunei, 10 parts of gynostemma pentaphylla, 10 parts of perilla stem, 5 parts of fructus forsythiae, 10 parts of liquorice, 10 parts of platycodon grandiflorum and 5 parts of radix sileris.
The preparation method of the composition comprises the following steps:
(1) Pulverizing the above materials into coarse powder respectively for use.
(2) Cordyceps militaris is prepared from 1g: adding 10 times of 10% ethanol water solution with volume percentage of 10% into 10ml solution, ultrasonic extracting for 30min for 3 times, filtering, mixing filtrates, concentrating at 70deg.C or below to obtain extract with relative density of 1.42 (55deg.C), drying at 70deg.C, and pulverizing into fine powder.
(3) 10 parts of eupatorium and 5 parts of radix sileris according to 1g: adding distilled water 10 times of the feed liquid ratio of 10ml solution, soaking for 1.5h, extracting by steam distillation for 12h, and collecting volatile oil for later use.
(4) Mixing the residue after volatile oil extraction of herba Eupatorii and radix Saposhnikoviae, volatilizing solvent, and mixing with 40 parts of Di-tip melon, 10 parts of Perilla stem, 5 parts of fructus forsythiae, 10 parts of herba Gynostemmatis, 10 parts of Glycyrrhrizae radix, and 10 parts of radix Platycodi according to 1g:15ml of solution is added with 15 times of distilled water, decocted for 3 times with slow fire for 1.5 hours each time, filtered, combined with filtrate, concentrated to the relative density of 1.12 (50 ℃), added with ethanol until the ethanol content reaches 70% (namely, the volume percent concentration of the ethanol is calculated to reach 70%), stirred evenly, stood for 30 hours, taken supernatant, concentrated, dried and crushed into fine powder for standby.
(5) Mixing the fine powder obtained in the steps (2) and (4) uniformly, and mixing the mixture with the volatile oil obtained in the step (3) before use, thus obtaining the sleep improving composition.
Example 2
A composition for improving sleep, which consists of the following raw materials in parts by weight.
25 parts of cordyceps militaris, 25 parts of ground tip melon, 15 parts of eupatorium fortunei, 12 parts of gynostemma pentaphylla, 8 parts of perilla stem, 4 parts of fructus forsythiae, 12 parts of liquorice, 12 parts of platycodon grandiflorum and 4 parts of radix sileris.
The preparation method of the above composition is described with reference to example 1.
Comparative example 1
A composition was substantially the same as the composition of example 1 except that in this comparative example, the Di Gua was omitted and the amount of Cordyceps militaris used was increased to 80 parts, which corresponds to the substitution of Di Gua with Cordyceps militaris.
Comparative example 2
A composition substantially identical to the composition of example 1 except that in this comparative example, petiole was replaced with eupatorium.
Comparative example 3
A composition substantially identical to the composition of example 1, except that in this comparative example, windbreaks were omitted.
Experimental example
This experimental example animal experiments were carried out on the compositions prepared in the above examples and comparative examples.
1. Animals and groups.
1) Experimental animals: male mice of 18-22 g were grown adaptively for 1 week.
2) Grouping and administration: randomly grouping 12 groups, specifically as follows:
the composition of example 1, designated Z-1, was set to three dosage groups of 1.2g/kg high, 0.6g/kg medium, and 0.3g/kg low;
the composition of example 2, designated Z-2, was given three dose groups of 1.2g/kg high, 0.6g/kg medium, and 0.3g/kg low;
the compositions of comparative examples 1-3, designated D-1, D-2, D-3, were set at a high dose of 1.2g/kg;
the blank group was filled with the same volume of distilled water.
The compositions of the examples were mixed immediately before use and then formulated with distilled water to give 8%, 4% and 2% suspensions, i.e. high, medium and low dose groups, respectively. Sleep experiments were performed 30 days after administration by divided gastric lavage.
2. Direct sleep experiment
After the last dose, each group of mice was observed for sleep. Sleep is indicated by the disappearance of the eversion and the reflection. When the mouse is placed in the dorsum position, the body position can be immediately turned over. If the person cannot turn over for more than 30-60 seconds, the person considers that the turning over reflection disappears and goes to sleep. The recovery of the eversion and the reflection is the arousal of the animal, and the period from the disappearance of the eversion and the reflection to the recovery is the sleep time of the animal. The experimental results are shown in Table 1.

From the above results, it was found that the mice in the administration group and the blank group had no direct sleep condition after administration by gastric lavage for 30 days, and that there was no difference between groups.
3. Experiment for prolonging sleeping time of pentobarbital sodium
After the last administration for 40min, 50mg/kg of pentobarbital sodium was intraperitoneally injected into each group of mice with an injection amount of 0.1mL/10g, and the composition was observed to determine whether the sleep time of the mice caused by pentobarbital sodium can be prolonged by taking the disappearance of the specular reflection as an index, and the results are shown in Table 2.

As can be seen from Table 2, the compositions of examples 1 and 2 of the present invention each had a prolonged sleep time in mice caused by sodium pentobarbital, and the effects showed an enhanced tendency with increasing doses, and the effects of the medium and high dose groups were remarkable. The compositions of comparative examples 1-3, however, were less effective even in the high dose group.
4. Sub-threshold dose hypnotic experiment with sodium pentobarbital
After the last administration for 40min, each group of mice was intraperitoneally injected with 28mg/kg of pentobarbital sodium, the number of animals falling asleep (the number of animals with the loss of the everlasting reflection reaching more than 1 minute) was recorded within 30 minutes, and whether the composition can improve the incidence of sleep of the mice caused by the subthreshold dose of pentobarbital sodium or not was observed. Experimental results showed that the incidence of sleep onset was significantly increased (p < 0.05) in mice given the compositions of examples 1-2 of the present invention with a sub-threshold dose of sodium pentobarbital compared to the placebo group, and the effect was enhanced with increasing dosing. The compositions of comparative examples 1-3, however, were less effective even in the high dose group.
5. Barbital sodium hypnotic latency experiment
After the last administration for 20min, 260mg/Kg of barbital sodium is injected into the abdominal cavity of each group of mice, the injection amount is 0.1mL/10g, the disappearance of the eversion and the specular reflection are taken as indexes, and the influence of the composition on the sleeping latency of the barbital sodium is observed. The results are shown in Table 3.

The above experimental results show that, compared to the blank control group, the barbituric sodium-induced mice have significantly shortened sleep latency (p < 0.05) and their effects exhibit a dose-dependent enhancement after administration of the compositions of examples 1-2 of the present invention. The compositions of comparative examples 1 to 3 were not good in the same effect.
The above experimental results show that the compositions of examples 1 and 2 can prolong the sleep time of pentobarbital sodium, increase the incidence of hypnosis in mice caused by the subthreshold dose of pentobarbital sodium, and shorten the sleep latency of the pentobarbital sodium after 30 days of continuous gastric administration, the effect is dose-dependent enhancement, and no obvious direct sleep effect exists, which indicates that the composition has good sleep improving function. Either omitting the medicinal odor or replacing the medicinal odor can result in the composition having reduced sleep improving effect.
The technical features of the above-described embodiments may be arbitrarily combined, and all possible combinations of the technical features in the above-described embodiments are not described for brevity of description, however, as long as there is no contradiction between the combinations of the technical features, they should be considered as the scope of the description.
The above examples illustrate only a few embodiments of the invention, which are described in detail and are not to be construed as limiting the scope of the invention. It should be noted that it will be apparent to those skilled in the art that several variations and modifications can be made without departing from the spirit of the invention, which are all within the scope of the invention. Accordingly, the scope of protection of the present invention is to be determined by the appended claims.

Claims (9)

1. The composition for improving sleep is characterized by being prepared from the following raw materials in parts by weight:
40+/-5 parts of cordyceps militaris, 40+/-5 parts of ground tip melon, 10+/-3 parts of eupatorium, 10+/-3 parts of gynostemma pentaphylla, 10+/-3 parts of perilla stem, 5+/-2 parts of fructus forsythiae, 10+/-3 parts of liquorice, 10+/-3 parts of platycodon grandiflorum and 5+/-2 parts of radix sileris.
2. A method of preparing a sleep improving composition as claimed in claim 1, characterized in that it comprises the steps of:
preparing cordyceps militaris: pulverizing Cordyceps militaris, adding ethanol water solution, performing ultrasonic extraction, filtering, collecting filtrate, concentrating to obtain extract, drying and pulverizing to obtain Cordyceps militaris extract;
extracting volatile oil: extracting volatile oil from herba Eupatorii and radix Saposhnikoviae by steam distillation;
preparation of the extract: decocting the above volatile oil-extracted herba Eupatorii and radix Saposhnikoviae residues and other materials in water, collecting filtrate, concentrating, adding ethanol, standing, collecting supernatant, concentrating, and drying to obtain mixed extract;
mixing: mixing the Cordyceps militaris extract and the mixed extract, and adding the volatile oil and mixing just before use to obtain the composition.
3. The method for preparing the composition for improving sleep according to claim 2, wherein in the step of preparing the cordyceps militaris, the cordyceps militaris is crushed, an ethanol aqueous solution with the volume percentage concentration of 10% is added according to the amount of 5-20ml/g cordyceps militaris, the ultrasonic extraction is carried out for 2-4 times, the time for each extraction is 15-60min, the filtrates are combined and concentrated under the condition of less than or equal to 70 ℃ to obtain an extract with the relative density of 1.40-1.50, and the extract is dried under the condition of less than or equal to 70 ℃ and crushed to obtain the composition.

The method for preparing the sleep improvement composition according to claim 2, wherein in the step of extracting the volatile oil, water is added according to the amount of 5-20ml/g of medicinal materials, the mixture is soaked for 0.5-3 hours, the volatile oil is extracted for 8-24 hours by a steam distillation method, and the volatile oil is collected.
5. The method for preparing the sleep improving composition according to claim 2, wherein in the step of preparing the extract, water is added according to the amount of 5-30ml/g of medicinal materials, the decoction is carried out for 2-4 times, each time for 0.5-3 hours, the filtrates are combined, the concentration is carried out until the relative density is 1.10-1.20, the ethanol is added until the alcohol content is 60% -90%, the mixture is stirred uniformly, the mixture is kept stand for 8-36 hours, the supernatant is taken, and the mixture is concentrated, dried and crushed into fine powder, thus obtaining the sleep improving composition.
6. Use of a composition according to claim 1 for the manufacture of a medicament for improving sleep.
7. A pharmaceutical composition for improving sleep, characterized by comprising the composition of claim 1 as an active ingredient, and pharmaceutically acceptable excipients.
8. The pharmaceutical composition of claim 7, wherein the composition is in a dosage form selected from the group consisting of: tablets, granules, oral liquids, soft capsules, hard capsules or powders.
9. The pharmaceutical composition according to claim 7, wherein the adjuvant is selected from the group consisting of: at least one of dextrin, lactose, citric acid, sodium hydroxymethyl starch, silica gel micropowder, starch, polyethylene glycol, magnesium stearate and vegetable oil.
