Abstract
本发明公开了一种新型快速超敏C‑反应蛋白试剂盒及应用,其特征在于:所述的试剂盒包括底板1、以及依次设在所述底板1上的样品垫2、结合垫3、NC膜4和棉浆板6;所述结合垫3上包被有时间分辨荧光微球标记的C反应蛋白(CRP)抗体;NC膜上有一条线质控线5,两条检测线;所述的质控线5包被有羊抗鼠抗体;所述的检测线分别是T1检测线,T2检测线;T1检测线包被抗体k1016(欧凯);T2检测线包被微球标记抗体K1017(欧凯);在底板1上依次粘贴样品垫2、结合垫3、NC膜4和棉浆板6,切割成检测试纸,封装即得试剂盒,本发明具有特异性强、灵敏度高、稳定性好、无需大型仪器及专业操作人员、检测时间快速等优点。

技术领域
本发明涉及蛋白检测技术领域,具体涉及一种新型快速超敏C-反应蛋白试剂盒及应用。
背景技术
只目前对人体CRP检测的方法有、毛细管沉淀、乳胶波板凝集、乳胶增强免疫比浊法、酶联免疫法、时光分辨分辨荧光免疫分析法、胶体金免疫层析法等;乳胶增强免疫比浊法、酶联免疫法、时间分辨荧光免疫分析法,操作过程复杂,需要大量仪器与设备及专业人员操作,检测时间较长,检测灵敏度高,胶体金免疫层析法操作简单,但灵敏度不高,无法精确定量。
发明内容
本发明提出一种新型快速超敏C-反应蛋白试剂盒及应用,以解决现有的技术问题。
本发明的技术方案是这样实现的:一种新型快速超敏C-反应蛋白试剂盒,其特征在于:所述的试剂盒包括底板、以及依次设在所述底板上的样品垫、结合垫、NC膜和棉浆板,
所述结合垫上包被有时间分辨荧光微球标记的C反应蛋白(CRP)抗体;NC膜上有一条线质控线,两条检测线;所述的检测线分别是T1检测线,T2检测线;T1检测线包被抗体K1016(欧凯);T2检测线包被微球标记抗体K1017(欧凯);
所述的质控线包被有羊抗鼠抗体;
所述时间分辨荧光微球载有镧系稀土元素或其螯合物,优选为铕;
所述的T2检测线靠近所述结合垫,所述质控线靠近所述棉浆板,所述T1检测线位于T2检测线和质控线之间,T1检测线 、T2检测线、质控线的间隔为3.5mm;
所述时间分辨荧光微球标记的C反应蛋白(CRP)抗体、羊抗鼠抗体、K1016(欧凯)和K1017(欧凯)抗体浓度均为0.45mg/ml。
本发明还提供了上述一种新型快速超敏C-反应蛋白试剂盒应用;其特征在于,所述试剂盒应用包括下列步骤:
(a)抗体包被:选用直径为120nm的荧光微球,将C反应蛋白(CRP)抗体、羊抗鼠抗体、K1016(欧凯)和K1017(欧凯)抗体标记到荧光微球上;
(b)结合垫包被:使用定量喷液装置将a步骤中的时间分辨荧光标记的C反应蛋白(CRP)抗体涂于处理过的结合垫上后48℃烘干2小时;
(c)NC膜的制备:使用包被K1016(欧凯)和K1017(欧凯)抗体分别稀释到120mmol/L;羊抗鼠抗体稀释到100mmol/L;使用定量喷液装置将上述抗体喷印到对应的T1检测线、T2检测线和质控线所在区域,避光,在45℃烘干12小时,加入干燥剂封存备用;
(d)试纸盒组装:将样品垫、结合垫、NC膜和棉浆板依次搭接并粘贴固定在底板上,底板的厚度为0.3mm,长度为6.5cm,宽度为3.5mm。
采用了上述技术方案,本发明的有益效果为:本发明试剂盒采用了一对灵敏度高的抗体通过双抗体夹心法检测血清血浆及全血;操作简便,不需要对样本进行复杂的处理,具有特异性强、灵敏度高、稳定性好、无需大型仪器及专业操作人员、检测时间快速等优点。

为了便于本领域技术人员的理解,下面结合实施例对本发明作进一步的说明,实施方式提及的内容并非对本发明的限定。
一种新型快速超敏C-反应蛋白试剂盒,其特征在于:所述的试剂盒包括底板、以及依次设在所述底板上的样品垫、结合垫、NC膜和棉浆板,
所述结合垫上包被有时间分辨荧光微球标记的C反应蛋白(CRP)抗体;NC膜上有一条线质控线,两条检测线;所述的检测线分别是T1检测线,T2检测线;T1检测线包被抗体K1016(欧凯);T2检测线包被微球标记抗体K1017(欧凯);
所述的质控线包被有羊抗鼠抗体;
所述时间分辨荧光微球载有镧系稀土元素或其螯合物,优选为铕。
所述的T2检测线靠近所述结合垫,所述质控线靠近所述棉浆板,所述T1检测线位于T2检测线和质控线之间,T1检测线 、T2检测线、质控线的间隔为3.5mm;
所述时间分辨荧光微球标记的C反应蛋白(CRP)抗体、羊抗鼠抗体、K1016(欧凯)和K1017(欧凯)抗体浓度均为0.45mg/ml。
【实施例1】
如图1所示,使用时,将100μL血清样样品滴入试剂盒样品垫上,进行免疫层析反应,并在荧光检测仪下进行检测读数。
反应原理:测试时,样本滴入检测试纸后,样本中的C反应蛋白(CRP)与预先包被在结合垫上的时间分辨荧光标记的C反应蛋白(CRP)抗体结合,样本中的T2检测线包被微球标记抗体K1017作为检测抗体;包被抗体K1016(欧凯)作为捕获抗体;利用时间分辨荧光免疫分析技术,以三价铕离子为标志物,镧系稀土元素荧光寿命较普通的荧光标记物寿命长,且Stokes 位移较大,待短寿命背景荧光消失后,测定长寿命的镧系稀土元素螯合物荧光,可避免背景荧光的干扰,达到定量分析的目的。
【实施例2】
一种新型快速超敏C-反应蛋白试剂盒应用;其特征在于,所述试剂盒应用包括下列步骤:
(a)抗体包被:选用直径为120nm的荧光微球,将C反应蛋白(CRP)抗体、羊抗鼠抗体、K1016(欧凯)和K1017(欧凯)抗体标记到荧光微球上;
(b)结合垫包被:使用定量喷液装置将a步骤中的时间分辨荧光标记的C反应蛋白(CRP)抗体涂于处理过的结合垫上后48℃烘干2小时;
(c)NC膜的制备:使用包被K1016(欧凯)和K1017(欧凯)抗体分别稀释到120mmol/L;羊抗鼠抗体稀释到100mmol/L;使用定量喷液装置将上述抗体喷印到对应的T1检测线、T2检测线和质控线所在区域,避光,在45℃烘干12小时,加入干燥剂封存备用;
(d)试纸盒组装:将样品垫、结合垫、NC膜和棉浆板依次搭接并粘贴固定在底板上,底板的厚度为0.3mm,长度为6.5cm,宽度为3.5mm。
使用时,将30-100μL血清样样品滴入试剂盒样品垫上,进行免疫层析反应,随后在在荧光检测仪下进行检测读数。
本发明试剂盒采用了一对灵敏度高的抗体通过双抗体夹心法检测血清血浆及全血;操作简便,不需要对样本进行复杂的处理,具有特异性强、灵敏度高、稳定性好、无需大型仪器及专业操作人员、检测时间快速等优点。

Claims (5)
1.一种新型快速超敏C-反应蛋白试剂盒;其特征在于:所述的试剂盒包括底板1、以及依次设在所述底板上的样品垫2、结合垫3、NC膜4和棉浆板6;所述结合垫3上包被有时间分辨荧光微球标记的C反应蛋白(CRP)抗体;NC膜上有一条线质控线5,两条检测线;所述的质控线5包被有羊抗鼠抗体;所述的检测线分别是T1检测线,T2检测线;T1检测线包被抗体k1016(欧凯);T2检测线包被微球标记抗体K1017(欧凯);在底板1上依次粘贴样品垫2、结合垫3、NC膜4和棉浆板6,切割成检测试纸,封装即得试剂盒。
2.根据权利要求1所述的一种新型快速超敏C-反应蛋白试剂盒,其特征在于:所述时间分辨荧光微球载有镧系稀土元素或其螯合物,优选为铕。
3.根据权利要求1所述的一种新型快速超敏C-反应蛋白试剂盒,其特征在于:所述的T2检测线靠近所述结合垫,所述质控线靠近所述棉浆板,所述T1检测线位于T2检测线和质控线之间,T1检测线 、T2检测线、质控线的间隔为3.5mm。
4.根据权利要求1所述的一种新型快速超敏C-反应蛋白试剂盒,其特征在于:所述时间分辨荧光微球标记的C反应蛋白(CRP)抗体、羊抗鼠抗体、K1016(欧凯)和K1017(欧凯)抗体浓度均为0.45mg/ml。
5.根据权利要求1-4任一项所述的一种新型快速超敏C-反应蛋白试剂盒应用;其特征在于,所述试剂盒应用包括下列步骤:
(a)抗体包被:选用直径为120nm的荧光微球,将C反应蛋白(CRP)抗体、羊抗鼠抗体、K1016(欧凯)和K1017(欧凯)抗体标记到荧光微球上;
(b)结合垫包被:使用定量喷液装置将a步骤中的时间分辨荧光标记的C反应蛋白(CRP)抗体涂于处理过的结合垫上后48℃烘干2小时;
(c)NC膜的制备:使用包被K1016(欧凯)和K1017(欧凯)抗体分别稀释到120mmol/L;羊抗鼠抗体稀释到100mmol/L;使用定量喷液装置将上述抗体喷印到对应的T1检测线、T2检测线和质控线所在区域,避光,在45℃烘干12小时,加入干燥剂封存备用;
(d)试纸盒组装:将样品垫、结合垫、NC膜和棉浆板依次搭接并粘贴固定在底板上,底板的厚度为0.3mm,长度为6.5cm,宽度为3.5mm。
