The invention has the beneficial effects that:
the calamus root extracting solution provided by the invention is derived from an aromatic medicinal plant with a long medicinal history, mainly contains alpha-, beta-asarone, cis-methyl iso-eugenol, methyl eugenol, calamus glycol, calamus ketone and the like, has obvious anti-inflammatory activity and acne-removing effect, can inhibit the generation of proinflammatory factors, and has anti-inflammatory effect on acute, chronic and some immune inflammation models; has strong bacteriostatic effect on saccharomycetes and propionibacterium acnes; meanwhile, the facial cleanser has obvious inhibition effect on microsporomycetes, aspergillus niger and white pearly fungi, and is suitable for being applied to facial cleansing products.
Detailed Description
The technical solutions in the embodiments of the present invention will be clearly and completely described below, and it is obvious that the described embodiments are only a part of the embodiments of the present invention, and not all embodiments. All other embodiments, which can be derived by a person skilled in the art from the embodiments given herein without making any creative effort, shall fall within the protection scope of the present invention.
Example one
An extractive solution of Amaranthus tricolor root comprises alpha-, beta-asarone, cis-methyl isoeugenol, methyl eugenol, Acorus calamus dilol and Acorus calamus ketone;
the application of Yandangshan calamus root extracting solution in cosmetics is amino acid facial cleansing honey which is composed of the following raw materials:
phase A: 20-23% of water, 0.03-0.1% of EDTA disodium, 3-5% of glycerol, 0.3-0.8% of glyceryl polyether-26 and 0.1-0.3% of xanthan gum;
phase B: cocoyl glutamic acid TEA salt-20-30%, CK 30-4-6%, RunmindTM10-18% of COP, 1-3% of polyglycerol-10, 7-9% of CHSB and 1-3% of PLANTACARE2000 UP;
and C phase: cetiol HE-0.1-0.3%, RunqatTM SMC18——0.4~0.6%;
Phase D: 1-3% of carbopol aqua SF-1 and 5-7% of water;
phase E: 0.5 to 0.7 percent of sodium hydroxide aqueous solution;
and (3) phase F: 0.3-0.6% of p-hydroxyacetophenone, 0.3-0.6% of hexanediol and 2-4% of butanediol;
phase G: 7-9% of calamus root extracting solution.
The sum of the mass fractions of the components of the phase A, the phase B, the phase C, the phase D, the phase E, the phase F and the phase G is 100 percent.
The cocoyl glutamic acid TEA salt active content was 30%.
The mass fraction of the sodium hydroxide aqueous solution was 10%.
The preparation method of the cosmetic comprises the following steps:
the method comprises the following steps: mixing the F phase, heating to 55-60 ℃ for dissolving, and cooling for later use;

step seven: cooling to room temperature, detecting and discharging to obtain the cosmetic amino acid facial cleansing honey.
The formula of the amino acid face-cleansing honey cosmetic is shown in table 1:

Example two
The physical and chemical indexes, efficacy and safety of the Yandangshan calamus root extract and the cosmetic containing the same (application example one) are tested as follows:
first, physicochemical index test
Sensory testing: visually observing the properties of the product to see whether the product is abnormal or not;
and (3) heat resistance test: putting the sample into an electric heating constant temperature incubator at the temperature of (40 +/-1) DEG C for 24 hours, and observing whether phenomena such as thinning, discoloration, layering, hardness change and the like exist after the temperature is recovered to judge the heat resistance of the sample;
cold resistance: putting the sample into a refrigerator at (-5-10) + -1 ℃ for 24 hours, and observing whether phenomena such as thinning, color change, layering, hardness change and the like exist after the room temperature is recovered to judge the cold resistance of the sample;
and (3) centrifugal test: and (3) placing the sample in a centrifuge, testing for 30min at the rotating speed of 2000-4000 r/min, and observing the separation and delamination conditions of the sample.
The results are shown in Table 2, and the Yandangshan Acorus calamus root extract and the cosmetic containing the same (application example one) are stable.

Second, efficacy test
1. Anti-inflammatory Activity assay
(1) Anti-inflammatory activity test of calamus root extract
In order to verify the anti-inflammatory activity of the calamus root extracting solution, the expression inhibition activity of proinflammatory cytokines (TNF-alpha and IL-6) is adopted, and the specific experimental steps are as follows:
RAW264.7 cells were inoculated in a 24-well plate at 1.8 × 105 cells/mL for 18 hours in DMEM medium supplemented with 10% fetal bovine serum, the medium was removed, 50 μ L of a sample prepared at 10-fold concentration (1mg/mL) and 450 μ L of a medium containing 1 μ g/mL LPS were treated, after 24 hours and 24 hours of culture, the medium was centrifuged (12000rpm for 3 minutes), the proinflammatory cytokine production content of the obtained supernatant was measured, all samples were subjected to cryopreservation (-20 ℃) until quantification, and the proinflammatory cytokine was quantified by mouse enzyme-linked immunosorbent assay (ELISA) kit, and the r2 value of the standard curve of the standard was 0.99 or more.
Comparative example 1: the difference from the calamus root extract is that comparative example 1 contains alpha-, beta-asarone, cis-methyl isoeugenol, methyl eugenol, calamus glycol, does not contain acolone (equal amount of acolone is replaced by water), and other components are the same as the calamus root extract.
Comparative example 2: the difference from the calamus root extract is that comparative example 2 contains alpha-, beta-asarone, cis-methyl isoeugenol, methyl eugenol, and calanolide, does not contain calanolic glycol (water is used to replace equivalent amount of calanolic glycol), and other components are the same as the calamus root extract.
Preparing 1%, 3% and 5% calamus root extract and the aqueous solutions of comparative example 1 and comparative example 2 according to the mass ratio, respectively, and testing the anti-inflammatory activity of the samples with different concentrations according to the above steps, wherein the results are as follows:
1) TNF-alpha expression inhibitory Activity
The inhibitory activity of pro-inflammatory cytokines (pro-inflammatory cytokines) TNF- α produced during the inflammatory reaction was measured, and the results are shown in table 3, whereby it was confirmed that the anti-inflammatory activity of the extract solution of Acorus calamus root was higher as the concentration was increased (1%, 3%, 5%); compared with the anti-inflammatory activity of the comparative examples 1 and 2, the calamus root extracting solution with the same concentration has obvious anti-inflammatory activity, which shows that the calamus root extracting solution compounded by alpha-, beta-asarone, cis-methyl iso-eugenol, methyl eugenol, calamus glycol and calamus ketone has the anti-inflammatory effect.

2) IL-6 expression inhibitory Activity
The result of measuring the IL-6 expression inhibition activity is shown in Table 4, the calamus root extract has obvious influence on the expression of one IL-6 in pro-inflammatory cytokines (pro-inflammatory cytokines).

(2) Application formula anti-inflammatory activity test of calamus root extracting solution
The amino acid facial cleansing honey of the application example I is used as an experimental group, and a blank group, a comparison group 1 and a comparison group 2 are added.
The blank group is an amino acid face-cleaning honey formula without adding calamus root extract.
Comparative group 1: the difference from the experimental group is that the added rhizoma Acori Calami extractive solution is different, the added rhizoma Acori Calami extractive solution of comparative group 1 does not contain Acorus calamus ketone (water is used to replace the same amount of Acorus calamus ketone), and other components are the same as those in the application example.
Comparative group 2: the difference from the experimental group is that the added calamus root extract is different, the comparative group 1 added calamus root extract does not contain calamus dilute glycol (water is used to replace the same amount of calamus dilute glycol), and other components are the same as the application examples.
The formulation process of the blank group, comparative group 1 and comparative group 2 was the same as that of the experimental group, the anti-inflammatory activity test was conducted according to the above procedure, and the results of the TNF-. alpha.expression inhibitory activity and IL-6 expression inhibitory activity are shown in Table 5.

According to the anti-inflammatory activity test results of the application formula of the calamus root extracting solution in the table 5, the amino acid facial cleanser prepared by adding the calamus root extracting solution has much higher anti-inflammatory activity than the amino acid facial cleanser without adding the calamus root extracting solution, and the calamus root extracting solution compounded by alpha-, beta-asarone, cis-methyl iso-eugenol, methyl eugenol, calamus glycol and calamus ketone has the anti-inflammatory effect.
2. Bacteriostasis test
(1) Bacteriostatic test of calamus root extract
In order to verify the bacteriostatic effect of the calamus root extracting solution, according to GB15979-2002 (hygienic Standard for Disposable sanitary articles) appendix C4 'test method for bacteriostatic performance of dissolution type anti (bacteriostatic) bacteria product', the method comprises the following steps:
a. test samples: rhizoma Acori Calami extractive solution.
b. Preparation of bacterial suspension: and (3) washing a 24-hour culture of the propionibacterium acnes by using 0.03mol/L phosphate buffer solution to prepare 104-106 CFU/mL bacterial suspension.
c. Respectively taking 1g of test sample, adding 0.1mL of the bacterial suspension, uniformly coating and mixing, starting timing, adding 9mL0.03mol/L of phosphate buffer solution after 20min, fully and uniformly mixing, properly diluting, taking 3 dilutions, respectively taking 1mL of the dilutions, placing in two parallel dishes, adding corresponding culture medium, culturing for 24h under the anaerobic condition of (36 +/-1) DEG C, counting colonies, repeating the above tests for 3 times, taking an average value, and setting a blank control group.
The calculation formula of the bacteriostatic rate is as follows: and X is (A-B)/A X100%, wherein X is the bacteriostasis rate, A is the average recovered colony of the control sample, and B is the average recovered colony of the test sample.
The evaluation criteria for efficacy were: (1) the bacteriostatic rate is more than or equal to 90 percent; (2) the bacteriostasis rate is 50-90% effective; (3) the bacteriostasis rate is less than 50 percent, which is ineffective.
The blank group was distilled water.
Comparative example 1: the difference from the calamus root extract is that comparative example 1 contains alpha-, beta-asarone, cis-methyl isoeugenol, methyl eugenol, calamus glycol, does not contain acolone (equal amount of acolone is replaced by water), and other components are the same as the calamus root extract.
Comparative example 2: the difference from the calamus root extract is that comparative example 1 contains alpha-, beta-asarone, cis-methyl isoeugenol, methyl eugenol, and acorone, does not contain calamus glycol (the same amount of calamus glycol is replaced by water), and other components are the same as the calamus root extract.
1 percent, 3 percent and 5 percent of calamus root extract, and the aqueous solutions of the comparative example 1(1 percent) and the comparative example 2(1 percent) are respectively prepared according to the mass ratio, and the bacteriostatic effect of samples with different concentrations is tested according to the bacteriostatic test steps, and the results are shown in table 6.

The results in table 6 show that the calamus root extract has better bacteriostatic effect with the increase of the concentration (1%, 3%, 5%); compared with the bacteriostatic effects of the comparative examples 1 and 2, the calamus root extracting solution with the same concentration has a remarkable bacteriostatic effect, and the effect of effectively inhibiting bacteria is shown by the calamus root extracting solution compounded by alpha-, beta-asarone, cis-methyl iso-eugenol, methyl eugenol, calamus glycol and calamus ketone.
(2) Application formula bacteriostasis test of calamus root extracting solution
The amino acid facial cleansing honey of example 1 was used as an experimental group, and a blank group, a comparative group 1 and a comparative group 2 were added.
The blank group is an amino acid face-cleaning honey formula without adding calamus root extract.
Comparative group 1: the difference from the experimental group is that the added extract of Acorus calamus root is different, the extract of Acorus calamus root added in the comparative group 1 does not contain Acorus calamus ketone (the same amount of Acorus calamus ketone is replaced by water), and other components are the same as those in the application example 1.
Comparative group 2: the difference from the experimental group is that the added Acorus calamus root extract solution is different, the added Acorus calamus root extract solution of the comparative group 1 does not contain Acorus calamus dilol (water is used to replace the same amount of Acorus calamus dilol), and other components are the same as the application example 1.
The formulation process of the blank group, the comparison group 1 and the comparison group 2 is the same as that of the experimental group, the bacteriostasis test is carried out according to the bacteriostasis test steps, and the results are shown in table 7.

According to the bacteriostatic test results of the application formula of the calamus root extracting solution shown in table 7, the amino acid facial cleanser prepared by adding the calamus root extracting solution has a better bacteriostatic effect than the amino acid facial cleanser without adding the calamus root extracting solution, and the calamus root extracting solution compounded by alpha-, beta-asarone, cis-methyl iso-eugenol, methyl eugenol, calamus glycol and calamus ketone has the bacteriostatic effect.
3. Acne removal test
(1) Oil secretion inhibition test of calamus root extract
Test samples: calamus root extract, blank, comparative example 1, comparative example 2.
The blank group was distilled water.
Comparative example 1: the difference from the calamus root extract is that comparative example 1 contains alpha-, beta-asarone, cis-methyl isoeugenol, methyl eugenol, calamus glycol, does not contain acolone (equal amount of acolone is replaced by water), and other components are the same as the calamus root extract.
Comparative example 2: the difference from the calamus root extract is that comparative example 1 contains alpha-, beta-asarone, cis-methyl isoeugenol, methyl eugenol and caladone, does not contain calamus glycol (the same amount of calamus glycol is replaced by water), and other components are the same as the calamus root extract.
The aqueous solutions of 1%, 3% and 5% calamus root extract and comparative examples 1 and 2 were prepared according to the mass ratio, and the acne removing effect was tested according to the following test methods.
The test method comprises the following steps: selecting 60 oily or mixed skin subjects (age 20-35 years), randomly dividing into 6 groups (10 in each group), and respectively smearing different test samples (rhizoma Acori Calami extractive solution, blank group, comparative example 1, and comparative example 2) on each group. The test samples were administered to the subjects once a day, morning and evening, for 28 consecutive days. Before and 28 days after the use, the oil content of the skin surface of the forehead part is respectively detected by an oil measuring instrument for 3 times, the average value is taken, the reduction of the oil content of the skin surface is calculated, and the test evaluation result is shown in table 8.

From the results in table 8, it can be seen that as the concentration of the calamus root extract increases (1%, 3%, 5%), the reduction of skin oil secretion increases and the acne removing effect is better; compared with the acne removing effects of the comparative examples 1 and 2, the rhizoma acori graminei extracting solution with the same concentration has an obvious acne removing effect, and the effect of removing acnes is shown by the rhizoma acori graminei extracting solution compounded by alpha-, beta-asarone, cis-methyl isoeugenol, methyl eugenol, calamus glycol and calamus ketone.
(2) Application formula of calamus root extracting solution for inhibiting oil secretion test
Taking the amino acid facial cleansing honey of example 1 as an experimental group, and adding a blank group, a comparison group 1 and a comparison group 2;
the blank group is an amino acid face-cleaning honey formula without adding calamus root extract.
Comparative group 1: the difference from the experimental group is that the added extract of Acorus calamus root is different, the extract of Acorus calamus root added in the comparative group 1 does not contain Acorus calamus ketone (the same amount of Acorus calamus ketone is replaced by water), and other components are the same as those in the application example 1.
Comparative group 2: the difference from the experimental group is that the added Acorus calamus root extract solution is different, the added Acorus calamus root extract solution of the comparative group 1 does not contain Acorus calamus dilol (water is used to replace the same amount of Acorus calamus dilol), and other components are the same as the application example 1.
The formula process of the blank group, the comparison group 1 and the comparison group 2 is the same as that of the experimental group, the acne removing test is carried out according to the test method, and the result is shown in a table 9.

According to the acne removing test result of the application formula of the calamus root extracting solution in the table 9, the amino acid facial cleanser prepared by adding the calamus root extracting solution has a better acne removing effect than the amino acid facial cleanser without adding the calamus root extracting solution, and the calamus root extracting solution compounded by alpha-, beta-asarone, cis-methyl iso-eugenol, methyl eugenol, calamus glycol and calamus ketone has the acne removing effect.
