The effect optimizing agent is a kalimeris indica extract.
The preparation method of the kalimeris indica extract comprises the following steps:
(1) cutting up the mangnolia into pieces, adding deionized water into the pieces, wherein the solid-liquid mass ratio of the pieces to the deionized water is 1: 8; keeping the temperature of the water bath constant to 60 +/-5 ℃ for 20 hours, then cooling the water bath constant to the normal temperature in air, and filtering the water bath constant to obtain a liquid phase A and filter residues; evaporating and concentrating the liquid phase A to 1/3 volume before concentration, and freeze drying at-80 deg.C to obtain extract powder A;
(2) extracting the filter residue by adopting ultrasonic-assisted ethanol water solution, wherein the ultrasonic-assisted ethanol water solution extraction process parameters are as follows: the volume ratio of ethanol in the ethanol aqueous solution is 75 percent; the ultrasonic power is 320W; the extraction temperature is 55 +/-3 ℃; the solid-liquid mass ratio of the filter residue to the ethanol aqueous solution is 1: 20. The comparative composition was prepared by:
1) weighing the raw materials in parts by weight;
2) mixing medlar, acanthopanax, rhodiola rosea, Chinese yam, rhizoma polygonati and dried orange peel, then pulverizing into mixed powder, adding water into the mixed powder, decocting for 1h, wherein the added water amount is 6 times of the mass of the mixed powder, then cooling to normal temperature, centrifuging, concentrating the supernatant under reduced pressure to 1/2 of the volume before concentration, then placing the mixture into an environment at the temperature of minus 80 ℃ for freeze drying to obtain powder C, adding the herba Kalimeridis extract into the powder C, and uniformly mixing to obtain the comparative composition of the comparative example.
Example 5
200 SPF-grade six-week-old healthy female ICR mice were selected and fed adaptively for seven days. During the feeding period, the mice had free diet and water. The environment is shady, cool and ventilated, the environment humidity is maintained at 50 to 60 percent, and the room temperature is 25 +/-2 ℃. After feeding, 108 mice which swim are selected and averagely divided into two groups, wherein one group is subjected to a load swimming test, and the other group is a biological index detection group. Each group was randomly divided into 9 subgroups of 6 each. Each group was administered one of the example or comparative compositions separately for gavage. The administration amount of each mouse by gavage is 200mg/kg d, and the administration lasts for 30 days. The weight swimming test method comprises the following steps: after the last administration, the test was carried out after 30 min. Lead pieces weighing 10% of the weight of the mice were fixed to the tails of the mice. Then the mice are placed in a swimming box with water depth of 40cm and water temperature of 25 +/-2 ℃ so as to be capable of swimming freely. The time from the start of swimming until the mice completely submerged in water were not able to float out of the water surface for 8 seconds was taken as the mouse weight swimming time. The results are shown in Table 1. The biological index detection method comprises the following steps: after the last administration, the test was carried out after 30 min. The mice are placed in a swimming box with water depth of 40cm and water temperature of 25 +/-2 ℃ so as to be capable of swimming freely for 30 min. Then rest for 60min and blood and liver were collected. Separating serum, and detecting liver with liver homogenate with liver mass fraction of 10% prepared from normal saline. The contents of serum urea nitrogen, liver glycogen and muscle glycogen and the activity of total superoxide dismutase in liver tissues are respectively detected according to the method in the kit instruction.

As can be seen from Table 1, the composition of the present invention has an effect of prolonging the time required for heavy swimming in mice. It is understood from comparative example 3 and comparative example 3 that the time for the mouse to swim with a load is significantly different from 45.8 + -3.2 s to 60.8 + -4.2 s after the addition of the effect-improving agent of the present invention. The effect optimizing agent can obviously improve the effect of the composition, prolong the exhaustive swimming time of mice and has anti-fatigue activity. As can be seen from the comparison of example 3 and comparative examples 1-2, the addition of ligularia fischeri leaf extract to the effect-optimizing agent can enhance the effect of the effect-optimizing agent, which means that the weight-bearing swimming time of mice is increased by about 5s, and the anti-fatigue activity of the composition can be further improved. As can be seen from FIGS. 1 to 4, the serum urea nitrogen content is significantly reduced after the effect optimizing agent is added to the composition. It is known that when mice swim under heavy load, energy is continuously consumed, glycogen and fat catabolism in the body are insufficient to provide sufficient energy, protein catabolism speed is increased, protein generates ammonia in the catabolism process and is converted into urea in the liver, and urea nitrogen enters blood circulation to form blood urea nitrogen. Therefore, the increase of the serum urea nitrogen content can reduce the load adaptability of the organism. The faster serum urea nitrogen is cleared after exercise, the faster fatigue is eliminated. After the effect optimizing agent is added into the composition, the serum urea nitrogen content is obviously lower than that of the composition without the effect optimizing agent, which shows that the optimized composition has good regulation effect on the serum urea nitrogen content of mice and can eliminate fatigue after exercise. In addition, as can be seen from the figure, after the effect optimizing agent is added into the composition, the liver glycogen content, the muscle glycogen content and the total superoxide dismutase activity in liver tissues of the mouse are improved to different degrees, which indicates that after the effect optimizing agent is added, the reserves of the liver glycogen and the muscle glycogen of the mouse are improved, and energy is supplemented for the loaded swimming, so that the fatigue occurrence degree is reduced, and the oxidative stress injury of the mouse caused by high-intensity exercise is reduced.
The technical solutions provided by the present invention are described in detail above, and for those skilled in the art, the ideas according to the embodiments of the present invention may be changed in the specific implementation manners and the application ranges, and in summary, the content of the present description should not be construed as limiting the present invention.
Claims (7)
1. The composition with the anti-fatigue effect is characterized by being a preparation prepared from the following raw materials: the Chinese wolfberry fruit-red sage root-wild ginger tea comprises Chinese wolfberry, acanthopanax root, rhodiola rosea, Chinese yam, rhizoma polygonati, dried orange peel and an effect optimizing agent, wherein the effect optimizing agent is a dictyophora indusiata-kalimeris extract.
2. The composition with the anti-fatigue effect according to claim 1, wherein the raw materials comprise, by weight: 10-20 parts of medlar, 8-12 parts of acanthopanax, 3-6 parts of rhodiola rosea, 1-5 parts of Chinese yam, 2-3 parts of rhizoma polygonati, 1-2 parts of dried orange peel and 0.1-0.3 part of an effect optimizing agent.
3. The composition with an anti-fatigue effect according to claim 1, wherein the dictyophora indusiata-malan extract is prepared by a method comprising the following steps:
(1) mixing the dictyophora indusiata and the mangnolia officinalis, chopping into small pieces, adding deionized water into the small pieces, keeping the temperature in a water bath at constant temperature of 60 +/-5 ℃, keeping the temperature, then cooling in air to normal temperature, and filtering to obtain a liquid phase A and filter residues; evaporating and concentrating the liquid phase A, and freeze-drying at-80 deg.C to obtain extract powder A;
(2) extracting the filter residue with ultrasonic-assisted ethanol water solution, centrifuging after extraction, collecting supernatant, evaporating for concentration, and freeze-drying at-80 deg.C to obtain extract powder B;
(3) mixing the extract powder A and the extract powder B to obtain the dictyophora indusiata-kalimeris indica extract.
4. The composition with the anti-fatigue effect according to claim 3, wherein the mixing mass ratio of dictyophora indusiata to mangrove is 10: 3-10; and adding deionized water into the broken dices, wherein the solid-liquid mass ratio of the broken dices to the deionized water is 1: 6-10.
5. The composition with the anti-fatigue effect according to claim 3, wherein the water bath is kept at the constant temperature of 60 +/-5 ℃ for more than 20 hours.
6. The composition with an anti-fatigue effect according to claim 3, wherein the ultrasonic-assisted ethanol aqueous solution extraction process parameters are as follows: the volume ratio of ethanol in the ethanol water solution is 70-75 percent; the ultrasonic power is 320W; the extraction temperature is 55 +/-3 ℃; the solid-liquid mass ratio of the filter residue to the ethanol aqueous solution is 1: 20.
7. The composition with anti-fatigue effect according to claim 3, wherein in step (2), ligularia fischeri is further added to the residue, and the mass ratio of ligularia fischeri to residue is: and (4) filtering residue, namely 4-5: 10.
